        Simulation Analysis of Cylinder Impact Performance Influenced by Shaft Sleeve Structure

        To study the cylinder motion characteristics, cylinder and shaft sleeve stress characteristics of cylinder stroke and return process influenced by different shaft sleeve structure, the cylinders with non-chamfering shaft sleeve, C0.3 chamfering shaft sleeve and R0.3 round corner shaft sleeve were used for simulation of cylinder stroke and return process. The results show that the piston velocity curves of three kinds of shaft sleeve are identical. Maximum stress of different shaft sleeves fluctuates and maximum stress elements of C0.3 and R0.3 shaft sleeve are similar. When cylinder strokes, maximum stress of cylinder occurs at the end of piston rod connected to the load; the stress of C0.3 shaft sleeve is the lowest. When cylinder returns, maximum stress of cylinder occurs at the shaft sleeve; the stress of C0.3 and R0.3 shaft sleeve differ little and are both lower than that of non-chamfering shaft sleeve. It provides a reference for structure optimization and service life improvement of the cylinder shaft sleeve.

        Layout Optimization Method of Railway Transportation Route Based on Deep Convolution Neural Network

        To improve the railway transportation capacity and maximize the benefits of railway transportation, a methodfor layout optimization of railway transportation route based on deep convolution neural network is proposedin this study. Considering the transportation cost of railway transportation and other factors, the layout modelof railway transportation route is constructed. Based on improved ant colony algorithm, the layout model ofrailway transportation route was optimized, and multiple candidate railway transportation routes were output. Taking into account external information such as regional information, weather conditions and actual informationof railway transportation routes, optimization of the candidate railway transportation routes obtained bythe improved ant colony algorithm was performed based on deep convolution neural network, and the optimalrailway transportation routes were output, and finally layout optimization of railway transportation routes wasrealized. The experimental results show that the proposed method can obtain the optimal railway transportationroute, the shortest transportation length, and the least transportation time, maximizing the interests of railwaytransportation enterprises.

        Law and Early Warning of Vertical Sluice Cluster Displacements in Soft Coastal Soil

        The vertical displacement, which is the product of natural sources and human activities, is the key factor affecting the sluice safety. This study provides a systematic approach used for analyzing the law and early warning of sluice cluster vertical displacements in coastal soft soil locations. Two important methods, including probability analysis and principal component analysis (PCA), are used to obtain the necessary information in this study. Among them, PCA is mainly used to identify the risk indices during vertical deformations of sluice cluster. As case studies, 27 sluices in a cluster in Northern Jiangsu Province's coastal area in China are chosen and 14 variables related to sluice uplift, settlement and differential settlement deformations are used. The PCA and additional evidence from the sluice deformation law are used to identify three variables as risk indices, including maximum differential settlement (MMDS), maximum cumulative vertical settlement (MCVS) and maximum cumulative vertical uplift (MCVU). This study divides the risk levels into five grades (i.e., Level 1 to Level 5) based on the selected risk indices and determines their risk thresholds based on the in-situ deformation data from 2010 to 2020. In general, the results demonstrate that the newly proposed approach exhibits an acceptable performance. However, the influence of epistemic and aleatory uncertainties on this study is worthy of further discussion in the future.

        Evaluation of Galactomannan Enzyme Immunoassay and Quantitative Real-Time PCR for the Diagnosis of Invasive Pulmonary Aspergillosis in a Rat Model

        Since there is no consensus about the most reliable assays to detect invasive aspergillosis from samples obtained by minimally invasive or noninvasive methods, we compared the efficacy of an enzyme-linked immunosorbent assay (ELISA) for galactomannan (GM) detection and quantitative real-time PCR assay (qRT-PCR) for the diagnosis of invasive pulmonary aspergillosis. Neutropenic, male Sprague-Dawley rats (specific pathogen free; 8 weeks old; weight, 200 ± 20 g) were immunosuppressed with cyclophosphamide and infected with Aspergillus fumigatus intratracheally. Tissue and whole blood samples were harvested on days 1, 3, 5, and 7 post-infection and examined with GM ELISA and qRT-PCR. The A. fumigatus DNA detection sequence was detected in the following number of samples from 12 immunosuppressed, infected rats examined on the scheduled days: day 1 (0/12), day 3 (0/12), day 5 (6/12), and day 7 (8/12) post-infection. The sensitivity and specificity of the qRT-PCR assay was 29.2% and 100%, respectively. Receiver operating characteristic curve (ROC) analysis indicated a Ct (cycle threshold) cut-off value of 15.35, and the area under the curve (AUC) was 0.627. The GM assay detected antigen in sera obtained on day 1 (5/12), day 3 (9/12), day 5 (12/12), and day 7 (12/12) post-infection, and thus had a sensitivity of 79.2% and a specificity of 100%. The ROC of the GM assay indicated that the optimal Ct cut-off value was 1.40 (AUC, 0.919). The GM assay was more sensitive than the qRT-PCR assay in diagnosing invasive pulmonary aspergillosis in rats.

      • Prognostic Significance of Annexin A1 Expression in Pancreatic Ductal Adenocarcinoma

        Annexin A1 is a 37-kDa calcium- and phospholipid-binding protein of the annexin superfamily considered to play an important role in tumorigenesis. However, associations with clinicopathological features in pancreatic ductal adenocarcinoma (PDAC) cases have yet to be fully defined. We therefore investigated the prognostic value of annexin A1 protein as a PDAC biomarker in 83 tumor and matched non-cancerous tissues or normal pancreas tissues. Expression was analyzed using real-time RT-PCR, Western blotting and immunohistochemistry. In non-tumor tissue, myoepithelial cells showed no or weak expression of annexin A1 while expression was strong and sometimes even located in the nuclei of endothelial cells in tumor tissue. High expression was significantly associated with advanced stage (P <0.05) and a worse overall survival (P <0.05). These results provide new insights to better understand the role of annexin A1 in PDAC survival, and might be relevant to prediction of prognosis and development of more effective therapeutic strategies aimed at improving survival.

        The Catechol-O-Methyltransferase Val158Met Polymorphism Contributes to the Risk of Breast Cancer in the Chinese Population: An Updated Meta- Analysis

        Purpose: Catechol-O-methyltransferase (COMT) enzyme plays acentral role in estrogen-induced carcinogenesis. Emerging evidencefrom association studies has revealed that the functionalVal158Met polymorphism (rs4680 G>A) of the Catechol-Omethyltransferasegene (COMT) has been implicated in susceptibilityto breast cancer in the Chinese population, while resultsof individual published studies remain inconclusive and inconsistent. To assess this association in the Chinese population, a meta-analysis was performed. Methods: Eligible studies weresearched on MEDLINE, Embase, Cochrane Library, China NationalKnowledge Infrastructure, and the Chinese BiomedicineDatabase. Odds ratios (ORs) with their corresponding 95% confidenceintervals (CIs) were pooled to assess the association betweenCOMT polymorphisms and the risk of breast cancer usingRevMan 5.2 and Stata 12.0 software. Results: The meta-analysisincluded 14 eligible studies, with a total of 4,626 breast cancercases and 5,637 controls. Overall, the COMT Val158Met polymorphism(rs4680 G>A) was significantly associated with an increasedrisk of breast cancer in several genetic models (A/A vs. G/G: OR, 1.59, 95% CI, 1.12–2.27; A/A vs. G/A+G/G: OR, 1.62,95% CI, 1.14–2.29; A vs. G: OR, 1.15, 95% CI, 1.00–1.32), and asubgroup analysis according to menopausal status showed thatthis association was especially evident among premenopausalChinese women (A/A vs. G/G: OR, 1.87, 95% CI, 0.99–3.54; A/Avs. G/A+G/G: OR, 1.94, 95% CI, 1.03–3.63). Conclusion: The resultsof this meta-analysis indicated that COMT Val158Met variantscontribute to breast cancer susceptibility in the Chinesepopulation, particularly among premenopausal women.

        Clinicopathologic and Prognostic Significance of the Zinc Finger of the Cerebellum Family in Invasive Breast Cancer

        Purpose: Five members of the zinc finger of the cerebellum (ZIC) family—ZIC1, ZIC2, ZIC3, ZIC4, and ZIC5—have been shown to be involved in various carcinomas. Here, we aimed to explore the clinicopathologic and prognostic roles of ZIC family members in invasive breast cancer patients using immunohistochemical analysis, western blotting analysis, and real-time quantitative polymerase chain reaction (RT-qPCR). Methods: A total of 241 female invasive breast cancer patients who underwent radical mastectomy between 2009 and 2011 were enrolled. ZIC proteins in 241 pairs of breast tumors and corresponding normal tissues were investigated using immunohistochemistry and the clinicopathologic roles of proteins were analyzed using Pearson’s chi-square test. Kaplan-Meier curves and Cox regression analysis were also used to analyze the prognostic value of the ZIC proteins. In addition, 12 pairs of fresh-frozen breast tumors and matched normal tissues were used in the western blotting analysis and RT-qPCR. Results: Only ZIC1 expression in normal tissues was obviously higher than that in tumors (p<0.001). On multivariate analysis, ZIC1 expression (in overall survival analysis: hazard ratio [HR], 0.405, 95% confidence interval [CI], 0.233–0.702, p=0.001; in disease-free survival analysis: HR, 0.395, 95% CI, 0.234–0.669, p=0.001) was identified as a prognostic indicator of invasive breast cancer. Conclusion: ZIC1, but not the other proteins, was obviously decreased in breast tumors and associated with clinicopathologic factors. Thus, ZIC1 might be a novel indicator to predict the overall and disease-free survival of invasive breast cancer patients.

        On-line Temperature Monitoring of the GIS Contacts Based on Infrared Sensing Technology

        Gas insulated switchgear (GIS) is widely used in the power systems, however, the contacts overheating of the inside circuit breaker or disconnector may be a potential cause of developing accidents. As the temperature of the contacts cannot be directly acquired due to existence of the metallic shield, an infrared sensor is adopted to directly measure the temperature of the shield and then the contacts temperature can be indirectly obtained by data fitting, based on which the on-line temperature monitoring technology specifically for GIS contacts based on infrared sensing is proposed in this paper. A real GIS test platform is constructed and experimental studies are carried out to account for the influential factors that affect the accuracy of the infrared temperature measurement. A heat transfer model of the GIS module is also developed, together with experimental studies, the nonlinear temperature relationship among the contacts, the metallic shield and the environment based on a neural network algorithm is established. Finally, an integrated on-line temperature monitoring system for the GIS contacts is developed for on-site applications.
