        Uncoupling Protein 3의 골격근 세포내 과발현이 OLETF 백서 및 배양된 골격근 세포에서 포도당대사에 미치는 영향

        연구배경:Uncoupling protein(UCP)는 미토콘드리아의 내막에 위치하는 단백질로 세포내의 과다한 에너지를 열로 발산시키는 기능을 가진다. 최근 동물의 갈색지방조직에만 존재하는 UCP와 유사성을 가진 아형들(UCP2,3)이 사람에게도 존재함이 알려져 큰 관심을 끌도 있는데 이중 UCP3는 그 발현이 골격근세포와 갈색지방조직에만 국한된다. 본 연구에서는 UCP3가 체내 인슐린 감수성을 결정하는데 가장 중요한 조직인 골격근에 국한되어 발현되는 점에 착안하여 UCP3를 골격근세포에 과발현시켰을 때 포도당 대사에 어떠한 영향이 나타나는 지를 조사하였다. 방법:25주령의 8마리의 OLETF 백서를 대상으로 하여 4마리는 골격근에 adenovirus 2mL(1×10¹²pfu/mL)를 주사하여 대조군으로 하였고 4마리는 골격근에 재조합법으로 제작된 adenovirus­UCP3 2mL(1×10¹²pfu/mL)를 주사하였다(UCP3 과발현군). UCP3를 투여한 백서에서 먹이섭취가 증가하는 경향이 있어 그 전날 대조군이 먹은 야의 먹이만큼 투여하였다. 골격근에 adenovirus를 주사한 10일 후에 euglycemic hyperinsulinemic clamp를 시행하였다. Adenovirus­UCP를 C2C12 골격근 세포에 transfection시켜 UCP3를 C2C12 골격근 세포에 transfection시켜 UPS3­C2C12를 만들고 C2C12 골격근 세포와 UPS3­C2C12 골격근 세포에서 포도당 수송 및 당원합성을 측정하였다. 결과:UCP3 과발현 OLETF에서 체중이 감소하는 경향을 보였고 인슐린 감수성이 증가하였다. C2C12세포에서 기저상태 포도당 수송은 1.28±0.17μmol/L/min였고 100nM 인슐린으로 2시간 처리한 후 2.67±0.20 μmol/L/min로 증가하였다. UCP3­C2C12 세포에서는 기저상태 포도당 수송이 3.98±0.13μmol/L/min로 증가되었고 인슐린 처리 후 5.74±0.44μmol/L/min로 증가하였다. 인슐린을 처리한 UCP3­C2C12 세포에 P13K 억제제인 wortmannin을 첨가하였을 때 포도당 수송활성이 3.81±0.20μmol/L/min로 감소하였다. 기저상태 당원합성은 C2C12 세포에서 0.25±0.01μmol/L/min였고 인슐린 처리 후 0.45±0.01μmol/L/min로 증가하였다. UCP3­C2C12 세포에서는 기저상태 당원합성이 0.62±0.01μmol/L/min였고 인슐린 처리 후 1.26±454μmol/L/min로 증가하였다. UCP3­C2C12세포에 wortmannin을 첨가하였을 때 당원합성율이 0.80±0.04μmol/L/min로 감소하였다. 결론:UCP3 과발현이 OLETF 백서에서 인슐린 감수성을 증가시켰고 골격근세포에서 포도당 수송 및 당원합성을 증가시켰다. wortmannin을 첨가하였을 때 포도당 수송 및 당원합성이 감소함으로 보아 이 과정이 인슐린 신호전달체계인 P13K에 일부 의존함을 알 수 있었다. Background : UC P3 is a mitochondrial membrane protein expressed selectively in the skeletal muscle and brown adipose tissue. Since the skeletal muscle is the main organ determining insulin sensitivity in the body, it was hypothesized that UCP3 overexpression in skeletal muscle cells would improve glucose metabolism. Methods : An adenovirus-UCP3 was produced by a recombinant DNA method. OLETF rats were divided into 2 groups. Four rats were injected with the adenovirus-UCP3 (UCP3 group) and others were injected with the adenovirus(control group) in the skeletal muscle. The UCP3 group was provided with the same quantity of food as that consumed by the control group on the previous day. Insulin sensitivity was evaluated by the euglycemic hyperinsulinemic clamp method. In a separate experiment, glucose transport and glycogen synthesis we evaluated in C2C212 cells transfected with ether an adenovirus or the adenovirus-UCP3. Results : The insulin sensitivity improved significantly and the body weight decreased in the UCP3 group. The glucose transport and glycogen synthesis were higher in the UCP3-C2C12 skeletal muscle cells at the basal state. After insulin treatment, glucose transport and glycogen synthesis were also higher in the UCP3-C2C12 cells but the increments were reduced after treatment with wortmannin, a PI3K inhibitor. Conclusion : Insulin sensitivity was higher in the UCP3-overexpressed OLETF rats in the in vivo study. UCP3 transfection also increased glucose transport and glycogen synthesis in the cultured skeletal muscle cells by a PI3K dependent mechanism(J Kor Diabetes Asso 25 :460~468, 2001).

        가열 조건을 달리한 단호박 페이스트와 검 종류별 단호박 라떼의 품질특성

        가열 조건에 따른 단호박 페이스트를 제조하기 위해, 단호박을 15분 간 초벌 증숙한 뒤, 고압가열 처리 0분(A), 10분(B), 20분(C), 40분(D) 실시하여 실험군의 품질특성 을 조사하였다. 그 결과 일반성분의 경우, 대체적으로 고 압가열 처리 유무에 따른 유의적 차이가 관찰되었으며, 고압가열 처리한 B, C, D 실험군의 수분함량, 조단백질, 조섬유가 고압가열 무처리군 A에 비해 감소하였고, 가용성 무질소물은 증가하는 것으로 나타났다. 가열 조건별 단호박 페이스트의 수용성식이섬유는 고압가열 20분 처리군인 C의 측정치가 2.02로 가장 높았으며 무처리군인 A의 1.60 보다 증가하였고, 고압가열 처리 40분의 경우 오히려 감소하는 것으로 나타났다. 색도의 L 값은 고압가열 무처리군인 A가 52.20에서 고압가열 처리 10분, 20분, 40분으로 시간이 증가함에 따라 각각 50.33, 49.46, 48.06으로 감소하였고, a 값과 b 값 또한 유의적인 차이를 보이며 감소하였다. 현미경을 통한 단호박 페이스트의 현탁액 입자를 관찰한 결과 카로티노이드를 포함하는 유세포가 관찰되었으며 고압가열 처리와 그 시간이 증가함에 따라 단호박 유세포의 변형이 뚜렷하게 관찰되었으나, 부유안정성 실험 결과 실험군 A, B, C, D 간 차이가 없었다. 이때, 가열조건의 선택은 수용성 식이섬유의 증가, 환원당 증가, 단맛의 관능특성이 유의적으로 높고, 전반적 기호도가 가장 우수한 것으로 나타난 고압가열 처리 20분인 C 실험군으로 결정하였다. 선택된 조건의 단호박 페이스트에 식품가공 시 널리 사용되는 검류인 xanthan gum, locust bean gum, guar gum을 단호박 페이스트에 종류별로 첨가하여 부유안정성을 확인하였는데, 이 결과 guar gum, locust bean gum, xanthan gum 순으로 부유안정성 효과를 나타냈으며 locust bean gum과 xanathan gum은 비슷한 정도의 효과를 보였다. 또한 관능검사를 통한 기호도 확인 결과 텍스쳐와, 전반적인 기호도가 가장 우수했으므로 locust bean gum(0.2%) 첨가 단호박 라떼의 품질이 가장 적절할 것으로 판단된다. For the production of pumpkin paste with respect to heating conditions, we steamed the pumpkin for roughly 15 min, heated it with high pressure treatment for 0 min (A), 10 min (B), 20 min (C), 40 min (D), and subsequently investigated the quality characteristics. Generally a significant difference was observed between the pumpkin paste treated with and without high-pressure heat. The values of water content, crude protein and crude fiber of the high-pressure heat-treated groups B, C, D were decreased compared with untreated group A. The soluble fiber in experimental group B sweet-pumpkin paste treated with high-pressure heat for 20 min was higher than the control, and the highest value at 2.02. Experimental group D sweet-pumpkin paste treated with high-pressure heat for 40 min was found to have a decreased soluble fiber content relative to the control. The L value for the color of the group A untreated control sweet-pumpkin paste (no high-pressure heating) decreased as the time increased from 10 min to 40 min, with L values of 50.33, 49.46, and 48.06, respectively. The b value for the color of the sweet-pumpkin paste also decreased, showing a significant difference. Taking into account all the results, we chose experimental group B in order to prepare sweet-pumpkin latte. We used 0.2% gum (xanthan gum, locust bean gum, guar gum) as a stabilizer. Sweet-pumpkin latte with xanthan and locust bean gum has a suspension stability effect that lasts 90 min. The L and b values of sweet-pumpkin latte with gums increase and a value decrease compared with the control. In terms of the overall acceptance of the sweet-pumpkin latte, the experimental group with xanthan gum scored the best.

      • Subcritical water extraction (SWE) of anthocyanin from blueberries (Vaccinium corymbosum)

        Subcritical water extraction (SWE) is an eco-friendly new extraction technology because it does not contain harmful organic solvent and has high extraction efficiency in a short time compared with conventional extraction methods. Blueberries (Vaccinium corymbosum) are widely known as superfood due to rich source of anthocyanin (malvidin-3-o-galgctoside) and antioxidant activity. In this study, optimal extraction condition of SWE from blueberries was determined and compared with the conventional extraction methods. SWE was carried out using a Dionex Accelerated Solvent Extractor (ASE, Model 350) under conditions of temperatures (110, 130, 150 and 170°C) and times (1, 3, 5 and 10 min). Total anthocyanin of SWE extracts was compared with hot water (60°C, 1 h) extract and pressed juice extract. The total anthocyanin content was determined by pH differential method. Considering both the extraction time and temperature conditions, the highest content of total anthocyanin content was 0.455 mg/g FW Vaccinium corymbosum at 130°C for 3 min. At high temperature and long extraction time, the anthocyanin in the blueberries will undergo thermal degradation due to low stability of anthocyanin at extreme condition. Besides, maximum yield of anthocyanin from blueberries using SWE was about 1.2 and 3.8 times more higher than hot water extract and pressed juice extract, respectively. Therefore, SWE is faster and more efficient method to extract anthocyanin from blueberries than conventional extraction methods. This study shows a possibility of SWE applied to food processing industry.

        대황의 항산화 효과 및 타이로시네이즈 저해 활성

        본 연구에서는 대황 추출물의 항산화 활성, 타이로시네이즈(tyrosinase) 저해 활성을 확인하였다. 대황의 50 % 에탄올 추출물, 에틸아세테이트(ethyl acetate) 분획, 아글리콘(aglycone) 분획으로 실험을 진행하였다. 대황 추출물들의 DPPH (1,1-diphenyl-2-picrylhydrazyl) 소거활성(FSC<sub>50</sub>)은 대표적인 항산화제인 (+)-α-tocopherol보다 낮은 것으로 나타났다. Luminol 발광법을 이용한 Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub> 계에서 생성된 활성산소종에 대한 아글리콘 분획의 소거활성(총 항산화능, OSC<sub>50</sub>)은 0.265 μg/mL로 매우 큰 활성을 나타내었다. 대황 추출물의 rose-bengal로 증감된 <sup>1</sup>O<sub>2</sub>에 의한 적혈구 파괴에 대한 세포보호 효과는 모든 분획에서 농도 의존적(1∼50 μg/mL)으로 증가하였으며, 특히 아글리콘 분획은 10 μg/mL 농도에서 τ<sub>50</sub>이 757.0 min으로 높은 세포 보호 활성을 나타내었다. 대황 추출물 중 아글리콘 분획의 타이로시네이즈 저해활성(IC<sub>50</sub>)은 11.20 μg/mL으로 226.88 μg/mL인 알부틴(arbutin)보다 큰 활성을 보여주었다. 이상의 결과들로부터 대황 추출물은 활성산소종을 소거하는 항산화제로 이용가능하며, 특히 아글리콘 분획의 현저한 항산화 작용 및 큰 타이로시네이즈 저해 효과로부터 이들 분획 또한 화장품원료로서 응용 가능성이 큼을 알 수 있었다. In this study, the antioxidative effects and inhibitory activities on tyrosinase of Rheum undulatum (R. undulatum) L. extracts were investigated. 50 % ethanol extract, ethyl acetate and aglycone fractions of R. undulatum L. were used in experiments. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activities (FSC<sub>50</sub>) of R. undulatum L. extracts was lower than (+)-α-tocopherol, known as a typical antioxidant. Reactive oxygen species (ROS) scavenging activities (OSC<sub>50</sub>) of aglycone fraction on ROS generated in Fe<sup>3+</sup>-EDTA/H<sub>2</sub>O<sub>2</sub> system using the luminol-dependent chemiluminescence assay showed the most prominent effect at a concentration of 0.265 μg/mL. The cellular protective effects of extract/fractions of R. undulatum L. on the rose-bengal sensitized photohemolysis of human erythrocytes were increased in a concentration dependent manner (1 ~ 50 μg/mL). Especially, aglycone fraction in 10 μg/mL concentration showed the most protective effect among extracts (τ<sub>50</sub>, 757.0 min). The inhibitory effects (IC<sub>50</sub>, 11.20 μg/mL) on tyrosinase of aglycone fraction was much higher than arbutin (226.88 μg/mL), known as a whitening agent. These results indicate that R. undulatum L. extracts can be used as antioxidant. Particularly, aglycone fraction of R. undulatum L. showed superior antioxdative activity and high inhibitory effect on tyrosinase. Therefore, aglycone fraction of R. undulatum L. could be applicable to new functional cosmetics.

        자몽껍질 유래 플라바논의 최적 추출 및 기능성 소재 캡슐화

        버려지는 자몽 껍질에 다량 함유되어있는 플라바논을 추출하여 기능성 성분을 재이용하였다. 친환경용매인 아임계수 추출기술을 이용하여 170℃, 10 min의 최적 조건에서 추출함으로써 무독성 용매로 빠르고 경제적으로 추출할 수 있었다. 자몽껍질 추출물을 β-cyclodextrin을 이용하여 처리함으로써 플라바논과 같은 비극성 물질을 캡슐화하여 소재의 가용화를 용이하게 하였다. 이것은 항산화 기능이 향상된 대체 소재의 개발 및 건강지향식품에 이용할 수 있으며, 이를 통하여 세계 기능성 식품 시장으로의 진출을 위한 발판으로도 삼을 수 있을 것으로 기대된다. The extraction of flavanones such as naringin, narirutin, naringenin, hesperidin, and hesperetin from grapefruit peels was performed using subcritical water extraction (SWE), hot water extraction, and conventional methods such as methanol and ethanol extraction. We analyzed the total flavanone content using high-performance liquid chromatography (HPLC) for each extracting method. Among the three methods, SWE was the optimal method with optimal operating conditions of 170℃ temperature and 10 min operating time. The maximum total flavanone extracted was 86.539±3.52 mg/g grapefruit peels. Moreover, we treated the extracts with 60% β-cyclodextrin and then analyzed the surface structure of the encapsulated compounds by field emission-scanning electron microscopy (FE-SEM). The results indicated that the encapsulation in β-cyclodextrin improved solubilization, and the inclusion complex could serve as food supplements.

        다문화가정 어머니의 언어 능력, 자발발화 특성과자녀의 자발발화 특성 분석

        15분의 자유놀이 상황에서 수집한 40쌍의 다문화가정 어머니와 자녀의 자발발화 특성을 평균발화길이와 문법적 형태소 사용의 측면에서 분석하였다. 다문화가정 어머니의 문법적 형태소의 사용빈도는 낮았으며 MLU는 3.53(.84)이었다. 다문화가정 아동의 MLU는 2.39(.79)였으며 한국아동의 수준보다 낮았다. 아동과 어머니의 언어능력과 서술 격조사, 용언접미사, 선어말어미간 사용간에는 유의한 정적 상관이 있었다. Forty multicultural mothers` language skills, language characteristics and those of children`s during 15 min. of free play were analyzed in terms of grammatical morphemes and MLU. The use of grammatical morphemes was low in multicultural mothers and the MLU of multicultural mothers was 3.53(.84) The MLU of children with multicultural mothers was 2.39(.79) which is low compared with that of Korean children. Mothers` language abilities and use of particle endings, suffix for a declinable word, suffix before endings was positively correlated with those of children`s.

        난치성 백반증에 대한 흡입물집을 이용한 비배양 표피세포이식술 20례

        Background: As nonsurgical interventions for vitiligo are not always successful, various surgical modalities have been used in patients with refractory vitiligo. Of these, non-cultured epidermal suspension transplantation (NCES) was recently introduced to treat large recipient sites using cells from small donor tissue. Objective: We assessed the effectiveness and safety of NCES as a surgical treatment for patients with refractory vitiligo. Methods: We retrospectively reviewed 20 cases in 17 patients (11 females; median age 25 years) who underwent NCES from July 2015 through March 2018. Suction blisters (20 mm in diameter) were collected from the patient’s inner thigh at a donor-to-recipient area ratio of 1:5. After the addition of 5 mL recombinant trypsin solution to the suction blisters, followed by incubation at 37°C for 60 min, epidermal cells were manually scraped off the blister surface, and epidermal cell suspension was obtained by centrifugation at 1,500 RPM for 5 min. The suspension was applied to the vitiligo regions after epidermal ablation of those regions. Phototherapy resumed 1 month later. Treatment success was defined as ≥75% repigmentation of the surgical site, and all adverse events were noted. Results: Overall, 85.0% of cases (17/20) exhibited treatment success. Adverse events included hyperpigmentation (20%) and surgical site infection (5%), but the treatment was tolerable in all cases. Conclusion: NCES is a reliable surgical option for patients with vitiligo refractory to nonsurgical treatment. Large areas of vitiligo can be treated by NCES, and use of this technique should be encouraged in Korea. (Korean J Dermatol 2018;56(7):426∼432)

        초등학교 저학년용 영양교육 소책자 개발 및 효과 분석

        The purpose of this study was to investigate effects of nutrition education program and pamphlet for the lower grades elementary students focused on individual daily needed food exchange units using Food Exchange System. Program consisted of four lessons (40 min/lesson), 5 major nutrients & function, 6 food group and sources, daily needed food exchange units for normal body weight, and smart snack choice and exercise. Pamphlet as activity book was developed for the program. The subjects were 3rd grade elementary students (educated group, 31 vs. non-educated group, 31). Educated group were lessoned as group and/or individual. We examined the differences in nutrition knowledge, dietary attitudes, dietary intakes and satisfaction of the program and pamphlet. In educated group, there were positive improvements on nutrition knowledge score function and foods of 5 nutrients and on dietary attitudes type of breakfast and snacks. In the evaluation of dietary intakes according to KDRI, there were positive improvements on intakes level of riboflavin, vit. C, folate, Ca, P, Fe and Zn in educated group. In satisfaction with the program and pamphlet, contents, font size, visual, figure, difficulty and program curriculum were over 2.90/3.0. It showed that the developed nutrition education program and pamphlet focused on individual daily needed food exchange units using Food Exchange System improved nutrition knowledge, dietary attitudes and nutrients intake level in the lower grades elementary students.         인체혈장 중 에탐부톨의 HPLC 분석법의 검증 및 단일용량 투여에 의한 약물동태 연구

        An HPLC method was employed for the determination of ethambutol in human plasma. After addition of internal standard (IS, octylamine, 2 μg/mL) and alkalinization of the plasma with 5 M sodium hydroxide, the drug and IS were extracted into the mixture of chloroform and diethyl ether (40:60, v/v). Following a 15-min vortex-mixing and a 10-min centrifugation, the organic phase was spiked with 100 pL of phenylethylisocyanate (2000 μg/mL) for chemical derivatization, mixed for 5 min and evaporated to dryness under a stream of nitrogen. The residue was reconstituted with 100 μL of mobile phase and 20 pL was injected into Cl8 column with a mobile phase consisting of methanol:water (70:30, v/v). The samples were detected utilizing an ultraviolet detector at 200 nm. The method was specific and validated with a limit of 0.15 μg/mL. Infra- and inter-day precision and accuracy were acceptable for all quality control samples including the lower limit of quantification. The applicability of this method was demonstrated by analysis of human plasma after oral administration of a single 1200-mg dose to 20 healthy subjects. From the plasma ethambutol concentration vs. time curves, the mean AUC was 9.61 ± 1.64 μg hr/mL. and Cmax of 2.68 μg/mL reached 2.73 hr after administration. The mean biological half-life of ethambutol was 3.46 ± 1.21 hr. Based on the results, this simple and validated assay could readily be used in any pharmacokinetic studies using humans.

      • Improving the subcritical water extraction of flavonoids narirutin and hesperidin from Citrus unshiu peel by pulsed electric fields

        Citrus fruit is important source of flavonoids, mainly flavanones which are narirutin and hesperidin. Those citrus flavonoids have been found to have health-related properties including antioxidant, anticancer, and anti-inflammatory. The main purpose of this study was to verify that the extraction of narirutin and hesperidin from Citrus peel can be more effective by combining pulsed electric field (PEF) pre-treatment and subcritical water extraction (SWE). Citrus unshiu peels were treated with PEF under conditions of electric field strength (3 kV/cm) and times (1 and 2 min). Subsequent SWE was conducted by using a Dionex Accelerated Solvent Extractor (ASE, Model 350) at extraction temperature 170°C for 10 min. The total flavonoids content was measured by using the aluminum chloride colorimetric method and the antioxidant capacity was analyzed by the Ferric reducing antioxidant power (FRAP) assay using spectrophotometer. The concentrations of narirutin and hesperidin were increased as PEF pre-treatment time increased. The highest concentrations of narirutin and hesperidin were 13.41 mg narirutin/g dry citrus peel and 141.16 mg hesperidin/g dry citrus peel at PEF pre-treatment condition of 3 kV/cm and 2 min. The total flavonoids contents of the extracts increased 105.2% and 123.1% for citrus peel PEF treated at 1 and 2 min, respectively. In addition, compared to the untreated sample, PEF pre-treatments of 1 and 2 min increased the antioxidant capacity of the extracts 109.2% and 160.8%, respectively. Therefore, the results demonstrate the potential of PEF pre-treatment to improve the SWE of flavonoids from citrus unshiu peel.
