The cytotoxicity of cantharidine(CTD), the main component of Cantharis, and the combined effect of CTD and anti-tumor drugs on HeLa. Hep G2,SK-OV3. KOHS-NP, BALB/c 3T3 cells, mouse splenocytes and human lymphocytes were estiated by MTT colorimetry assay. CTD inhibited the proliferation of anti-tumor cells, BALB/C 3T3, mouse spleen cells and human lymphocytes. CTD increased the cytotoxicity of mitomycin C, cisplatin or mercaptopurine on tumor cells and BALB/c 3T3 cells. These results indicate that cantharidine has the cytotoxicity of tumor cells, fibroblast cells and immunocytes.

The tumor suppressor gene p53, located on the short arm of chromosome 17, encodes for nuclear protein which regulates cell proliferation by inhibiting cells entering S-phase. Mutation in p53 gene are the most frequent genetic alteration found in human cancers to date. This study was examined mutant p53 gene mutation using PCR-SSCP method and mutant p53 oncoprotein expression using avidin biotin peroxidase complex method immunohistochemically, in 2 seborrheic keratosis, 3 basal cell carcinoma, 3 squamous cell carcinoma and 1 metaststic squamous cell carcinoma cases. With PCR-SSCP methods in exon 5 and 7, no genetic mutation of p53 gene was observed in 2 seborrheic keratosis, 3 basal cell carcinoma, 3 squamous cell carcinoma, and 1 metaststic squamous cell carcinoma cases. With immunohistochemical staining with ABC method, a case of squamous cell carcinoma developed on lower lip was positive but another were all negative. This result means that p53 gene mutation on skin tumor may develope in squamous cell carcinoma developed on sun exposed area and this mutation on squamous cell carcinoma can develope other exonal site except exon 5 and 7.

The purpose of this research was to investigate the mechanism of inhibitory action of Glycyrrhizin(GZ) on the proliferation of mouse leukemia cell-line, L1210 cells. The cytotoxic activity was tested using a colorimetric tertrazolium assay(MTT assay), the apoptosis was tested using flow cytometry. Nitric oxide(NO) production form mouse peritoneal macrophage was tested using a Griess method and the phagocytosis of human polymorphonuclear cells was tested using a lucigenin chemiluminescence. GZ ingibited the proliferation of L1210, BALB/c 3T3 cells and mouse thymocytes at 50 ug/ml/ GZ did not affect nitric oxide production from mouse peritoneal macrophages in vitro, but ingibited nitric oxide production from lipopolysaccharide and y-interferon treated macrophages. Macrophages of GZ-administered mice accelerated NO production. The proliferation of L1210 cells apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. The apoptosis of L1210 cells were induced by co-culture with macrophage of GZ-administered mice. GZ increased the phagocytosis of human polymorphonuclear cells. These results suggest that GZ inhibit the proliferation of L1210 cells not only as a direct cytotoxic agent o tumor cells, but also by the enhancement of NO production and phagocytic activity.

The purpose of this research was to investigate effects of Sa-Kunja-Tang(SKT) on immune cells of L1210 cell-transplanted mice. The apoptosis and T lymphocytes subopoulation were tested using a flow cytometry, and the proliferation was tested using a MTT assay. Nitric oxide production from mouse peritoneal macrophage was tested using a Griess reagents, and the phagocytic activity of mouse peritoneal macrophage was tested using a lucigenin chemiluminescence. SKT suppressed apoptosis of T-lymphocytes induced by L1210 transplantation. SKT decreased nitric oxide production from mice peritoneal macrophages increased by L1210 transplantation, and the phagocytic activity decreased by L1210 transplantation. These results suggest that SKT suppresses T lymphocyte apoptosis and macrophage activity in L1210 transplanted mice.

The purpose of this research was to investigate effects of Sa-Kunja-Tang(SKT) on immune cells of antitumor drugs administered mice. The apoptosis and T lymphocytes subpopulation were tested using a flow cytometry, and the proliferation was tested using a MTT assay. The administration of etoposide. vincristine or doxorubicin increased the apoptosis of T-lymphocytes, but the action of doxorubicin was decreased by the administration of SKT. The administration of etoposide or vincristine decreased helper T and cytotoxic T cells population of T lymphocytes, but the action of vincristine was recovered by the administration of SCT. The administration of etoposide, vincristine or doxorubicin decreased the proliferation of T-lymphocytes, but the action of doxorubicin was increased by the administration of SKT. These results suggest that SKT has a regulative function of T-lymphocytes in anti-tumor drugs administered mice.

Lupus vulgaris, which is a progressive form of postprimary tuberculosis in patients with a moderate to high degree of immunity and tuberculin sensitivity, is the most common, most serious, and most variable type of cutaneous and mucous membrane tuberculosis. Domonstration of M.tuberculosis directly or in culture in some of these eruptions can be difficult. It is essential to demonstrate the presence of M.tuberculosis in a clinical sample for a definitive diagnosis of tuberculosis. With the discovery of the polymerase chain reaction (PCR), the DNA diagnostic approach for infection has developed in a short period. With the dual purpose of definitive and rapid diagnosis of tuberculosis, this study examined the usefulness of a nested PCR for detection of M. turberculosis. Compared with a single-step PCR, the two-step PCR was able to enhance sensitivity approximately 1,000-fold on the basis of bacterial counts and DNA quantity. We concluded that two-step nested PCR appeared to be the most useful PCR protocol for early and sensitive diagnostic method detecting M.tuberculosis in acid-fast stain negative specimens.

The acoustic emission (AE) method as a passive non-destructive monitoring technique is proposed for real-time monitoring of mechanical degradation in underground structures, such as deep geological disposal of high-level nuclear waste (HLW). This study investigates the low-frequency characteristics of AE signals emitted during the fracturing of meter-scale concrete specimens; uniaxial compression tests (UCT) in a lab scale and Goodman jack (GJ) tests in a 1.3 m-long concrete block were conducted while acquiring the AE signals using low-frequency AE sensors. The results indicate a sharp increase in AE energy emission at approximately 60% and 80% of the yield stresses in the UCT and GJ tests, respectively. The collected AE signals were primarily found in two frequency bands: the 4-28 kHz range and the 56-80 kHz range. High-frequency AE signals were captured more as the stress increased in the GJ tests, which was in contrast to the UCT tests. Furthermore, the AE signals obtained from the Goodman jack tests tended to lower RA values than the UCT results. This study presents unique experimental data with low-frequency AE sensors under different loading conditions, which provides insights into field-scale AE monitoring practices.

Deep geologic disposal of high-level nuclear wastes (HLW) requires intensive monitoring instrumentations to ensure long-term security. Acoustic emission (AE) method is considered as an effective method to monitor the mechanical degradation of natural rock and man-made concrete structures. The objectives of this study are (a) to identify the AE characteristics emitted from concretes as concrete materials under different types of loading, (b) to suggest AE parametric criteria to determine loading types and estimate the failure stage, and finally (c) to examine the feasibility of using AE method for real-time monitoring of geologic disposal system of HLW. This study performs a series of the mechanical experiments on concrete samples simultaneously with AE monitoring, including the uniaxial compression test (UCT), Brazilian tensile test (BTT) and punch through shear test (PTST). These mechanical tests are chosen to explore the effect of loading types on the resulting AE characteristics. This study selects important AE parameters which includes the AE count, average frequency (AF) and RA value in the time domain, and the peak frequency (PF) and centroid frequency in the frequency domain. The result reveals that the cumulative AE counts, the maximum RA value and the moving average PF show their potentials as indicators to damage progress for a certain loading type. The observed trends in the cumulative AE counts and the maximum RA value show three unique stages with an increase in applied stress: the steady state stage (or crack initiation stage; < 70% of yield stress), the transition stage (or damage progression stage; 70–90% of yield stress) and the rising stage (or failure stage; > 90% of yield stress). In addition, the moving average PF of PTST in the early damage stage appears to be particularly lower than that of UCT and BTT. The loading in BTT renders distinctive responses in the slope of the maximum RA–cumulative AE count (or tan ?). The slope value shows less than 0.25 when the stress is close to 30% of BTT, 60% of UCT and 75% of PTST and mostly after 90% of yield stress, the slope mostly decreases than 0.25 in all tests. This study advances our understanding on AE responses of concrete materials with well-controlled laboratoryscale experimental AE data, and provides insights into further development of AE-base real-time diagnostic monitoring of structures made of rocks and concretes.
