        Bacillus sp. FF-7에 의한 항산화물질 생산조건과 항산화 활성

        발효식품 시료로부터 분리된 세균을 DPPH(α,α¹-diphenyl-β-picrylhydrazyl) 전자공여능으로 항산화 활성을 측정하여 가장 활성이 강한 균주를 선별하여 형태학적, 생화학적, 생리학적 특성 및 16S rRNA 염기서열을 조사한 결과 Bacillus sp.으로 판명되어 FF-7로 명명하였다. DPPH 전자공여능법에 의한 Bacillus sp. FF-7이 생산하는 항산화물질의 최적 생산 배지조건은 탄소원 2% galactose와 질소원 1% tryptone 첨가였다. Bacillus sp. FF-7에 의해 생성된 항산화 물질의 활성을 DPPH 전자공여능, 흰쥐 각 조직 microsomal 실험계 및 linoleic acid 과 산화지질 실험계에서 malondialdehyde를 thiobarbituric acid(TBA)방법으로 측정하였다. 흰쥐 각 조직 microsomal 실험계에서 지질과산화에 대한 항산화 효과는 뇌(97.50%)>심장(79.95%)>신장(77.84%)>비장(77.47%)>고환(69.96%)>간장(62.45%) 순이였다. Linoleic acid의 과산화지질률 TBA법으로 측정한 결과 반응 4일째까지 억제 효과가 강하게 나타났으며, 동시에 대조구로 사용한 0.05% BHT 첨가구에서도 실험종료시까지 항산화 활성이 강하게 나타났다. The antioxidative activity of antioxidative substances produced from several bacterial strains isolated from fermented foods were tested by DPPH (α,α'-diphenyl-β-picrylhydrazyl) free radical scavenging activity. One of the strains showing the highest antioxidative activity was identified as Bacillus sp. based on the morphological, biochemical, physiological characteristics, and 165 rRNA sequence, and named FF-7. The molt optimal medium condition for the production of antioxidative substance from Bacillus sp. FF-7 was 2% galactose as carbon source and 1% tryptone as nitrogen source. The antioxidative substance produced from FF-7 in these cultural medium was also tested by in vitro experimental models, the peroxidation of linoleic acid and the peroxidation of rat tissues microsomes by using thiobarbituric acid (TBA) for assay of free rnalondialdehyde production. The antioxidative avtivity against lipid peroxidation of rat tissues microsomes was shown in the following order; brain 97.50% > heart 79.95% > kidney 77.84% > spleen 77.47% > testis 69.96% > liver 62.45%. The antioxidative substance produced from FF-7 on linoleic acid peroxidation by TBA method was effectively inhibited during four days, and 0.05% BHT(butylated hydroxytoluene) used comparative control was also effectively inhibited. Results showed that the highest antioxidative activity by DPPH method of antioxidative substance produced from Bacillus sp. FF-7 was obtained by supplementing 2% galactose as carbon source and 1% tryptone as nitrogen source in cultured medium, this substance effectively inhibited the formation of TBARS in brain microsome in vitro system and in linoleic acid peroxidation.

        Serial Ultrasonographic Appearance of Normal Uterus during Estrous Cycle in Miniature Schnauzer Dogs

        Serial ultrasonography was performed to measure the normal appearance of uterine during estrous cycle and to determine whether the unterine appearance was related to the sex hormone, progesterone and estrogen. The uterine appearances, shape, diameter and echogenicity were daily monitored with ultrasonography in 9 Miniature Schnauzer dogs undergoing II estrous cycles. During proestrus and estrous, the uterus became hypoechoic but developed hyperechoic luminal echo. In the longitudinal view, the shape of the uterus occasionally changed from rectangular to coiled or serpentine, compared to other stages of the cycle. The diameter of the uterus during proestrus and estrous was larger (range: 0.600.86 mm) than other stages (range: 0.480.62 mm) of the cycle. The rising estrogen concentrations (range: 14.5116.86 pg/ml) in plasma during proestrus correlated with changes in the uterus (p<0.05). Progesterone concentrations were 0.080.15 ng/ml at the onset of proestrus, but rose 1.061.26 ng/ml at the end of proestrus. There was no relation to progesterone concentration from onset of estrus (p>0.05). There was dramatical changes in normal uterus and sex hormone during estrous cycle. Especially, the appearance, shape and diameter of uterus were related to plasma estrogen concentration during proestrus, correlated with other stages of the cycle.

        <P>Crinum asiaticum Linne var. japonicum has long been used as a rheumatic remedy, as an anti-pyretic and as an anti-ulcer treatment, and for the alleviation of local pain and fever in Korea and Malaysia. In order to investigate the possibility of Crinum asiaticum Linne var. japonicum extract as a cosmetic ingredient, we measured its anti-inflammatory effect by its inhibition of iNOS (inducible nitric oxide synthase) and the release of PGE2, IL-6, and IL-8. We also measured its anti-allergic effect by its inhibition of beta-hexosamidase release. An HPLC experiment after extraction with 95% EtOH at pH 3.5 showed that Crinum asiaticum Linne var. japonicum was mainly composed of lycorine (up to 1%), a well-known immunosuppressor. The content of lycorine varied, depending on the type of plant tissue analyzed and the extraction method. In an anti-inflammatory assay for inhibition of nitric oxide formation on lipopolysaccharide (LPS)-activated mouse macrophage RAW 264.7 cells, the ethanol extract of Crinum asiaticum showed an inhibitory activity of NO production in a dose-dependent manner (IC50 = 58.5 microg/ml). Additional study by RT-PCR demonstrated that the extract of Crinum asiaticum significantly suppressed the expression of the iNOS gene. Moreover, the extract of Crinum asiaticum did not show any cytotoxicity, but did show a cell proliferation effect against LPS (a 10 approximately 60% increase in cell viability). In an assay to determine inhibition of the H2O2-activated release of PGE2, IL-6, and IL-8 in human normal fibroblast cell lines, the release of PGE2 and IL-6 was almost completely inhibited above concentrations of 0.05% and 1%, respectively. Moreover, the release of IL-8 was completely inhibited over the entire range of concentration (>0.0025%). In order to investigate the skin-sensitizing potentials of the extract of Crinum asiaticum, a human clinical test was performed after repeated epicutaneous 48-h applications under an occlusive patch (RIPT). The repeated and single cutaneous applications of Crinum asiaticum Linne var. japonicum extract under the occlusive patch did not provoke any cumulative irritation and sensitization reactions. The result showed that the extract of Crinum asiaticum Linne var. japonicum has a sufficient anti-inflammatory effect. Therefore, Crinum asiaticum Linne var. japonicum extract may be useful for development as an ingredient in cosmetic products.</P>

        시스템 다이내믹스를 이용한 적정 법정 간호인력 산정에 관한 연구

        Purpose: The Purpose of the study is to lay foundation for reasonable legal nursing manpower staffing by building the nursing manpower-staffing model. Method: The System dynamics approach was adapted to build a simulation model for calculating reasonable legal nursing manpower-staffing. The model was built up on Vensim 5.0b DSS. Result: The results in the basic circumstance are in following. The level of nursing service quality, patient satisfaction, and nursing organization proficiency maintains higher and turnover intention also maintains lower at the tertiary hospitals with the ratio of bed to nurse 2.5:l-3.0:l than at any other situations. The level of nursing service quality, patient satisfaction, and nursing organization proficiency maintains higher, and turnover intention maintains lower at the secondary hospitals with the ratio of bed to nurse 3.0:l-3.5:l than at any other situations in the fundamental form. The results in the simulation circumstance are in following. As author raises the operating rate of bed into 95-100%, and cut skilled nursing staff down, the level of nursing service quality, patient satisfaction, and nursing organization proficiency drops slightly, and turnover intention also rises slightly. However with time the results show improvement. Conclusion: In terms of economic efficiency on tertiary hospitals, the ratio of bed to nurse for the best reasonable legal nursing manpower-staffing is 3.0:1(2.6:1 if calculating in terms of the ratio of patient to nurse). In terms of economic efficiency on secondary hospitals, the ratio of bed to nurse for the best reasonable legal nursing manpower-staffing is 3.5:1(3.0:1 if calculating in terms of the ratio of patient to nurse).

      • The Characterization of PLGA/Small Intestinal Submucosa Composites as Scaffolds for Intervertebral Disc

        <P>In order to application for the tissue engineered intervertebral disc (IVD), we designed the synthetic/natural hybrid scaffolds with poly(lactide-co-glycolide) (PLGA) and small intestine submucosa (SIS). SIS has been widely used as a biomaterial because SIS consists of various collagens and cytokines. SIS, however, possesses disadvantages such as their weak mechanical properties and uncontrolled degradation. Novel composite scaffolds of PLGA/SIS were manufactured by simple immersion method of PLGA scaffolds in SIS solution under vacuum. Then SIS was crosslinked. Also, PLGA scaffolds and SIS sponges were manufactured by solvent casting/salt leaching and freeze-dried methods, respectively. We evaluated pore structure, porosity, water absorption ability and cell viability of three types of scaffolds for the application of IVD.</P>

      • Functional characterization of a bacterial expansin from Bacillus subtilis for enhanced enzymatic hydrolysis of cellulose

        <P>Expansin is a plant protein family that induces plant cell wall-loosening and cellulose disruption without exerting cellulose-hydrolytic activity. Expansin-like proteins have also been found in other eukaryotes such as nematodes and fungi. While searching for an expansin produced by bacteria, we found that the BsEXLX1 protein from Bacillus subtilis had a structure that was similar to that of a β-expansin produced by maize. Therefore, we cloned the BsEXLX1 gene and expressed it in Escherichia coli to evaluate its function. When incubated with filter paper as a cellulose substrate, the recombinant protein exhibited both cellulose-binding and cellulose-weakening activities, which are known functions of plant expansins. In addition, evaluation of the enzymatic hydrolysis of filter paper revealed that the recombinant protein also displayed a significant synergism when mixed with cellulase. By comparing the activity of a mixture of cellulase and the bacterial expansin to the additive activity of the individual proteins, the synergistic activity was found to be as high as 240% when filter paper was incubated with cellulase and BsEXLX1, which was 5.7-fold greater than the activity of cellulase alone. However, this synergistic effect was observed when only a low dosage of cellulase was used. This is the first study to characterize the function of an expansin produced by a non-eukaryotic source.         Vaginal prolapse by ovarian follicular cysts in a female Jin-do dog

        A six-year-old, female Jin-do dog was referred for the recurrence of vaginal prolapse. Less than 7 months previously, the dog with the vaginal prolapse had been treated with hormone therapy because ultrasonography had identified a single follicular cyst in the left ovary. Three months after the first visit, the dog came into heat and the vaginal prolapse recurred. Ultrasonography showed multiple follicular cysts in both ovaries and radioimmunoassay detected a plasma estradiol-$17{\beta}$ concentration of 13.3 pg/ml. Treatment involved the repositioning of the vaginal prolapsed, ovariohysterectomy and the resection of the protruding tissue. The dog had been completely recovered two months later after the treatment.

        We developed the demineralized bone particle(DBP) impregnated poly(lactide-co-glycolide)(PLGA) scaffolds(PLGA/DBP) to investigate the effect of adhesion, growth, viability and inflammatory reaction of the cells. PLGA/DBP scaffolds were prepared by solvent casting/salt leaching method and were characterized by porosimeter, and scanning electron microscopy. NIH/3T3 fibroblast cells were cultured in the PLGA/DBP scaffolds as well as film and MTT assay was used to assess the viability of cells. Also, human promyelocytic leukemia cells(HL-60) were cultured in the PLGA/DBP scaffolds. We observed IL-1ß? and TNF-α expression of HL-60 cells seeded in PLGA/DBP scaffold by RT-PCR. NIH/3T3 fibroblast cell seeded on PLGA/DBP film were more adhere and spread with increasing DBP content due to increasing hydrophilicity and bioactivity. The fluorescence intensity of the band of TNF-α and IL-1ß? gene was decreased with increasing the concentration of DBP. It seems that the DBP affected on the improvement of physicochemical properties of PLGA such as biocompatibility, wettability and inflammatory response.
