        안식향이 인체상피암종세포에 미치는 효과에 관한 연구

        Objective : This study was done to examine the cytotoxicity of styrax benzoin (SB) on human oral epithelioid carcinoma cell line (KI3). Methods : The cytotoxic effect of SB was measured by MTT assay or XTT assay. The microscopic study was performed to observe the mophorlogical changes of KI3 cells. Results : The cytotoxic effect of SB on KI3 cells showed a significant increase at 50 uM or 100 uM SB by MTT assay or XTT assay compared with control after cells were treated with various concentrations of SB for 48 hours. MTT_(50) and XTTg were 419.9 uM and 375.5 uM, respectively. In microscopic study, KB cells exposed to SB showed both the decreased cell number and the morphological changes. Conclusion : From these results, SB has a midcytotoxicity by the showing the decrease of cell number and the morphological changes in KJ3 by MTT and XTT assays.

      • 배추(Brassica campestris L. var. pekinensis Makino)의 cDNA library 구축 및 상동성 비교

        Expressed sequence tag(EST) has a good value to discover a new gene or to study its structure. Some 20 ESTs were generated to obtain new genetic resources of chinese cabbage(Brassian campestris L. var. pekinensis Makino). Poly A+ RNAs were isolated from 10-day-old seedlings grown at 25℃ under the day light. cDNA gene bank was constructed using λ ZAP /cDNA synthesis /Giga Pack Gold Packing kit. About a million clones were able to obtain from the library. All the clones examined so far had insert DNAs. Nucleotide compositions of randomly selected clones were determined by the Sanger mthod. The DNA sequences were compared with those deposited in the GenePept and GenBank database to figure out nucleotide homologies. Two ESTs showed significant similarities to the enlisted sequences. They are chloroplast GADPH subunit of Arabidopsis thaliana and carbonic anhydrase of A. thaliana. The full DNA sequences of the two clones are being determined. The cDNA gene bank constructed. in this experiment will being used to isolate more genes induced by the light in the plant.

        太陰調胃湯이 白鼠의 肥滿症 및 誘導肥滿細胞에 미치는 效果

        太陰調胃湯은 四象體質中 太陰人에 적용되는 處方으로 그 구성은 주로 祛濕, 祛痰하는 藥物로 이루어져있다. 韓醫學에서 肥滿의 병리기전은 주로 濕痰이 체내에 전체된 것으로 보고 있는데 太陽調胃湯의 處方구성은 이와같은 體內濕痰을 제거하는데 알맞음을 볼 수가 있다. 이에 太陽調胃湯이 gold thioglucose와 高脂肪食餌로 誘發한 肥滿 마우스에 미치는 效果와 前指肪細胞의 增殖과 分化에 미치는 影響을 실험하고 관찰한 결과 다음과 같은 결과를 볼 수 있다. 1. 太陽調胃湯은 前脂肪細胞(3T3-L1 세포)의 增殖과 分化를 抑制한다. 2. 細胞內 脂質蓄積을 誘導하는 酵素와 中性脂質의 細胞內 蓄積을 抑制하는 效果가 認定된다. 以上의 結果로 보아 太陰調胃湯은 四象體質中 가장 肥濕하기 쉬운 太陽人의 肥滿治療에 有用할 것으로 思料된다. Effects of Taeyeumjoweetang on the obesity of mouse induced by gold thioglucose. It is researched to elucidate the effects of Taeyeumjoweetang on the obesity of mouse induced by gold thioglucose and the diifferentiation and growth of preadipocyte, 3T3-L1 cell. The result were as follows. 1. Taeyeumjoweetang extract improved the blood level of transaminase changed by the obesity of mouse induced by gold thioglucose. 2. Taeyeumjoweetang extract inhibited the increase of liver fat and body fat induced by the obesity of mouse induced by gold thioglucose. 3. Taeyeumjoweetang extract inhibited the increase of body weight induced by the obesity of mouse induced by gold thioglucose. 4. Taeyeumjoweetang extract inhibited the growth of undifferentiate preadipocyte 3T3-L1. 5. Taeyeumjoweetang extract showed the effects of inhibition on the differentiation of preadipocyte 3T3-L1. The above results suggested that the Taeyumjoweetang extract may be used on the obesity induced by the overgrowth and differentiation of adipocyte, and the accumulation of fat in liver and body.

        黃連의 Streptococcus Mutans 10449의 성장 및 pH변화에 미치는 영향

        연구배경 : 최근에는 의학, 약학, 생물학 등의 여러 분야에서 한의학, 특히 한약을 이용한 새로운 치료제의 개발에 많은 관심을 가지게 되었다. 한의학계에서도 한의학의 국제화, 또는 제재의 다양화를 위해서 이러한 노력을 하여야 할 때이라고 사료된다. 이러한 연구의 일환으로 구강세정 및 치아우식증을 예방하기 위한 한약제의 개발을 위하여 항미생물작용을 지닌 생약제가 구강내 세균에 미치는 영향을 연구하게 되었다. 방법 : 黃連抽出液을 제조하여 Streptococcus mutans 10449의 성장에 미치는 영향을 측정하기 위하여 Todd-Hewitt배지를 이용하여 균집락 형성에 미치는 영향을 측정하였다. 결과 : 黃連이 Streptococcus mutans 10449의 성장을 억제하는 효과를 보였으며 рH의 감소를 유발하여 산생성을 억제함을 알 수 있었다. 결론 : 위의 결과를 근거로 하여 黃連이 치아우식증의 새로운 구강세정제 및 치아우식증예방에 치료제로서 개발할 수 있을 것으로 생각된다. It has been reported that Coptidis Rhizama have an antibacterial effect. The purpose of this study is to know the growth rate &the рH change of Streptococcus mutans 10449 by Coptidis Rhizama. Growth rate was measured by optical densith and рH change was measured by рH meter. The growth of Streptococcus mutans 10449 in the absence of Coptidis Rhizama was more than that in the presence of Coptidis Rhizama irrespective of their concentration. The рH of Streptococcus mutans 10449 in the 10% of Coptidis Rhizama was less than that in the control and 1% of Copyidis Rhizama. These results suggest that Coptidis Rhizama might have an inhibitory effect of growth and acid production of S. mutans 10449. Further study is needed to know the effect onb human gingival fibroblast by Coptidis Rhizama.

      • APCVD법을 사용한 보론 포스파이드의 증착 및 특성에 관한 연구

        Boron Phosphide films were deposited on the glass substrate at the low temperature, 550℃, by the reaction of B₂H_6 with PH₃ using CVD. N₂ was employed as carrier gas. The reactant gas rates were 50 ㎖/min for B₂H_6, 50 ㎖/min for PH₃ and 1.5ℓ/min for N₂. The films were annealed for 1hour, 3hours in N₂ambient at 550℃. The deposition rate was 1000Å/min and the refractive index of film was 2.6. The measurement of XRD shows that the films have the preferred orientation of (1 0 1). The data of VIS spectrophotometer show 75.49% for as-deposited, 76.71% for 1hr-annealed and 86.4 % for 3 hrs-annealed. The data of AFM show 73Å for as-deposited, 88.9Å for 1hr-annealed and 220Å for 3hrs-annealed. Also, The data of the secondary electron emission rate(γ) s how 0.317 for as -deposited, 0.357 for 1hr-annealed and 0.537 for 3hrs-annealed. And, The film shows the surface deposited thin film, bu using FT-IR, has the O-H bond and this bond is greatly decreased as annealing.

      • 상표자산과 구매의도와의 관계에 관한 국제비교연구 : 아시아와 유럽의 의류시장을 중심으로

        본 연구는 상표자산의 구성요소에 대한 선행요인을 분석하고 이를 토대로 하여 상표자산 구성요소와 상표자산 영향요인간의 관계 그리고 상표자산 구성요소와 구매의도간의 관계를 규명하고 한국과 영국을 중심으로 비교하여 상표자산관리 방안을 도출해보고자 하였다 연구결과, 정보탐색은 제품지식에 정(+)의 영향을 미치며, 상표태도와 상표지식은 상표 충성도와 상표 가치에 정(+)의 영향을 미치는 것으로 나타났다 그리고 상표충성도와 상표가치는 구매의도에 정(+)의 영향을 미치는 것으로 나타났다. 정보원천의 경우, 한국에서는 제품지식에 정(+)의 영향을 미치고 있는 반면, 영국에서는 영향을 미치지 않는 것으로 나타났다. Brand equity is one of the most important concepts in business practice as well as in academic research. Successful brands can allow marketers to gain competitive advantage (Lassar et al.,1995), including the opportunity for successful extensions, resilience against competitors' promotional pressures, and the ability to create barriers to competitive entry (Farquhar, 1989). Branding plays a special role in service firms because strong brands increase trust in intangible products (Berry, 2000), enabling customers to better visualize and understand them. They reduce customers' perceived monetary, social, and safety risks in buying services, which are obstacles to evaluating a service correctly before purchase. Also, a high level of brand equity increases consumer satisfaction, repurchasing intent, and degree of loyalty. Brand equity can be considered as a mixture that includes both financial assets and relationships. Actually, brand equity can be viewed as the value added to the product (Keller, 1993), or the perceived value of the product in consumers' minds. Mahajan et al. (1990) claim that customer-based brand equity can be measured by the level of consumers’ perceptions. Several researchers discuss brand equity based on two dimensions: consumer perception and consumer behavior. Aaker (1991) suggests measuring brand equity through price premium, loyalty, perceived quality, and brand associations. Viewing brand equity as the consumer’s behavior toward a brand, Keller (1993) proposes similar dimensions: brand awareness and brand knowledge. Thus, past studies tend to identify brand equity as a multidimensional construct consisted of brand loyalty, brand awareness, brand knowledge, customer satisfaction, perceived equity, brand associations, and other proprietary assets (Aaker, 1991, 1996; Blackston, 1995; Cobb-Walgren et al., 1995; Na, 1995). Other studies tend to regard brand equity and other brand assets, such as brand knowledge, brand awareness, brand image, brand loyalty, perceived quality, and so on, as independent but related constructs (Keller, 1993; Kirmani and Zeithaml, 1993). Walters(1978) defined information search as, "A psychological or physical action a consumer takes in order to acquire information about a product or store." But, each consumer has different methods for informationsearch. There are two methods of information search, internal and external search. Internal search is, "Search of information already saved in the memory of the individual consumer"(Engel, Blackwell, 1982) which is, "memory of a previous purchase experience or information from a previous search."(Beales, Mazis, Salop, and Staelin, 1981). External search is "A completely voluntary decision made in order to obtain new information"(Engel & Blackwell, 1982) which is, "Actions of a consumer to acquire necessary information by such methods as intentionally exposing oneself to advertisements, taking to friends or family or visiting a store."(Beales, Mazis, Salop, and Staelin, 1981). There are many sources for consumers’ information search including advertisement sources such as the internet, radio, television, newspapers and magazines, information supplied by businesses such as sales people, packaging and in-store information, consumer sources such as family, friends and colleagues, and mass media sources such as consumer protection agencies, government agencies and mass media sources. Understanding consumers’ purchasing behavior is a key factor of a firm to attract and retain customers and improving the firm’s prospects for survival and growth, and enhancing shareholder’s value. Therefore, marketers should understand consumer as individual and market segment. One theory of consumer behavior supports the belief that individuals are rational. Individuals think and move through stages when making a purchase decision. This means that rational thinkers have led to the identification of a consumer buying decision process. This decision process with its different levels of involvement and influencing factors has been widely accepted and is fundamental to the understanding purchase intention represent to what consumers think they will buy. Brand equity is not only companies but also very important asset more than product itself. This paper studies brand equity model and influencing factors including information process such as information searching and information resources in the fashion market in Asia and Europe. Information searching and information resources are influencing brand knowledge that influences consumers purchase decision. Nine research hypotheses are drawn to test the relationships among antecedents of brand equity and purchase intention and relationships among brand knowledge, brand value, brand attitude, and brand loyalty. H1. Information searching influences brand knowledge positively. H2. Information sources influence brand knowledge positively. H3. Brand knowledge influences brand attitude. H4. Brand knowledge influences brand value. H5. Brand attitude influences brand loyalty. H6. Brand attitude influences brand value. H7. Brand loyalty influences purchase intention. H8. Brand value influence purchase intention. H9. There will be the same research model in Asia and Europe. We performed structural equation model analysis in order to test hypotheses suggested in this study. The model fitting index of the research model in Asia was X2=195.19(p=0.0), NFI=0.90, NNFI=0.87, CFI=0.90, GFI=0.90, RMR=0.083, AGFI=0.85, which means the model fitting of the model is good enough. In Europe, it was X2=133.25(p=0.0), NFI=0.81, NNFI=0.85, CFI=0.89, GFI=0.90, RMR=0.073, AGFI=0.85, which means the model fitting of the model is good enough. From the test results, hypotheses were accepted. All of these hypotheses except one are supported. In Europe, information search is not an antecedent of brand knowledge.         扶正抗癌湯의 抗腫瘍效果에 關한 實驗的 硏究

        腫瘍으로 인한 死亡率은 多樣한 名種 治療에도 不拘하고 循環器疾患 다음으로 2위를 차지하고 있다. 이에 著者는 韓醫學과 西醫學의 結合治療를 通해서 抗腫瘍藥物의 效果를 增進시키고 副作用을 줄이는 藥物의 開發을 爲하여 健脾 益氣 祛痰 補腎 杭癌效能을 갖고 잇는 扶正杭癌湯抽出液을 使用하여 Colony 形成抑制實驗, Sulforhoda-mine B(SRB) assay를 觀察하였고, 扶正抗癌湯抽出液과 抗癌化學療法劑인 Mitomycin C(MMC)를 Ehrlich carcinoma의 solid tumor와 ascites tumor에 各各 單獨 및 倂用 投與하여 抗腫瘍效果 및 Iysosomal enzymes의 活性에 대한 效果를 觀察하였다. 먼저 扶正抗癌湯抽出液을 Caki-1 cell, Hep 3B 및 A549에 투여한 후 增殖抑制作用을 colony 形成抑制實驗과 SRB assay를 통하여 觀察한 결과 濃度依存的으로 腫瘍細胞의 成長을 抑制하였다. Ascites form of Ehrlich carcinoma에 대한 抗腫瘍效果 實驗에서 扶正抗癌湯抽出液과 MMC를 병용투여한 결과, 扶正抗癌湯抽出液에 의하여 MMC의 抗腫瘍效果가 增加하였다. Solid form of Ehrlich carcinoma에 대한 抗腫瘍效果 實驗에서 扶正抗癌湯抽出液과 MMC를 병용 투여하였을 경우, MMC를 단독 투여하였을 경우보다 腫瘍의 크기가 현저하게 減少하는 결과를 보였다. Ehrlich ascites carcinoma cell에 扶正抗癌湯抽出液과 MMC를 병용 투여하였을 경우, MMC를 단독 투여하였을 경우보다 lysosomal enzymes의 活性이 强하게 나타났다. 本 實驗을 통하여 扶正抗癌湯抽出液은 MMC의 抗腫瘍效果를 증가시키는 效果를 보였다. 또한 扶正抗癌湯抽出液은 마우스에서 solid form of Ehrlich ascites carcinoma의 크기를 감소시키는 效果를 나타냈으며, 생존기간 實驗에서는 扶正抗癌湯抽出液과 MMC를 병용투여시 ascites tumor에 MMC를 단독으로 투여하였을 때와 비교하여 유의하게 생존기간을 증가시키는 결과를 보였다. 이상의 결과를 종합하여 볼 때 扶正抗癌湯抽出液은 用量에 依存的으로 直接的인 抗腫瘍效果가 있으며, 抗癌療法劑인 MMC의 效果를 亢進시키는 것으로 보아 間接的인 抗腫瘍效果도 있을 것으로 思科되나 正確한 效果를 究明하기 위해서는 보다 正確한 抗腫瘍機轉에 대한 硏究와 藥物學的 硏究 및 臨床的 硏究가 追加되어야 할 것이다. In order to investigate the effects of Bujeonghangamtang Extract(扶正抗癌湯抽出液) on antitumor effects after human cell lines(A549, Hep3B, Caki-1, Ehrlich) transplantation into the peritoneal cavity or right groin in mice induced by RPMI 1640 and DMEM etc, the extracts of its herbal medicines were orally administered for 10 or 12 days. Experimental studies were performed for measurement of antitumor effect of Mitomycin C(MMC) and lysosomal enzyme's activities using colony forming efficiency, SRB assay which were regarded as a valuable method for the measurement of antitumor effects of unknown compound on tumor cell lines. The results obtained in this studies were as follows: 1. The change of colony-forming efficiency and SRB assay of Caki-1 cells, Hep3B and A549 Cells after exposure to the extract of Bujeonghangamtang extract depressed the growth of tumor cells by concentration of Bujeonghangamtang. 2. Antitumor activity of the ethanol extract of Bujeonghangamtang extract and MMC on ascites form of Ehrlich carcinoma in mice is slightly improved. Especially the mean of survival times in the group of 200㎎/㎏ and MMC 0.1㎎/㎏ is improved over 30.9%. 3. When Bujeonghangamtang extract and MMC are administered together, the weight of tumor is more decreased than MMC alone. 4. The lysosomal enzyme's activities of the Bujeonghangamtang extract and MMC are more significantly improved than MMC alone. According to the above result, it could be suggested that Bujeonghangamtang extract has indirect antitumor effect by the increase of MMC uptake.

      • 산국 (Chrysanthemum boreale) 휘발성 추출액 및 EDTA가 미생물 및 인체 치은 섬유아세포에 미치는 영향

        Chrysanthemum boreale (C. boreale) has been used for the treatment of furuncles, carbuncles, sores, conjunctivitis, headache, vertigo and swollen throats as a folk medicine in Korea, but there is little report about experiment evidence of antimicrobial activity. To investigate the antifungal and antibacterial activity of Chrysanthemum boreale(C. boreale) volatile substance and EDTA, the growth inhibition against Aspergillus nidulans (A. nidulans), Fusarium oxysporum (F. oxysporum), Alternaria mali(Al. mali), Candida albicans (C. albicans), staphylococcus aureus (S. aureus), Streptococcus pyogenes(S. pyogenes), streptococcus mutans (S. mutans), Escherichia coli(E. coli), Helicobacter pylori (H. pylori) and Porphyromonas gingivalis (P. gingivalis) was monitored by agar dilution and broth microdilution method. The antifungal effect of C. boreale on A. nidulans, F. oxysporum, Al. mali and C. albicans was shown at the concentration of 0.04%. The antibaterial effect of C. boreale on S. aureus, S. pyogenes, S. mutans and H. pylori was shown at the concentration of 0.04%. However, the growth was inhibited at the concentration of 0.04% irrespective of the presence of EDTA 0.125% was much higher than higher than that of several concentration of volatile substance. The minimum inhibitory concentrations of volatile substance were 0.16% against S. aureus, 0.04% against S. pyogenes, 0.02% against S. mutans, 0.40% against E. coli, and 0.18% against C. albicans. The growth of human gingival fibroblasts was not affected by C. boreale volatile substances at the concentration of 0.04%. These results indicate that volatile substance from C. boreale may have biologically toxic activities to the microorganisms.

        Physico-chemical Properties of Lactobacillus casei 00692 during Fermenting for liquid-type Yogurt

        This study was carried out to find the physico-chemical attributes of yogurt base with Lactobacillus case; 00692 during 72 hr fermentation at $37^{\circ}C$. The pH decreased up to 44 hr and plateaued thereafter, and the titratable acidity increased up to 40 hr. The number of lactic acid bacteria sharply increased with $1.0\times10^7cfu/mL$ up to 48 hr of fermentation and slowly increased thereafter. The free amino acids produced during the fermentation reached the maximum value at 40 hr and gradually decreased thereafter. In the result of electrophoresis, the band was the thickest at 44 hr and mostly disappeared at 72 hr fermentation. The present data showed that the range of optimum fermentation time for yogurt base using Lactobacillus casei 00692 was from 40 to 44 hr.
