      • Chlorin e6 Prevents ADP-Induced Platelet Aggregation by Decreasing PI3K-Akt Phosphorylation and Promoting cAMP Production

        <P>A number of reagents that prevent thrombosis have been developed but were found to have serious side effects. Therefore, we sought to identify complementary and alternative medicinal materials that are safe and have long-term efficacy. In the present studies, we have assessed the ability of chlorine e6 (CE6) to inhibit ADP-induced aggregation of rat platelets and elucidated the underlying mechanism. CE6 inhibited platelet aggregation induced by 10 <I>µ</I>M ADP in a concentration-dependent manner and decreased intracellular calcium mobilization and granule secretion (i.e., ATP and serotonin release). Western blotting revealed that CE6 strongly inhibited the phosphorylations of PI3K, Akt, c-Jun N-terminal kinase (JNK), and different mitogen-activated protein kinases (MAPKs) including extracellular signal-regulated kinase 1/2 (ERK1/2) as well as p38-MAPK. Our study also demonstrated that CE6 significantly elevated intracellular cAMP levels and decreased thromboxane A<SUB>2</SUB> formation in a concentration-dependent manner. Furthermore, we determined that CE6 initiated the activation of PKA, an effector of cAMP. Taken together, our findings indicate that CE6 may inhibit ADP-induced platelet activation by elevating cAMP levels and suppressing PI3K/Akt activity. Finally, these results suggest that CE6 could be developed as therapeutic agent that helps prevent thrombosis and ischemia.</P>

        Mutations in AP22.65 Accelerate Flowering in Arabidopsis thaliana

        Identification of the gene(s) responsible for floweringtime in Arabidopsis has significant implications. We used theT-DNA insertion library of Arabidopsis thaliana to screen anearly-flowering mutant that exhibits accelerated floweringunder short-day conditions. AP22.65, a novel flowering-timegene in that species, was isolated and identified via genomewalkingand bioinformatics analysis. The flowering time ofAP22.65-complementing plants was similar to that of theCol-0 wild type (WT). Conversely, its overexpression delayedflowering. Consistent with this phenotype, expression ofAP22.65 was decreased in the ap22.65-1 mutant, recoveredin AP22.65-complementing plants, and increased in AP22.65-overexpressing plants. Compared with the WT, expressionlevels of critical genes in different flowering pathways, i.e.,SPY, FLC, GI, CO, FT, and LFY, were down-regulated inloss-of-function mutants. Expression of AP22.65 was distributedin flowers, siliques, rosette leaves, and whole seedlings. Therefore, this gene may be a negative regulator of Arabidopsisflowering.

        Two cases of methyl alcohol intoxication by sub-chronic inhalation and dermal exposure during aluminum CNC cutting in a small-sized subcontracted factory

        Background: Methyl alcohol poisoning has been mainly reported in community. Two cases of methyl alcohol poisoning occurred in a small-sized subcontracted factory which manufactured smartphone parts in Korea. Case presentation: One young female patient presented with dyspnea and visual disturbance. Another young male patient presented with visual disturbance and myalgia. They treated with sodium bicarbonate infusion and hemodialysis for metabolic acidosis. In addition, he received ethyl alcohol per oral treatment. Her and his urinary methyl alcohol concentration was detected as 7.632 mg/L, 46.8 mg/L, respectively, although they were treated hemodialysis. Results of the working environment measurement showed that the concentration of methyl alcohol (1030.1–2220.5 ppm) in the air exceeded the time weighted average (200 ppm). They were diagnosed with optic neuropathy due to methyl alcohol poisoning and still have visual impairment. Conclusions: Workers who hired as dispatched employees in a small-sized subcontracted factory were exposed to high concentrations of methyl alcohol. The workplace had poor ventilation system. In addition, workers did not wear proper personal protect equipment. Working environment measurement and annual chekups for workers were not performed. They were in a blind spot to occupational safety and health. More attention is needed to protect vulnerable workers’ health.

      • Effect of 5-aza-2'-deoxycytidine on Cell Proliferation of Non-small Cell Lung Cancer Cell Line A549 Cells and Expression of the TFPI-2 Gene

        Objective: The present study employed 5-aza-2'-deoxycytidine (5-Aza-CdR) to treat non-small cell lung cancer (NSCLC) cell line A549 to investigate the effects on proliferation and expression of the TFPI-2 gene. Methods: Proliferation was assessed by MTT assay after A549 cells were treated with 0, 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR, a specific demethylating agent, for 24, 48 and 72h. At the last time point cells were also analyzed by flow cytometry (FCM) to identify any change in their cell cycle profiles. Methylation-specific polymerase chain reaction (MSPCR), real time polymerase chain reaction(real-time PCR) and western blotting were carried out to determine TFPI-2 gene methylation status, mRNA expression and protein expression. Results: MTT assay showed that the growth of A549 cells which were treated with 5-Aza-CdR was significantly suppressed as compared with the control group (0 ${\mu}mol/L$ 5-Aza-CdR). After treatment with 0, 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR for 72h, FCM showed their proportion in G0/G1 was $69.7{\pm}0.99%$, $76.1{\pm}0.83%$, $83.8{\pm}0.35%$, $95.5{\pm}0.55%$ respectively (P<0.05), and the proportion in S was $29.8{\pm}0.43%$, $23.7{\pm}0.96%$, $15.7{\pm}0.75%$, $1.73{\pm}0.45%$, respectively (P<0.05), suggesting 5-Aza-CdR treatment induced G0/G1 phase arrest. MSPCR showed that hypermethylation in the promoter region of TFPI-2 gene was detected in control group (0 ${\mu}mol/L$ 5-Aza-CdR), and demethylation appeared after treatment with 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR for 72h. Real-time PCR showed that the expression levels of TFPI-2 gene mRNA were $1{\pm}0$, $1.49{\pm}0.14$, $1.86{\pm}0.09$ and $5.80{\pm}0.15$ (P<0.05) respectively. Western blotting analysis showed the relative expression levels of TFPI-2 protein were $0.12{\pm}0.01$, $0.23{\pm}0.02$, $0.31{\pm}0.02$, $0.62{\pm}0.03$ (P<0.05). TFPI-2 protein expression in A549 cells was gradually increased significantly with increase in the 5-Aza-CdR concentration. Conclusions: TFPI-2 gene promoter methylation results in the loss of TFPI-2 mRNA and protein expression in the non-small cell lung cancer cell line A549, and 5-Aza-CdR treatment could induce the demethylation of TFPI-2 gene promoter and restore TFPI-2 gene expression. These findings provide theoretic evidence for clinical treatment of advanced non-small cell lung cancer with the demethylation agent 5-Aza-CdR. TFPI-2 may be one molecular marker for effective treatment of advanced non-small cell lung cancer with 5-Aza-CdR.

      • <i>Pistacia chinensis</i> Methanolic Extract Attenuated MAPK and Akt Phosphorylations in ADP Stimulated Rat Platelets <i>In Vitro</i>

        <P><I>Pistacia chinensis</I> (Chinese pistache) is a widely grown plant in southern China where the galls extract is a common practice in folk medicine. However, extracts from this plant have never been attempted for their cardiovascular protective effects in experimental setting. Here therefore we aimed to investigate the antiplatelet activity of <I>Pistacia chinensis</I> methanolic extract (PCME) in ADP stimulated rat platelets <I>in vitro</I>. PCME (2.5–20 <I>μ</I>g/mL) inhibited ADP-induced platelet aggregation. While PCME diminished [Ca<SUP>2+</SUP>]<I>i</I>, ATP, and TXA2 release in ADP-activated platelets, it enhanced cAMP production in resting platelets. Likewise, PCME inhibited fibrinogen binding to <I><I>α</I></I>IIb<I><I>β</I></I>3 and downregulated JNK, ERK, and Akt phosphorylations. Thus, PCME contains potential antiplatelet compounds that could be deployed for their therapeutic values in cardiovascular pathology.</P>

        Detection of Powdery Mildew of Bitter Gourd Based on NIR/Fluorescence Spectra

        Purpose Powdery mildew as one of the common vegetable diseases has very rapid infection. Its outbreak will bring about disastrous consequences to vegetable output; thus, it is of key importance to do rapid identifi cation and prevention of powdery mildew. Methods In this test, 100 bitter gourd leaves were collected as research samples, and the data of near-infrared spectra, fl uorescence spectra, and chromatic values L*a*b* , and the classic K-S algorithm was adopted to divide the sample sets; then, the quantitative forecasting and qualitative discrimination models were established. First, Pearson’s correlation analysis was carried out to fi nd the feasibility of taking a * as the modeling parameter, through cross-validation; the quantitative forecasting model was optimized by the PLSR (partial least squares regression) method. The model is also optimized by extracting the spectral feature bands using the continuous projection SPA method. Results The optimization results showed that the MSC + SPA + PLSR quantitative forecasting model of near-infrared spectra could eff ectively improve model precision, which was signifi cantly higher than that of fl uorescence spectra. Classifi cation Leaner was used to establish the quantitative forecasting model. Compared with the model of near-infrared spectra, the SPA + SVM qualitative discrimination model of fl uorescence spectra could improve the identifi cation precision of powdery mildew of bitter gourd as high as 98% through training. Conclusion This study proposed diff erent combination methods based on quantitative forecasting and qualitative discrimination and could provide a method and reference to the identifi cation of powdery mildew of bitter gourd.

      • Therapeutic Effects and Adverse Drug Reactions are Affected by Icotinib Exposure and CYP2C19 and EGFR Genotypes in Chinese Non-Small Cell Lung Cancer Patients

        Background: The aim of this study was to evaluate how CYP2C19 affects icotinib and metabolite' exposure, and to determine whether the exposure and EGFR genotype influences survival time, tumor metastasis and adverse drug reactions. Materials and Methods: 274 NSCLC patients who accepted 125mg icotinib/t.i.d. were chosen from a phase III study. Blood samples were obtained in $672^{nd}$ ($4^{th}$ week) and $1,680^{th}$ hours ($10^{th}$ week), and plasma was used to quantify the concentration of icotinib and blood cells were sampled to check the genotypes. Clinical data were also collected at the same time, including EGFR genotypes. Plasma concentrations were assessed by HPLC-MS/MS and genotype by sequencing. All data were analyzed through SPSS 17.0 and SAS 9.2. Results: CYP 2C19 genotypes affected bio-transformation from icotinib to M24 and M26, especially in poor-metabolisers. Higher icotinib concentrations (>1000 ng/mL) not only increased patient PFS and OS but also reduced tumor metastasis. Patients with mutant EGFR experienced a higher median PFS and OS (234 and 627 days), especially those with the 19del genotype demonstrating higher PR ratio. Patients who suffered grade II skin toxicity had a higher icotinib exposure than those with grade I skin toxicity or no adverse effects. Liver toxic reactions might occur in patients with greater M20 and M23 plasma concentrations. Conclusions: CYP2C19 polymorphisms significantly affect icotinib, M24 and M26 exposure. Patients with mutant EGFR genotype and higher icotinib concentration might have increased PFS and OS and lower tumor metastasis. Liver ADR events and serious skin effects might be respectively induced by greater M20, M23 and icotinib concentrations.

        Optimization of image reconstruction method for dual-particle time-encode imager through adaptive response correction

        Time-encoded imagers (TEI) are important class of instruments to search for potential radioactive sources to prevent illicit transportation and trafficking of nuclear materials and other radioactive sources. The energy of the radiation cannot be known in advance due to the type and shielding of source is unknown in practice. However, the response function of the time-encoded imagers is related to the energy of neutrons or gamma-rays. An improved image reconstruction method based on MLEM was proposed to correct for the energy induced response difference. In this method, the count vector versus time was first smoothed. Then, the preset response function was adaptively corrected according to the measured counts. Finally, the smoothed count vector and corrected response were used in MLEM to reconstruct the source distribution. A one-dimensional dual-particle time-encode imager was developed and used to verify the improved method through imaging an AmeBe neutron source. The improvement of this method was demonstrated by the image reconstruction results. For gamma-ray and neutron images, the angular resolution improved by 17.2% and 7.0%; the contrast-to-noise ratio improved by 58.7% and 14.9%; the signal-to-noise ratio improved by 36.3% and 11.7%, respectively.
