        Preparation of Poly(N-vinyl-2-pyrrolidone) Microparticles Using Supercritical Anti-solvent

        화장품, 의약품, 전자소재 분야에서 생체적합, 생분해성 고분자로 널리 사용되는 폴리비닐피롤리돈(PVP)을 에어로젤 용매추출 방법에 의해 미세입자를 제조하였다. 용매로는 이염화메탄올, 반용매로는 초임계 이산화탄소가 사용되었다. 온도, 압력, 이산화탄소 유량, 용액유량의 작업조건에 따라 0.184 - 0.249 ㎛입자크기를 얻었고, 그 입자크기에 영향을 미치는 초기구형 입자크기와의 상관관계를 규명하였다. Poly(N-vinyl-2-pyrrolidone) (PVP) has been used as biocompatible and biodegradable polymer in cosmetics, pharmaceuticals and electronics. Micro-particles of PVP were produced using an aerosol solvent extraction system (ASES). Dichloromethane (DCM) and supercritical carbon dioxide were used as solvent and antisolvent, respectively. The mean diameter of the obtained polymer particles ranged from 0.184 to 0.249㎛. The relationship between particle size and initial drop size was also considered.

        An Effective Antidandruff Agent 一 IPBC

        The purpose of this study was to determine the effect of IPBC(3-lodo-2-propynylbutyl carbamate) on dandruff caused by the anthropophilic fungus Malassezia furfur. The effects of IPBC on dandruff were examined by evaluating (a) the MIC value of IPBC using broth dilution method ; (b) the remnant antimicrobial activity of IPBC containing shampoo on skin disc ; (c) the antidandruff efficacy of 1.0 % IPBC containing shampoo in double blind clinical trial. To investigate the remnant antimicrobial activity of IPBC against Malassezia furfur, guinea pig-skin disc was washed with antidandruff shampoo and then the diameter of inhibition zone per disc was measured. For clinical trial, thirty healthy volunteers, aged 25-35, participated in 4 week study. At 0, 2, 4 weeks, examinations of scaling, itching on scalp were carried out. The MIC(Minimun Inhibition Concentration) values of IPBC range from 0.10 to 1.00μ g/ml and it seems that IPBC is more effective in the MIC values than zinc pyrithione, selenium disulphide, piroctone olamine and comparable to ketoconazole, climbazole. When the remnant antimicrobial activity of IPBC shampoo on skin disc was determined, 5% IPBC shampoo and 2.0% Ketoconazole shampoo resulted in similar antimicrobial effect. In addition, 1.0%, 2.0% IPBC shampoo was more effective than 2.0% ketoconazole shampoo. After two and four-weeks of 1.0% IPBC shampoo treatment, there was significant reduction of scaling, itching in test group compared to control group. On the basis of these results, it can be concluded that 1.0% IPBC is more effective.than 2.0% Ketoconazole in reducing dandruff. It seems that strong capacity of drug binding to the stratum corneum plays a role in its antidandruff effect since adsorption of active ingredients on scalp is very important factor in reducing dandruff.

        Solubility of Triclosan in Supercritical Carbon Dioxide and its Application to Micronization Process

        여드름 치료제인 트리클로산의 초임계이산화탄소에 대한 용해도를 가변부피셀(Variable volume cell)을 이용하여 온도 313.15, 323.15와 333.15 K에 대해서 10 ? 40 ㎫ 압력범위에서 측정되었다. QLF (Quasichemical nonrandom lattice fluid) 상태방정식을 이용하여 측정된 용해도를 잘 계산할 수 있었고, 이 자료를 미세입자공정에 활용하였고, 입자크기에 미치는 온도와 압력의 효과를 해석하였다. The solubility of triclosan, an anti-acne agent was measured in supercritical carbon dioxide (scCO₂) with a variable volume view cell at 313.15, 323.15, and 333.15 K and at pressures between 10 and 40 ㎫. We successfully correlated triclosan solubility in scCO₂ using the quasi-chemical nonrandom lattice fluid (QLF) equation of state. Triclosan was micronized using the rapid expansion of supercritical solutions (RESS) process. The effects of temperature and pressure on particle size were investigated using phase behavior data and correlated results from the QLF model.

      • A Lattice Model Based on Molecular Clusters for Supercritical Fluids

        A semi-empirical fluctuation term is presented to improve a classical equation of state (EOS) for volumetric properties in the critical region. The term is based on the two assumptions: (1) The Helmholtz energy is individually divided into classical and long-range density fluctuation contribution (2) All molecules form cluster near the critical region due to long-range density fluctuation. To formulate such molecular cluster, we extended the Veytsman statistics originally developed for the cluster due to hydrogen bonding. The probability function in the statistics is modified to represent the characteristics of long-range density fluctuation vanishing far from critical region. The proposed fluctuation contribution was incorporated into the Sanchez-Lacombe EOS and the combined model with 6 adjustable parameters has been tested against experimental VLE data. The combined model is found to well represent flatten critical isotherm for methane and top of the coexistence curve for the tested components. The prediction results for caloric data are in good agreement with the experimental data.

        계면활성제를 함유한 혼합물에서 내외부 분자회합을 위한 격자모델

        분자내부 회합은 계면활성제계, 특히 콜로이드와 생체물질에 있어서 수소결합 중에서 매우 중요한 역할을 나타낸다. 미셀과 마이크로에멀젼을 형성하는 양친매성 계는 분자간 회합뿐만 아니라 분자내부 회합 때문에 매우 비 이상적인 거동을 나타낸다. 본 연구의 목적은 계면활성제를 함유한 혼합물의 상거동을 계산하기 위하여, 내외부 분자 회합을 설명할 수 있는 수정된 베이츠만 통계역학이론과 결합된 유사화학반응 비무질도서 격자모델을 제시하는 것이 다. 본 논문에서 제시된 격자모델은 알칸과 계면활성제의 실험결과를 매우 잘 설명할 수 있다. Intra-molecular association is an important contribution to the overall hydrogen bonding in surfactant systems, especially in systems of colloidal and biological interest. Amphiphile systems, especially micelle and microemulsion systems, showed highly non-ideal behavior due to the intermolecular association and intra-molecular association. The objective of this research is to present a lattice fluid equation of state that combines the quasi-chemical nonrandom lattice fluid model with modified Veytsman statistics for intra + inter molecular association to calculate phase behavior for mixture containing surfactant systems. The lattice model could describe the literature data well for alkane and surfactant systems.

        유화제형 바디클렌징 시스템에서 보습효과

        최근 불용성의 결정성 수산화 왁스, 불용성의 결정성 계면활성제 구조, 캡슐 입자, 스멕타이트 점토를 이용하여 고함량의 친유성 보습제를 함유할 수 있는 유화제형 바디클렌징 시스템이 개발되었다. 본 논문에서는 유화제형 바디클렌징 시스템에서 바세린, 트리글리세라이드와 같은 다양한 밀폐제의 보습효과를 연구하였다. 본 실험결과에 따르면, 유화제형 바디클렌저에서 바세린이 트리글리세라이드에 비하여 보습력이 떨어지는 것을 알 수 있었다. 로션, 크림과 같은 바르는”제형에서는 바세린이 최고의 보습력을 가지지만, 본 연구의 “씻어내는”유화바디클렌징 제형에서는 다른 결과가 나타났다. 고함량의 음이온, 양성이온 계면활성제가 함유된 유화제형 바디클렌징시스템에서, 바세린은 트리글리세라이드에 비해 용해도가 커서, 헹굼과정에서 피부로의 부착량이 떨어짐으로 인해, 보습력이 부족함을 밝혀내었다. Emulsion body cleansing systems containing high levels of occlusives have recently been developed such as using insoluble crystalline hydroxy waxes, insoluble crystalline surfactant structures, encapsulated particles and smectite clay. In this study, moisturizing effect of various occlusives such as petrolatum and triglycerides was researched in emulsion body cleansing systems. The experiment shows that petrolatum has less moisturizing capacity in emulsion body cleansing systems than triglycerides. In “leave-on” products such as lotion and cream formulations, the moisturizing capacity of petrolatum is more than that of triglycerides, but in “wash-off” emulsion body cleanser of this research, different results appeared. Emulsion body cleansing systems contained high levels of anionic and amphoteric surfactants, therefore petrolatum has more solubility and less deposited amount into the skin during washing process than triglycerides.

        흰쥐에서 심혈관 조절기전에 대한 척수의 Adenosine A2 수용체의 역할

        The present study was designed to assess the role of spinal adenosine A2 receptor in the regulation of cardiovascular functions such as mean arterial pressure (MAP) and heart rate (HR) in male Sprague-Dawley rats. Rats (250∼300 g) were anesthetized with urethane and paralyzed with d-tubocurarine and artificially ventilated. blood pressure and HR were continuously monitored via a femoral catheter connected to a pressure transducer and a polygraph. Drugs were administered intrathecally using injection cannula through guide cannula which was inserted inthrathecally at lower thoracic level through a puncture of an atlantooccipital membrane. Intrathecal injection of an adenosine A₂ receptor agonist, 5`-(N-cyclopropyl)-carboxamaidoadenosine (CPCA; 1, 2 and 3 nmol, respectively), produced a dose-dependent decrease in MAP and HR. Pretreatment with N^G-nitro-L-arginine methyl ester (L-NAME), a nitric oxide synthase inhibitor or 10 nmol of MDL-12,330, an adenylate cyclase inhibitor blocked significantly the depressor and bradycardic effect of 2 nmol of CPCA. But, Pretreatment with 3 nmol of bicuculline, gamma-aminobutyric acid A (GABA_A) receptor antagonist, or 50 nmol of 5-aminovaleric acid, GABA_B receptor antagonist did not inhibit the depressor and bradycardic effect of 2 nmol of CPCA. These results indicate that adenosine A₂ receptor in the spinal cord plays an inhibitory role in the regulation of cardiovascular function and that the depressor and bradycardic action of adenosine A₂ receptor are mediated via the synthesis of nitric oxide and the activation of adenylate cyclase in the spinal cord of rats.

        초임계 이산화탄소를 이용한 미립 항비듬제 제조

        화장품, 의약품에서 항비듬제로 널리 사용되는 요도프로피닐 부틸카바메이트와 클림바졸을 초임계 유체를 이용하여 미세입자를 제조하였다. 초임계 유체로는 임계온도와 임계압력이 상대적으로 낮은 이산화탄소가 사용되었다. 초임계 이산화탄소에 용해력이 있는 요도프로피닐 부틸카마메이트와 클림바졸을 항비듬제로 선택되었다. 초임계 용액 급속 팽창법을 이용하여 압력과 온도를 변화시켜 입자크기와 형상에 미치는 영향을 분석하였다. Iodopropynyl butylcarbamate and climbazole as anti-dandruff agents widely used in cosmetics and pharmaceutics were micronized using supercritical fluid. Supercritical carbon dioxide was selected due to relatively low critical temperature and critical pressure. Iodopropynyl butylcarbamate and climbazole were chosen because of their solubility in supercritical carbon dioxide. The rapid expansion of supercritical solution (RESS) experiments involved investigations of the effect of pressure, temperature on particle size and morphology.
