        Floristic characteristics of vascular plants and first distributional report of Pseudostellaria baekdusanensis M. Kim in Yongneup wetland protected area

        Yongneup wetland protected area, the only high moor in Korea, is a core area to conserve biodiversity. Even though the Yongneup wetland protected area is relatively small, various plant species are distributed in the Yongneup wetland protected area because it includes various habitats showing different environmental gradients. Vascular plants distributed in the Yongneup wetland protected area were identified as a total of 376 taxa that is composed of 73 families, 217 genera, 322 species, 3 subspecies, 44 varieties and 7 forms. For endangered plants designated by the Ministry of Environment, 5 species including Trientalis europaea var. arctica, Lilium dauricum, Halenia corniculata, Lychnis wilfordii and Menyanthes trifoliata were found and 34 taxa were confirmed to be distributed only in the mountainous wetland habitats. Regarding naturalized plants, a total of 11 taxa were distributed, but most of them were distributed in the areas where artificial interference has occurred. And in areas inside the wetlands that are relatively well preserved, 2 species of Bidens frondosa and Erigeron annuus were observed. In this study, the occurrence and distribution of Pseudostellaria baekdusanensis M. Kim, which was recently found in Mt. Baekdu and reported as a new species, were identified in the Yongneup wetland protected area. A wetland is a very vulnerable area to drastic environmental changes and damages to its ecosystem could cause the extinction of rare plant species which are distributed only in the wetlands. Therefore, it is mandatory that current status of the Yongneup wetland protected area is evaluated and actions to prevent rapid environmental changes are taken. Fourteen separate investigations were conducted in 2013 and another four in 2014, to evaluate current status of the Yongneup wetland protected area. These investigations have provided us the basic information for future actions of conservation and restoration.

        Today, it has been taking a serious view to make comfortable circumstances that are free from pollution, traffic problems, energy shortage, etc. Therefore, when we plan to improve the road condition in city, we should give priority to building familiar community road. Taking account of this, in the study I have studied the case and policies of foreign countries to community road. The results of this study can summarized as follows. 1 It should be accepted to equip large humps or round shaped humps (length:3.7m, height:l0.cm) that ate generally used in Austria. There is no doubt that traffic signs should be equipped before these humps. 2 Each zone of daily life should be setted up by range of about 1km2 and it should be given various traffic regulations or safety facilities for pedestrians, bicycles (especially, children or the old.l) 3 As living environment is getting worse by inflowing of passing cars, it is important to protect pedestrians, bicycles from cars. Generally, the policy to prohibit large cass(like trucks)into living aces has been taken. In addition to this, from now all the cars(like trucks) into living area has should be excluded from the area, except special cases. 4 The policy that pedestrians should be prior to cars must be enforced in Taegu city, and community road for both pedestrians and cars should be designed. 5 When plan to organize the network of pedestrian lane or bicycle road, we should collect public opinion and policies of executive organizations. Especially, in the case of giving great influence on community transportation as redevelopment of around railroad station, the schemes should be included how to connect pedestrian with car or public transit. Doing all this, the planner should consider the synthetic transportation planning for the small district in urban area.

        Inhibitory Effect of Scutellaria baicalensis Root Extract on Chemical Mediator Release and Immune Response

        Inhibitory effect of Scutellaria baicalensis ethanol extracts (SR) on chemical mediator release and immunoglobulin (Ig) production from Sprague-Dawley rats originated cells as type I allergic reaction was examined. SR showed concentration-dependent inhibition on basal and concanavalin A (ConA)-stimulated Ig production. In the mesenteric lymph node lymphocytes, the inhibitory effect of SR on the IgE production in the presence of Con A was stronger than these on IgA and IgG production. Moreover, tumor necrosis factor-alpha $(TNF-{\alpha})$ production-inhibiting effect of SR in the presence ConA was observed. However, SR did not affect the production of $interferon-{\gamma}$. SR also inhibited histamine release from the peritoneal exudate cells stimulated with a calcium ionophore A23187. In the case of leukotriene B4, SR markedly inhibited it at the concentration of 100 mg/ml. From these results, ethanol extracts obtained from Scutellaria baicalensis may have an anti-allergic effect on the intestinal system of rats.

      • Process Innovation Improves Trial Operation Efficiency

        <P>Background: Despite the fact that unaddressed delays in clinical trial operation could severely compromise the overall effort invested, there seems to be a lack of concerted effort in reforming such delays. This study evaluated the composite effect of initiatives in reforming trial operation efficiency. Methods: A high-volume academic medical center in Korea has implemented various initiatives to improve the trial operation efficiency by expediting times from institutional review board (IRB) submission to approval, from IRB submission to trial open for subject enrollment, and from trial open to first patient-in. The initiatives include implementation of the protocol preliminary review, parallel processing of the clinical trial agreement review in line with the protocol submission to the IRB, and involvement of project manager for operational risk management. Times from IRB submission to approval, from IRB submission to trial open, and from trial open to first patient-in before and after implementation of initiatives were compared. Results: The median time required in IRB approval was meaningfully shorter in the postinitiative group (19 vs 14 days; P < .001). The median times from IRB submission to trial open for subject enrollment and from trial open to first patient-in were reduced significantly in the postinitiative group (trial open: 25 vs 18 days, P < .001; first patient-in: 111.5 vs 100 days, P = .014). Conclusions: The initiatives were effective in reforming trial operational efficiency. Additional studies to address the cause of operational delay and modifiable factors influencing subject enrollment are needed to further improve operational efficiency.</P>

      • Inverse Relationship between Adipocyte Differentiation and Ceramide Level in 3T3-L1 Cells

        <P>Adipocyte differentiation has been a target in anti-obesity strategies and is known to be closely related to lipid metabolism. Ceramide, a major sphingolipid metabolite, has been implicated in differentiation. In this study, we investigated whether ceramide biosynthesis is related to adipogenesis in 3T3-L1 cells. Preadipocytes can be differentiated synchronously by a mixture of adipogenic inducers including 3-isobutyl-1-methylxanthine, dexamethasone and insulin. The number of lipid droplets and the triglyceride content, which are differentiation biomarkers, gradually increased during adipogenesis. Interestingly, ceramide and sphingosine contents in the differentiated cells were decreased compared to those in preadipocytes. When the preadipocytes were treated with an 3-isobutyl-1-methylxanthine- or dexamethasone- or insulin-deficient mixture of inducers, the cellular ceramide levels were significantly increased compared with those in cells treated with the complete set of inducers. When preadipocytes were treated with 0, 0.1 or 1 μg/ml insulin along with 3-isobutyl-1-methylxanthine and dexamethasone, the ceramide levels were decreased and the triglyceride content was increased in a concentration-dependent manner. When the cells were treated with epigallocatechin gallate, an adipocyte differentiation inhibitor, during adipogenesis, the ceramide levels of adipocytes were increased and the fat content was decreased. In conclusion, our findings demonstrate that cellular ceramide levels are inversely correlated with adipocyte differentiation.</P>

        Developmental Competence of Intrafollicular Oocytes Derived from Preantral Follicle Culture with Different Protocols after Parthenogenetic Activation

        This study was conducted to improve efficiency of a follicle culture system without reducing developmental competence of intrafollicular oocytes. Preantral follicles (100 to $125{\mu}m$ in diameter) of F1 hybrid (B6CBAF1) mice were cultured singly for 216 h in modified ${\alpha}$-MEM-glutamax medium, to which 2.5 IU/ml hCG and epidermal growth factor was added 16 h prior to the end of culture. Medium change was either performed three times (54 h interval), twice (72 h interval), once (108 h interval), or not at all (216 h interval). Maturation (progression to the metaphase II stage) of intrafollicular oocytes was detected from 4 days after culture in the three-times change treatment, while all treatments yielded mature oocytes from day 5 of culture. Compared with the three-times change, decreasing the change frequency to once did not reduce the capacity to begin maturation (germinal vesicle breakdown of 82 to 86%), to mature (78 to 79%) and to develop into blastocysts after parthenogenetic activation (29 to 32%). Morphological parameters were similar among these treatments. Except for the no medium change treatment, similar colony-forming activity of inner cell mass cells after culturing of blastocysts in leukemia inhibitory factor-containing medium was detected, while the morphology of the colony-forming cells deteriorated in the change-once treatment compared with the change twice or three-times. In conclusion, the efficiency of the preantral follicle culture system could be improved by reducing frequency of medium change up to a 72 h interval (three times in total 216 h culture) without decreasing developmental competence of oocytes.

        Exercise With a Novel Digital Device Increased Serum Anti-influenza Antibody Titers After Influenza Vaccination

        It has been reported that some exercise could enhance the anti-viral antibody titers after vaccination including influenza and coronavirus disease 2019 vaccines. We developed SAT-008, a novel digital device, consists of physical activities and activities related to the autonomic nervous system. We assessed the feasibility of SAT-008 to boost host immunity after an influenza vaccination by a randomized, open-label, and controlled study on adults administered influenza vaccines in the previous year. Among 32 participants, the SAT-008 showed a significant increase in the anti-influenza antibody titers assessed by hemagglutination-inhibition test against antigen subtype B Yamagata lineage after 4 wk of vaccination and subtype B Victoria lineage after 12 wk (p<0.05). There was no difference in the antibody titers against subtype “A.” The SAT-008 also showed significant increase in the plasma cytokine levels of IL-10, IL-1β, and IL-6 at weeks 4 and 12 after the vaccination (p<0.05). A new approach using the digital device may boost host immunity against virus via vaccine adjuvant-like effects.
