      • Seeing the World: Photoreaction and Photosensory Mechanisms of Cordyceps militaris

        Light is a necessary environmental factor for stroma formation and development of Cordyceps militaris, a well-known edible and medicinal fungus. In this study, photo morphogenesis and the blue-light receptor gene were studied using five representative strains of C. militaris. The results suggest that light was essential for colony pigmentation and could promote conidia production. Cmwc-1 gene inactivation results in thicker aerial hyphae, disordered fruit body development, a significant reduction in conidial formation, and carotenoid and cordycepin production. These characteristics were restored when the ΔCmwc-1 strains were hybridized with wild-type strains of the opposite mating type. A genomewide expression analysis revealed that there were 1042 light responsive genes in the wild-type strain and only 458 in the ΔCmwc-1 strain. Among five putative photoreceptors identified, Vivid, cryptochrome-1, and cyclobutane pyrimidine dimer photolyase are strongly induced by light in a Cmwc-1- dependent manner, while phytochrome and cryptochrome-2 were not induced. The transcription factors involved in the fungal light reaction were mainly of the Zn2Cys6 type. CmWC-1 regulates adenylosuccinate synthase, an important enzyme for adenosine de novo synthesis, which could explain the reduction in cordycepin production. Some G proteincoupled receptors that control fungal fruit body formation and the sexual cycle were regulated by CmWC-1, and the cAMP pathway involved in light signal transduction in N. crassa was not critical for the photoreaction in the fungus here. A transcriptional analysis indicated that steroid biosynthesis was more active in the ΔCmwc-1 strain, suggesting that CmWC-1 might switch the vegetative growth state to primordia differentiation by suppressing the expression of related genes.

        Development of Simple and Effective Dual-Readout Sensor Based on Gold Nanoparticles and Cadmium Telluride Quantum Dots for Cartap Analysis

        Cartap is among the most popular pesticides that are widely used to boost food production, however, its residues pose a great threat to human health and the environment. We presented simple and effective colorimetric sensor based on aggregation of gold nanoparticles (AuNPs) with appropriate diameter and fluorescent sensor via inner filter effect (IFE) of AuNPs on cadmium telluride quantum dots (CdTe QDs), respectively. Firstly, effect of diameter of AuNPs was investigated and the results indicated that AuNPs with diameter of 13 nm (AuNPs13nm) showed better sensitivity toward cartap. Thus, AuNPs13nm was chosen for colorimetric and fluorescent cartap assay. On one hand, mercaptoacetic acid (MA) was functionalized on AuNPs13nm for the improvement of anti-disturbance ability for discrimination study. A decent linear relationship for cartap was obtained in the range from 1 µM to 50 µM with detection limit of 1.32 µM for colorimetric readout; on the other hand, fluorescent sensor based on AuNPs13nm displayed excellent linear relationship with cartap concentration varying from 20 nM to 100 nM with low detection limit of 4.02 nM. And the above-mentioned sensor showed acceptable anti-disturbance ability for cartap discrimination. These results demonstrated that the sensor as-developed would be a great potential candidate for cartap detection in real application.

        Variations of SSU rDNA Group I Introns in Different Isolates of Cordyceps militaris and the Loss of an Intron during Cross-Mating

        Cordyceps militaris, the type species of genus Cordyceps, isone of the most popular mushrooms and a nutraceutical ineastern Asia. It is considered a model organism for thestudy of Cordyceps species because it can complete its lifecycle when cultured in vitro. In the present study, the occurrenceand sequence variation of SSU rDNA group I introns,Cmi.S943 and Cmi.S1199, among different isolatesof C. militaris were analyzed. Based on the secondary structurepredictions, the Cmi.S943 intron has been placed insubgroup IC1, and the Cmi.S1199 intron has been placedin subgroup IE. No significant similarity between Cmi.S943and Cmi.S1199 suggested different origins. Three genotypes,based on the frequency and distribution of introns, were describedto discriminate the 57 surveyed C. militaris strains. It was found that the genotype was related to the stromacharacteristics. The stromata of all of the genotype II strains,which possessed only Cmi.S943, could produce perithecium. In contrast, the stromata of all genotype III strains, whichhad both Cmi.S943 and Cmi.S1199, could not produce perithecium. Cmi.S1199 showed the lowest level of intra-specificvariation among the tested strains. Group I introns can belost during strain cross-mating. Therefore, we presumed thatduring cross-mating and recombination, intron loss could bedriven by positive Darwinian selection due to the energeticcost of transcribing long introns.

        Levonorgestrel-releasing intrauterine system-based therapies for early- stage endometrial cancer: a systematic review and meta-analysis

        Objective: To conduct a systematic review and meta-analysis of studies evaluating the oncological and fertility outcomes of early-stage endometrial cancer (EC) treated with the levonorgestrel-releasing intrauterine system (LIUS)-based regimens. Methods: The Meta-analyses Of Observational Studies in Epidemiology statement for meta-analyses was followed. Searches were conducted on MEDLINE, Embase, PubMed, Preprints, and the Cochrane Central Register of Controlled Trials from January 1990 to August 4, 2022. The Joanna Briggs Institute Critical Appraisal Checklist was used for quality assessment. The primary endpoint was the complete response (CR) rate and the secondary endpoints were relapse, pregnancy, and live birth rate. Results: A total of 25 studies (821 women) were included. The CR rate of LIUS-based regimens was 63.4% (95% confidence interval [CI]=52.3%–73.2%), with 29.6% (95% CI=23.3%–36.8%) of cases experiencing recurrence during follow-up. In sensitivity analyses, patients younger than 45 years of age with a body mass index <30 kg/m2 who were treated with LIUS-based regimens achieved a high CR rate of 84.6% (95% CI=80.3%–88.1%) over a median follow-up of more than 24 months. Overall pregnancy and live birth rates were 37.9% (95% CI=24.1%–53.9%) and 39.3% (95% CI=24.0%–57.0%), respectively. No statistical differences were apparent in CR or relapse rates among the LIUS+GnRH agonist, LIUS+oral progesterone, or hysteroscopic resection followed by LIUS subgroups. Conclusion: LIUS-based therapies are viable for the conservative management of early-stage endometrioid EC on CR and fertility outcome.
