      • 잡초성벼의 superoxide dismutase cDNA cloning과 재배벼로의 형질전환

        냉해나 한발등의 환경스트레스에 대해 저항성을 유발하는 유전자를 환경스트레스에 강한 잡초성벼로부터 선발하고 이들 유전자를 재배벼에 도입하여 도입유전자 산물의 과량 발현을 통해 냉해나 한발 등에 대한 저항성이 향상된 벼를 선발하고자 하였다. 잡초성벼인 Bhutan 14Ad로부터 냉해 및 한발 저항성 유전자로 알려진 superoxide dismutase (SOD) cDNA를 분리하고자 mRNA를 분리하고 이 분리된 mRNA를 이용해 reverse transcriptase PCR방법으로 SOD cDNA를 cloning 하였다. 그 결과 2종의 SOD cDNA가 cloning되어 SOD-A, SOD-B로 명명하였다. 이들 cDNA의 염기서열을 결정한 결과 이들은 아미노산 서열 상동성이 88.4%를 나타내었으며, SOD-A는 oryza sativa 계열의 Cu/Zn SOD유전자인 GenBank accession No. L36320와 99.3%로 동일하였으며, SOD-B는 accession No. D01000과 100% 동일하였다. 이들 SOD-A와 SOD-B cDNA를 재배벼인 낙동벼에 형질전환하여 형질전환체 벼를 선발하였으며, 이들 형질전환체 벼의 냉해저항성 및 한발저항성 검정을 통해 저항성이 향상된 형질전환체 벼를 선발하고 있다. Two different cDNA clones for superoxide dismutase (SOD) were isolated from an weedy rice variety (Oryza sativa, cv. Bhutanl4Ad) and were introduced into a cultivated rice variety (Oryza sativa, cv. Nakdong) in order to develop the environmental stress-resistant rice plants. Sequence analysis of the cloned cDNAs indicated that the deduced amino acid sequence of SOD-A is 88.4% identical to that of SOD-B. Furthermore, the nucleotide sequence of SOD-A is 99.3% identical to that of a Cu/Zn SOD gene of Oryza sativa (GenBank accession No. L36320). The nucleotide sequence of SOD-B was identical to that of the previously published SOD gene (Accession No. D01000). A cultivated rice variety, Nakdong-byeo, was transformed with chimenc SOD genes containing a actin promoter of rice and pin2 terminator using a particle bombardment technique. Transformed calli were selected on an selection medium containing phosphinothricin (PPT). Transgenic rice plants were regenerated from the PPT-resistant calli. PCR analysis with genomic DNAs from transgenic plants revealed that transgenes are introduced into rice genome.

        Silaethene Cl₂Si=CHCH₂?Bu 1, generated as a reaction intermediate by the thermal elimination of LiX from Cl₃SiCHLiCH₂?Bu 2, combines with ene-component containing compounds(propene, 2-methylpropene, DMB) to give 40∼90% yield ene-products. They can be isolated and spectroscopic identified.

      • 온산만의 퇴적물과 총알고둥(Littorina brevicula)내 중금속 분포

        To investigate the distribution of heavy metal concentrations in Onsan bay, concentrations of cadmium, copper, zinc and lead in surface sediments and the periwinkles (Littorina brevicula) were determined in February and July, 1996. The ranges of cadmium, copper, zinc, and lead concentrations in the surface sediments were 0.08∼3.72, 3∼307, 49∼1273 and 15∼399㎍/g dry weight, respectively. The decrease of concentration in heavy metals with distance from Daejeong stream indicated that this stream is the pollutant source of heavy metals in Onsan Bay. The concentrations of cadmium, copper, zinc and lead in L. brevicula were 0.11∼11.81, 21∼212, 30∼96 and 0.26∼4.12㎍/g dry weight in February and 1.01∼24.9, 66∼325.4, 54∼225 and 0.68∼8.41㎍/g dry weight in July, respectively. These concentrations in L. brevicula were the significantly different(p<0.001) with the season of sampling. However, overall distribution of concentrations of heavy metals in L. brevicula had very similar tendency that concentrations of heavy metal decreased with the distance from the pollutant source irrespective of the two seasons. Therefore, L.brevicula is considered as a useful indicator for heavy metals pollution. According to analysis of the organs, tissues such as the digestive gland, gill and viceral mussel accumulate high concentrations of heavy metals.

        Y₁_(x)Ca_(x)Ba₂Cu₃O_(7-8) powder was synthesized by a coprecipitation method. This process is very easy to control the chemistry of the cation's solutions and coprecipitation with oxalic acid. Starting materials Of nitrates such Y(NO₃)₂ · 6H₂0, Ca(NO₃)₂ · 4H₂0, Ba(NO₃)₂and Cu(NO₃)₂ · xH₂O were dissolved into distilled water. Each solution was mixed and pH-controlled with oxalic acid The best condition of coprecipitation was pH 1.3 at room temperature. The yield of precursor was up to 92%. After calcining at 850℃ in air, 123 single phase was obtained. The size of 123 phase powder was about 90nm.

        Two saturated and four unsaturated glycerol ethers : 3-hexadecyloxy-1, 2-propanediol (chimyl alcohol)(1), 3-octadecyloxy-1,2-proapnediol (bathyl alcohol)(2), 3-dodecenyloxy-1,2-propanediol(3), 3-tridecenyl-1,2-propanediol(4), 3-tetradecenyloxy-1,2-propanediol(5), 3-hexa-decenyloxy-1,2-propanediol(6) were isolated from marine mollusca Fusitriton oregonensis from Samchuck of the East cost of Korea. Their structure were identified from ^1H-, ^13C-NMR, FT-IR and GC-MS.

        한국 남해안 소흑산도에서 채집한 미동정된 해면(동정을 의뢰중임)으로부터 일반적인 tetradecanoic, hexadecanoic methyl ester 이외에 탄소수가 홀수인 pentadecanoic, heptadecanoic와 가지가 달린 12-methyltetradecanoic, 14-methylpentadecanoic, 15-methylhexadecanoic, 14-methylhexadecanoic methyl ester가 발견되었으며, 주 sterol 성분으로는 cholesterol과 소량의 ergost-25-ene-3,5,6-triol이 검출되었다. The Korea sea unknown marine sponge was shown to contain odd-numbered pentadecanoic, heptadecanoic, and branched 12-methyltetradecanoic, 14-methylpentadecanoic, 15-methylhexadecanoic, 14-methylhexadecanoic methyl ester, in addition to tetradecanoic and. hexadecanoic methyl ester. Futher elution with more polar solvent gave 5α-cholest-3β-ol along with minor amount of ergost-25- ene-3,5,6-triol.

      • 해면 Hymeniacidon sinapium의 Lipid 성분 연구

        일반적으로 해양 생물에는 육지 생물에 비해 훨씬 다양하고 특이한 구조의 sterol과 지방산이 나타나고 있다. 본 연구에서는 한국산 해면(Hymeniacidon sinapium)으로부터 2개의 sterol 성분을 얻었는데 주 sterol 성분은 탄소가 27개인 5α-Cholestan-3β-ol였으며 cholesterol도 소량 발견되었다. 또한 팔미틴산, 팔미토레인산, 올레인산, 스테아린산, 리놀레인산, arachidonic acid ethyl ester가 발견되었다. 이외에도 dioctyl phthalate, bis(2-ethylhexyl) adipate가 검출되었는데 이것은 일반적으로 흔히 사용되는 plastic의 가소제이기 때문에 순수한 대사물질로 보기는 어렵고 해양 오염에 인한 것으로 추정된다. There has been a continuing interest in the sterols and fat related compounds of marine organisms. The most exciting results of recent studies have been the characterization of polyunsaturated fatty acids and a host of novel sterols, many with unique alkylation patterns in the side chain. The Korean Sea marine sponge Hymeniacidon sinapium was shown to contain 5α-Cholestan-3β ol along with minor amount of cholesterol. Further elution with more polar solvent gave palmitic, palmitoleic, oleic, stearic, linolenic, and arachidonic acid ethyl ester. Dioctyl phthalate and bis(2-ethylhexyl) adipate were also identified from the extracts of Hymeniacidon sinapium, but they may be not de nevo metabolites, rather from the contaminants from plasticizer.

        Single phase induction motor(SPIM) is one of the most widely used type of low power AC motors in the world, especially for domestic or commercial applications where a three phase power supply is not available. Fractional horse power single phase induction motors have no starting torque their own. So there are several ways of starting single phase induction motors. The most common type is the starting capacitor installed in series with the auxiliary winding to increase the starting torque. Also, the auxiliary winding is disconnected once the speed of the motor reaches 70 to 80% of the rated speed. In the conventional systems, this function is conducted by a centrifugal switch. But the mechanical centrifugal switch has many problems such as switch malfunction. This paper presents the new methods of universal starting switch by digital controller to overcome these shortcomings of centrifugal switch.

      • 민챙이(Bullacta exatrata)의 화학성분 연구

        한국 서해 상낙월도에서 채집한 연체 동물인 Bullacta exatrata 로부터 통상적인 9-hexadecenoic ethyl ester 이외에 불포화된 5, 8, 11, 14-eicosatetraenoic, 7, 10, 13-hexadecatrienoic, 10-13-octadecadienoic ethyl ester 및 상당량의 glyceryl ether인 chimyl alcohol이 검출되었다. 주 sterol 성분인 cholesterol이었으며 지방산 ester 형태로도 소량 존재하였다. Unsaturated 5, 8, 11, 14-eicosatetraenoic, 7, 10, 13-hexadecatrienoic, 10, 13-octadecadienoic ethyl ester and a lot of chimyl alcohol were isolated along with common 9-hexadecenoic ethyl ester from marine mollusca Bullacta exatrata collected from sangnackworl island of the Korea sea. In addition, cholesterol and its fatty acid ester were obtained by futher elution in more polar solvent. Their structures were deduced from ^1H-and ^13C-NMR, GC-ms, FT-IR spectra.

        Renal ischemia is one of the most common causes of acute renal failure. Four factors related to the pathogenesis of acute renal failure are vasoconstriction, decreased glomerular filtration rate, tubular back leak of filtrate, and intratubular obstruction. The cellular response to renal ischemic insults include decreased content of adenosine trihosphate, lipid peroxidation induced membrane degradation, alteration in cellular pH, and calcium or phospholipase induced mitochondrial dysfunction. Much attention has been given to the role of increased cellular calcium as a pathogenetic contributor to cell injury during ischemia. Author studied the protective effects of calcium antagonists on cellular injury during renal ischemia in rat. To investigate the protective role of these agents, author measured the amount of malondialdehyde(MDA) and the enzyme activities of free radical scarvengers-superoxide dismutase(SOD), catalase and glutathione peroxidase from non-pretreated group and calcium antagonists pretreated groups in control, ischemia and reflow subgroups. The results were summerized as follows: 1) The amount of MDA in non-pretreated group was higher in the reflow compared with the control(<p<0.01). But, in all pretreated groups, there was no statistically difference in the amount of MDA. 2) The SOD activity in non-pretreated group was lower in both the ischemia and the reflow compared with the control (P<0.05). But, in both verapamil and trifluoperazine-pretreated groups, there was no statistically difference in the SOD activity. 3) Both catalase and glutathione peroxidase activities in non-pretreated group were lower in both the ischemia and the reflow compared with the control (P<0.05). But in all pretreated groups, there was no statically difference in both catalase and glutathione peroxidase activities. These results suggest that free radical mediated ischemic injury by renal artery clamp in rat can be protected by intraperitoneal pretreatment with calcium antagonists. As trifluoperazine has a protective effect in renal ischemia, the calcium activated calmodulin dependent enzyme may play a role in renal ischemic injury.
