        Ethanol Extract of Ganoderma lucidum Augments Cellular Anti-oxidant Defense through Activation of Nrf2/HO-1

        Objectives: The mushroom Ganoderma lucidum has been widely used as a traditional herbal medicine for many years. Although several studies have focused on the anti-oxidative activity of this mushroom, the molecular mechanisms underlying its activity have not yet been clearly established. The present study investigated the cytoprotective effect of ethanol extract of Ganoderma lucidum (EGL) against oxidative stress (hydrogen peroxide, $H_2O_2$) and elucidated the underlying mechanisms in a C2C12 myoblast cell line. Methods: Oxidative stress markers were determined by using the comet assay to measure reactive oxygen species (ROS) generation and deoxyribonucleic acid (DNA) damage. Cell viability and Western blotting analyses were employed to evaluate the cellular response to EGL and $H_2O_2$ in C2C12 cells. Transfection with nuclear factor erythroid 2-related factor 2 (Nrf2)-specific small interfering ribonucleic acid (siRNA) was conducted to understand the relationship between Nrf2 expression and $H_2O_2$-induced growth inhibition. Results: The results showed that EGL effectively inhibited $H_2O_2$-induced growth and the generation of ROS. EGL markedly suppressed $H_2O_2$-induced comet-like DNA formation and phosphorylation of histone H2AX at serine 139 ($p-{\gamma}H2AX$), a widely used marker of DNA damage, suggesting that EGL prevented $H_2O_2$-induced DNA damage. Furthermore, the EGL treatment effectively induced the expression of Nrf2, as well as heme oxygenase-1 (HO-1), with parallel phosphorylation and nuclear translocation of Nrf2 in the C2C12 myoblasts. However, zinc protoporphyrin IX, a HO-1 inhibitor, significantly abolished the protective effects of EGL against $H_2O_2$-induced accumulation of ROS and reduced cell growth. Notably, transient transfection with Nrf2-specific siRNA attenuated the cytoprotective effects and HO-1 induction by EGL, indicating that EGL induced the expression of HO-1 in an Nrf2-dependent manner. Conclusion: Collectively, these results demonstrate that EGL augments the cellular anti-oxidant defense capacity through activation of Nrf2/HO-1, thereby protecting C2C12 myoblasts from $H_2O_2$-induced oxidative cytotoxicity.

        Morin exerts cytoprotective effects against oxidative stress in C2C12 myoblasts via the upregulation of Nrf2-dependent HO-1 expression and the activation of the ERK pathway

        <P>In the present study, we investigated the cytoprotective efficacy of morin, a natural flavonoid, against oxidative stress and elucidated the underlying mechanisms in C2C12 myoblasts. Our results indicated that morin treatment prior to hydrogen peroxide (H2O2) exposure significantly increased cell viability and prevented the generation of reactive oxygen species. H2O2-induced comet-like DNA formation and gamma H2AX phosphorylation were also markedly suppressed by morin with a parallel inhibition of apoptosis in C2C12 myoblasts, suggesting that morin prevented H2O2-induced cellular DNA damage. Furthermore, morin markedly enhanced the expression of heme oxygenase-1 (HO-1) associated with the induction and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the inhibition of Kelch-like ECH-associated protein 1 (Keapl) expression. Notably, these events were eliminated by transient transfection with Nrf2-specific small interfering RNA. Additional experiments demonstrated that the activation of the Nrf2/HO-1 pathway by morin was mediated by the extracellular signal-regulated kinase (ERK) signaling cascade. This phenomenon was confirmed with suppressed Nrf2 phosphorylation and consequently diminished HO-1 expression in cells treated with a pharmacological inhibitor of ERK. Collectively, these results demonstrated that morin augments the cellular antioxidant defense capacity through the activation of Nrf2/HO-1 signaling, which involves the activation of the ERK pathway, thereby protecting C2C12 myoblasts from H2O2,-induced oxidative cytotoxicity.</P>

        Objective: Occupational medicine specialists in the Busan, Ulsan and Kyung-Nam areas established an area-based occupational disease surveillance system and used this system to collected case information for the purpose of preventing occupational diseases Methods: l l hospital participated in this system. The authors selected five main diseases, which were hand-arm vibration syndrome (HAVS), work-related musculoskeletal disorder (WRMSD), occupational asthma, occupational skin disease and occupational and occupational lung cancer and established their case definitions. All cases were reported on the web, and real time analysis was conducted. Results: Between April 2001 and April 2003, 192 cases of HAVS, 118 cases of WRMSD, 33 cases of occupational asthma, 17 cases of occupational skin disease and 17 cases of occupational lung cancer (for a total of 377 cases of these five main diseases) were reported. most of the HAVS cases came from shipbuilding, and 172 of them (92.7%) were associated with grinding. Of the four main types of WRMSD, carpal tunnel syndrome (CTS) was the most prevalent with 46 cases and shipbuilding was also the main industry involved (83.9%). The ergonomic risk factors involved mainly associated with the hand. In 19 (57.6%) and 4 (12.1%) cases of occupational asthma, the agents involved. The causative agents of occupational lung cancer included Cr, welding fume, PAH, Ni, etc. 10(58.8%) of the cases were diagnosed as squamous cell carcinoma and 5(29.4%) as adenoma. Conclusion: This result showed that an area-based occupational disease surveillance system might provide an effective method of evaluating the prevalence of such diseases, however the Busan, Ulsan and Kyung-Nam provinces are too large to be treated as individual areas. Therefore, the authors suggest that each province should establish its own surveillance system.

        This study was designed to develop the service quality assessment scale to evaluate tourist perception and satisfaction toward service in buddhist temple as tourist attractions. The data(N=329) were collected Haein-sa temples that is the largest buddhist temple in Korea. Using exploratory factor analysis and confirmatory factor anaysis the scale with 17 items was generated : attractiveness(4 items), interpretation(5 items), education(2 items), consumables(3 items), facilities(3 items). The fit indexes suggested that the final version of the model provided an adequate fit to the data(e.g., SRMR=.049, CFI=.903, GFI=.941, and AGFI=.918).

        스케일이 있는 급수관내의 유도초음파의 전파 특성에 관한 연구

        배관 내의 스케일은 통수비를 감소시키므로 배관을 적절히 관리하기 위해서는 스케일량에 대한 정량적인 평가가 필수적이다. 상수도 급수관내의 스케일량을 평가하기 위해서 유도초음파를 사용하였다. 가변각 웻지를 사용하여 유도 초음파의 여러 모드를 발생시키고, 노후 배관을 따라서 전파하는 특성을 분석하였다. 실험적으로 관내의 스케일이 증가함에 따라서 f×d=1.5㎒·㎜인 경우에 F(M,2) 모드들의 진폭이 크게 감소하는 것을 알 수 있었다. 본 연구의 결과로 상수도 급수관의 스케일량 평가에 적합한 유도 초음파는 F(M,2) 모드들임을 알 수 있었다. Since the scale in pipes reduces the flow rate, a quantitative evaluation of the scale is essential for the proper maintenance of pipes. Guided waves were employed to estimate the amount of scale in water supplying pipes. Using variable angle wedge, several modes of guided waves were generated and their propagation charcteristics along the pipes with scale were analyzed. It was experimentally observed that the amplitude of F(M,2) modes at f×d=1.5㎒·㎜ decreased significantly with increasing amount of scale. The present study showed that F(M,2) modes were optimal to evaluate the scale in water supplying pipes.

      • 수도권 매립지의 매립가스 발생량 예측과 활용방안 연구

        수도권 매립지에서의 매립가스 발생량 평가를 위해 국내에 적용되었었던 몇 개의 모델을 사용한 결과 SCS model과 국가 온실가스 배출량 조사에 사용되었던 모델이 실제 포집하여 사용했던 양과 비교하였을 때 가장 유사한 결과를 나타내었다. 수도권 매립지의 경우 플라스틱, 합성고무 등 유기성 폐기물이 상당수 존재하기 때문에 예상했던 것 보다 매립가스의 생성속도가 느린 것으로 나타났다. 또한 매립지의 활용방안에 있어서도 기존의 매립장 포화 후 후속 매립장의 운영하는 방안보다, 단계적으로 매립장을 병행 운영하는 방안을 통해 매립가스의 원활한 수습과 매립지의 수명연장에 있어 보다 효율적인 것으로 나타났다.

      • Sloshing을 고려한 액체 추진제 로켓의 모델링 및 자동조종장치 설계에 관한 연구

        액체 추진제 로켓에 있어서 조종안정성에 미치는 영향을 해석하기 위하여 추력 5톤급의 과학관측 로켓의 기본적인 제원을 중심으로 강체운동, 공력, Sloshing 등을 고려하여 추력방향제어를 하는 자동조종장치를 설계하였다. 유체 Sloshing은 고정된 질량과 진동하는 질량으로 나누어서 생각할 수 있고, 진동하는 질량이 진자-감쇠기로 연결되어 있는 기구학적 모델로 생각할 수 있다. 로켓은 시간에 따라서 고도와 속도, 질량 등이 변하므로 시간을 일정 간격으로 나누어 비례-미분-적분 제어 이득을 조절함으로써 자동조종장치를 설계하고 전산모의 실험을 실시하였다. Sloshing의 극점과 영점을 S-영역의 허수축상에 쌍으로 나타나며, 극점이 영점보다 위에 있으면 안정하다는 것을 알았다. 따라서 로켓이 안정하기 위해서는 진자 중심의 위치가 로켓 무게중심보다 뒤에 있어야 하는데, 본 모델의 전산모의실험 결과 진자 중심의 위치가 초기에 무게중심 전방에 있다가 곧바로 후방으로 이동함에 따라 시스템 전체의 양상이 바뀌게 되고 그에 따른 PD이득을 한번 조절함으로써 안정한 상태를 끝까지 유지하는 것으로 나타났다. 또한 Sloshing 과 발음각의 영향을 모두 고려하고 초기 발사각을 80도로 주었을 때에는 정상상태 오차가 발생함에 따라 이를 보상하기 위해서 PI이득을 조절함으로써 정상상태 오차가 없어졌다. In this work modeling and design of an autopilot for liquid propelleant rocket with sloshing effect is considered. In the modeling the aerodynamic forces and sloshing of the liquid fuel as well as rigid body dynamics are included. The design parameters for controller are varied due to mass, altitude, and velocity changes. Also the center of sloshing mass is changed aft as time goes. Therefore a PID controller is constructed with gain schedule technique based on time slices. The simulation results show that the proposed controller makes the rocket attitude follow the command in areasonable way.

      • 間歇的 運動에 의한 脫水 및 水分攝取의 生理的 效果

        This study aims at revealing the physiological responses of dehydration (DH) and rehydration (RH) in intermittent exercises at the work intensity of 70% HR max. First, with general student public, three times of exercise (each for 20 minutes) at the given work intensity were taken, with a break for 25 minutes respectively. Their heart rate, rectal temperature and plasma electrolyte concentration both DH and RH were measured. Second, in two groups of athletes and non-athletes, 9 rounds of exercise (for 100 minutes) and 8 breaks (for 60 minutes) were taken, with RH forced 10 times to compensate for the subjects' water loss. Their change in sweat loss and plasma electrolyte concentration as well as the IVDP(Intensive Voluntary dehydration Phenomenon) were experimented. The summary of this research follows. 1) Mean heart rate and rectal temperature in intermittent exercises were higher at DH than at RH, growing up at the increasing level of DH. 2) RH saw no change in the plasma concentration of Na+, K+ and Cl-, while DH observed a significant increase. 3) HCO₃made a significant decrease at DH before growing up. 4) Mg++ had no change, while Ca++ increased significantly at RH and DH. 5) Lactate showed an increase with significance at RH and DH, especially greater at DH. 6) Osmotic pressure increased meaningfully at DH. 7) Anion gap increased significantly at DH and RH. 8) On the other hand, when the forced RH was held, the group of athletes had greater sweat loss per body surface area than that of non-athletes. 9) The athletes' plasma concentration of Na+ and Cl- revealed a clear decrease. 10) There was seen no significant change in the plasma concentration of K+, Mg++ and Ca++ in both groups. 11) The IVDP occurred at the RH of 2.3ℓfor athletes and more than 2.5ℓfor no-athletes.

      • 施肥量 Yacon의 生育 및 收量에 미치는 效果

        This experiment was conducted to investigate the effect of the amount of fertilizer on the growth and yield characters of yacon(Polymnia sonchifolia POEPP) and to establish the cultivation method. An introduced variety of yacon was used. Seeding dates were from April 5 to April 7, and seedlings were transplanted on May 25. Planting density was 80㎝(width)×45㎝(distance). The minimal amounts of fertilizers of N-P-K were 3.5-5.0-3.5㎏/10a. And twice, three times and four times as much as the minimal amounts were applicated. The results were summarized as follows. 1. As the amounts of fertilizer were increased, plant height, leaf width, no. of leavesm no. of nodes, stem diameter and fresh weight of top part increased but leaf length was not different among treatments. 2. Tuber yield was highest in the 10.5-15.0-10.5㎏/10a treatment, whose total yield increased because of the heavy amount of fertilizer, and yield of commercial goods was highest in 7.0-10.0-7.0㎏/10a treatment(36.9%). 3. Plant height(r=0.973^**), leaf width(r=0.968^**), no. of leaves(r=0.994^**), and no. of nodes(r=0.949^**) had higher positive correlation with tuber yield, respectively.
