Transcription activator-like effector nucleases (TALENs) are powerful tools for targeted genome editing in diverse cell types and organisms. However, the highly identical TALE repeat sequences make it challenging to assemble TALEs using conventional cloning approaches, and multiple repeats in one plasmid are easily catalyzed for homologous recombination in bacteria. Although the methods for TALE assembly are constantly improving, these methods are not convenient because of laborious assembly steps or large module libraries, limiting their broad utility. To overcome the barrier of multiple assembly steps, we report a one-step system for the convenient and flexible assembly of a 180 TALE module library. This study is the first demonstration to ligate 9 mono-/dimer modules and one circular TALEN backbone vector in a one step process, generating 9.5 to 18.5 repeat sequences with an overall assembly rate higher than 50%. This system makes TALEN assembly much simpler than the conventional cloning of two DNA fragments because this strategy combines digestion and ligation into one step using circular vectors and different modules to avoid gel extraction. Therefore, this system provides a convenient tool for the application of TALEN-mediated genome editing in scientific studies and clinical trials.

The hindgut of horses is an anaerobic fermentative chamber for a complex and dynamic microbial population, which plays a critical role in health and energy requirements. Research on the gut microbiota of Mongolian horses has not been reported until now as far as we know. Mongolian horse is a major local breed in China. We performed high-throughput sequencing of the 16S rRNA genes V4 hypervariable regions from gut fecal material to characterize the gut microbiota of Mongolian horses and compare them to the microbiota in Thoroughbred horses. Fourteen Mongolian and 19 Thoroughbred horses were used in the study. A total of 593,678 sequence reads were obtained from 33 samples analyzed, which were found to belong to 16 phyla and 75 genera. The bacterial community compositions were similar for the two breeds. Firmicutes (56% in Mongolian horses and 53% in Thoroughbred horses) and Bacteroidetes (33% and 32% respectively) were the most abundant and predominant phyla followed by Spirochaete, Verrucomicrobia, Proteobacteria, and Fibrobacteres. Of these 16 phyla, five (Synergistetes, Planctomycetes, Proteobacteria, TM7, and Chloroflexi) were significantly different (p<0.05) between the two breeds. At the genus level, Treponema was the most abundant genus (43% in Mongolian horses vs 29% in Thoroughbred horses), followed by Ruminococcus, Roseburia, Pseudobutyrivibrio, and Anaeroplasma, which were detected in higher distribution proportion in Mongolian horses than in Thoroughbred horses. In contrast, Oscillibacter, Fibrobacter, Methanocorpusculum, and Succinivibrio levels were lower in Mongolian horses. Among 75 genera, 30 genera were significantly different (p<0.05) between the two breeds. We found that the environment was one of very important factors that influenced horse gut microbiota. These findings provide novel information about the gut microbiota of Mongolian horses and a foundation for future investigations of gut bacterial factors that may influence the development and progression of gastrointestinal disease in horses.

A convolution neural networks (CNNs) has demonstrated outstanding performance compared to otheralgorithms in the field of face recognition. Regarding the over-fitting problem of CNN, researchers haveproposed a residual network to ease the training for recognition accuracy improvement. In this study, a novelface recognition model based on game theory for call-over in the classroom was proposed. In the proposedscheme, an image with multiple faces was used as input, and the residual network identified each face with aconfidence score to form a list of student identities. Face tracking of the same identity or low confidence weredetermined to be the optimisation objective, with the game participants set formed from the student identitylist. Game theory optimises the authentication strategy according to the confidence value and identity set toimprove recognition accuracy. We observed that there exists an optimal mapping relation between face andidentity to avoid multiple faces associated with one identity in the proposed scheme and that the proposedgame-based scheme can reduce the error rate, as compared to the existing schemes with deeper neural network.
Stability Analysis and Evaluation of ESR for Hysteresis Controlled Buck Converter

Buck converter using hysteresis controller has been widely used for power supply in industry applications owing to its instantaneous load transient. However the equivalent series resistance (ESR) of output capacitor has a significant effect for the stability performance. In this paper, the critical value of ESR is analyzed and calculated in the hysteresis controlled buck converter. Finally, simulation results indicate that the larger output capacitor ESR is necessary to make sure the converter operate normally, otherwise, the converter will operate abnormally.

As load-bearing material in biomedical-applications as cartilage replacement, artificial meniscus and tendons,etc., poly(vinyl alcohol) (PVA)/graphene oxide (GO)-tannic acid (TA) nano-composite hydrogelswith multiple-crosslinking network were fabricated by establishing freezing/thawing-annealing-swellingmethod. By TA anchoring, PVA molecules were grafted onto GO surface efficiently, and strong interfacialinteraction led to exfoliation and uniform distribution of GO in matrix. By introducing annealing process,the crystallinity and crystallite size of PVA increased and introducing GO-TA led to more perfect and densercrystalline structure, while physical crosslinking network centered on GO-TA and hydrogen bondabundantcrystalline phase formed, resulting in increasing crosslinking density of hydrogel. By furtherswelling in CaCl2 aqueous solution, hydroxyl-Ca2+ coordination formed, and multiple-crosslinking networkwas constructed with high crosslinking density. The tensile strength and fracture toughness of compositehydrogel were remarkably improved, reaching 14.38 MPa/27.93 MJ/m3, approximately 11-/26-foldhigher than those of neat PVA hydrogel, while tearing strength was significantly enhanced, attributed tohigh energy dissipation through unzipping multiple interactions.

A method for detecting foreign substances in mould based on scatter grams was presented to protect mouldsautomatically during moulding production. This paper proposes a wavelet transform foreign detection methodbased on Monte Carlo analysis mechanism to identify foreign objects in the tube. We use the Monte Carlomethod to evaluate the image, and obtain the width of the confidence interval by the deviation statistical grayhistogram to divide the image type. In order to stabilize the performance of the algorithm, the high-frequencyimage and the low-frequency image are respectively drawn. By analyzing the position distribution of the pixelgray in the two images, the suspected foreign object region is obtained. The experiments demonstrate theeffectiveness of our approach by evaluating the labeled data.
