        본 연구는 영양표시에서 1회분량 및 영양소함량표시의 기준으로 사용되는 "식픔창고량"에 관한 연구를 수행하므로써 합리적이고 일관성있는 영양표시기준 개선을 위한 기초자료로 활용코자 하였다. 이를 위하여 미국 및 캐나다를 중심으로 제외국의 현황 및 식품참고량 설정방법을 조사 ·연구 하으며 제도도입 및 개선 방안을 강구하였파. 또한, 시판 제품의 영양료시현황 및 음료류 둥의 포장단위 등을 피악하여 식품참고량설정을 위한 기초자료로 활용코자 하였다. 앞으로 국민건강 ·영양조사자료의 체계적인 통계분석, 가공식품의 섭취량조사 등을 통하여 우리나라 국민의 1인 1회 섭취량자료의 축적이 선행되어야 할 것이다. The purpose of this ?roject is to study on the reference amounts customarily consumed per eating occasion to improve the nutrition labelling standards. We Performed to study on the methodology to establish the reference amounts on the basis of the review on the advanced counITy's status. And we investigated the packaged unit ofprocessed foods. Further study is needed to establlsh the customarily consumed amount per eating occasionthrough the statistical analysis of nationwide food consumption data. Our results will be helpful information to improve nutrition labelling system in Korea.

      • BATF와 ATF2의 이량체 형성에 있어서의 transactivation potential 분석

        BATF는 Jnu과 이량체를 형성하는 염기성 루신 지퍼(bZIP) 전사인자이다. 이들이 이량체를 이루어 AP-1 DNA 결합 복합체를 이루는 경우 고전적 AP-1 이량체(c-Fos:c-Jun)를 형성하였을 경우 보다 비교적 전사 잠재력이 감소된다. AP-1은 발생과 세포의 stress 반응의 과정에 널리 관련되어 있다. ATF2도 세포의 전사인자이며 CREB/ATF class에 속하는 루신지퍼 단백질이다. 이것은 암의 변형과 같은 무수한 세포의 표현형의 생성에 관계한다. pET 표현 vector는 6xHis 융합단백질을 생성하여 affinity column을 사용하여 융합단백질의 분리를 용이하게 한다. Nickel chelating 순수분리법은 대장균 BL 21 균주로부터 표현된 ATF2를 분리하는데 사용되어 졌다. 이 연구는 EMSA(Electrophoretic Mobility Shift Assay)에 의해서 BATF와 ATF2 사이의 vitro 상호작용을 나타낸다. C3H1OT1/2세포 transfection으로서 이량체 형성을 위한 전사인자들의 표현은 immunoblot로서 확인되었다. 전사 잠재력은 AP-1를 위하여는 TRE를 가진 reporter 유전자에 의해서, 그리고 ATF2를 위하여는 CRE를 가진 reporter 유전자에 의해 조사되었다. BATF is a basic leucine-zipper(bZIP) transcription factor that dimerizes with Jun to form AP-l DNA binding complexes with reduced transactivadon potential relative to classical AP-l dimers(c-Fos: c-Jun). AP-l has been linked to a wide variety of developmental and cellular stress-response processes. ATF-2 is a cellular transcription factor which belongs to the CREB/ATF class and is leucine zipper protein, It also can be participate in the generation of numerous cellular phenotypes such as oncogenic transformation. The pET expression vector was used since it produced 6xHis fusion protein for easy purification using affinity column. The Nickel chelating chromatography was used for purifying the expressed ATF2 from E. coli BL 21. This study presents the vitro interaction between BATF and ATF2 by EMSA(Electro-phoretic Mobility Shift Assay). The expression of transcription factors for dimerization by transfection from C3H10T1/2 cells were analyzed by immunoblot. The transactivation potential was investigated by using reporter gene containing either TRE sites for AP-1 or CRE sites for ATF2. BATF negatively regulates expression of AP-l and CRE reporter genes.

      • 살아있는 세포에서의 bZIP 전사인자들의 상호작용의 분석

        Activator protein 1 (AP-1) belongs to the basic region leucine zipper (bZIP) family of transcription factors. It had examined the predicted interaction between BATF, a member of the AP-1 transcription factor family and ATF2 a member of the CREB/ATF family of bZIP proteins. To determine if the mechanism of BATF inhibition involves dimer formation with Jun, ATF2 or both proteins, it was examed by multicolor BIFC (multicolor bimolecular fluorescence complementation) assay in this study. This assay allows us to study multiple protein interactions simultaneously in the same cell. The expression of transcription factors for dimerization were analyzed by immunoblot. The transactivation potential was investigated for dimerization of transcription factors by transfection from Hela cells by using plasmid containing reporter gene for AP-1 target sequences. Activator protein 1 (AP-1)는 전사인자들의 basic region leucine zipper (bZIP) family에 속한다. AP-1 전사인자 family에 속하는 BATF와 bZIP 단백질의 CREB/ATF member에 속하는 ATF2와의 예측된 상호작용은 조사되었다. BATF와 억제기능을 내포하는 mechanism에 있어서 Jun, ATF2 또는 두 종류의 단백질 모두와의 이량체를 이루는지를 알아보기 위 하여 이 연구에서는 multicolor BIFC (multicolor bimolecular fluorescence complementation) assay 로 조사하였다. 이 assay는 같은 세포에 있어서 동시에 여러 가지 단백질의 상호작용들을 연구하도록 해 준다. 이량체를 형성 하는 전사인자 단백질의 표현은 immunoblot로서 analyze 하였다. 그리고 이량체 형성의 전사잠재력 분석은 AP-1 표적서열의 reporter gene을 가진 plasmid를 이용하여 cell에 transfection을 시킨 후 transactivation potential을 조사하였다

        반딧불이의 발광유전자는 루시퍼라제는 유전자인데 이 유전자를 최근 생명과학 분야의 연구에 많이 활용되고 있다. 반딧불이의 발광 유전자의 유용성은 현재 cell-based assay 방법으로 개발하여 많은 리포터 유전자 시스템으로 사용되어 발현벡터로서 상용화되고 있다. 본 논문은 발광유전자를 이용하여 전사활성 및 그 이외의 생리적 활성을 측정하는 분자생물학적 연구의 유용성에 관하여 알아본다.

        산 혹은 알칼리 음독시 상부 위장관의 주 손상 부위

        Background: It is generally known that while alkali ingestion injures principally the esophagus, acid usually spares the esophagus and damages the stomach mainly. The purpose of this study is to evaluate the most common site of upper gastrointestinal tract injury in acid/alkali ingestion based on endoscopic findings. Methods: We retrospectively evaluated 40 patients who ingested acid/alkali for types and amounts of product ingested, time required to undergo endoscopy, and initial endoscopic findings. Endoscopic lesions were graded according to the criteria suggested by Zagar and were graded at the authors' discretion to compare acid injury and alkali injury. Results: In the acrid ingestion group(n=27), the esophagus injury score was 2.55±1.18, the stomach injury score was 2.62±1.78, and the difference was -0.07±1.13, therefore, no significant difference was present(p=0.939). In the alkali ingestion group(n=10), the esophagus injury score was 1.63 ± 1.50 and the stomach injury score was 2.63 ± 1.20. Stomach injury was significantly more severe than esophageal injury(difference : -1.00± 1.18, p=0.026, Wilcoxon signed ranks test). Conclusion: While no difference exists between injuries of the esophagus and the stomach due to acid ingestion, the stomach was the most common site in alkali injury.

        미용종사자의 재교육 실태조사 및 문제점

        The purposes of this study were, a more systemic and desirable impmvenient method of retraining program was researched for cultivation of field beauticians through acknowledgement of the environmental change and phenomenon of beauty shops by time and through recognition of current retraining situation. 1) In terms of acknowledgement of the necessity for beautician retraining, the item on the necessity for beautician retraining showed beauticians' high requirement fur retraining. 2) The main problem of reeducation was that reeducation curricular of each organization have not been programed. 3) Presence education was the main thing to be completed for reeducation program improvement. We also could know that the improvement and complement of work environment, the education extension besides technology education, and the rapid acquisition of information as improvement were recognized importantly.

      • 영아기 자녀를 양육하는 어머니와 조모의 상호작용활동

        The purpose of this study is to consider the interaction between two generations carrying on actual bring-up children through investigation of its interaction between mother and grandmother caring their infant. The subjects were 60 pairs of mother as a housewife who has the first child under 24 monthes and grandmother on mother's side or father's side using the inquiry paper method and interview. To take answer, it was distributed/explained directly to each person and then filled out the inquiry. Total 120 sheets were used for final analysis. The measuring instruments were interaction between grandmother and mother according to delivery before and after, it was used the modified 4 questions from barbara J. Myers and Margaret G. Williams petersen(1991)'s study through Grandmother-Mother-Infant Interaction (Jarvos & Creaesy, 1987). For analyzing this matter, it was generated the frequency, percentage, average and value of standard deviation through SPSSWIN program. The results of analyses for this study are as follows. The interaction between grandmother and mother was shown that father-side grandmother often interacted more than mother-side grandmother. It was shown that while mother-side grandmother interacted with mother 'everyday', mother-side grandmother interacted with mother just 2 to 3 times per week, or 1 to 2 times per month.

      • 학령기 아동의 언어적 특성 : 영재아동과 일반아동의 비교

        The Linguistic Properties Comparison between nongifted children and Gifted children. These purposes are to have a necessity to be educated for the linguistic gifted among many gifted in speedy changable age, and to find out what differences are of properties between the linguistic gifted children and nongifted children through comparing/analysing to gifted children any nongifted children using performance evaluation on writer's ability. Therefore, it intends to use as a assisting material in order to develope ability and properties of the linguistic gifted children and nongifted chi ldren as well. The studying details are 1) to compare/analyze thinking ability between the linguistic gifted chi ldren and nongifted children 2) to find out differences of thinking ability for unrealistic reasoning between the linguistic gifted children and nongifted children. The studying subjects had been chosen 3 children as a first grade in 'C' Gifted Academy and 3 children as a first grade in an elementary school from June 3, 1999 to June 12, 1999. The studying instrument was an evaluation of linguistic properties certification(Project Spectrum : Krechevsky, '1994). It had got a frequency calculation, average and standard deviation through the material anylzing with the program SPSSWIN. The conclusions are as belows, First, as a result of performance evaluation on writer's ability to gifted children and nongifted children, the gifted children were outstandingly shown the linguistic ability getting much higher score than nongifted children in respect of vocabulary level, structure of writing, and consistency/logicality of theme. Second, it was shown the gifted children had diffusing thought than nongifted children through the esthetic question and imformation memories with listening to the realistic juvenil story and the unrealistic juvenile story.

      • Transgenic mouse model system을 위한 BATF knock out vector를 이용한 ES cell의 transfaction

        Transgenic vector that is knocked out of exon 111 of BATF was transfected to ES cells to investigate the function of leucine zipper of BATF transcription factor. The ES cells that was transfected by transgenic vector was selected from grown in the selected medium. The DNA from ES cells that was selected from selected medium was performed PCR and the product was analyzed by 1% agarose gel to get positive clone. The probe for southern blot was prepared to analyze for transgenic mouse model system. 전사인자 BATF의 leucine zipper 기능을 알기위하여 이를 encode 한다고 알려져 있는 exon 111 부분을 knock out 시킨 transgenic vector를 ES cell에 transfection 시켰다. 그 후 transgenic vector를 가진 ES cell은 선택적인 medium에서 선택되었다. 선택되어진 ES cell의 DNA을 이용한 PCR product는 1% agarose gel에서 분석되어 졌으며 positive clone을 확인 하였다. 또한 knock out mouse를 확인할 수 있도록 Southern blot를 위한 probe를 만들었다.

      • In vivo biotinylation system에 의한 BATF 결합 단백질의 정제를 위한 전략

        This paper presents the construction of vector BATF for purification of binding protein with BATF with in vivo biotinylation system from cell extracts. Constructed vector contains BT/BATF gene and another vector which contains HA -BirA gene were cotransfected to Hela cells. The expression of BT/BATF protein and the detection of biotinylated BT/BATF were confirmed by Immunoblot. The result of immunoblot was confirmed by using Myc, HA antibodies and streptavidin HRP antibody. 본 논문은 in vivo biotinylation system을 사용하여 cell extracts로 부터 BATF와 결합하는 단백질을 정제하기 위하여 필요한 vector를 construct 하였다. Constructed 된 plasmids 중 하나는 2개의 분리된 cDNs 를 포함하는 pcDNA3-BT/BA TF-Myc 이다 Constructed 된 plasmid 와 HA-BirA 유전자를 가진 또 다른 하나의 pcDNA3-HA-Bir A 라는 2개 의 plasmid들을 Hela cell 에 cotransfection 하였다. BT/BATF등의 각각의 단백질의 expresslon과 biotinylated BT/BATF의 탐지는 lmmunoblot를 사용하였으며 Myc 와 HA antibody 그리고 streptavidin HRP antibody에 의해 확인 되었다.
