In order to find a relationship between firefly luciferases structure and bioluminescence spectra, we focus on excited substrate geometries which may be affected by rigid luciferases. Density functional theory (DFT) and time dependent DFT (TDDFT) were employed. Changes in only six bond lengths of the excited substrate are important in determining the emission spectra. Analysis of these bonds suggests the mechanism whereby luciferases restrict more or less the excited substrate geometries and to produce multicolor bioluminescence.

A novel non-viral gene delivery system, Procationic-Liposome-Protamine-DNA complexes (PLPD) which could further adsorb transferrin on the surface as a targeting ligand to form Tf-PLPD, was prepared and characterized before and after lyophilization. The size distribution of Tf-PLPD was in the range of $240{\pm}12nm$ and the zeta potential was $-24.10{\pm}2.5mV$. The transfection efficiencies of PLPD and Tf-PLPD were $12.18{\pm}3.8\;and\;24.26{\pm}2.6mU\;{\beta}-galactosidase/mg$ protein respectively. The lyophilization and the presence of serum didn't affect the tansfectivities of PLPD or Tf-PLPD. Compared to $Lipofectamine^{TM}$ 2000 (Invitrogen, U.S.A.), the procationic liposomes had less cytotoxicity to cells. In summary the procationic lipoplex described here, combining the condensing effect of protamine and the targeting capability of Tf, was a perspective non-viral vector for gene delivery system.

Weather Forecasting Using Ensemble of Spatial-Temporal Attention Network and Multi-Layer Perceptron

Weather forecasting is a challenging task, which is especially suited for artificial intelligence due to the large amount of data involved. This paper proposed an end-to-end hybrid regression model, called Ensemble of Spatial-Temporal Attention Network and Multi-Layer Perceptron (E-STAN-MLP), to forecast surface temperature, humidity, wind speed, and wind direction at 24 automatic weather stations in Beijing. Combining the data from historical observations with the data from the numerical weather prediction (NWP) system, our proposed model give better results than the NWP system or previously reported algorithms. Our E-STAN-MLP model consists of two parts. One is to use the spatial-temporal attention based recurrent neural network to model the time series of meteorological elements. The other is a simple but efficient multilayer perceptron architecture forecasts the regression value while ignoring time dependence. Results at each time stamp are integrated together using a step-wise fusion strategy. Moreover, we use a joint loss step integrating both the regression loss function and the classification loss function to simultaneously forecast the wind speed and direction. Experiments demonstrate that our proposed E-STAN-MLP model achieves state-of-the-art results in weather forecasting.

Background: The role of surgical resection for patients with single large (${\geq}5cm$) and/or multinodular (${\geq}2$) hepatocellular carcinoma (HCC) is still controversial. This systematic review was performed to evaluate the safety and efficacy of resection for patients with single large and/or multinodular HCC. Materials and Methods: Databases (the PubMed, Web of Science, Embase, and Cochrane databases) were systematically searched to identify relevant studies exploring the safety and efficacy of resection for single large and/or multinodular HCC, published between January 2000 and December 2014. Perioperative morbidity and mortality, overall survival, and disease-free survival of the resection group were calculated. In addition, these outcome variables were also calculated for the control group in the included studies. Results: One randomized controlled trial and 42 nonrandomized studies involving 9,580 patients were eligible for analysis. Eight (1,594 patients) of the 43 studies also reported the outcomes of transarterial chemoembolization (TACE). Although 51.4% of patients featured cirrhosis, 90.7% of them demonstrated Child-Pugh A liver function in the resection group. The median rates of morbidity (24.5%) and mortality (2.5%) after resection were significantly higher than that of TACE (11.0%, P<0.001; 1.9%, P<0.001). However, patients who underwent resection had significantly higher median one-, three-, and five-year overall survival (76.1%, 51.7%, and 37.4%) than those who underwent TACE (68.3%, 31.5%, and 17.5%, all P<0.001). The median 1-, 3-, and 5-year DFS rates after resection were 58.3%, 34.6%, and 24.0%, respectively. Conclusions: Although tumor recurrence after resection for patients with single large and/ or multinodular HCC continues to be a major problem, resection should be considered as a strategy to achieve long-term survival.

Background/Aims: Pyogenic liver abscess (PLA) is a serious, life threatening condition with a high mortality rate that represents a diagnostic and therapeutic challenge. The aim of this study was to collect demographic data and clinical, laboratory and microbiological characteristics of PLA patients treated between 2000 and 2010. We also aimed to collect information regarding our management experience of these cases. Methods: As a retrospective review, 47 patients with PLA in a tertiary referral center were examined to determine their demographic characteristics, clinical features, and laboratory, imaging, and microbiologic findings as well as the treatment outcome. Results: Cryptogenic PLA was the most frequently identified type of PLA, while benign biliary tract disease was the most frequently identifi able cause of PLA (18/47 patients; 38.3%). Leukocytosis and elevated alanine transaminase were common laboratory findings and were observed in 35 (74.5%) and 22 (46.8%) patients, respectively. Increased fibrinogen was also detected in 11 of 15 investigated cases (73.3%). Notably, infection-induced thrombocytopenia occurred in 8 patients (17%). Diabetes mellitus was associated with the occurrence of infection induced shock when compared to the non-diabetic group (p<0.05). Patients with two or more comorbid diseases had longer hospitalizations when compared to patients with one comorbid disease or those without comorbidities (p<0.001). The number of days needed to establish diagnosis was correlated with the length of hospitalization (p<0.001). The overall hospital mortality rate was 2.1% (1/47). Conclusions: Characteristics of PLA patients from the past 10 years are presented. The number of days needed to establish a PLA diagnosis was correlated with the length of the hospital stay. The hospital stay of PLA patients can be further improved by early diagnosis and effective treatments during the early stages of PLA progression. (Gut Liver 2011;5:221-227)
