      • 민어(Miichthys miiuy)의 卵發生過程 및 仔稚魚 발육단계에 따른 형태발달

        본 연구는 2000년 8월 19일 전라남도 신안군 임자도 연안에서 2각망으로 어획된 민어 어미를 인위적인 방법으로 수정하여 부화 사육하면서 관찰한 卵發生 過程 및 仔稚魚 형태변화는 다음과 같다. 1. 민어의 受精卵은 구형의 분리 부성난으로 알의 크기는 0.96~1.08㎜(평균 1.01㎜, n=50)였고, 크고 작은 油球가 1~5개(0.06~0.34㎜) 존재하였다. 2. 사육수온 25.2~28.4℃에서 受精 후 17시간 10분에는 배체가 卵膜을 뚫고 머리부터 孵化하기 시작하였다. 3. 孵化 직후 仔魚의 전장은 2.36~2.63㎜(평균 2.48㎜, n=10)로 전장의 45.0~47.0%에 달하는 큰 卵黃(장경 1.11~1.20㎜, 단경 0.92~0.96㎜)을 가지고 있으며 筋節은 7~8+ 17~19=24~27개였다. 4. 孵化 후 3일째 仔魚는 전장 3.98~4.32㎜(평균 4.16㎜, n=10)로 입은 열려있었고, 이 시기에 섭이활동이 활발하게 관찰되면서 卵黃이 완전히 흡수되어 後期仔魚期로 이행되었다. 5. 孵化 후 10일째 後期仔魚는 전장이 5.88~6.74㎜(평균 6.30㎜, n=10)로 꼬리의 말단이 굽어지기 시작하는 시기로 黑色素胞가 기저후두골 뒷부분의 체측에서 점모양 및 나뭇가지모양 으로 출현하였다. 6. 孵化 후 31일째 개체의 전장은 9.62~12.76㎜(평균 11.12㎜, n=10)로 모든 지느러미가 정수에 달하였고, 체형이나 반문이 成魚와 닮아 種魚期로 이행하였다. Egg development and morphological change of larvae of the brown croaker, Miichthys miiuy, were investigated after obtaining samples by artificial insemination. The spawning season extends from the middle to the end of August in the seas off Lim-ja Island, Cheollanam-do. The fertilized eggs were transparent, spherical, separate and they floated. Egg diameter ranged 0.96~1.08㎜ with several oil globules of 0.06~0.34㎜. Hatching occurred 17 hours 10 minutes after fertilization in 25.2~28.4℃ water temperature. The newly-hatched larvae were 2.36~2.63㎜(mean:2.48㎜, n=10) in total length(TL), had a large yolk, and 7~8+17~19=24~27 myotomes. Melanophores were distributed on the eye lens. Three days after hatching the larvae had attained 3.98~4.32㎜ in TL(mean:4.16㎜, n=10), the yolk sac was completely absorbed, and they had transformed into the postlarva stage. The larva reached the juvenile stage with all fins formed and with a complete set of fin rays(D.Ⅹ-28~31;A.Ⅱ-7~8;P. 21~23;C. 19~21;V. 25~26) on the 31st day after hatching. The juveniles were 9.62~12.76㎜(mean:11.12㎜, n=10) in total length. At this point the juvenile was similar to the adult in body form and pigmentation pattern.

        사업장 소음 폭로에 의한 일과성 역치 상승과 회복

        To determine the recovery time from noise-induced temporary threshold shift(TTS), a prospective field study was conducted at three worksites where workers are known to be exposed high level of noise. Subjects were selected according to answers on a questionnaire which inquired about otological history and previous noise exposure, including avocational, military and occupational exposures. After excluding employee with past otologic problems, recent exposure to high level noise, and under medications, total 92 employees participated in the study. Among 92 participants, complete consecutive audiometric examinations were carried out at 0∼2 hours, 5∼7 hours, 14∼16 hours after worktime noise exposure on 26 participants wearing hearing protectors and 22 participants wearing no protective devices. The difference between the hearing level 0∼2 hours after noise exposure and 5∼7 hours is statistically significant by paired t-test(p<0.01). The median recovery times calculated from the data of 22 participants wearing no protective hearing devices are 15.6 hours at 4000Hz, and 7.7 hours, 10.3 hours, 8.4 hours at 1000Hz, 2000Hz and 8000Hz respectively. These data suggest that when measuring the pure tone audiometry for noise exposed workers, at least 16 hours noise-free interval is required.

        In general, the viscosity of fluid increases as temperature decreases, and decreases as temperature increases. This phenomenon can be explained by decreasing mobility of molecules as temperature decreases. But, we found the opposite phenomenon that viscosity decreases as temperature decreases in particular mixed surfactant system. To analyze this phenomenon, various experiments have been proceeded such as composition (anionic, nonionic, zwitterionic surfactant), variation of zwitterion surfactants, structure of hydrophobic chains, structure of Hydrotrope, variation of pH. Conclusively, controlling the complex of zwitterion and anionic surfactant, we could control the viscosity of mixed surfactant system on variation of temperature. We analyzed this phenomenon theoretically, and then can complete "The technology of viscosity control by temperatre in micellar phase".

      • 임상재료에서 분리된 녹농균(綠膿菌)의 혈청형과 약제감수성(藥劑感受性)에 대하여

        Two hundred and fourty eight strains of Pseudomonas aeruginosa isolated from clinical materials at Department of Bacteriology in National Medical Center and Han-iI Hospital during January to November in 1973, were typed serologically by Hommos agglutination method utlizing a routine set of 13 standard sera. In addition, their susceptiblity to several kinds of antibiotics were determind. The following results were obtained; One hundred seventy eight strains (71,77%) were typable with an occurrence of type T5 in 41 strains (16. 58%), type T5, in 36 (14.52%), type T3, in 24 strains (9. 68%) and smalI numbers of strains were distributed in other types. Seventy strains (28. 23%) were nontypable. The rate of isolation of Pseudomonas by clinical meterials was shown as 49. 19% in ous, 16. 53% in sputum and 8. 87% in urine; the isolation rate of 1. 21-3. 15% was shown in other clinical meteraIs and the definite distribution rate could not be observed in the serotype by different materials. Majorities of strains used in this experiment of isolates were resistant to common antibiotics but Gentamycin and Carbenicillin, known relativeIy as sensitive antibiotics to Pseudomonas aeruginosa, were observed resistance of 2. 44-10. 5% and 16. 69-57. 8%. Moreover any particular relationship between serotype and the sensitivity of antibiotics was not identified.

      • 유도 허벅다리 걸기 동작시 주동근의 동원양상 분석

        The purpose of this study was to verify what major muscles are during Judo uchimata, and analyze major muscle type on recruitment during repeated uchimata. 16 university students(male) were participated in the exercise test. The subject were divided into two groups; expert(N=8) and non-expert(N=8). Electromyogram test was analyzed muscle type on recruitment during repeated uchimata by surface electrode. Major muscle was measured by Integrated EMC in pre-test while seoi-otoshi 3 times. The results of this study as follows: 1. In expert goop on Judo uchimata showed that major muscle of upper limb was measured Left Brachi oradialis, Left Trapezeius, Right Brachi oradialis, Left Deltoid, and major muscle of lower limb was Left Rectusfemoris, Left Castrocnemius. 2. Non-expert group on Judo Uchimata showed that major muscle of upper limb was measured Left Brachi oradialis, Left Trapezeius, Left Biceps brachii , Left Deltoid, and major muscle of lower limb was Left Rectusfemoris, Left Biceps femoris.

      • 관찰학습에 따른 아동의 운동수행에 관한 효과

        The present study purposed to examine the effects of children's learning of physical education activities in elementary school according to model type and to provide basic strategic materials for observational learning through presenting demonstrative models in the field of physical education for children. The participants in this study are 40 male sixth graders. They were assigned to the non model group, the skillful model observation group, the learning model observation group or the mixed model observation group at random, and 10 students to each group. The task used in this study was shooting, a basic motion in soccer, and the experiment was conducted at a mini soccer field suitable for children. The shooting motion pattern score was given for dribble shoot, pass short and penalty shoot, and the score ranged from 1 to 5 points. In this study, statistical processing used SPSS (Ver 12.0) for Windows, and AxB two way ANOVA with repeated measures on blocks was performed to see differences between the acquisition and retention stages, among the observation groups, and among the divisions, Statistics related to the research task were analyzed at significance level a=0.05, focused on main effects and interaction effects among the groups and among the blocks, and posttest on the significance of difference among the groups and among the blocks was performed using Tukey's HSD. The results of this study are summarized as follows. First, no statistically significant difference was observed in the score of shooting pattern at the acquisition stage and the retention stage between the control group bservation groups. Second, no statistically significant difference was observed in the score of shooting pattern at the acquisition stage and the retention stage according to model type. Third, in the children s learning of shooting as a physical education activity, the score of shooting pattern at the acquisition stage according to the type of model presented (no model, skillful model, learning model, mixed model) showed the tendency of increasing through the learning compared to that before the learning. That is, statistically significant difference was observed among the division at the acquisition stage but not at the retention stage.

      • 성남시 일부 성인의 건강지각과 건강행위에 관한 연구

        The purpose of this study was to investigate the health perception and health behavior of adults in Sung Nam area, and the correlation between this variable. The subjects were 354 adults who participated voluntary in health examination program. The instrument for this study were health perception scale and health practice performance scale. Health perception scale were developed by Ware(1976) and the health practice performance scale developed by Chang et al(1999). The results of tills study as follows: 1. Of a possible maximum score of 44 points on the 11 question about health perception, the mean score of health perception was 28.21±5.23. And of a possible maximum score of 26 points on the 13 question about health behavior. the mean score of health behavior was 17.58±2.27. So it was found that the level of health perception and health behavior of adults in Sung Nam area was low. 2. Health perception and Health behavior were showed statistically positive correlation (r=.184.p=.000). 3. According to the result of multiple regression analysis, health perception and general charactenstics(age, vocation, religion) predicted 13.9% of health behavior.

        Random Amplified Polymorphic DNA Typing에 의한 동물과 사람 유래 Vancomycin 내성 장구균의 유전학적 비교

        It has been recently reported the possibility in the transfer of antimicrobial resistant to other animals and humans. In particular, the occurrence of vancomycin resistant enterococci (VRE), which have been known as a principal antimicrobial resistant bacteria in humans, has been increased as a pathogen of nosocomial infections. And then animal VRE were suspected as an origin of human VRE. In this work, we investigated the relatedness between VRE from animals and humans through random amplified polymorphic DNA (RAPD) typing. The genetic relatedness of vanA, vanB, vanC-1 and vanC-2 genotypes was examined by RAPD typing, respectively. van4 genotypes have not been shown high genetic relationship each other, containing VRE from humans (n = 8) and chickens (n = 5). In vanB genotypes from humans (n = 3), 2 isolates were made up one cluster, shown 77.8% homology. Chicken isolates of vanC-1 genotypes (n = 11) were constituted 2 clusters and the homology of 2 clusters was 81.8% and 80.0%, respectively. In case of vanC-2 genotypes (n = 19), 2 isolates from pigs have been shown 76.9% homology. But, between VRE from animals and humans, the isolates of high genetic relationship could not be found and RAPD typing was a useful epidemiological method that could be confirmed the genetic relationship among VRE.

      • Immobilized metal ion affinity chromatography(IMAC)를 이용한 His-tagged B^30-homoserine 인슐린 전구체의 rapid one-step 분리 정제

        To purify His tagged B^30-homoserine insulin precursor from recombinant E. coli using a rapid single-step purification method, immobilized Ni^2+ -affinity chromatography which has been widely used to purify genetically fused to the interest protein was used. His tagged B^30-homoserine insulin precursor, molecular weight 9KDa, expression was accomplished by IPTG induction on pellet centrifuged of flask and batch broth and confirmed by SDS-PAGE either. It showed that recombinant protein is expressed as inclusion body. His-tagged B^30-homoserine insulin precursor which chelated with Ni^2+ metal ion was eluted lX Elution buffer, pH7.9 including 6M Urea with 1000mM imidazole. His tagged B^30-homoserine insulin precursor band was ascertained by SDS-PAGE with freeze dried powder obtained from lX Elution buffer extract.
