This study aims at the effective estimation of water supply capacity of small scale reservoir and the proposal of the data which is necessary to establish the water resources management plan of down stream area of the reservoir in the future by comparison and examination about reservoir operation technique for the security of agricultural water in small scale reservoir. The result of flow calculation by Tank model is used for the input data as the inflow data which is needed for the analysis of water supply capacity. Stochastic method, simulation method, and optimization method are used to examine the water supply capacity, and water security amount is compared with each method. From the analyses of water supply capacities by each method, slightly different results are shown in spite of the effort to compare them equally using input data such as inflow data under equal conditions, and the comparison of water supply capacities by each method are follows; linear planning method, and transition probability matrix method in the order of amount from the largest. It is thought that the simulation method in which comparatively reasonable application of the inflow data is possible and is simulated in successive time series dam operation of the three methods used in this study thus, simulation model is proper to estimate the water supply capacity of agricultural small scale reservoir. And it is judged that the heightening of efficiency of water resources utilization according to the development of downstream area of dam may be possible using the upward readjusted water supply amount of 55.18×10^6ton at 95% and 90% supply reliability respectively which are above the planning water supply amount of 50.0×10^6ton when the simulation method is introduced as the standard.
Syntheis of an Aspartame Precursor Using Immobilized Thermolysin in an Organic Solvent

The synthesis of N-(benzyloxycarbonyl)-L-aspartyl-L-phenylalanine methylester (Z-APM), a precursor of aspartame, from N-(benzyloxycarbonyl)-L-aspartic acid (Z-Asp) and L-phenylalanine methylester hydrochloride(L-PM·HCl) was investigated in a saturated-ethylacetate single phase system using immobilized thermolysin. Among the various supports tested, glyceryl-CPG was found to be most efficient for retaining enzyme activity. The enzyme immobilized onto glyceryl-CPG also showed the highest activity for Z-APM synthesis in saturated ethyl acetate. Z-APM conversion yield in saturated ethylacetate was half of that obtained in an ethylacetatebuffer two-phase system under the same reaction conditions. However, as the mole ratio of L-PM·HCl to Z-Asp was increased to 4.0, the conversion yield reached 95 %. When continuous synthesis of Z-APM was carried out in a plug flow reactor (PFR) with 80mM of L-PM·HCl and 20mM of Z-Asp in saturated ethylacetate (pH 5.5), more than 95 % of Z-Asp was converted to Z-APM with a space velocity of 1.16 hr^-1 at 40℃. Although the operational stability in PFR was reduced rapidly, more than 80% of initial activity was maintained in CSTR even after a week of operation.

Detection of Anticancer Activity from the Root of Angelica gigas in Vitro

Anticancer activity of a fraction of the ethanol extract from the root of Korean angelica (Angelica gigas Nakai) was recognized in human cancer cell lines HeLa S_3 K-562, and Hep G_2. The extract blocked the phorbol ester-inducing megakaryocytic differentiation of K-562 cells, which indicated the modification of protein kinase C (PKC) activity. In vitro assay showed the activation of PKC by the extract. An effective fraction of the Angelica gigas extract, of which R_f value was 0.64 in a thin layer chromatography, was a different component from those of European angelicas. The ED_50 value of the fraction was 8, 9, and 16 ㎍/㎖ against HeLa S_3 Hep G_2 and K-562 cells, respectively, while the fraction showed higher ED_50 values against normal cell lines.

In order to study the effect of GA_3 on the phosphorylation of ribosomal proteins during germination in Zea mays, ribosomal proteins were labelled with ^32P, extracted, electrophoresed and autoradiographed. There are five phosphorylated ribosomal proteins. One of these is in 40S subunit and has molecular weight of 33,000 daltons. Others are in 60S subunit and have molecular weights of 37,000, 16,000, 15,200 and 13,500, respectively. Phosphorylation of ribosomal proteins was increased maximum 47.7% in shoots of Zea mays treated with GA_3.

A total of 5,000 ethanolic and methanolic extracts of different plant species from 23 nations including Costa Rica, Vietnam, Philippines, India, South Africa, Pakistan and Peru were evaluated for their larvicidal activities against Aedes aegypti, the major vector of dangue, dangue hemorhagic fever and yellow fever. The larval mortalities were observed 24h after treating the larvae to the extracts. At 500 ppm, 179 extracts showed >80% larval mortality in the 24h exposure. Among the extracts tested, the highest larval mortality was observed in the extracts of Piper guianense, Piper nigrum, Piper mocropodum, Piper sem-immersum, Piper magen and Piper pubicatulum. The LC50 value of extract P. guianense, P. nigrum, P. mocropodum, P. sem-immersum, P. magen and P. pubicatulum were 8.84, 11.48, 8.84, 13.86, 9.48 and 10.12 ppm against Ae. aegypti. It is suggested that P. guianense, P. nigrum, P. mocropodum, P. sem-immersum, P. magen and P. pubicatulum can be developed as potent larvicidal agents against Ae. aegypti.
