<P>Mesenchymal stem cells (MSC) have emerged as a new therapeutic tool for a number of clinical applications, because they have multipotency and paracrine effects via various factors. In the present study, we investigated the effects of adipose-derived MSC (Ad-MSC) transplantation via intrathecal injection through the cisterna magna on cell proliferation and differentiation of endogenous stem cells in the hippocampal dentate gyrus (DG) using Ki-67 (a marker for proliferating cells), and doublecortin (DCX, a marker for neuroblasts). The transplanted Ad-MSC were detected in the meninges, not in the hippocampal parenchyma. However, the number of Ki-67-immunoreactive cells was significantly increased by 83% in the DG 2 days after single Ad-MSC injection, and by 67% at 23 days after repeated Ad-MSC treatment compared with that in the vehicle-treated group after Ad-MSC transplantation. On the other hand, the number of DCX-immunoreactive cells in the DG was not changed at 2 days after single Ad-MSC injection; however, it was significantly increased by 62% 9 days after single Ad-MSC injection. At 23 days after repeated Ad-MSC application, the number of DCX-immunoreactive cells was much more increased (223% of the vehicle-treated group). At this time point, DCX protein levels were also significantly increased compared with those in the vehicle-treated group. These results suggest that the intrathecal injection of Ad-MSC could enhance endogenous cell proliferation, and the repeated Ad-MSC injection could be more efficient for an enhancement of endogenous cell proliferation and differentiation in the brain.</P>

<P><B>Abstract</B></P> <P>The chromenone derivatives (<B>1</B>–<B>4</B>) from the root part of <I>Flemingia philippinensis</I> showed a significant inhibition against bacterial neuraminidase (NA) which plays a pivotal role in a cellular interaction including pathogenesis of bacterial infection and subsequent inflammation. The compounds <B>1</B> and <B>2</B> were the new compounds, philippin D (<B>1</B>) and philippin E (<B>2</B>). In particular, compounds (<B>1</B>–<B>3</B>) exhibited sub micromolar levels of IC<SUB>50</SUB> values with 0.75, 0.54, and 0.07 μM. This is the first report that chromenone skeleton emerged as a lead structure of bacterial NA inhibition. In kinetic study, 8,8-diprenyl compounds displayed competitive inhibitory mode, whereas 4a,8-diprenyl ones showed noncompetitive behavior. It was manifested that all competitive inhibitors (<B>1</B> and <B>2</B>) were simple reversible slow-binding against bacterial NA. The binding affinities (<I>K</I> <SUB>SV</SUB>) of inhibitors to enzyme were agreement with their respective inhibitory potencies. Molecular docking data confirmed that the position of 3-methyl-2-butenyl substituent affects inhibitory mechanism against <I>Cp</I>NanI. The tri-arginyl cluster of R266, R555, and R615 and D291 in NanI tightly interact with the competitive inhibitors.</P>

봄철 관상식물로 활용되는 할미꽃의 종자는 실온보관 수명이 6주 정도로 알려져 있으며 장기저장이 어려운 것으로 알려져 있다. 본 연구는 할미꽃 종자의 실온보관 수명을 밝히고 장기저장 기술 개발을 하고자 실시하였다. 상온보관 기간에 종자 무게 변화는 5.158g에서 8주 후 4.988g, 16주 후 4.793g으로 변화하였고, 발아율은 각각 82±4.9%, 72.5±13.42%, 70.7±8.08%였다. 기건 기간에 따라 수분함량과 발아율은 모두 감소하였으나 처리 모두 70% 이상의 높은 발아율을 보였다. 15℃, RH 15%, 4주간 건조처리 한 결과 무게는 4.0714g 감소하였으며 발아율은 73.3±1.15%였고, 건조 후 -18℃, 12주 저장 후발아율은 72.0±8.16%였다. 건조 단독처리, 건조 및 저온 복합처리 결과 수분함량과 발아율 모두 감소하였으나 70% 이상의 높은 발아율을 보였다. 할미꽃 종자는 실온 보관 수명이 6주 정도로 알려져 있었으나, 본 실험 결과 16주의 실온보관이 가능하다. 또한, 10℃ 이하의 저온건조저장이 어려운 것으로 알려져있었으나, 본 실험 결과 12주의 저온건조저장(-18℃)이 가능하다. 따라서, 할미꽃 종자에 대하여 저온건조저장법에 의한 장기저장기술 개발연구가 요구된다. For long-term storage possibility of Pulsatilla koreana(Yabe ex Nakai) Nakai ex Nakai seeds according to drying and keeping at cold temperature, water content and germination ratio were compared at room temperature drying and cold-temperature storage after drying. Depending on storage condition at room-temperature(25℃), average weight were changing from 5.158g to 4.988g, 4.798g and germination ratios were studied 82±4.9%, 72.5±13.42%, 70.7±8.08%. Although water content was decreasing, germination ratios were showing more than 70%. Drying treatment was oven-drying(15℃, RH 15%) for 4 weeks. After drying, the weight was 4.0714g that was decreased. Germination of drying seeds was 73.3±1.15%, Germination of .18℃ 12 weeks storage seeds with drying was 72.0±8.16%. According to drying and low-temperature treatment, water content were decreasing but still germination ratios were more than 70%. Pulsatilla koreana(Yabe ex Nakai) Nakai ex Nakai seeds were known that life expectancy was 6 weeks at room temperature, but 16 weeks life was possible was shown in this study. Also the seeds were known that was difficult to be storage at less 10℃ and long-term storage, but it was possible to preserve 12 weeks at drying low-temperature and to be long-term storage in this study.
