A new androstane derivative, 4β-methyl-15-oxa-14β-androstane-7-ene-4α-carboxylic acid (1) and a known one 4β-methyl-15-oxa-14β-androstane-7-ene-4α-hydroxyl (2) were isolated from the EtOAc extract of the cultures of the fungus Marasmiellus ramealis (Bull.) Singer. Their structures were elucidated on the basis of 1D and 2D NMR as well as MS spectroscopic data analysis. The inhibitory activity of two isolates against acetylcholinesterase (AChE) revealed that compound 1 exhibited definitely inhibitory activity.

Objective: Although endoscopic drill has the advantages in manipulation and hemostasis, whose low efficiency and blurred vision reduce the efficacy of lumbar endoscopic unilateral laminotomy with bilateral decompression (LE-ULBD). The present study was designed to evaluate the safety and efficacy of full-visualized trephine/osteotome in the LE-ULBD surgery for severe lumbar stenosis. Methods: Fifty-seven severe lumbar stenosis patients who underwent LE-ULBD between January 2020 to January 2023 were enrolled, who were divided into drill and visualized trephine groups. The medical records including demographics, operative duration, intraoperative electrophysiological findings, postoperative hospital stay or hospital stay, postoperative outcomes and complications were retrospectively reviewed and analyzed. Results: A total of 57 patients included 15 in drill and 42 in trephine group were enrolled in the study. There was significant difference in the pre- and postoperative visual analogue scale and Oswestry Disability Index scores in both groups (p < 0.05). The mean operative duration in the trephine group (101.05 ± 12.18 minutes) was shorter than that in the drill group (134.67 ± 9.68 minutes) (p < 0.05). There was no statistical difference between the 2 groups in electrophysiological monitoring, posthospital stays, postoperative outcomes and complications. Abnormal free-electromyography (EMG) were recorded in 2 (13.3%) and 5 patients (11.9%) in the drill and trephine group. Intraoperative somatosensory evoked potential changes occurred in 3 (20%) and 3 patients (7.1%) in the drill and trephine group and all patients recovered immediately when surgery ended. No serious complications and recurrence occurred in all the patients. Conclusion: Full-visualized trephine/osteotome has been approved to be convenient, safe and efficient in our study, which combined with translaminar inside-out technique and EMG monitoring especially free-EMG may offer a new choice in LE-ULBD surgery for lumbar stenosis patients.

Background: Postherpetic neuralgia (PHN) is the most common complication of acute herpes zoster. The treatment of PHN remains a challenge for clinical pain management. Despite the effectiveness of anticonvulsants, antidepressants, and lidocaine patches in reducing PHN, many patients still face intractable pain disorders. In this randomized controlled study, we evaluated whether hydromorphone through intravenous patient-controlled analgesia (IV PCA) was effective in relieving PHN. Methods: Patients with PHN were randomly divided into two groups, one group received oral pregabalin with IV normal saline, another group received oral pregabalin with additional IV PCA hydromorphone for two weeks. Efficacy was evaluated at 1, 4, and 12 weeks after the end of the treatments. Results: Two hundred and one patients were followed up for 12 weeks. After treatment, numerical rating scale (NRS) score of patients in the hydromorphone group was significantly lower than that of the control group, and the difference of NRS scores between the two groups was statistically significant at 4 and 12 weeks after treatment. The frequency of breakthrough pain in the hydromorphone group was significantly lower than that in the control group 1 and 4 weeks after treatment. After treatment, the quality of sleep in the hydromorphone group was significantly improved compared with the control group. The most common adverse reactions in the hydromorphone group were dizziness and nausea, with no significant respiratory depression. Conclusions: IV PCA hydromorphone combined with oral pregabalin provides superior pain relief in patients with PHN, which is worthy of clinical application and promotion.

Guided by bioisosterism and pharmacokinetic parameters, we designed and synthesized a series of novel benzamide derivatives. Preliminary in vitro studies indicated that compounds 10b and 10j show significant inhibitory bioactivity in HepG2 cells (IC50 values of 0.12 and 0.13 μM, respectively). Compounds 10b and 10j induced the expression of HIF-1α protein and downstream target gene p21, and upregulated the expression of cleaved caspase-3 to promote tumor cells apoptosis.

Starting from the relativistic equation of motion governing quantum collision processes, we shall formulate the relativistic quantum collision theory in an ab-initio manner. Quantum electrodynamic effects are, however, incorporated perturbatively. Because heavy projectiles or ultra-high incident energies are considered, the recoil of the target is also treated. Electron-impact ionization of the uranium ion U^(91+) and proton-impact ionization of hydrogen will be given as examples.

An Efficient Reactive Power Regulation based DPC Method for AC/DC Converters

To eliminate the cyclical fluctuation problems of the reactive power existing in the traditional direct power control (DPC), a novel method is proposed for the threephase voltage source AC/DC converter. It employs two switching tables to select the proper switching states according to the instantaneous reactive power deviations. The design principle of the switching tables is provided to ensure the efficient reactive power regulation. Moreover, it should guarantee the steady characteristic of the converter system. The simulation model established under the MATLAB/Simulink environment and the experiment were presented to compare the characteristics obtained by the proposed strategy with that obtained by the traditional DPC strategy. The results show that the ac current obtained by the proposed strategy has the much lower total harmonic distortion rate (THD), and the fluctuation range of the reactive power has the more than two times reduction.

<P>The evolutionarily conserved CONSTITUTIVE PHOTOMORPHOGENESIS 1 (COP1) is a RING and WD40 protein that functions as a substrate receptor of CULLIN4–DAMAGED DNA BINDING PROTEIN 1 (CUL4–DDB1)–based E3 ubiquitin ligases in both plants and animals. In <I>Arabidopsis</I>, COP1 is a central repressor of photomorphogenesis in the form of COP1–SUPPRESSOR OF PHYA (SPA) complex(es). CUL4–DDB1–COP1–SPA suppresses the photomorphogenic program by targeting the transcription factor ELONGATED HYPOCOTYL 5 for degradation. Intriguingly, under photomorphogenic UV-B light, COP1 reverses its repressive role and promotes photomorphogenesis. However, the mechanism by which COP1 is functionally switched is still obscure. Here, we demonstrate that UV-B triggers the physical and functional disassociation of the COP1–SPA core complex(es) from CUL4–DDB1 and the formation of a unique complex(es) containing the UV-B receptor UV RESISTANCE LOCUS 8 (UVR8). The establishment of this UV-B–dependent COP1 complex(es) is associated with its positive modulation of ELONGATED HYPOCOTYL 5 stability and activity, which sheds light on the mechanism of COP1’s promotive action in UV-B–induced photomorphogenesis.</P>
Baseline-free damage detection method for beam structures based on an actual influence line

The detection of structural damage without a priori information on the healthy state is challenging. In order to address the issue, the study presents a baseline-free approach to detect damage in beam structures based on an actual influence line. In particular, a multi-segment function-fitting calculation is developed to extract the actual deflection influence line (DIL) of a damaged beam from bridge responses due to a passing vehicle. An intact basis function based on the measurement position is introduced. The damage index is defined as the difference between the actual DIL and a constructed function related to the intact basis, and the damage location is indicated based on the local peak value of the damage index curve. The damage basis function is formulated by using the detected damage location. Based on the intact and damage basis functions, damage severity is quantified by fitting the actual DIL using the least-square calculation. Both numerical and experimental examples are provided to investigate the feasibility of the proposed method. The results indicate that the present baseline-free approach is effective in detecting the damage of beam structures.
