        위도상사화(Lycoris uydoensis M. Kim)의 효율적인 기내 급속 대량 증식체계를 확립시키고자 잎과 인편을 배양하였다. 5℃에서 60일간 저온 처리한 모구를 구고를 중심으로 상부, 중부 및 하부로 구분하여 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA, 0.5 mg·L-1 NAA와 1.0 mg·L-1 BA 또는 3.0mg·L-1 IBA와 3.0 mg·L-1 BA 혼용처리된 MS 배지에 배양한 결과 식물생장조절물질과는 관계없이 디스크가 부착된 인편의 하부가 캘러스 및 자구 형성에 가장 효과적인 부위였다. 캘러스는 잎절편이나 인편조직에서 쉽게 형성되었으나 소자구 및 신초는 인편에서만 형성되어 기내번식에 적합한 절편체는 인편이었다. 자구 형성 및 신초분화는 배지내 첨가된 식물생장조절물질에 따라 치상체에서 직접 자구를 형성하거나 캘러스를 통해 식물체로 분화되는 2가지 경로로 형성되었는데 디스크가 부착된 인편을 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA, 0.5 mg·L-1 NAA와 1.0 mg·L-1 BA 또는 3.0 mg·L-1 IBA와 3.0 mg·L-1 BA 혼용처리된 MS 배지에 배양하여 30일 이내에 60%-100%까지 유도하였다. 다량의 자구형성 및 유지는 MS배지에 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA가 혼용되거나 또는 3.0 mg·L-1 IBA와 3.0mg·L-1 BA 혼용처리 되었을 때 0.5 mg·L-1 NAA와 1.0mg·L-1 BA 혼용처리 되었을 때 보다 효과적이었다. 6% sucrose는 자구형성 및 비대를 촉진시켰으며 특히 3.0 mg·L-1 IBA와 3.0 mg·L-1 BA 혼용처리배지에 6% sucrose를 첨가시키므로 0.1 mg·L-1 2,4-D와 1.0 mg·L-1 BA가 혼용 배지보다 구의 비대가 1-2배 증가하였다. 인편유래 캘러스로부터 기관분화 또는 인편에서 직접 분화된 자구에서 성공적으로 식물체가 재분화되었다. This study was conducted to establish an effective in vitro mass propagation system using leaf and bulb scale explant sources of Lycoris uydoensis M. Kim. For the bulb scale culture, the bulbs treated at 5℃ for 60 days were dissected into three parts according to bulb height (upper, middle, and bottom), and allowed for a culture on the MS medium with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA, 0.5 mg ? L-1 NAA plus 1.0 mg ? L-1 BA, and 3.0 mg ? L-1 IBA plus 3.0 mg ? L-1 BA. Regardless of plant hormone treatments, the bottom part of the bulb including the disk proved to be most effective in forming the callus and bulblets. Callus was readily produced on the young leaf segment and bulb scale tissues, whereas, bulblets and shoots were formed only on bulb scale explants. Bulb scales with disk appeared to the best optimal explant for in vitro propagation. Two distinct pathways, either organogenesis through callus or direct bulblet formation, could be recognized in bulb scale culture. Bulblet formation and shoot organogenesis from the bulb scales with disk were effectively induced up to the 60%~100% on the MS medium supplemented with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA, 0.5 mg ? L-1 NAA plus 1.0 mg ? L-1 BA, and 3.0 mg ? L-1 IBA와 3.0 mg ? L-1 BA within 30 days of culture. On MS media supplemented with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA or 3.0 mg ? L-1 IBA와 3.0 mg ? L-1 BA were more effective than 0.5 mg ? L-1 NAA plus 1.0 mg ? L-1 BA for induced and maintained a large number of bulblets. Six percent of sucrose enhanced bulblet formation and expanded the bulblets. Especially, adding 6% of sucrose on MS media supplemented with 3.0 mg ? L-1 IBA와 3.0 mg ? L-1 BA was one or two times of bulblets expanted than on MS mediun with 0.1 mg ? L-1 2,4-D plus 1.0 mg ? L-1 BA plus 6% of sucrose. Plantlet regeneration was successfully achieved from bulb scale-derived callus, via shoot bud induction or direct bulblet formation.

        전통누룩 진균류를 이용한 입국의 제조 및 입국곰팡이의 동정

        다양한 향미를 가진 막걸리의 개발을 위해 전통누룩으로부터 분리한 곰팡이로 입국을 제조한 후 품질특성을 분석하여 입국의 규격에 적합하며 이취가 없고 관능이 우수한 9균주를 입국 제조용 우수균주로 최종 선발하였다. 선발된 균주는 Aspergillus oryzae(C1-5-2-2, C20-7-3, CN1.3.1-4, CN16.19.1-1, N152-1, N220-1), Mycocladus corymbiferus (N162-2), Rhizopus oryzae(N20), Lichtheimia corymbifera (N21)로 동정되었으며, 제조한 입국의 산도는 5.0~6.8, 당화력은 128~241sp이었다. Various koji were prepared by fungi isolated from traditional nuruk and their quality characteristics were investigated. Acidity and saccharification power of their koji were ranged in 5.0~6.8 and 128sp~241sp. Nine fungi which were showed good quality and sensory evaluation were identified by analysis of their nucleotide sequences with PCR-amplified 18S rDNA internal transcribed spacer-1(ITS-1) and ITS-4 genes. Among them, six strains were identified as Aspergillus oryzae and the other strains were identified as Mycocladus corymbiferus, Rhizopus oryzae, Lichtheimia corymbifera.

      • Structural and functional studies of SAV0551 from <i>Staphylococcus aureus</i> as a chaperone and glyoxalase III

        <P>The DJ-1/ThiJ/<I>Pf</I>pI superfamily of proteins is highly conserved across all biological kingdoms showing divergent multifunctions, such as chaperone, catalase, protease, and kinase. The common theme of these functions is responding to and managing various cellular stresses. DJ-1/ThiJ/<I>Pf</I>pI superfamily members are classified into three subfamilies according to their quaternary structure (DJ-1-, YhbO-, and Hsp-types). The Hsp-type subfamily includes Hsp31, a chaperone and glyoxalase III. SAV0551, an Hsp-type subfamily member from <I>Staphylococcus aureus</I>, is a hypothetical protein that is predicted as Hsp31. Thus, to reveal the function and reaction mechanism of SAV0551, the crystal structure of SAV0551 was determined. The overall folds in SAV0551 are similar to other members of the Hsp-type subfamily. We have shown that SAV0551 functions as a chaperone and that the surface structure is crucial for holding unfolded substrates. As many DJ-1/ThiJ/<I>Pf</I>pI superfamily proteins have been characterized as glyoxalase III, our study also demonstrates SAV0551 as a glyoxalase III that is independent of any cofactors. The reaction mechanism was evaluated via a glyoxylate-bound structure that mimics the hemithioacetal reaction intermediate. We have confirmed that the components required for reaction are present in the structure, including a catalytic triad for a catalytic action, His<SUP>78</SUP> as a base, and a water molecule for hydrolysis. Our functional studies based on the crystal structures of native and glyoxylate-bound SAV0551 will provide a better understanding of the reaction mechanism of a chaperone and glyoxalase III.</P>

        On Asymmeticity for Power Transformed TARCH Model

        Zokian(1993) and Li and Li(1996) developed TARCH(Threshold ARCH) model, considering the asymmetries in volatility. The models are based on Engle(1982)'s ARCH model and Bollerslev(1986)'s GARCH model. However, two TARCH models can be expressed a common model through Box Cox Power transformation, which was used by Higgins and Bera(1992) for developing NARCH(nonlinear ARCH) model. This article shows the PTARCH(Power transformation TARCH) model is necessary in some condition, and it checks the fact that PTARCH model has better performance comparing estimates and RMSE(Root Mean Square Error) with those of Zakoian's TARCH model and Li and Li's TARCH model. PTARCH model would give contribution in asymmetric study as well as heteroscedastic study.

        CO-Dependent H<sub>2</sub> Production by Genetically Engineered <i>Thermococcus onnurineus</i> NA1

        <P>Hydrogenogenic CO oxidation (CO + H<SUB>2</SUB>O → CO<SUB>2</SUB> + H<SUB>2</SUB>) has the potential for H<SUB>2</SUB> production as a clean renewable fuel. <I>Thermococcus onnurineus</I> NA1, which grows on CO and produces H<SUB>2</SUB>, has a unique gene cluster encoding the carbon monoxide dehydrogenase (CODH) and the hydrogenase. The gene cluster was identified as essential for carboxydotrophic hydrogenogenic metabolism by gene disruption and transcriptional analysis. To develop a strain producing high levels of H<SUB>2</SUB>, the gene cluster was placed under the control of a strong promoter. The resulting mutant, MC01, showed 30-fold-higher transcription of the mRNA encoding CODH, hydrogenase, and Na<SUP>+</SUP>/H<SUP>+</SUP> antiporter and a 1.8-fold-higher specific activity for CO-dependent H<SUB>2</SUB> production than did the wild-type strain. The H<SUB>2</SUB> production potential of the MC01 mutant in a bioreactor culture was 3.8-fold higher than that of the wild-type strain. The H<SUB>2</SUB> production rate of the engineered strain was severalfold higher than those of any other CO-dependent H<SUB>2</SUB>-producing prokaryotes studied to date. The engineered strain also possessed high activity for the bioconversion of industrial waste gases created as a by-product during steel production. This work represents the first demonstration of H<SUB>2</SUB> production from steel mill waste gas using a carboxydotrophic hydrogenogenic microbe.</P>

        Acupuncture Treatment of Adhesive Capsulitis of the Shoulder: A Randomized Controlled Pilot Trial

        Background: Adhesive capsulitis (AC) is a common condition that includes shoulder pain and limited movement. Despite more than 100 years of AC treatment, the most efficacious treatment remains unclear. The aim of this study was to evaluate the feasibility of a randomized controlled trial (RCT) using acupuncture for AC. Methods: Thirty participants with AC were randomly assigned to acupuncture (A) or sham acupuncture (SA) groups. The participants received 15 acupuncture sessions over 6 weeks, and follow-up occurred for an additional 4 weeks thereafter. The primary clinical outcome was the numeric rating scale (NRS) for shoulder pain 6 weeks from the baseline. Secondary outcomes included range of motion (ROM) in the shoulder, the shoulder pain and disability index (SPADI), the EuroQol-5 dimensions (EQ-5D), the Pittsburgh sleep quality index (PSQI), and the patient global impression of change (PGIC). Results: Thirty participants were enrolled out of 37 screened individuals. Recruitment was conducted between August 2014 until May 2015. A total of 28 participants (93%) completed the 6-week intervention, and 26 participants (87%) completed the study. NRS, ROM, SPADI, EQ-5D, PSQI, and PGIC scores improved in both the experimental group and the sham group after 6 weeks, but the difference between the groups was not statistically significant. Adverse events were reported by 12 participants, although these events were not associated with acupuncture. Conclusion: A future RCT for AC may be feasible with some modifications to the recruitment plan and the secondary outcome measurement methods.

        Variation in Trichothecene and Zearalenone Production by Fusarium graminearum Isolates form Corn and Barley in Korea

        강원도와 남부지방에서 각각 옥수수와 보리를 채집하여 두 곡류로 부터 총 110 균주의 Fusatium graminearum을 분리한 후 쌀배지에서의 trichothecene과 zearalenone(ZEA)의 생성능을 측정하였다. 옥수수로 부터 분리한 51개의 F. graminearum 균주중 trichothecene 생성빈도는 deoxynivalenol(Don)이 64.5%, 3-acetyldeoxynivalenol (3-ADON)이 7.8%, 15acetyldeoxynivalenol (15-ADON)이 33.3%, nivalenol(NIV)이 21.6%, 4-acetylnivalenol (4-ANIV)이 13.7%였다. DON을 생성하는 균주들은 3-ADON보다는 15-ADON을 동시에 생성하였다. 한편, 보리로부터 분리한 59개의 F. graminearum 균주중 trichothecene 생성빈도는 NIV가 71.2%, 그리고 4-ANIV가 61.0%였으며, 한 균주만이 DON과 3-ADON을 동시에 생성하였다. 옥수수로부터 분리한 균주들의 ZEA의 생성빈도와 생성량은 각각 32.0%, 71㎍/g였으며, 보리로부터 분리한 균주는 각각 29%, 745㎍이었다. 이와같이 우리나라 강원도지방과 남부지방의 F. graminearum 균주들의 trichothecene 생성양상은 지역적인 큰 차이를 나타내었다. A total of 110 Fusarium graminearum isolates were obtained from corn and barley samples which were collected from Kangwon province and the southern part of Korea, respectively. The isolates were tested for trichothecene and zearalenone (ZEA) production in rice culture. The incidences of trichothecene production by 51 isolates of F. graminearum from corn were 64.7% for deoxynivalenol (DON), 7.8% for 3-acetyldeoxynivalenol (3-ADON),33.3% for 15-acetylde-oxynivalenol (15-ADON), 21.6% for invalenol (NIV), and 13.7% for 4-acetylnivalenol (4-ANIV). DON producers frequently co-produced 15-ADON rather than 3-ADON. On the other hand, the incidences of trichothecene production by 59 isolates of F. graminearum from barley were 71.2% for NIV, 61.0% for 4-ANIV, and only one isolate produced DON and 3-ADON. The incidences and mean levels of ZEA producers were 32.0% and 71.$\mu$g/g for the isolates from corn, and 29.0% and 74 .$\mu$g/g for the isolates from barley. There was a great regional difference in trichothecene production of F. graminearum isolates between Kangwon province and the southern part of Korea.
