Histone deacetylase 3 is selectively involved in L3MBTL2-mediated transcriptional repression

<P><B>Abstract</B></P><P>This is the first report that L(3)mbt-like 2 (L3MBTL2) specifically interacts with the histone deacetylase domain of histone deacetylase 3 (HDAC3) via its MBT domain. Here, we show that L3MBTL2 selectively interacts with HDAC3, but not other class I HDACs. An in vitro peptide-binding assay demonstrated the specific association of HDAC3 with methylated histone-K20 tail and L3MBTL2. Furthermore, depletion of HDAC3 resulted in a decrease of methylated K20-H4, as well as an increase in acetylated histone H3. Consequently, HDAC3 knock-down selectively suppressed L3MBTL2-mediated transcriptional repression. Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

Reactive oxygen species(ROS) are implicated in the pathogenesis of a wide variety of human diseases. Numerous studies indicate that ROS may serve as common intracellular molecules that contribute to the process of nuclear factor(NF)κB activation in response to a diverse stimuli. In our laboratory, we have demonstrated tat antioxidants could reverse the decline of immune function caused by exercise-induced ROS. Furthermore, it in necessary to understand a mechanism underlying ROS-dependent disorder in biological system. Recent studies have been shown that several gene expression were regulated by oxidants, antioxidants and other determinants of the intracellular reduction-oxidation(redox) state. In this process, NF-κB have been shown to play a important role. The purpose of the present investigation was to evaluate the effect of exercise-induced oxidative stress and antioxidnt supplementation on NF-κB activation in peripheral mononuclear cells. Forty male SD rats(4 weeks old) were randomly divided into noraml diet group and antioxidants(ATO) supplement group, and then ATO groups were treated with antioxidants(VE: 2001U/kg, VC: 50mg/rat, β-carotene: 300mg/kg, vitamin B6: 250㎍/100g, selenomethionine: 0.1mg/kg) for 16 weeks. After 16 weeks breeding at each condition, each group divided into two groups: Control group(CR) fed generally, Exercise group(CE) fed generally and followed by acute exercise 16 weeks later. Antioxidant Antioxidants and Exercise group(AE) fed with antioxidants and followed by acute exercise. The activation of NF-κB binding activity was increased after exhaustive exercise bout in both group. In addition, pretreatment of ATO group with the antioxidants mixture lead to the inhibition of NF-κB binding activity. This results suggest that NF-κB activation should be further studies in response to a variety of exercise.

Background: Intestinal ischemia remains a devastating event despite improvements in clinical recognition and in diagnostic and therapeutic modalities. The ischemic bowel diseases encompass a wide clinical spectrum from mild, reversible disease to severe, irreversible injury. The clinical picture is characterized initially by poorly localized visceral pain. Thus, it is difficult to diagnose this ischemia at an early stage. This study investigated whether an increased serum lactate level is a recognized danger signal marker for intestinal ischemia in patients who present at the emergency department because of abdominal complaints. Methods: Patients who came to our emergency department with abdominal pain and the risk factors of intestinal ischemia between Apr. 1999 and Nov. 1999 were included in this study. The data analysis included age, sex, final diagnosis, pathogenesis of bowel ischemia, and serum lactate level. Results: The serum lactate level in the intestinal ischemia group was 28.54±22.51 mg/dl; in non-ischemia group, it was 15.49±22.52 mg/dl. This difference between the two groups was significant(p<0.05). An increased serum lactate level had a sensitivity of 88.2% and specificity of 59.2%, a positive likelihood ratio of 4.92, and a negative likelihood ratio of 0.47 as a marker of bowel ischemia. These results do not represent a very meaningful revision of bowel ischemlc provability, but may make a small contribution to management of the disease, depending upon their magnitude and the clinical context in which they are applied. Conclusion: In patients with abdominal complaints, an increased serum lactate level is usually a useful aid as a diagnostic marker of bowel ischemia.

Kochujang was prepared with different raw materials, and with and without sake. Quality changes during fermentation of Kochujang were measured in terms of pH, titratable acidity and amino nitrogen content. pH of Sunchang Kochujang gradually decreased as the increase of fermentation time, while that of Sachun remained almost constant. Titratable acidity was the highest in Sunchang, and was similar in Sachun regardless of glutinous rice and wheat flour as a starch source. Amino nitrogen of Sunchang increased drastically by 10 days of fermentation and then increased gradually. Amino nitrogen of Sachun with glutinous rice was gloater than that with wheat flour. pH of Kochujang without sake decreased drastically by 10 days and then stayed almost the same by 60 days of fermentation, while that with sake increased slightly as the increase of aging time. Titratable acidity of Kochujang with sake decreased sharply by 10 days, increased gradually by 30 days and then stayed at that level. Kochujang without sake showed greater decreases in titratable acidity by 5 days, increased gradually by 20 days and then reached equilibrium. Amino nitrogen was a little higher in Kochujang without sake than that with sake.
