        방선균 분리주가 생산하는 Phospholipase C 저해물질인 MT2617-2B의 분리 및 특성

        방선균 분리주 MT2617-2의 배양액으로 부터 phospholipase C (PLC) 저해물질인 MT2617-2B를 n-butanol 추출 및 column chromatography 법을 이용하여 분리하였다. MT2617-2B는 IR ^13C- 및 ^1H-NMR 그리고 ESI-MS에 의한 구조분석 결과, 한 개의 hemiketal ring, polyhydroxyl 및polymethyl groups으로 구성되었으며 side chain으로 한 개씩의 malonate 및 guanidine group을 가지는 분자량 1057의 macrolide 화합물이었다. 따라서, MT2617-2B를 기존의 macrolide 항생제인 copiamycin 및 niphithricin A로 동정하였다. 한편, MT2617-2B는 methanol 용액에서 실온에서 방치하였을 때 도일한 분자량을 가진 두 개의 이성질체를 생성하였다. PLC γ1과 -β1에 대해 각각 25 및 50㎍/㎖의 IC_50 값을 가지며, Staphylococcus aureus 와 Candida albicans에 대해서는 항균활성을 나타내지만 Escherichia coli에는 나타내지 않았다. A phospholipase C (PLC) inhibitor (MT2617-2B) was isolated from the culture broth of actionmycetes isolate MT2617-2 by the extraction with n-butanol and column chromatographic techniques. The molecular weight of the inhibitor was 1057, by the spectroscopic analyses of IR ^13C- and ^1H-NMR and ESI-MS. The chemical structure of MT2617-2B was found to be a macrolide compound consisted of a hemiketal ring, polyhydroxyl and polymethyl groups, which had a malonate and guanidine group as its side chain. MT2617-2B produced its two isomers having the same molecular weight by standing in methanol solution at room temperature. Therefore, MT2617-2B was identified as copiamycin and niphithricin A, macrolide antibiotics. The values of IC_50 against PLC γ1 and PLC-β1 were 25 and 50㎍/㎖, respectively. MT2617-2B had antimicrobial activities against Staphylococcus aureus and Candida albicans, but not against Escherichia coli.

      • 전기영동법을 이용한 제주재래마유 단백질의 특성연구

        This study was carried out to determined the physico-chemical properties of milk protein of Cheju native mare milk using polyacrylamide gel and sodium dodecyl sulfate -polyacrylamide gel elrctroporesis and to obtain fundamental information for the preservation of Cheju native horse. The results obtained are summarized as follows; 1. Whey protein of Cheju native mare's milk were separated on polyacrylamide gel electrophoresis into immunoglobulin, serum albumin, α-lactalbumin. β-lactoglobulin , whey protein. 1) The whey protein showed four variable bands which were considered to be different types (A,B, C and D). 2) Seven whey phenotypes (AA,BB,AB,AC,BD,BC and CD) were observed. The distribution of AA, AB and BB was 13.4% 40% and 13.4%, respectively, and that of the other individual phenotypes was less than 10%. 2. Casein of Cheju native mare's milk was separated into three malor components α_(s)-.β-and k-casein on polyacrylamide gel electrophoresis. 1) α_(s)-casein zone was found in as hort distance wlth 4-7 zones. 2) The ratio of α_(s)-casein to β-casein was 1:1.3 and simllar to that reported on thorughbred mare milk (1:1.6) but higher than that β-casein found in Holste in cow's milk (1:0.6) 3) k-casein was found the form of a polymer held together by intermolecular dlsulfide bond. 3. Whey protein of Cheju native mare's milk were separated on sodium dodecyl sulfate-polyacrylamide gel electrophoresis in to 7 bands. The molecular weights of correspond to approximately 66,500, 66,000, 65,000, 59,500, 19,500, 16,500, 12,000 daltons, respectively.

      • NP계 비이온 계면활성제/물/용제 시스템의 상거동에 관한 연구

        본 연구에서는 polyoxyethylene nonylphenyl ether(NP) 계의 비이온 계면활성제, 물, 용제의 3성분 계에 대하여 계면활성제, 용제, 온도 등의 변화에 따른 상평형 실험을 수행하였다. 단일상의 microemulsion은 NP-8 시스템의 경우 50℃ 부근의 조건에서 가장 넓은 영역에 걸쳐서 형성되었으며, 계면활성제의 친수성이 증가할수록 저농도의 계면활성제 영역에서 형성되었다. NP series의 계면활성제 시스템에서 생성된 1 phase microemulsion은 bicontinuous microemulsion 보다는 oil-continuous microemulsioin에 가까운 것으로 전기전도도 측정으로부터 확인할 수 있었다. 동일한 계면활성제 시스템을 온도에 따라 비교하였을 때 온도가 증가함에 따라 상전이가 계면활성제의 고농도 방향으로 진행하였으며, 이때 형성된 1 phase microemulsion은 용제 apex 쪽에서 형성되었다. 즉, 비이온 계면활성제의 친수성이 증가할수록 1 phase microemulsion이 생성되는 온도는 증가하며, 계면활성제 농도는 낮아지는 것을 알 수 있었다. 용제로 D-limonene 대신에 YK series의 탄화수소를 사용했을 경우에는 middle-phase microemulsion을 포함한 3 phase 영역이나 1 phase의 microemulsion이 형성되지 않음을 알 수 있었다. Phase behavior for systems containing polyoxyethylene nonylphehyl ether(NP) nonionic surfactant, water, and solvent was studied. One-phase microemulsion was found over a wide range of compositions at 50℃ in NP-8 surfactant system and surfactant concentration necessary for one-phase microemulsion decreased with an increase in surfactant hydrophilicity. Electrical conductivity measurements showed that the one-phase microemulsion formed with NP series nonionic surfactants used during this study was oil-continuous rather than bicontinuous. Increasing temperature shifted the same phase transitions to the direction of increase in surfactant concentration and one-phase microemulsion was formed near the apex of a solvent. It was also found that temperature necessary for one-phase microemulsion increased with an increase in surfactant hydrophilicity. Neither three-phase region containing middle-phase microemulsion nor one-phase microemulsion was found in the same surfactant system except that YK series hydrocarbon solvent was used instead of D-limonene.

      • 농산물 전자상거래 교육을 위한 농업용 E-Business 시스템 구현

        World wide spread of internet technologies has resulted in economic development and social change in many countries. In particular, it has contributed in creating a cyber shopping system in which fast transactions in commerce are made possible. The electronic marketing system in the Korean agricultural sector is still in its infant stage of development. But in the near future I-business will become popular in agricultural marketing. The professional agrirultural industry managers have to be well prepared to adapt themselves to the challenges of E-commerce that is now being used in the global market. Thus, we established the I-business sys-tem that will open a virtual shop and operate it fer education and practice in Korea National Agricultural Col-lege(KNAC). The E-business system's main organization is made up of five modules. information management of agricultural products, marketing management, marketing analysis, consumers and stores' management and fifteen sub modules of these modules. Complementary to these, we included Webpage designing for agricultural products. We constructed DB with Microsoft Access DB and SQL. We used DHTML(based on ASP, Java Script and CSS on Windows NT and 98) to make our home page. Also we gave EC's decicated server and If address and connected interact with "shop.kn.ac.kr" by E1 Line(2048Mbps). This system is designed primarily for stu-dents after graduation and practicing in their own farms. As such, it is also envisioned to become a big farmers' association shopping mall where individual Homepages of former students are connected with KNAC's main Homepage to facilitate easy access to various agricultural products. When the EC system expands, we plan to add more useful skills and develop new versions.

      • 비혈연간 동종골수이식에 의해 치료된 중증 재생불량성빈혈 1예

        비혈연간 동종골수이식은 혈연간 동종골수이식이 불가능한 환자에서 기존의 면역역제요법으로 효과가 없었던 경우나 재발한 경우에 적응증이 될수 있다. 이식 거부와 이식편대숙주병의 발생이 골수 이식의 성공률을 감소시키므로, 보다 강력한 conditioning regimen의 사용과 T cell이 제거된 골수를 사용하는 것이 필요하다. 기존의 high dose corticosteroid나 cyclosporine에 대하여 불응성인 이식편대숙주병의 치료를 위해서 ATG와 Mycophenolate mofetil (Cellcept®) 의 사용이나 Tacrolimus, Methotrexate와 같은 다른 면역억제제치료를 고려해 볼 수 있다. It is known that allogenic bone marrow transplantation is the most effective treatment for aplastic anemia. However, this treatment is only applicable to a minority of patients because the proportion of patients who have HLA-matched siblings about 25% to 30% of all the aplastic anemia patients. In the case of the absence of HLA-matched donor, unrelated allogenic bone marrow transplantation is considered. However, (because of severe acute and chronic GVHD and graft failure) the success rate of this approach is only 20~30%. Recently, it was reported that increased immunosuppressive condition combined with T cell depletion of the marrow graft would result in greater success. There was no case of successfully treated aplastic anemia patient by unrelated allogenic bone marrow transplantation in Korea. Here, we report a case of severe aplastic anemia successfully treated with unrelated allogenic bone marrow transplantation.

      • Filton과 활성탄에 의한 Emulsified Oil흡착에 관한 연구

        This study is an experimental research to determine the adsorption capacity of emulsified oil of Filton(FA) in comparison with that of Activated Carbon(AC). Batch type study was conducted with varying the following factors; pH, temperature, adsorbent size, and dosage. The results are as follows. 1. Freundlich isotherms of emulsified oil were; q = ?? for FA and q = ?? for AC. 2. Adsorption capacity of FA was 18.4 mg of Oil/g of FA, while that of AC was 12.8mg of Oil/g of AC. 3. Above 50% of FA adsorption was occurred within 30 minutes of contact time, the equilibrium state was reached in three hours of contact time both FA and AC. 4. Removal efficiency of emulsified oil by adsorbents was increased with the smaller size of adsorbents, the larger amount of dosage, the higher temperature, and the lower value of pH.

        생체용 Ti₁-x-Zrx합금의 기계적 성질과 내부식성

        While titanium and its alloys has been well studied for a long time, new interest in these materials and their properties has arisen in recent years with respect to their application as biomedical metallic materials. For practical purposes, the following performances are essential for biomaterials. (1) low specific gravity (2) high corrosion resistance (3) good biostability and biocompatibility (4) sufficient mechanical properties (5) non-toxicity To develop biomedical implant materials having all the properties mentioned above, new titanium-zirconium alloys were prepared in this study. Mechanical properties and corrosion resistance were then investigated for as-cast and as-homogeneized Ti₁_x-Zrx alloys in order to reveal their possible use for biomaterials. From the experiments, the following results are obtained: 1. No martensite structure are observed in the specimens made of pure Ti and Zr. In contrast them, Ti-Zr alloys containing 18wt%Zr, 24wt%Zr and 48wt%Zr show a kind of martensite structure regardless heat treatment. 2. Fineness of structures for as-cast-alloys seems to vary with alloy composition. Ti-Zr alloy containing 48wt%Zr shows the finest microstructure. 3. Hardness of Ti-Zr alloys shows continuous change through the system and the alloys containing 48 to 60wt%Zr indicate higher hardness compared with those of pure Ti and other alloys. It is well known that hardness is usually changed with substitutional atoms, and effect of solute atom on hardness is greatest with an equi-atomic composition. The greatest hardness which was obtained in Ti-Zr alloys containing 48 to 60wt%Zr is therefore reasonable. Also, enhanced hardness can be explained on elastic interaction between dislocation and substitutional solute atoms in crystal structure and on fineness of structure. 4. Pure Ti and Zr alloys containing 6wt%Zr, 12wt%Zr, 18wt%Zr, 36wt%Zr show better corrosion resistance compared with those of pure Zr, stainless steel(SUS 316L) and the Ti-Zr alloy containing 48wt%Zr. 5. Comparing hardness, microstructure and corrosion of Ti₁_x-Zrx alloys, no large differences between as-cast and as-homogenized alloys are seen.
