Twelve Suffolk${\times}$Small-tail-Han male sheep (body weight 21-26 kg), aged four months, were used to study the effects of volatile fatty acids (VFA) on IGF-I (insulin-like growth factor-I), IGFBP-3 (insulin-like growth factor binding protein-3), GH (growth hormone), insulin and glucagon in plasma, and IGF-I and IGFBP-3 in different tissues. The sheep were randomly divided into four groups with 3 sheep in each group. The sheep were sustained by total intragastric infusions and four levels of mixed VFA (the molar proportion of acetic acid, propionic acid and butyric acid was 65:25:10), which supplied 333, 378, 423 and 468 KJ energy/kg $W^{0.75}$/d, were infused into the rumen as experimental Treatments I, II, III and IV, respectively. The experiment lasted 12 days, of which the first 8 days were for pretreatment and the last 4 days for collection of samples. At the end of the experiment, blood samples were taken and then the sheep were slaughtered and tissue samples from the rumen ventral sac, rumen dorsal sac, liver, duodenum and Longissimus dorsi muscle were obtained. IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in different tissues were analysed. Results showed that the concentration of IGF-I, IGFBP-3, GH, insulin or glucagon in plasma and the content of IGF-I and IGFBP-3 in the rumen dorsal sac, rumen ventral sac, liver or Longissimus dorsi muscle were increased with VFA infusion level (p<0.05). No significant differences were found in duodenum IGF-I between Treatments I and II and in rumen dorsal sac IGFBP-3 between Treatments II and III (p>0.05). It was concluded that IGF-I, IGFBP-3, GH, insulin and glucagon in plasma and IGF-I and IGFBP-3 in rumen dorsal sac, rumen ventral sac, liver and Longissimus dorsi muscle were increased significantly with increasing level of ruminal infusion of mixed VFA.
The Impact Force of Large Boulders with Irregular Shape in Flash Flood and Debris Flow

The impact force of large boulders carried by flash floods and debris flows is one of the main causes of structural damage. The elastoplastic modification model of the impact force was derived, and it was found that the impact force was significantly affected by large boulders with irregular shapes. However, a large boulder with an irregular shape is often simplified as an isovolumetric sphere or ellipsoid, which may lead to inaccurate calculation of the impact force. In this paper, a method to obtain the irregular shape of a large boulder in the field is proposed by combining field investigation, image processing, and graphic analysis. The irregular shape is described by a nonuniform rational B-spline (NURBS) curve. The curvature radii corresponding to the potential impact contact points on the surface of a large boulder, which can reflect the influence of the irregular shape, are extracted according to the concavity and convexity analysis. The results demonstrate that NURBS curves can describe irregular shapes both conveniently and accurately. The impact force was corrected by the elastic–plastic model, the impact force increased with increasing curvature radius, and the increase ratio of the impact force gradually decreased with increasing velocity. Compared with the isovolumetric sphere model and ellipsoid model, the impact force calculated by the ellipsoid model is closer to the results obtained in this paper. The reduction factor of the impact force is 0.03 − 0.16, which first increases significantly and then linearly increases with increasing curvature radius. In addition, the reduction factor of the impact force initially exhibits a significant decline with increasing velocity and then gradually stabilises. To simplify parameter selection, we suggest using the maximum curvature radius in the ellipsoid model as the calculation parameter in calculating the impact force of large boulders.
Involvement of MicroRNA-198 Overexpression in the Poor Prognosis of Esophageal Cancer

Objective: This study aimed to investigate whether the miR-198 expression level is related to clinicopathological factors and prognosis of esophageal cancer. Methods: MicroRNA was extracted from esophageal cancer patients who underwent surgery for assessment using the Taqman@ MicroRNA assay. The correlation between miR-198 expression and clinicopathological features was analyzed, and the significance of miR-198 as a prognostic factor and its relationship with survival was determined. Results: MicroRNA-198 (miR-198) expression was higher in patients with poor prognosis than those with good prognosis (P<0.05). Kaplan-Meier analysis results showed that the miR-198 expression level had a significant correlation with survival time (P=0.030) and that patients with a higher expression of miR-198 had a shorter survival time. Cox multi-factor model analysis showed that patient prognosis (P=0.014), tumor length (P=0.040) and expression (P=0.012), and survival time had a significant correlation; the corresponding risks were 7.268, 1.246, and 3.524, respectively. Conclusion: miR-198 overexpression is involved in the poor prognosis of esophageal cancer and can be used as a biomarker for selection of cases requiring especial attention.
Thermogravimetric analysis and pyrolysis of waste mixtures of paint and tar slag

We describe thermogravimetric analyses and pyrolysis kinetic studies carried out on hazardous waste mixtures of tar slag, paint slag, paper, sodium sulfate and calcium oxide. Both thermogravimetric (TG) and differential thermogravimetric (DTG) profiles were measured by a thermogravimetric analyzer at different final temperatures, particle sizes and heating rates. Pyrolysis kinetic parameters were calculated by the Coats-Redfern method. Influences of particle size, heating rate and final temperature on pyrolysis yields and kinetic parameters are also discussed. The results show that final temperature and particle size have a great effect on pyrolysis yields. We find that with increasing temperature the activation energy initially increases to a maximum value and then decreases.

A cDNA encoding a cysteine protease inhibitor (CPI) was isolated from the cDNA library of clamworm Perinereis aibuhitensis Grube. The deduced amino acid sequence analysis showed that the protein had 51%, 48%, and 48% identity with Zgc: 153129 from Danio rerio, cystatin B from Theromyzon tessulatum, and the ChainA, stefin B tetramer from Homo sapiens, respectively. The gene was cloned into the intracellular expression vector pET-15b and expressed in Escherichia coli. The recombinant CPI (PA-CPI) was purified by affinity chromatography on Nicharged resin and ion-exchange chromatography on DEAE-Sepharose FF. The relative molecular mass of PACPI was 16 kDa as deduced by SDS-PAGE. Activity analysis showed that the recombinant protein could inhibit the proteolytic activity of papain. A constitutive and secretive expression vector was also constructed, and the cDNA encoding CPI was subcloned into the vector for extracellular expression. Western blotting analysis results showed that the PA-CPI was secreted into the medium. Bioassay demonstrated that E. coli DH5α harboring pUC18ompAcat-CPI showed a significant difference in mortality to the Asian longhorned beetle Anoplophora glabripennis compared with untransformed E. coli DH5α and control.

Liver cancer is one of leading digestive malignancies with high morbidity and mortality. There is an urgent need for the development of novel therapies for this deadly disease. It has been proven that asparagus polysaccharide, one of the most active derivates from the traditional medicine asparagus, possesses notable antitumor properties. However, little is known about the efficacy of asparagus polysaccharide as an adjuvant for liver cancer chemotherapy. Herein, we reported that asparagus polysaccharide and its embolic agent form, asparagus gum, significantly inhibited liver tumor growth with transcatheter arterial chemoembolization (TACE) therapy in an orthotopic hepatocellular carcinoma (HCC) tumor model, while significantly inhibiting angiogenesis and promoting tumor cell apoptosis. Moreover, asparagine gelatinous possessed immunomodulatory functions and showed little toxicity to the host. These results highlight the chemotherapeutic potential of asparagus polysaccharide and warrant a future focus on development as novel chemotherapeutic agent for liver cancer TACE therapy.
