      • Mitochondrial dysfunction and calcium deregulation by the RanBP9-cofilin pathway

        <P>Mitochondrial dysfunction and synaptic damage are important features of Alzheimer's disease (AD) associated with amyloid β (Aβ) and tau. We reported previously that the scaffolding protein RanBP9, which is overall increased in brains of patients with AD and in mutant APP transgenic mice, simultaneously promotes Aβ generation and focal adhesion disruption by accelerating the endocytosis of APP and β1-integrin, respectively. Moreover, RanBP9 induces neurodegeneration <I>in vitro</I> and <I>in vivo</I> and mediates Aβ-induced neurotoxicity. Here we show in primary hippocampal neurons that RanBP9 potentiates Aβ-induced reactive oxygen species (ROS) overproduction, apoptosis, and calcium deregulation. Analyses of calcium-handling measures demonstrate that RanBP9 selectively delays the clearance of cytosolic Ca<SUP>2+</SUP> mediated by the mitochondrial calcium uniporter through a process involving the translocation of cofilin into mitochondria and oxidative mechanisms. Further, RanBP9 retards the anterograde axonal transport of mitochondria in primary neurons and decreases synaptic mitochondrial activity in brain. These data indicate that RanBP9, cofilin, and Aβ mimic and potentiate each other to produce mitochondrial dysfunction, ROS overproduction, and calcium deregulation, which leads to neurodegenerative changes reminiscent of those seen in AD.—Roh.         한국인 직무 스트레스 측정도구의 개발 및 표준화

        Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

        폐기 목질 재료로 제조한 학생용 책상 천판의 성능에 미치는 성형조건의 영향

        폐기 목질재료를 사용하여 학생용 책상 천판의 성형시 각 제조조건에 따른 천판의 성능을 검토하였다. 책상 천판의 파티클 구성은 고운 파티클과 거친 파티클을 적절히 혼합한 것이 고운 파티클이나 거친 파티클만으로 제조한 것보다는 전체적으로 성능이 양호하였으며, 특히 표층에는 고운 파티클을 중층에는 거친 파티클을 구성하는 것이 표면 마감성이 가장 우수하였다. 그리고 밀도가 높을수록 박리강도, 휨 강도는 우수하나, 흡수두께 팽창율은 증가하였으며, 수지 도포량이 많을수록 박리강도, 흡수두께 팽창율은 양호하나, 포름알데히드의 방출량은 높았다. 가압온도는 높을수록 대부분의 성능이 향상되었으며, 190℃가 가장 적절하였다. 가압시간은 1단계 2분과 2단계 2분이 비교적 양호하나 2단계의 가압시간을 1분으로 단축하여도 성능에는 큰 문제가 없어 앞으로 생산성의 향상을 기대할 수 있을 것으로 생각된다. This research was performed to investigate the effect of moulding conditions on properties of the table top for student desk manufactured by wood waste materials. The table top manufactured using the mat mixed properly with fine particles and coarse particles generally showed better physical properties than those manufactured using either fine particles or coarse particles did. Especially, highest performance for overlaying was observed when fine particles were used for face layer and coarse particles were used for core layer. The internal bond (IB) strength, the bending strength and thickness swelling increased with the increase of density. Most of physical properties were improved with the increase of hot pressing temperature. The best physical properties were found at 190℃ when the first hot pressing and the second hot pressing times were two minutes. However, the effect of the second hot pressing time of two minutes on the physical properties was not markedly different from that of one minute. This result suggests that the second hot pressing time of one minute is enough to increase the productivity in manufacturing the table top with the least decrease of the physical properties.

      • 側腦室內 d-Tubocurarine이 家兎 腎臟機能에 미치는 影響

        Effect of d-tubocurarine administered intracerebroventricularly on the renal function of rabb-its was investigated. 1) Intraventricular d-tubocurarine induced decrease of urine flow, renal plasma flow, glomerular filtration, and urinary sodium and potassium excretion, but increase of reabsorption of urinary sodium and potassium. 2) Correlation coefficient between changes of two parameters among urine volume, renal plasma flow, glomerular filtration and urinary sodium excretion was statistically significant. 3) Changes of various parameters of renal function induced by intraventricular d-tubocurarine were inhibited by phenol treatment on the renal artery except change of sodium. From the above results, it is suggested that the effect of intraventricular d-tubocurarine on the renal function of rabbit is due to renal hemodynamic changes by central sympathetic activation.

        Background and Objectives : Overexpression of cyclooxygenase-2 (COX-2), a rate-limiting enyme in the generation of prostanoids from arachidonic acid, has known to be closely related to tumorigenesis, tumor growth, angiogenesis, and metastasis. Selective or non-selective COX-2 inhibitors have been used for the growth inhibition of cancers with preventative intents ; however, it has been suggested recently that cancer cells have COX-2-independent mechanisms. Materials and Method : Using MTT assat and cell counts, we observed the growth inhibition of SCC VII, CT-26 and B16F10 murine cancer cell lines when treated by celecoxib and indomethacin. SNU-1041 and HOK 16B were used as controls for comparing with the murine cell lines. The COX-2 expression of these cell lines was analyzed by western blotting and compared with the degree of inhibition by the drugs. Results : The growth inhibition of the cell lines by the drugs was clearly demonstrated in a concentration-dependent manner and depended on the type of cell lines and test drug. The in vitro viability assay revealed that CT-26 expressing COX-2 protein was slightly inhibited but SCC Vn and B16F10 without COX-2 expression were moderate-to-highly inhibited by the drug treatment. Celecoxib and indomethacin appeared to have no close relation with the COX-2 expression of cell lines in their growth inhibition, HOK 16B showed a resistance by concentrations less than 25 μM of celecoxib, which implies that celecoxib has a more selective effect on tumor cells and is safer than indomethacin Conclusion : The growth of cancer cells was inhibited by celecoxib and indomethacin treatment, which depends on the type of cancer, treated drug, and its concentration.         최근 모자이크의 시대 이후 인터넷의 폭발적인 발달로 인해 Client-Sever의 방식의 네트웍 서비스 방식뿐만 아니라 P2P(Peer to Peer) 등 분산 서비스 환경이 각광을 받고 있다. 사용자의 직접적인 자료의 공유를 위해서는 Peer to Peer 환경을 설정하여 분산되어진 자료의 효과적인 관리를 위한 통신의 주체가 되는 각각의 에이젼트들 간의 통신과 Sever와 Agent의 통신에 있어 악의적인 침입에 의한 정보누출이 문제가 되고 있다. 본 논문에서는 이러한 문제의 해결방안으로 환경에 적합한 PKI(공개키 기반구조)를 기반을 이용하여 보안 메커니즘을 설계하였다. By greatly development of internet since age of mosaic recently, P2P (Peer to Peer) back breakup service environment as well as the network service way of Client-Sever's way is getting into the spotlight. Information leakage by invasion that is enemy of evil in communication of communications division Sever and Agent between each agents that become burden of communication for effective administration of become discrete data establishing Peer to Peer environment for share of direct data of user is becoming an issue. In environment by solution way of these problem in this treatise suitable PKI (PKI) using base security mechanism design.

        엔진의 토크 조절 능력과 니켈-티타늄 파일의 삭제 성향이 근관 성형에 미치는 영향

        이 연구의 목적은 니켈-니타늄 파일의 삭제 성향과 엔진의 토크 조절 능력이 근관 성형에 미치는 영향을 평가하는 것이다. 레진 모형 40개를 사용하였으며, Hero Shaper^(®)와 Endo-Mate2^(®)을 이용한 HE군, Hero Shaper^(®)와 Tecnika^(®)를 이용한 HT군, Profile^(®)과 Endo-Mate2^(®)를 이용한 PE군, Profile^(®)과 Tecnika^(®)를 이용한 PT군으로 구분하여 근관성형 하였다. 각 군별 근관 성형 시간을 기록하였다. 근단공으로부터의 수직 이동 거리 1, 3 그리고 5 ㎜ 위치에서의 근관 삭제량, 중심이 동률을 산출하여 다음의 결론을 얻었다. 1. 근관 성형 시간은 HT군이 가장 빨랐다 (p < 0.05). 2. 삭제량은 1 ㎜ 지점에서 HE군이 PT군보다 컸고 (p < 0.05), 3 ㎜ 지점에서는 PT 군이 다른 군들보다 작았다 (p < 0.05). 5 ㎜에서는 PE군이 PT군보다 컸다 (p < 0.05). 3. 중심 이동률은 1㎜ 지점에서 HE군이 다른 군들보다 컸으며 (p < 0.05), 5 ㎜ 에서는 HT군이 PE군과 PT군보다 컸다 (p < 0.05). 따라서 토크 조절이 되는 엔진을 사용하는 것이 근관의 변위를 적게 유발하고 과도한 삭제량을 예방 할 수 있으며 근관 성형 시간도 단축 할 수 있는 것으로 보이며 따라서 양호한 근관 성형 결과에 영향을 준다고 볼 수 있다. The purpose of this study was to compare the shaping ability of two different Ni-Ti file systems and the two different engine systems in simulated canals. A total of four groups of each 10 were tested. Each group was instrumented with HeroShaper^(®) and Endo-Mate2^(®) (Group HE), HeroShaper^(®) and Tecnika^(®) (Group HT), ProFile^(®) and Endo-Mate2^(®) (Group PE), and ProFile^(®) and Tecnika^(®) (Group PT). Canal preparation time was recorded. The images of pre- and post- instrumented root canals were scanned and superimposed. The amounts of increased width and centering ratio were measured and calculated at apical 1, 3 and 5 ㎜ levels. These data were statistically analyzed with one-way ANOVA and Duncan's multiple range test The results of this study were as follows; 1. Canal preparation time of HT group was the shortest (p < 0.05). 2. The amount of increased canal width in HE group was significantly larger than PT group at apical 1 ㎜ level (p < 0.05). At apical 3 ㎜ level, PT group was significantly smaller than other groups (p < 0.05). At apical 5 ㎜ level, PE group was significantly larger than PT group (p < 0.05). 3. The amount of centering ratio in HE group was significantly larger than other groups (p < 0.05). At apical 5 ㎜ level, HT group was significantly larger than PE group and PT group (p < 0.05). Under the condition of this study, torque-controlled endodontic motor is safer than no torque controlled motor, especially when the active file is used.

      • 우수선수와 비우수선수의 투포환 동작에 대한 운동학적 비교분석

        This study was conducted to identify the effective teaching methods of short put. The subjects who participated in this study comprised the high performance players(N=3) and the low performance players(N=4). A three-dimensional cinematographic method using DLT was used to analyze motions in shot put. The results of this study included the following important things: 1.Making glide long within a very limited time was contributing to the improvement of the record. 2.The large gait in shot put was considered as an unappropriated factor because it makes the reduction of using a flat force. 3.Releasing high on the condition that the high of central gravity of the body is stable is one of the ways to make good distance. 4.The releasing angles of the high performance players were smaller than those of the low performance players. 5.The knee angles of the high performance players were 95.7±2.517˚ while the glided foot landed in thee center of the circle. And, the knee angles of these players were 135±3.000˚ while the other foot landed completely. 6.The quickly releasing velocity of the shot put was found to be a better chance to make a good distance.

      • 당귀와 천궁 및 시호의 병용 경구 투여에서 흰쥐 혈중 Decursinol 농도의 비교 검토

        This study was performed to elucidate the influence on the plasma decursinol concentration after administration of combined decursin and Cnidii Rhizoma or decurisin and Bupleuri Radix extracts in comparison with single decursin administration. The identification of decursinol in the plasma after oral administration was determined by HPLC. The value of area under the plasma concentration (AUC) and the maximal concentration (Cmax) of decursinol after administration of methanol and ether Angelicae Gigantis Radix extract was higher than those of single decursin-treated group. It was also found that the AUC and the Cmax of decursinol after combined administration with decursin and Cnidii Rhizoma were higher than those of decursin administration, whereas those of combined with decursin and Bupleuri Radix was lower. Moreover, the studies were performed with decursinol only or with combined decursinol and Cnidii Rhizoma/Bupleuri Radix, both of combined treatment increased the plasma decursinol concentrations compared with decursinol-treated group in rats. On the basis of these results, it is concluded that co-existing substances or co-administration with other drugs may influence the plasma levels of decurisinol after oral administration.
