        모기업 협력업체 산업안전보건관리에 대한 인식

        This study is planned to investigate the attitude toward the safety and health management of contractor comparry. Under the contract based production system, all of activities including safety and health management in the contractor company are depended upon the contract. How to make contract influence the worker's health of contractor company. Work's health of contractor company can be protected by efforts of company of contract-out and contractor company, especially their safety managers. The modelling of the effective safety and health management system for contractor company should consider the need of safety manager of each company and employer of contractor company. Data is collected from safety managers of 3 contract out companies as electronic and electrical manufacturing industry and 55 safety managers, 57 employers of their contractor companies using self administered survey with structured questionnaire. The most of all respondents want to support from the contract out companies. The most important items supported from contract out company is the information based on the safety information network between each company. Safety manager and employer of contactor company also itemized safety education and training in the supporting system from contract out company. These results can be generalized to survey on the other industries.

      • Hippocalcin increases phospholipase D2 expression through extracellular signal-regulated kinase activation and lysophosphatidic acid potentiates the hippocalcin-induced phospholipase D2 expression

        <P>We have previously isolated a 22 kDa protein from a rat brain which was found to be involved in activating phospholipsae D (PLD), and identified the protein as hippocalcin through sequence analysis. Nevertheless, the function of hippocalcin for PLD activation still remains to be resolved. Here, we proposed that hippocalcin was involved in extracellular signal-regulated kinase (ERK)-mediated PLD2 expression. To elucidate a role of hippocalcin, we made hippocalcin transfected NIH3T3 cells and showed that the expression of PLD2 and basal PLD activity were increased in hippocalcin transfected cells. We performed PLD assay with dominant negative PLD2 (DN-PLD2) and hippocalcin co-transfected cells. DN-PLD2 suppressed increase of basal PLD activity in hippocalcin transfected cells, suggesting that increased basal PLD activity is due to PLD2 over-expression. Hippocalcin is a Ca<SUP>2+</SUP>-binding protein, which is expressed mainly in the hippocampus. Since it is known that lysophosphatidic acid (LPA) increases intracellular Ca<SUP>2+</SUP>, we investigated the possible role of hippocalcin in the LPA-induced elevation of intracellular Ca<SUP>2+</SUP>. When the intracellular Ca<SUP>2+</SUP> level was increased by LPA, hippocalcin was translocated to the membrane after LPA treatment in hippocalcin transfected cells. In addition, treatment with LPA in hippocalcin transfected cells markedly potentiated PLD2 expression and showed morphological changes of cell shape suggesting that increased PLD2 expression acts as one of the major factors to cause change of cell shape by making altered membrane lipid composition. Hippocalcin-induced PLD2 expression potentiated by LPA in hippocalcin transfected cells was inhibited by a PI-PLC inhibitor, U73122 and a chelator of intracellular Ca<SUP>2+</SUP>, BAPTA–AM suggesting that activation of hippocalcin caused by increased intracellular Ca<SUP>2+</SUP> is important to induce over-expression of PLD2. However, downregulation of PKC and treatment of a chelator of extracellular Ca<SUP>2+</SUP>, EGTA had little or no effect on the inhibition of hippocalcin-induced PLD2 expression potentiated by LPA in the hippocalcin transfected cells. Interestingly, when we over-express hippocalcin, ERK was activated, and treatment with LPA in hippocalcin transfected cells significantly potentiated ERK activation. Specific inhibition of ERK dramatically abolished hippocalcin-induced PLD2 expression. Taken together, these results suggest for the first time that hippocalcin can induce PLD2 expression and LPA potentiates hippocalcin-induced PLD2 expression, which is mediated by ERK activation. J.       • Expression of Anti-Colorectal Cancer Monoclonal Antibody CO17-1A in Insect Cells

        Insect cell expression system using baculovirus has several benefits from high capacity, flexibility, safety to humans and glycosylation capability. Thus, the baculovirus insect cell system has been widely used for production of recombinant protein. In this study, the insect cell expression system was designed to produce anti-cancer mAb CO17-1A, which recognizes the antigen GA733 highly expressed on the surface membrane of colorectal carcinoma cells. The heavy (HC) and light chain (LC) genes of the mAb CO17-1A were cloned under the control of two different promoters, P10 and PPH, respectively, on baculovirus expression pFastBacTM Dual vector. The gene expression cassettes carrying HC and LC genes were transferred into a parent bacmid in E.coli (DH10Bac). The bacmid was transfected to Sf9 insect cells to generate the baculovirus expressing mAb CO17-1A. Western blot and immunoflouorescence analyses confirmed expression and secretion of mAb CO17-1A in the virus infected insect cells. The optimum condition for mAb expression was optimized at 24, 48 and 72hr after the virus infection with MOI (optimum virus concentration) ranging (0.2, 1 and 5). Expression of mAb CO17-1A in insect cells significantly increased at both 48 and 72hr after transfection with the MOI 1. HPLC chromatography revealed that the insect-derived mAb CO17-1A had insect specific glycan structures. Cell ELISA showed that the purified mAb from insect cell cultured media had a specific binding activity to SW948 human colorectal cancer cell. These results indicated that the baculovirus insect cell system is able to express, assemble, and secrete functional full size monoclonal antibody with insect specific glycosylation.

        Identification of Irradiated Granule-Type Ramen Soup Powder by Pulsed Photostimulated Luminescence and Thermoluminescence during Storage

        This study was carried out to establish a method for determining if granule-type Ramen soup powder has been irradiated. Thermoluminescence (TL) and pulled photostimulated luminescence (PPSL) were used as the detection methods through observed changes of TL and PPSL intensities after storage under differing conditions. PPSL intensities increased with increases in irradiation doses. The threshold level of PPSL was below 412$\pm$58 photon counts regardless of storage conditions (room and darkroom) after 10 months. TL intensities also increased with increasing irradiation doses. The coefficients ($R^2$) of PPSL (0.74~0.94) and TL intensities (0.92~0.58) were very highly correlated with irradiation dose. The PPSL and TL intensities were decreased after 10 months of storage. These results indicate that discrimination of irradiated from non-irradiated granule-type Ramen soup powder is possible using TL and PPSL methods despite the decrease in intensities of TL and PPSL with increasing storage times.

        한국판 Hamilton 우울증 평가 척도의 신뢰도, 타당도 연구

        Objectives : The reliability and validity of the Korean version of Hamilton Depression Rating Scale (K-HDRS) were examined in Korean patients depressive symptoms. Methods : 33 inpatients and 70 outpatients diagnosed as major depressive disorder or depressive episode of bipolar I disorder according to the DSM-IV criteria were assessed with K-HDRS, Clinical Global Impression score (CGI), Beck Depression Inventory (BDI) and Montgomery-Aberg Depression Rating Scale (MADRS). Results : Internal consistency (Cronhach's alpha coefficeint=0.76) and interrater reliability (r=0.94, p<0.001) were Statistically significant. Principal axis factoring analysis revealed 4 factors that accounted for 50.4% of the total variance. The Correlations of K-HDRS with CGI, BDI and MADRS were 0.84, 0.54, 0.58 respectively. Conclusion : These results showed that the K-HDRS could be a reliable and valid tool for the assessment of depressive Korean patients. The K-HDRS will be a useful tool for assessing depressive symptoms in Korea.

      • Role of phospholipase D2 in anti-apoptotic signaling through increased expressions of Bcl-2 and Bcl-xL

        <P>We have previously reported that Fas-resistant A20 cells (FasR) have phospholipase D (PLD) activity upregulated by endogenous PLD2 overexpression. In the present study, we investigated how overexpressed PLD2 in FasR could generate survival signals by regulating the protein levels of anti-apoptotic Bcl-2 and Bcl-xL. To confirm the effect of PLD2 on Bcl-2 protein levels, we transfected PLD2 into wild-type murine B lymphoma A20 cells. The transfected cells showed markedly the increases in Bcl-2 and Bcl-xL protein levels, and became resistant to Fas-induced apoptosis, similar to FasR. Treatment of wild-type A20 cells with phosphatidic acid (PA), the metabolic end product of PLD2 derived from phosphatidylcholin, markedly increased levels of anti-apoptotic Bcl-2 and Bcl-xL proteins. Moreover, PA-induced expressions of Bcl-2 and Bcl-xL were enhanced by propranolol, an inhibitor of PA phospholydrolase (PAP), whereas completely blocked by mepacrine, an inhibitor of phospholipase A<SUB>2</SUB> (PLA<SUB>2</SUB>), suggesting that PLA<SUB>2</SUB> metabolite of PA is responsible for the increases in Bcl-2 and Bcl-xL protein levels. We further confirmed the involvement of arachidonic acid (AA) in PA-induced survival signals by showing that 1,2-dipalmitoyl-sn-glycero-3-phosphate (DPPA), PA without AA, was unable to increase Bcl-2 and Bcl-xL proteins. Moreover, PA notably increased cyclooxygenase (COX)-2 protein expression, and PA-induced expression of both Bcl-2 and Bcl-xL was inhibited by NS-398, a specific inhibitor of COX-2. Taken together, these findings demonstrate that PA generated by PLD2 plays an important role in cell survival during Fas-mediated apoptosis through the increased Bcl-2 and Bcl-xL protein levels which resulted from PLA<SUB>2</SUB> and AA-COX2 pathway.       • Development of polymeric gene delivery carriers: PEGylated copolymers of l-lysine and l-phenylalanine

        <P> Block copolymers consisting of poly(ethylene glycol) (PEG) and poly(amino acid)-based random copolymers were successfully synthesized by the ring opening polymerization of the N-carboxy anhydrides (NCA) of l-lysine and l-phenylalanine. The synthesized copolymers had a molecular weight of around 30,000 and contained l-lysine and l-phenylalanine residues with molar ratios of 10/0, 9/1, 8/2, 7/3 and 6/4. The complex formation of the copolymer and pCMV-luc plasmid DNA was confirmed by the gel retardation assay and zeta potential measurement. Complete neutralization was achieved at an N/P ratio of more than 1.0 and the size of the complex was determined to be around 150 nm by dynamic light scattering. The cytotoxicity and transfection efficiency were tested on the HEK 293T cell line. The synthesized copolymers displayed negligible cytotoxicity, resulting in a cell viability of more than 95%, while those of the poly(l-lysine) (PLL) and poly(ethylenimine) (PEI) homopolymer were around 65 and 55%, respectively, under comparable conditions. The introduction of the hydrophilic PEG is believed to reduce the toxicity of the copolymer, due to its enhanced biocompatibility, and to impart improved stability to the complex under physiological conditions. The transfection efficiency at the optimized charge ratio of 7 was dramatically improved as the molar content of the l-phenylalanine residues in the copolymers increased and reached a maximum value at an l-phenylalanine content of 30 mol%. The transfection efficiency of the PEGK7/plasmid DNA complex was around 80 times higher than that of PLL, despite the presence of neutral PEG as a block segment.</P>

      • Extremely small multimode-interference coupled triangular resonator with sharp angle of incidence

        <P>Novel triangular ring resonators combining extremely small multimode-interference (MMI) coupler, low loss total internal reflection (TIR) mirrors, and semiconductor optical amplifiers are reported for the first time. The MMI length of 90 microm is among the shortest reported. The incidence angle of the TIR mirror inside the resonator is 22 degrees. A free-spectral range of approximately 2 nm is observed near 1550 nm along with an on-off ratio of 17 dB. The triangular resonators with a sharp angle are very attractive components due to their promise of compact size and high levels of integration. Therefore, large numbers of resonators can be integrated on a chip to increase functionality in future optical wavelength division multiplexing system.</P>
