<P>Endothelial inflammation is regulated by a complex molecular mechanism. TC1(C8orf4) is a novel regulator implicated in cancer and inflammation. It is a small protein conserved well among vertebrates. In zebrafish embryos, it is mostly expressed in angio-hematopoietic system and the overexpression induces edema. In human aortic endothelial cells and umbilical vein endothelial cells, TC1 transfection up-regulates key inflammatory cytokines, enzymes, and adhesion proteins including IL-6, IL-1alpha, COX-2, CXCL1, CCL5, CCL2, IL-8, ICAM1, VCAM1, and E-selectin, while TC1 knockdown down-regulates them. TC1 also enhances inflammatory parameters such as monocyte-endothelial adhesion and endothelial monolayer permeability. TC1 is up-regulated by IL-1beta, TNF-alpha, LPS, and phorbol ester, and the up-regulation is inhibited by I-kappaB-kinase inhibitors. TC1, in turn, enhances the nuclear translocation of RelA and the DNA binding activity, suggesting a biological role of amplifying NF-kappaB signaling via a positive feedback. Our findings suggest that TC1 is a novel endothelial inflammatory regulator that might be implicated in inflammatory vascular diseases.</P>
A Smart Bench Press Machine: Automatic Weight Control Sensitive to User Tiredness

In order to provide a safe free-weight-training environment to people without workout trainers, we suggest a smart bench press machine with an automatic weight control system sensitive to user tiredness. Physical weight plates on the machine are replaced with a hydraulic cylinder as a press load and the cylinder knob is coupled with a step motor to change its tensile force automatically in-between lifting exercises. Three subjects participated to verify the usability of the smart bench press machine. They were asked to lift a 6-RM press load 10 times with 3 different lifting conditions: 1) no assistance, 2) a human assistance, and 3) the automatic weight control. All subjects were not able to complete the 10 sets without assistance due to tiredness, but they finished the full sets under the two assistive conditions. Average lifting speeds under the automatic weight control condition showed the most consistent level. Normalized quasi-tension data based on surface electromyogram signals of both Pectoralis Majors revealed that the subjects maintained the target muscle activation level above 50% but not more than 80% throughout the 10 sets. Therefore, the smart bench press machine is expected to both keep pace with the lifting exercise and reduce risk of injuries due to excessive muscle tensions.

Background: Pharmacopuncture therapy and acupotomy are commonly used in combination for Conventional Korean Medicine Treatments (CKMT) for the treatment of patients with lumbar spinal stenosis (LSS). The aim of this study is to evaluate the effect and safety of combining pharmacopuncture therapy and acupotomy in the treatment of LSS. Methods: This study is designed as a pragmatic, assessor-blinded, randomized controlled trial with two parallel arms in a 1:1 ratio. A total of 104 participants diagnosed with LSS will be randomly assigned to an experimental group (pharmacopuncture therapy and acupotomy in addition to CKMT) or a control group (only CKMT). Patients in both groups will receive treatment two times weekly for 6 weeks. The primary outcome will be the mean change on the 100-mm visual analog scale (VAS) from the baseline to the end of the treatment (week 6). The secondary outcomes will include the mean change in the 100-mm VAS from baseline to week 10 and week 14, respectively. Proportion of patients who achieve the clinically important difference, Zurich Claudication Questionnaire, Roland-Morris disability questionnaire, self-reported maxium walking distance, EuroQol 5-dimension 5-level, and Patients’ Global Impression of Change will also be assessed. Adverse events will be assessed at each visit. The outcomes will be measured for a total of 14 weeks, including a treatment period of 6 weeks and follow-up of 4, 8 weeks. Discussion: The results of this trial will confirm the effect and safety of combining pharmacopuncture therapy and acupotomy in the treatment of patients with LSS.

<P>Next-generation sequencing (NGS) panels are widely used for defining tumor mutation profiles and determining treatment approaches. We performed targeted NGS with 382 colorectal cancer genes with known microsatellite instability (MSI). After exclusion of germline alterations, the load of somatic mutations and small insertion/deletion (indel) alterations were determined. In the test set, 79 patients with 41 microsatellite-stable (MSS) and 38 MSI tumors were included. There were 120 MSS and eight MSI-high tumors in the validation set. The number of somatic mutations of whole samples were distinguished into three groups: mutant functional polymerase epsilon catalytic subunit, MSI, and MSS tumors. The median numbers of somatic and indel mutations in MSI tumors were higher. The indel mutation to whole mutation ratio (I index) was higher in MSI tumors. Hypermutation and low I index of polymerase epsilon catalytic subunit mutant tumors, a somatic mutation load cut-off of ≥40, and an I index of ≥9% were selected as the criteria for detecting MSI tumors with high sensitivity and specificity. With the analysis of alteration patterns of homopolymer genes, a higher median number of homopolymer mutations in MSI tumors was observed. Mutated homopolymer ≥5 was selected as the criterion for detecting MSI tumors. MSI in colorectal cancer can be detected by targeted NGS panels with high sensitivity and specificity using somatic mutation load and I index.</P>

A Study on the Effect of Group Commitment on Improving Awareness of Recycling through Gamification

Raising awareness of recycling and the seriousness of environmental pollution is the fundamental resort of influencing individuals to engage in a sustainable lifestyle. By utilizing gamification of recycling with the use of an OptiTrack motion capture system, this study aims to conceptualize contributions to the main factors of improving awareness of recycling of individuals. In this paper, we present a comparison model based on the survey results before and after the influence of group commitment of the individuals by means of extending and contemplating findings from the existing literature. The experiment result reveals that the influence of group commitment contributes to the willingness to recycle by 28.9%, and decrement of 10.7% in prioritizing self-convenience when it comes to recycling, thus providing a basis to explore the impact of group commitment and other factors on environmental awareness and behavior.

The surface flatness of metal meshes in a deflector of particle beam mass spectrometer (PBMS) required ideally flat, and this can specify the particle trajectories which goes through the detector. In this research, charged particle current was measured using the different surface roughness deflectors. NaCl particles were generated monodispersed in its size by using differential mobility analyzer and the whole processes were followed the way calibrating PBMS. The results indicate that the mesh surface morphology in the deflector can affect to the particle size and the concentration errors, and sensitivity of PBMS.
