        This study was conducted to investigate the volatile free fatty acid contents in the domestic market cheeses. The results are summarized as follows : 1. There was an average of pH 5.46, and nonsignificant. 2. Total volatile free fatty acid showed an average of 6.33ml, the highest (7.2ml) in sample D. 3. Volatile free fatty acid showed the highest (55.92%) in acetic acid, lowest (10.92%) in caprylic acid. Butylic acid and caproic acid represented 15.94%, 17.23%, respectively.

        Cell differentiation is a very important process in morphogenesis, and epithelial-mesenchymal interaction may play an important role in cell differentiation. Therefore, during development of root dentin, arcllular cementum and periodontal ligament, the changes of the morphology of dental mesenchymal cells and of the distribution of extracellular matrix proteins - type I collagen, type ID[ collagen, and fibronectin(FN) - are investigated using 14-day-old Sprague-Dawley rats. 1. In the zone of Hertwig's epithelial root sheath, intact epithelial root sheath which is formed with two layers of epithelial a -Rs and covered with basement membranes is observed. Cells of dental follicle and of dental papilla adjacent to the epithelial root sheath are not differentiated yet. In the zone of differentiation, odontoblasts and cementoblasts are differentiated. Cells in the middle of the dental follicle are differentiated into fibroblasts. In the zone of formation, dentin and acellular cementum are observed. The fibroblasts in the forming periodontal ligament are oriented obliquely ard synthesize the collagen fibrils parallel to their direction. 2. In the zone of HERS, immunoreactivity of FN is very strong at the inner basement membrane of the HERS. In this zone, immunoreactivity of type Ill collagen in the extracellular matrix is stronger than in other zones. 3. In the zone of differentiation, cell body of odontoblast shows little of immunoreactivity to FN and collagens. But cell membrane of proximal surfaces of odontoblasts shows very strong immunoreactivity to FN. Along the cementoblasts attached root dentin, strong immunoreactivity to FN is shown. 4. In the zone of formation, acellular cementum matrix is stained strongly to FN. Periodontal ligament shows strong immunoreactivity to FN, type I collagen, and type III collagen with their fibrils' direction. Predentin is stained moderately with FN, type I and type III collagen, but immunoreactivity of mineralized dentin is weaker. 5. Hertwig' s epithelial root sheath ;plays an important role in cell differentiation by the epithelial-mesenchymal interaction. Extracellular matrix of the mesenchymal cells seems to affect the cell differentiation directly and/or indirectly.

        시험관내 A-253 세포주의 방사선 및 항암제 감수성에 관한 연구

        The purpose of this study was to aid in the prediction of tumor cell tolerance to radiotherapy and/or chemotherapy. For this study, cell surviving curves were obtained for human epidermoid carcinoma A-253 cell line using semiautomated MTT assay. 2, 4, 6, 8, 10 Gy were irradiated at a dose rate of 210 cGy/min using 60Co Irradiator ALDORADO 8. After irradiation, A-253 cell lines(2×10⁴ cells/ml) were exposed to bleomycin or cisplatin for 1 hour. The viable cells were determined for each radiation dose with/without 2 ㎍/ml of drug at the 3rd day. And they were compared to control values. The results were obtained as follows: 1. The surviving curve with gentle slope was obtained after irradiation of 2,4,6,8,10 Gy on A-253 cell line. 2. The cytotoxicity of bleomycin or cisplatin at the concentration of 2㎍/ml was great on A-253 cell line. But, there was no significant difference between the cytotoxicity of bleomycin and that of cisplatin. 3. There were significant differences of surviving fractions after irradiation with 2㎍/ml of bleomycin compared with irradiation only on A-253 cell line. 4. There were significant differences of surviving fractions after irradiation with 2㎍/ml of cisplatin compared with irradiation only on A-253 cell line. 5. There were no significant differences of surviving fractions between the groups of irradiation with bleomycin and the groups of irradiation with cisplatin on A-253 cell line

        The prevalence of PPNG among pretreated gonorrhea cases isolated at the STD clinic of Choong-Ku Public Health Center in Seoul has been studied and reported annually since 1981. In 1991, 123 strains of N.gonorrhoeae were isolated, among which 58(47.1%) were PPNG. In 1992, 98 starains of N.gonorrhoeae were isolated, among which 51(52.0%) were PPNG. In all, 109(49.3%) strains were found to be PPNG among 221 strains isolated between 1991-1992. The prevalence of PPNG in Seoul showed increased tendency till 1989, thereafter, it has been stationary or slightly decreasing.

        Alteration in Phenolic Compounds and Antioxidant Activities of Aronia melanocarpa Ethanol Extracts following Fermentation Using Different Strains of Leuconostoc mesenteroides to Develop Natural Antibiotic Alternative

        Antioxidant activity is important for reducing oxidative stress that causes various metabolic disorders. Metabolic disorders are highly related to loss of productivity in livestock. Therefore, development of effective antioxidant compounds originating from plants is important for organic agriculture. Phenolic compounds in edible plants are regarded as major components relevant to antioxidant activity. The present study investigated the changes in antioxidant activity and phenolic compound profiles of Aronia (Aronia meloncarpa) by fermentation using different strains of Leuconostoc mesenteroides. A total of 5 strains of L. mesenteroides were used as starter cultures and their β-glucosidase activities were measured. A total of 6 experiment runs were prepared, one for control (uninoculated) and the others (inoculated) for treatments. For biological activity, antioxidant and antibacterial activities were measured. For phenolic compound profiling, TLC and HPLC analysis were performed. The strains of KACC12313 and KACC12315 showed greater enzyme activity than others. Treatment with KCCM35046 showed strong and broad antibacterial activity against to Listeria monocytogenes. Treatments with KCCM35046 and KACC12315 showed the highest total polyphenol content. The highest antioxidant activity was found in KACC12315 treatment. No remarkable alteration was found in thin layer chromatography (TLC) analysis. In phenolic compound profiling analysis, KCCM35046 showed notable alteration in compound area ratio compared to others and also showed the highest caffeic acid content. In chlorogenic acid, treatments with KCCM35046 and KACC12315 showed great content than others. Treatment with KACC12315 showed the greatest content of trans-ferulic acid. As a result of relative performance indexing analysis, L. mesenteroides KCCM35046 and KACC12315 were selected as the best strain for the fermentation of Aronia.

      • Evaluation of multi-walled carbon nanotubes-induced fibrosis in lung tissue from Fischer 344 rats

        Multi-walled carbon nanotubes (MWCNTs) are nanomaterials consisting of concentric layers of cylindrical carbon nanotubes (CNTs) with a diameter less than 100 nm, light weight, extreme tensile strength, thermal conductivity, and electrical property that are potential applications in the engineering, electronics, and medical industries. Because of their unique physicochemical properties, MWCNTs are easily aerosolized and inhalation exposure during production, use, and disposal.A recent study showed that MWCNTs induce many toxicological effects in mouse lungs after both aspiration and inhalation exposure between in vivo and in vitro. Exposure to MWCNTs resulted in not only pulmonary inflammation and fibrotic reactions .Thus, to evaluate the fibrosis of MWCNTs on lung tissues in vivo, eight-week-old rats were divided into four groups, a fresh air control (0 mg/m3), low (0.20 mg/m3), middle (0.50 mg/m3), and high (1.00 mg/m3) dose group, and exposed to MWCNTs via nose-only inhalation for 28 days. Lung tissues were isolated from five rats in each group on day 0 and 28 following the 28-days exposure. To investigate the mode of fibrosis, proteinase (MMP-9), proteinase inhibitor (TIMP)-1), hematopoietic cytokine levels (G-CS), GM-CSF, and VEGF) and their gene expression in the lung tissue were measured. As a results, there was no significant difference in inflammatory cytokine levels and their gene expression between control and exposure groups. Therefore, these results suggest concentration-dependent inflammatory response by exposure to MWCNTs are mimicked.

      • Effects of Interleukin-6 T15A Single Nucleotide Polymorphism on Baseline Peritoneal Solute Transport Rate in Incident Peritoneal Dialysis Patients

        <B>Objective</B><P> To study the genetic effects of various inflammatory cytokines on peritoneal solute transport rate (PSTR) in incident Korean peritoneal dialysis (PD) patients. </P><B>Design</B><P> Case-control association study. </P><B>Methods</B><P> 132 patients with baseline peritoneal equilibration test within 1 - 3 months after starting PD were enrolled. We analyzed the influence of single nucleotide polymorphisms (SNPs) of interleukin-6 (IL-6; -572G/C, T15A), tumor necrosis factor-alpha (TNF-α; -1031C/T, -863C/A, -308G/A), and IL-10 (-1082A/G, -592A/C) on baseline PSTR. Clinical parameters such as age, gender, presence of diabetes mellitus, comorbidity, C-reactive protein, and residual renal function were also included as covariates. </P><B>Results</B><P> The T15A SNP of IL-6 (rs13306435) was associated with PSTR. Patients with TA genotype ( n = 18) had significantly lower D4/P creatinine (0.65 ± 0.087 vs 0.73 ± 0.110, p = 0.0046) and higher D4/D0 glucose (0.39 ± 0.174 vs 0.31 ± 0.119, p = 0.027) than patients with TT genotype ( n = 114). The log value of the dialysate appearance rate of IL-6 had a strong positive correlation with D4/P creatinine ( r<SUP>2</SUP> = 0.1294, p @@<@@ 0.0001) and was significantly lower in the TA genotype than the TT genotype (201.7 ± 14.42 vs 116.8 ± 88.91 pg/minute, p = 0.0358). By multiple logistic regression, TA genotype was negatively associated with a higher PSTR (high or high average; odds ratio 0.18; 95% confidence interval 0.048 - 0.666). </P><B>Conclusions</B><P> In incident Korean PD patients, T15A polymorphism of IL-6 is associated with dialysate IL-6 concentration and baseline PSTR. </P>

        Optimization of Carbon Sources to Improve Antioxidant Activity in Solid State Fermentation of Persimmon peel Using Lactic Acid Bacteria

        The present study was conducted to develop persimmon peel, a by-product of dried persimmon manufacturing, as a feed additive via lactic acid bacteria fermentation. Pediococcus pentosaceus, Lactobacillus plantarum, and three strains of Leuconostoc mesenteroides were used as a starter culture in the solid state fermentation of persimmon peel, and antioxidant activity and total polyphenol content were assessed. Leuconostoc mesenteroides KCTC 3100 showed high antioxidant activity (p<0.05), whereas Pediococcus pentosaceus showed high total polyphenol content (p<0.05). These two strains were thus selected as starter culture strains. Glucose, sucrose and molasses were used as variables for optimization and a total 15 experimental runs were produced according to Box-Behnken design. Regarding significant effects of variables, molasses showed linear and square effects on antioxidant activity of persimmon peel fermentation (p<0.05). In conclusion, optimum concentrations of glucose, sucrose, and molasses were determined to be 4.2, 3.9 and 5.3 g/L, respectively, using a response surface model. Antioxidant activity was also improved 2.5 fold after optimization.

        Molecular Analysis of Isoniazid-Resistance Related Genes of Mycobacterium tuberculosis Isolated from Korea

        Resistance to isoniazid (INH), which is one of the most important drugs in Mycobacterium tuberculosis chemotherapy, has been associated with mutations in genes encoding the mycobacterial catalse-peroxidase (katG), the enoyl acyl carrier protein (ACP) reductase (inhA), alkyl hydroperoxide reductase (ahpC), beta-ketoacyl acyl carrier protein synthase (kasA), and NADH dehydrogenase (ndh). In this study, we examined INH-resistance related genes in 50 INH-resistant and 24 INH-susceptible isolates by PCR-sequence analysis. In brief, mutations at the katG gene were found at codon 315 alone (2/50), at codon 463 alone (19/50), and both at 315 and 463 (29/50). However, while mutations at codon 315 were only detected in INH-resistant isolates, mutations at codon 463 were also detected in INH-susceptible isolates indicating mutations at 463 alone do not seem to confer resistance to INH. Similar to the case of katG 463, some of inhA mutations were also found among INH-susceptible isolates. For example, whereas mutations at 8 upstream of the start codon (UPS) and 15 UPS of the inhA gene were detected only in INH-resistant isolates, mutations at 101, 115, and 125 UPS were detected only in INH-susceptible isolates. Many different kinds of mutations were detected in INH­resistant isolates at ahpC, oxyR gene, and intergenic region of the oxyR-ahpC genes. Howerver, the mutations were not found oxyR and the intergenic regions in INH-susceptible isolates. No mutations were found at either kasA or at ndh gene among INH-resistant isolates. In conclusion, some of mutations such as katG 315, inhA promotor region, and oxyR-ahpC seem to be strongly related to INH-resistance. Currently we are developing a molecular diagnostic method based on these results.
