      • Imidazole이 내독소에 의한 Shock폐에 미치는 효과

        Thromboxane synthetase 억제제인 imidazole이 내독소에 의한 쇽폐에 미치는 영향을 규명하기 위해 토끼에 내독소 4.0㎎/㎏을 정맥으로 투여한 후 1, 4 및 8시간에, 그리고 내독소 4.0㎎/㎏을 정맥으로 투여한 직후 imidazole 50㎎/㎏/2hr의 양으로 2시간마다 정맥으로 투여하면서 내독소 투여후 1, 4 및 8시간에 각각 3마리씩 전자현미경으로 관찰하였다. 그 성적을 요약하면 다음과 같다. 내독소 투여후에는 Ⅰ형 폐포세포 표면의 불규칙화, 내피세포의 종창 및 간질의 부종이 일어났으며 이는 내독소 투여후 4∼8시간에 가장 현저하였다. 내독소 및 imidazole 병합투여군에서는 4시간 이후 내피세포의 종창과 간질의 부종이 일어났으며 그 정도는 내독소 단독투여군에 비해 경하였다. 이상의 소견으로 보아 thromboxane A_2의 합성 억제는 내독소에 의한 폐상해를 상당히 경감시킨다고 인정된다. This study was carried out to investigate the effect of imidazole, known as a inhibitor of thromboxane synthetase, on shock lung induced by E. coli endotoxin. White rabbits were intravenously injected 4.0㎎/㎏ of E. coli O26:B6 lypopolysaccharide. The lungs were observed 1, 4, 10 hours after injection. The other rabbits were given same dose of endotoxin simultaneously with imidazole, 50㎎/㎏ continously injections. The results were summarized as follows: After endotoxin injection, irregularity of the surface of type Ⅰ appeared. These changes were more pronounced 4 to 8 hours after injection. In the lungs of endotoxin and imidazole injected group, endothelial swelling and interstitial edema occurred 4 hours after injections. But these changes were mild in degree compared to endotoxin injected group. From these results, it is concluded that inhibition of thromboxane A_2 synthesis can suppress the endothelial injury and pulmonary edema induced by endotoxin.

      • 중금속 오염 농경지토양에서 중금속을 제거하는 방법

        This study was carried out to find the methods to remove or to make them inactivate from soil saturated with heavy metal cations. The chemical form of heavy metals was influenced by the physicochemical properties of soils, and the adsorption of heavy metals by soils was comformed to the Langmuir equations. The order of Cu contents in the chemical fractions extracted by several reagents was NaOH>EDTA>HNO_3 but that of Zn was EDTA>HNO_3>NaOH. Flooding, liming and addition of chelating reagents and clay minerals to the contaminated soil seems to be desirable methods to alleviate the harmful effect of heavy metals on plant by reducing the heavy metal contents in KNO_3 fraction.

      • 임상 가검물에서 분리된 균(대장균과 황색 포도균)의 약제 내성

        1981년 1월에서 12월말까지 임상 제료에서 분리한 Escherichia coli (E. coli)와 Staphylococcus aureus (S. aureus) 중에서 무작위로 각 50주 씩 선택하여 원판 확산법과 평판 희섭법으로 그 내성주를 조사하여 다음과 같은 성적을 얻었다. 전체적으로 볼 때에 Teteracyclin (TET), Chloramphenicol (CM) 및 Ampicillin (AMP)에 대한 내성주가 가장 많았고, Amikacin (AMK)에 대한 내성주는 가장 적어다. 이것을 균종별로 보면 E. coli에 있어서는 TET, AMP 및 CM는 원판 확산법으로서 74∼76%, 평판 희석법으로서는 78∼94%가 내성주임을 나타내었고, 또 Cephalothin(CEP)은 원판 확산법에서는 8%가 내성주로 나타났으나, 평판 의석법에서는 52%가 내성주로 나타났다. S. aureus에 있어서는 TET는 원판 확산법으로 82%, 평판 희석법으로서는 78%가 내성주로 나왔고 Penicillin (P)은 원판 확산법에서는 56% 평판 희석법으로는 22%의 내성주가 나왔으며, GM과 CM은 원판확산법에서는 18∼44%, 평판 희석법에서는 56∼58%가 내성주로 나왔다. Each 50 strains of Escherichia coli and Staphylococcus aureus were selected at random out of the stock cultures which were isolated from the clinical materials in the bacteriological laboratory of the department of clinical pathology. Kyungpook National University Hospital from the first of January to the end of December in 1982, and their resistance against eight antibiotics were as follows; Escherichia coli and Staphylococcus aureus had the most resistant pattern against tetracyclin, chloramphenicol, and ampicillin, while they had the most sensitive pattern against amikacin in general. Escherichia coli revealed resistant pattern to tetracyclin, ampicillin, and chloramphenicol 74-76% by the disk diffusion method and 78-94% by the plate dilution method, and revealed resistant pattern to cephalothin 8% by the disk diffusion and 52% by the plate dilution. Staphylococcs aureus disclosed resistant pattern to tetracyclin 82% by the disk diffusion and 78% by the plate dilution, and disclosed resistant pattern to penicillin 56% by the disk diffusion and 22% by the plate dilution, and to gentamicin and-chloramphenicol 18-44% by the disk diffusion and 56-58% by the plate dilution.

      • 耕作地의 重金? 汚染度 調査

        This investigation was carried out in order to know the heavy metal (Zn,Pb,Cu,Cd) contents in soil, stream water and plant grown in the area near a closed zinc mine at Sucksan Dong, Goro Myun, Gunwui Gun, Kyungpook Province. The results obtained were as follows: The heavy metal pollution was caused to the waste ore which had been accumulated around the closed zinc mine and the stream water which has sprung out through the waste ore deposit. Total contents of heavy metals in soils, plants and stream water under the influence of waste ore were in the order of Zn, Pb, Cu, Cd. The nearer from the waste ore deposit and stream, the more the contents of heavy metals in soil. The heavy metal contents is higher in paddy field than in dry land, in top soils than in subsoils, and in inlet water than in outlet. The heavy metal contents of the plant was higher in upper part than in underground part. And it was clear that the more contents of heavy metals was determined from the plants grown in soils high in heavy metal contents. The heavy metal contents of the rice plant was highest among the rice, tobacco, and sesame plants in this experiment. The nearer the distance from the waste ore deposit, the higher the contents of heavy metal in stream water.

        E-rosette法에 의한 T cell의 百分率과 螢光抗體法, 感作牛赤血球擔體法, 酵素抗體法 및 마우스赤血球 rosette法에 의한 B cell 의 百分率을 檢査하여 다음과 같이 요약할 수 있었다. 정상성인군의 active T cell은 26±8.2%이었고 total T cell은 72±4.9%이었다. B cell 는 螢光抗體法으로 22±5.4%, 感作牛赤血球擔體法으로 15.8±4.9%, 酵素抗體法으로 15±5.2% 그리고 mouse RBC-rosette法으로 17.2±4.1%이었다. 惡性腫瘍에 있어서 active T cell은 27.±10.4%이었고 total T cell 은 59±11.2%로서 정상인군에 비하여 유의한 감소를 보였다. 良性疾患에 있어서 T cell 은 정상인군에 비하여 유의한 차이를 볼 수 없었다. T cell percent by E-rosette and B cell by fluorescent antibody, senitized ox RBC rosette, enzyme antibody and mouse RBC rosette tests were performed. In healthy adult group the active T cell was 26±8.2% and total T cell was 72±4.9%, B cell by fluorescent antibody was 22.±5.40%, by ox RBC rosette 15.8±4.9%, by enzyme antibody 15±5.2% and by RBC-rosette 17.2±4.1%. In malignant tumor the active T cell was 27±10.4% and total T cell was 59±11.2%. In benign diseases T cell count showed no significant difference from normal adult group.

      • 결핵의 진단법으로서 중합효소 연쇄반응 검사의 가치

        목적 : 결핵균 동정을 위한 중합효소 연쇄반응을 임상병리검사로서 적용하기 위한 기초 실험으로 분석학적 예민도와 특이도를 알아보고 검토된 조건에 의한 중합효소 연쇄반응을 실제 임상 가검물에 적용하기 위함이다. 재료 및 방법 : Ziehl-Neelsen염색 양성인 객담, 뢰벤스타인-젠센(Loewenstein-Jensen) 배지에서 분리된 결핵균 및 M. tuberculosis H37Rv를 비롯한 항산균 표준균주 11주와 비항산균 10주를 사용하여 2가지 DNA의 분리법, 3set 프라이머, 3종류 중합효소, 2가지 미량 원심시험관 및 Easy-Cycler^R(Ericomp사, 미국)와 수조형으로 검사를 시행하여 DNA 증폭 산물을 비교하였다. 또한 이미 지일-닐슨 염색법에 의한 선별검사에서 음성을 나타낸 객담 53예, 소변 101예, 삼출액 128예, 뇌척수액 54예 등 총 370예의 임상가검물을 대상으로 하여 위에서 검토된 중합효소 연쇄반응 조건으로 검사한 성적과 지일-닐슨 염색법, 배양법 성적과 비교하였다. 결과 : DNA분리방법 2가지와 3가지 중합효소는 분석학적 예민도와 5fg로 나타나 차이를 발견할 수 없었으며, 프라이머 P1, P2쌍은 예민도가 5fg, 프라이머 INS1, INS2쌍과 프라이머 Pt3, Pt6쌍의 예민도는 0.5fg이었다. Easy-Cycler^R가 수조형에 비해 높고, 미량 원심시험관은 Robbins사 제품이 Sarstedt사 제품에 비해 높은 분석학적 예민도를 나타냈다. 프라이머 P1, P2쌍과 프라이머 INS1, INS2와 Pt3, Pt6 2쌍의 프라이머의 특이도에 있어서는 항산균 표준균주 중 M. tuberculosis H37Rv, M. tuberculosis ATCC 27294, M. bovis ATCC 29312의 3주에서 특이 증폭 띠를 관찰할 수 있었다. 그러나 비결핵성 항산균 8주에는 특이 증폭 띠를 나타내지 않았다. 또 비항산균 10주에도 특이 증폭 띠가 관찰되지 않았다. 따라서 Robbins사의 미량 원심시험관과 proteinase-K법에 의한 DNA분리방법에 Easy-Cycler^R를 사용하여 프라이머 INS1, INS2쌍으로 일차 중합효소 연쇄반응을 시행한 후 프라이머 Pt3, Pt6쌍으로 이차중합효소 연쇄반응을 시행하는 방법을 370예의 임상가검물에 시행하였고 지일-닐슨 염색법과 결핵균배양검사도 동시에 시행한 결과 중합효소 연쇄반응 양성은 106예(28.6%), 배양 양성은 27예(7.3%) 및 염색 양성은 18예(4.9%)였으며, 결핵환자는 141명(38.1%)였다. 370예의 임상 가검물로 시행한 중합효소 연쇄반응의 진단학적 예민도와 특이도, 양성결과 예측치 및 음성결과 예측치는 68.8%, 96.1%, 91.5% 및 83.3%로 각각 나타났다. 결론 : 결핵균 동정을 위한 중합효소 연쇄반응은 배양검사에 비해 신속하며, 분석학적인 예민도와 특이도가 뛰어나므로 결핵 진단을 위한 임상병리검사로서 유용하다고 사료됨. In this study, We investigated the optimal conditions of polymerase chain reaction(PCR) assay for the rapid identification of Mycobacterium tuberculosis in clinical specimens as routine laboratory test. So we have compared two DNA isolation methods, 3 polymerases, 2 set of primer, 2 kinds of thermocycler and 2 microtubes. We evaluated two DNA isolation methods, bead-beating method and proteinase-K method, the latter was more sensitive than the former. In comparision of two sets of primers, P1 and P2 primers detecting 123-base pair fragment of IS6110 made from Biosnthesis(U. S. A.) and INS1 and INS2 primers detecting 245-base pair fragment of IS986 showed equally sensitive results ie. 5fg. Specificity of primers were tested and INS1 and INS2 primers gave 245-base pair product from M. tuberculosis H37Rv, M. tuberculosis ATCC 27294 and Mycobacterium bovis ATCC 29312 but not from 8 nontuberculous mycobacterial strains such as M. kansasii ATCC 12478, M. terrae ATCC 15755, M. intracellurae ATCC 13950, M. avium ATCC 25281, M. gordonae ATCC 14470, M. fortuitum ATCC 6841, M. smegmatis ATCC 19420, M. scrofulaceum ATCC 19981. Coagulase-positive staphylococcus, coagulase-negative staphylococcus, streptococcus, Esccherichia coli, Serra-tia marcescens, Klebsiella pneumoniae, Enlerobacter aerogenes, Enterobacter cloacae, Acinetobacter calcoaceticus, Pseudomonas aerugonosa showed no 245-base pair amplification product. PCR by the P1 and P2 primers showed 123-base pair amplification product from M. tuberculosis H37Rv, M. tuberculosis ATCC 27294 and Mycobacterium bovis ATCC 29312 but not from 8 nontuberculous mycobacterial strains such as M. kansasii ATCC 12478, M. terrae ATCC 15755, M. intracellularae ATCC 13950, M. avium ATCC 25281, M. gordonae ATCC 14470, M. fortuitum ATCC 6841, M. smegmatis ATCC 19420, M. scrofulaceum ATCC 19981. Coagulase-postive staphylococcus, coagulase-negative staphylococcus, streptococcus, Escherichia coli, Serratia marcescens, Klebsiella pneumoniae, Enlerobacter aerogenes, Enterobacter cloacae, Acinelobacler calcoaceticus, Pseudomonas aeruginosa showed no 123-base pair amplification product. In comparision of polymerase, Taq DNA polymerase^R(Promega Co., U. S. A.) was more cost effective than Ampli Tag^R(Perkin-Elmer Cetus, U. S. A.), but these two were equally sensitive and specific in the detection of M. tuberculosis-specific DNA. Tac polymerase (Korea'Biotech, Korea) showed many nonspecific bands. In comparision of thermocycler, Easy-Cycler^R(Ericomp, U. S. A.) was more sensitive than water-bath type and in that of microtubes Robbin's were more sensitive than Sarstedt's PCR results were compared with results of culture for M. tuberculosis and Ziehl-Neelsen stain in 370 clinical specimens that were negative by initial Ziehl-Neelsen stain. There were 27 specimens(7.3%) that were positive for M. tuberculosis by culture, 18(4.9%) specimens that were positive by Ziehl-Neelsen stain, and 106 specimens(28.6%) that were positive for PCR and 141 cases that were positive for tuberculosis. Overall sensitivity, specificity, positive predictive value and negative predictive value were 68.8, 96.1, 91.5 and 83.3% respectively, for PCR; 19.1, 100, 100, and 66.8%, respectively for culture; and 12.8, 100, 100, and 59.0% respectively for Ziehl-Neelsen stain.

      • 유효미생물에 의한 하수슬러지의 효율적 처리

        This study was conducted to evaluate the effects of microbial inoculation on sewage sludge composting. The number and species of microorganisms in sewage sludge sampled on February were higher than those sampled on August. The composting of sewage sludge is inhibited by the polyacrylamide cation, which was used as a coagulant and known to repress the growth of microorganisms. The growth of all microorganisms was inhibited by the addition of the polyacrylamide cation at a concentration of more than 0.8%. The species and viable counts of microorganisms were observed to increase during composting sewage sludge by inoculation of the effective microorganisms and addition of the pine tree sawdust as a bulking agent, compared with those without inoculation. A variety of organisms in compost(sewage sludge plus sawdust) were observed after composting for 30 days, such as Fragilaria sp., Proales sp., Vorticella sp., Schizothrix sp., Anabaena sp., Zoothaminium sp., Epstylis sp., Arcella sp., Balantidium sp., Actinophrys sp., Synedra sp., Euglypha sp., Ulothrix sp., Anacystis sp., and Clostium sp.
