        Impaired phagocytosis of apoptotic cells causes accumulation of bone marrow-derived macrophages in aged mice

        Accumulation of tissue macrophages is a significant cha-racteristic of disease-associated chronic inflammation, and facilitates the progression of disease pathology. However, the functional roles of these bone marrow-derived macrophages (BMDMs) in aging are unclear. Here, we identified age-dependent macrophage accumulation in the bone marrow, showing that aging significantly increases the number of M1 macrophages and impairs polarization of BMDMs. We found that age-related dysregulation of BMDMs is associated with abnormal overexpression of the anti-inflammatory interleukin-10. BMDM dysregulation in aging impairs the expression levels of pro-inflammatory cytokines and genes involved in B-cell maturation and activation. Phagocytosis of apoptotic Jurkat cells by BMDMs was reduced because of low expression of phagocytic receptor CD14, indicating that increased apoptotic cells may result from defective phagocytosis of apoptotic cells in the BM of aged mice. Therefore, CD14 may represent a promising target for preventing BMDM dysregulation, and macrophage accumulation may provide diagnostic and thera-peutic clues. [BMB Reports 2017; 50(1): 43-48]

      • Molecular Cloning and Expression of DMRT Gene in Protogynous Wrasse, Halichoeres tenuispinis

        The sex differentiation of fishes occurs under the control of genetic and various environmental factors. DM-domain containing genes are novel zinc finger transcription factors and play key roles in sex determination. In order to isolate the wrasse DMRT (wDMRT) cDNA from the protogynous wrasse (Halichoeres tenuispinnis), the wrasse testis cDNA library was screened using the P-labeled PCR products, which were amplified with the degenerate primers from conserved DM-domain regions of several DMRT genes. Among a few positives obtained through screening, the full length wDMRT cDNA of 2.9kb size encoding a predicted 300 amino acid residues was isolated. The sequence analysis exhibited 60%, 43% sequence identity with rainbow trout and tilapia DMRT1, respectively. RT-PCR assay showed that wDMRT was expressed specifically in male testis. Also, wDMRT gene was strongly expressed in May during reproductive season, when the reproductivity of wrasse is most active. This results suggested that wDMRT gene function in testis differentiation The conserved DM-domain regions were amplified using PCR from DMRT genes of several species among Labridae, and their sequences were determined. The sequence of DM-domain region of Halichoeres. tenuispinis was identical to those of Pseudolabrus japonicus, Pteragogus flagellifera, and showed 94% identity with that of Halichoeres poecioptrerus.

        Synergistic effect of washing and cooking on the removal of multi-classes of pesticides from various food samples

        <P><B>Abstract</B></P><P>The principal objective of this study was to investigate the effect of household processing, including washing and cooking on pesticide residue levels in various food samples. For this study, 31 food materials were selected and 44 pesticide residues were monitored using the “quick, easy, cheap, effective, rugged, and safe” QuEChERS extraction-based and liquid chromatography–tandem mass spectrometry (LC–MS/MS) methods. Eight pesticides, including acetamiprid, azoxystrobin, fenobucarb, fosthiazate, iprobenfos, lufenuron, propiconazole, and trifloxystrobin were detected in nine food samples including colored rice, glutinous rice (white rice), glutinous rice (unpolished rice), green chili, ginger, butterbur, chinamul, spinach, and perilla leaf. Results indicated that residue levels in positive food commodities declined substantially following washing and cooking. However, the residual level of acetamiprid increased in green chilis after boiling and stir-frying. In sum, household processing (washing and cooking) tended to substantially reduce or eliminate pesticide residues in a synergistic manner. These applications are necessary to protect consumers from the negative health effects of pesticide residues detected in food commodities.</P> <P><B>Highlights</B></P><P>► The effects of household processing on pesticide residue levels were investigated. ► Residue levels were substantially declined following washing and cooking. ► The household processing tend to remove the residues in a synergistic manner.</P>

        Affinity Maturation of an Anti-Hepatitis B Virus PreS1 Humanized Antibody by Phage Display

        In a previous study we generated an anti-Hepatitis B Virus (HBV) preS1 humanized antibody (HzKR127) that showed in vivo HBV-neutralizing activity in chimpanzees. However, the antigen-binding affinity of the humanized antibody may not be sufficient for clinical use and thus affinity maturation is required for better therapeutic efficacy. In this study, phage display technique was employed to increase the affinity of HzKR127. All six amino acid residues (Glu95-Tyr96-Asp97-Glu98-Ala99-Tyr100) in the heavy (H) chain complementary-determining region 3 (HCDR3) of HzKR127 were randomized and phage-displayed single chain Fv (scFv) library was constructed. After three rounds of panning, 12 different clones exhibiting higher antigen-binding activity than the wild type ScFv were selected and their antigen-binding specificity for the preS1 confirmed. Subsequently, five ScFv clones were converted to whole IgG and subjected to affinity determination. The results showed that two clones (B3 and A19) exhibited an approximately 6 fold higher affinities than that of HzKR127. The affinity-matured humanized antibodies may be useful in anti-HBV immunotherapy.

        The Proximal Optimization Technique Improves Clinical Outcomes When Treated without Kissing Ballooning in Patients with a Bifurcation Lesion

        Background and ObjectivesThere are limited data regarding the clinical efficacy of the proximal optimization technique (POT) in the treatment of coronary bifurcation lesions. We investigated the influence of POT on the clinical outcomes of patients with coronary bifurcation lesions. MethodsWe enrolled a total of 1,191 patients with a bifurcation lesion with a side branch (SB) diameter ≥2.5 mm treated with a drug-eluting stent from 18 centers between January 2003 and December 2009. The primary outcome was major adverse cardiac events (MACEs: cardiac death, myocardial infarction or target lesion revascularization [TLR]). We performed one-to-many (1:N) propensity score matching with non-fixed matching ratio. ResultsPOT was performed in 252 patients. During follow-up (median 37 months), the incidence of MACE was lower in the POT group than it was in the non-POT group (adjusted hazard ratio, 0.43; 95% confidence interval [CI], 0.24–0.79; p=0.006). After propensity score matching, these were 0.34; 95% CI, 0.17–0.69; p=0.003 for MACE and 0.37; 95% CI, 0.17–0.78; p=0.01 for TLR. The use of POT was associated with significantly lower TLR in patients treated without kissing ballooning, but was not in those who underwent kissing ballooning (p for interaction=0.03). ConclusionsIn coronary bifurcation lesions with a large SB, POT may be beneficial to improve long-term clinical outcome, particularly in patients treated without kissing ballooning during the procedure.       • <i>Microbulbifer gwangyangensis</i> sp. nov. and <i>Microbulbifer pacificus</i> sp. nov., isolated from marine environments

        <P>Two novel Gram-stain-negative, chemoheterotrophic and strictly aerobic bacteria, strains GY2<SUP>T</SUP> and SPO729<SUP>T</SUP>, were isolated from a tidal flat at Gwangyang Bay in Korea and a marine sponge sample from the Pacific Ocean, respectively. The two strains were halotolerant, catalase- and oxidase-positive, and non-motile rods. Optimum temperature and pH for growth of both strains were observed to be 35 °C and pH 7.0–7.5, but optimum salinity for strain SPO729<SUP>T</SUP> [2–3 % (w/v)] was slightly higher than that for strain GY2<SUP>T</SUP> (1–2 %). The major cellular fatty acids of both strains were C<SUB>16 : 0</SUB>, iso-C<SUB>15 : 0</SUB>, iso-C<SUB>17 : 0</SUB>, iso-C<SUB>17 : 1</SUB>ω9<I>c</I>, C<SUB>18 : 1</SUB>ω7<I>c</I>, iso-C<SUB>11 : 0</SUB> and iso-C<SUB>11 : 0</SUB> 3-OH. The genomic DNA G+C contents of strains GY2<SUP>T</SUP> and SPO729<SUP>T</SUP> were 55.1 and 57.9 mol%, respectively, and ubiquinone 8 (Q-8) was detected as the sole respiratory quinone from the two strains. Phylogenetic analysis based on 16S rRNA gene sequences showed that strains GY2<SUP>T</SUP> and SPO729<SUP>T</SUP> formed tight phyletic lineages with members of the genus <I>Microbulbifer</I>. Strain GY2<SUP>T</SUP> was closely related to <I>Microbulbifer okinawensis</I> ABABA23<SUP>T</SUP> (98.2 %), strain SPO729<SUP>T</SUP> (98.0 %) and <I>Microbulbifer donghaiensis</I> CN85<SUP>T</SUP> (97.0 %); strain SPO729<SUP>T</SUP> was closely related to <I>M. okinawensis</I> ABABA23<SUP>T</SUP> (98.3 %) and <I>M. donghaiensis</I> CN85<SUP>T</SUP> (98.2 %). The DNA–DNA relatedness values of strain GY2<SUP>T</SUP> with <I>M. okinawensis</I> ABABA23<SUP>T</SUP>, strain SPO729<SUP>T</SUP> and <I>M. donghaiensis</I> CN85<SUP>T</SUP> were 40.0±2.1 %, 13.1±3.9 % and 16.2±5.8 %, respectively, whereas those of strain SPO729<SUP>T</SUP> with <I>M. okinawensis</I> ABABA23<SUP>T</SUP> and <I>M. donghaiensis</I> CN85<SUP>T</SUP> were 48.0±4.0 % and 34.6±9.3 %, respectively. On the basis of phenotypic and molecular features, it is concluded that the two strains GY2<SUP>T</SUP> and SPO729<SUP>T</SUP> represent two novel species of the genus <I>Microbulbifer</I>, for which the names <I>Microbulbifer gwangyangensis</I> sp. nov. and <I>Microbulbifer pacificus</I> are proposed; the type strains are GY2<SUP>T</SUP> ( = KACC 16189<SUP>T</SUP> = JCM 17800<SUP>T</SUP>) and SPO729<SUP>T</SUP> ( = KCCM 42667<SUP>T</SUP> = JCM 14507<SUP>T</SUP>), respectively.</P>

        메타버스(가상 · 증강 · 혼합 · 확장현실)를 이용한 운동재활의 활용 방안

        Objective: The aim of this study was to analyze domestic and foreign studies using virtual reality or metaverse for exercise rehabilitation in order to help the disabled or elderly patients with exercise rehabilitation, and suggest a method for using metaverse for exercise rehabilitation. Method: In this study, after analyzing and discussing various information related to the metaverse and exercise rehabilitation through electronic search of recently published papers, academic journals, books, and internet websites, the exercise rehabilitation plan using the metaverse was proposed. Results: In the case of domestic research, the diversity of virtual reality application technology for the rehabilitation of disabled and elderly patients was not secured, but recently, virtual reality or metaverserelated technologies were developed and specialized in a form suitable for exercise rehabilitation. In the case of overseas studies, it was analyzed that exercise rehabilitation using virtual reality and metaverse games for the rehabilitation of disabled and elderly patients can help improve brain, physical ability, and anti-aging by activating the body and mind. Conclusion: Smart metaverse health care is actively introduced to exercise rehabilitation, metaverse telemedicine business is applied to exercise rehabilitation programs, and digital twin games and exercise rehabilitation programs developed by metaverse related companies take into account the characteristics of disabled and elderly patients. If customized smart metaverse healthcare is used for exercise rehabilitation, it is analyzed that it can lead this field.

      • Development of Cultural Enterprise in the Marine City Busan by Hosting Mega Sports Events

        The purpose of this was to analyze how the international events hosted so far affected the development of local society through economic, cultural, and social ripple effects and how infrastructure of facilities and human capitals as the result of hosting sports events affected sports culture. Also developmental strategies to ensure local identity and host mega sports events for local economy and cultural development was discussed in the light of growing cultural enterprise in the marine city Busan. Busan city decided to invite the Olympic Games in 2020 as a part of Visions and Strategies for Busan Development 2020 and planned to submit the application for candidate cities to the government and KOC until November. If the Olympics is invited, the direct ripple effect on the national economy is estimated 20 trillion 290 billion won and the number of jobs created will be 115 thousand. According to the result from the feasibility study of hosting the Olympic Games in 2020, the direct ripple effects was estimated 12 trillion 743.8 billion won of production, 115 thousand of jobs, 2 trillion 329.2 billion won of incomes, and 5 trillion 232.2 billion won of added value. Hosting mega sports events is the communication media with strong ripple effects like economic effects, industrial development, regional development, international exchange and the activation of culture and art. Also it can create own local color through the forming local identity. The local cultural assets created can give history and pride to its people and also have the function to transfer local value industrially.
