      • NOD/SCID 마우스 모델을 이용한 인간 제대혈 혼합이식에서의 생착 양상 분석

        연구배경: 제대혈은 골수에 비해 많은 장점을 가지고 있음에도, 이식 후 성적에 있어 총세포수에 의해 좌우되며, 한 개의 태반으로부터 얻을 수 있는 총세포수의 한계가 있어 주로 소아에 국한되고 있다. 따라서 제대혈 응용의 확대를 위해 생착량을 늘이기 위한 방법으로 다중공여자에 의한 제대혈 혼합이식을 늘일 수 있는지 연구하기 위하여, 본 연구에서는 주조직적합성 형별로 조합된 제대혈을 NOD/ SCID 생쥐에 이식하는 방법으로, 혼합생착의 가능성을 연구하였다. 방법: 조합된 2개 공여자로부터 얻어진 제대혈들을 단일이식 및 혼합이식하여, 혼합이식된 숙주에 생착된 세포를 PCR-SSOP 방법을 사용하여 공여자 별로 추적하였다. 또한 각 공여자세포의 상대적 생착비율을 대조군인 단일제대혈 이식에서 얻어진 생착량과 비교하는 정량적 비교도 함께 하였다. 결과: 총단핵구를 혼합이식한 실험군에서는 주조직적합성의 일치도에 관계없이 한쪽공여자의 세포가 다른쪽에 비해 우세한 편향생착을 보였다. 그러나 임파구를 비롯한 Lineage 양성세포를 제거한 실험군에서는 주조직적합성의 불일치에도 불구하고 뚜렸한 생착의 공존이 관찰되었다. 결론: 본 연구는 제대혈의 생착량을 증가를 목적으로 총단핵구를 혼합이식할 경우 한쪽공여자의 세포에 의해 편중되며, 이것을 이식 후 생착과정에서의 두 종류의 세포의 경쟁에 의해 초래된다는 것을 보이고 있다. 또한 이러한 경쟁은 제대혈에 포함되어 있던 임파구들에 의한 것이며, 생착공존을 위하여는 공여자간 주조직적합성의 일치도 보다, 임파구의 제거가 더 중요하다는 것을 보여주고 있다. Background: Although umbilical cord blood (UCB) has been an attractive alternatives for bone marrow cells, application is limited to pediatric case due to limitation in total number of nucleated cells, that is a single most important factor for outcomes of UCB transplantation. Therefore, increasing overall engraftment by mixed transplantation of UCB derived from multiple donor should comprise one strategy to circumvent the posed limitation of UCB transplantation. In order to investigate the feasibility of establishing co-engraftment by multi-donor UCB cells, we carried out a xenotransaplantation study using NOD/SCID mice for systemic analysis of results on the input-based control of single unit transplantation. Methods: UCB units with various extent of HLA-matchings were co-transplanted into NOD/SCID mice along with single unit transplantation control. The relative contributions of engraftment by cells from each donor-derived were analyzed by HLA polymorphism using PCR-SSOP. Results: In all HLA-based mixed transplantation of total nucleated UCB cells, engraftment of one donor predominated over the other despite that equivalent amount of engraftments were achieved by single donor transplantation. When lineage depleted UCB units were co-transplanted, significant degree of co-engraftments were observed regardless of HLA disparity. Conclusion: Our result show that one donor cells dominate over the other in mixed UCB transplantation and that it is due to competition between donor cells during post-transplantation process. Our results suggest that immune cells contained in UCB unit mediate such competitioin and that 6 locus HLA matching would not be sufficient to prevent the competition.

        푸드뱅크 기탁식품의 안전성 확보를 위한 HACCP 제도 적용

        The purpose of this study was to analyze the problems arising from the actual conditions of the Foodbank, and to implement the HACCP system as a solution in terms of increasing the safety of donated food within the Foodbank. In order to apply HACCP system, the entire Foodbank working process such as preparation, collection, transportation, division, and distribution was considered and analyzed to decide the application point for CCPs. Donated foods mainly consisted of processed foods, raw materials, lunch boxes, and cooked foods from mass catering establishments, which dominated over the others in terms of quantity. Cooked foods were divided into three groups based on menu-types and processing methods. Temperature, pH, and aw were measured on cooked foods, and Total Plate Count, Coliforms, E. coli, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, and E. coli O157:H7 were conducted in order to apply a HACCP plan. Form these experiments, temperature, pH, and aw of donated food were likely contributed to microbial growth. Donated foods before HACCP implementation showed high numbers in terms of total plate count and Coliforms, both well over the acceptable standard levels. By setting the CCPs on maintenance of donated food below 10℃ and using a 75 ℃ reheating method, microbiological hazard levels were able to be controlled and lowered. From these results, it is concluded that in order to guarantee food safety, foods donated to the Foodbank must not only maintain a reasonable level of initial microbiological growth, but also must be handled properly through time and temperature controls within the Foodbank system. Furthermore, in terms of implementing the HACCP plan within the Foodbank management structure, basic food safety and sanitation measures, such as reheating facilities and various cold chain systmes such as refrigerated vehicle for food transportation are importantly needed. The training and education of Foodbank personnel and management in areas such as awareness of hygiene and safe food handling and practice are also required and necessary.

        NIHSS 활용을 통한 뇌졸중 환자 간호의 질향상 전략

        문제: 뇌졸중 환자를 돌보는 간호현장에서는 급성기 뇌졸중 환자의 증상악화에 대한 조기사정과 적절한 대처가 매우 중요한 문제이다. 그러나 임상현장에서는 이를 객관화하고 수치화하여 모든 의료진이 의사소통할 수 있는 유용한 도구를 사용하고 있지는 못하는 실정이다. 이러한 문제를 해결하기위해 NIHSS(National Institute of Health Stroke Scale)를 신경과 진료의사를 중심으로활용하고 있지만 아직 간호현장에서는보편적으로사용하지 않고있다. 이에 뇌졸중환자간호에 NIHSS를 활용하여 뇌졸중 증상악화의 조기사정과 빠른 대처로 궁극적으로는 뇌졸중환자 간호의 질을보다 향상시키기 위하여 본활동이 시도되었다. 목적: 첫째, 간호사의 NIHSS 측정 신뢰도의 현수준을 점검하고 이를향상시키기 위한 프로그램을 개발·적용한다. 둘째, 간호사가급성기 뇌졸중환자에게 NIHSS 측정을 통하여 뇌졸중악화를 얼마나 예측할 수 있는지 분석하고 이를 향상시킬수있는 방법을 모색한다. 셋째, 궁극적으로 급성기 뇌졸중 환자의 증상악화의 조기 발견과치료를위한의료진의 올바른 대처지침을 마련하여 이를 적용한다. 의료기관: 경기도에 소재한 대학병원의 뇌졸중 집중치료실 질 향상 활동: 첫째, 간호사의 NIHSS 측정 신뢰도 향상을 위해 「NIHSS 간호사 교육 프로그램」을 마련하였다.

        Synthesis and characterization of β-carotene-loaded albumin nanoparticles by high-speed homogenizer

        β-Carotene (BC) is a strong antioxidant, but its therapeutic application has been limited by its poor aqueous solubility. Protein-based nanoparticles (NPs) have been used to increase the aqueous solubility of hydrophobic drugs. In this study, bovine serum albumin (BSA) was used to prepare NPs to encapsulate BC using a high-speed homogenizer. After homogenization, BC-BSA-NPs were formed and the encapsulated BC was dissolved in phosphate-buffered saline at pH 7.4. Various characterization methods, including dynamic light scattering, transmission electron microscopy, and spectrophotometry, indicated that BCBSA- NPs were spherical with an average size of 112 nm and encapsulated BC with high efficiency. BC encapsulated in NPs was stable during the preparation process and was continuously released in PBS at 37 C for 3 days. The 2,2- diphenyl-1-picrylhydrazyl and ferric reducing antioxidant power assays demonstrated good antioxidant activity of BC-BSA-NPs. These results suggest that BSA-NPs are a promising nanocarrier for the delivery of BC.

      • Comparative Evaluation of Sulfur Compounds Contents and Antiobesity Properties of <i> Allium hookeri</i> Prepared by Different Drying Methods

        <P>Despite the nutritional and medicinal values of<I> Allium hookeri</I>, its unique flavor (onion or garlic taste and smell) coming from sulfur containing compounds limits its usage as functional food. For comparative study,<I> A. hookeri</I> roots were prepared under two different drying conditions, namely, low-temperature drying that minimizes the volatilization of sulfur components and hot-air drying that minimizes the garlic odor and spicy taste of<I> A. hookeri</I>. In GC/MS olfactory system, the odorous chemicals and organosulfur compounds such as diallyl trisulfide, dimethyl trisulfide, and dipropyl trisulfide were significantly decreased in hot-air drying compared to low-temperature drying. The spiciness and saltiness taste were noticeably reduced, while sourness, sweetness, and umami taste were significantly increased in hot-air dried<I> A. hookeri</I> according to electronic tongue. Although the content of volatile sulfur components was present at lower level, the administration of hot-air dried<I> A. hookeri</I> extract (100 mg/kg<I> p.o.</I>) apparently prevented the body weight gain and improved insulin resistance in C57BL/6J obese mice receiving high fat diet. Results suggested that the hot-air dried<I> A. hookeri</I> possessing better taste and odor might be available as functional crop and bioactive diet supplement for the prevention and/or treatment of obesity.</P>

        Anti-allergic Activity of an Ethanol Extract from Salviae miltiorrhiza

        As part of an ongoing investigation aimed at the discovery of novel bioactive medicinal herbs with anti-inflammatory properties, the effects of an ethanolic extract from the parts of Salviae miltiorrhiza Bunge (ESM) were evaluated using in vitro and in vivo animal model analysis. ESM inhibited cyclooxygenase-2 (COX-2) and COX-1-dependent phases of prostaglandin $D_2$ ($PGD_2$) generation in bone marrow-derived mast cells (BMMC) in a concentration-dependent manner with $IC_{50}$ values of $3.96{\mu}g/mL$ and $21.54{\mu}g/mL$, respectively. Furthermore, ESM inhibited leukotriene $C_4$ ($LTC_4$) production with an $IC_{50}$ value of $2.6{\mu}g/mL$. These results clearly demonstrated the dual COX-2 selective/5-lipoxygenase inhibitory activity that ESM possessed. ESM strongly inhibited a degranulation reaction in a dose dependent manner within a BMMC system, with an $IC_{50}$ value of $22.4{\mu}g/mL$. Additionally, ESM was tested in a rat passive cutaneous anaphylaxis (PCA) reaction assay by oral administration (25 to 100 mg/kg). ESM dose-dependently inhibited the PCA reaction, which was activated by anti-dinirophenyl (DNP) IgE. These results suggested that ESM might be beneficial in regulating various allergic reactions.

        Roles of Host Nonhematopoietic Cells in Autoimmunity and Donor Cell Engraftment in Graft-versus-host Disease

        Background: Graft-versus-host disease (GVHD) is initiated when alloreactive donor T cells are primed by host APCs to undergo clonal expansion and maturation. Since there is a controversy regarding the role of nonhematopoietic cells in GVHD, we wanted to investigate the influence of MHC disparity on nonhematopoietic cells on the pathogenesis of GVHD in the MHC-haplomismatched C57BL/6 ($H-2^b$) or DBA/2 $(H-2^b){\rightarrow}$unirradiated ($C57BL/6{\times}DBA/2$) $F_1(BDF_1;\;H-2^{b/d})$ murine model of acute GVHD (aGVHD) or chronic GVHD (cGVHD). Methods: We generated ($BDF_1{\rightarrow}C57BL/6$), ($BDF_1{\rightarrow}DBA/2$), and ($BDF1{\rightarrow}BDF_1$) chimeras and examined GVHD-related parameters and donor cell engraftment in those chimeras. Results: Using this experimental system, we found that 1) severe aGVHD across MHC Ag barrier depends on the expression of nonhematopoietically rather than hematopoietically derived alloAgs for maximal GVHD manifestations; 2) host APCs were sufficient to break B cell tolerance to self molecules in cGVHD, whereas host APCs were insufficient to induce autoimmunity in aGVHD; 3) donor cell engraftment was greatly enhanced in the host with MHC-matched nonhematopoietic cells. Conclusion: Taken together, our results provide an insight into how MHC disparity on GVHD target organs contribute to the pathogenesis of GVHD.

        모바일 디바이스의 베젤이 디바이스 내에서 보여지는 상품의 가치 평가에 미치는 영향 -베젤 이펙트-

        본 연구는 경험적 연구를 통해 모바일 디바이스의 베젤 때문에 화면에 나타나는 상품의 가치 평가가 달라 질 것이라는 새로운 관점을 제안한다. 이를 위해 실험 1에서는 A사와 B 사에서 출시된 핸드폰과 태블릿 디바이스의 베 젤에 대한 디자인 평가를 실시하였다. 그 결과 A사 제품의 디자인이 B사제품의 디자인보다 우수하며, 핸드폰의 디자인이 태블릿 제품의 디자인보다 우수하다고 평가되었다. 실험 2에서는 이러한 디자인 평가의 차이가 각 디바이스에서 보여지는 제품의 가치 평가에 영향을 미치는지 살펴보았다. 실험 2의 결과, 모든 브랜드에서 핸드폰에서 보여진 상품의 가치가 태블릿에서 보여진 상품의 가치보다 높았다. 브랜드의 효과는 유의하지 않았다. 본 연구의 결과는 모바일 디바이스 디자인은 해당 제품의 가치를 향상시키는 효과를 넘어, 디바이스에서 제공되는 상품의 가치에까지 영향을 준다는 사실을 밝힘으로써 학문적, 산업적 중요한 함의를 가진다. The current study offers a new perspective of importance of bezel design in mobile device:bezel design in mobile device is important in that it affects the perceived value of product displayed in the devices. To verify this claim, we carried out a design assessment of cell phones and tablet PCs of Company A and B. In addition, we investigated how such a difference in the valuation of design influences the assessment of the value of products shown in each device. The results showed that products displayed in cell phones were valued higher than those shown in tablet PCs. The effect of the brand of company was not significant. The findings imply that design of bezel in mobile device has an effect not only on the value of the device itself, but also on the value of the products shown in that device. Possible limitations were discussed.
