        태음인(太陰人) 청심연자탕(淸心蓮子湯)이 Hydrogen Peroxide에 손상(損傷)된 백서(白鼠)의 대뇌신경세포(大腦神經細胞)에 미치는 영향(影響)

        1. 연구목적 본 실험은 청심연자탕이 대뇌신경세포의 산화적 손상에 대한 효능을 밝히기 위한 것이다. 2. 연구방법 신생 생쥐에서 분리 배양한 대뇌신경세포에 여러 농도의 hydrogen peroxide가 포함된 배양액에서 6시간 동안 처리하여 hydrogen peroxide가 배양 대뇌신경세포에 미치는 영향을 조사하였으며 hydrogen peroxide의 독성효과에 대한 청심연자당의 영향을 조사하였다. 3. 결과 및 결론 산소자유기인 hydrogen peroxide는 NR assay와 MTT assay에 의한 세포생존율을 감소시켰고 lipid peroxidation의 증가 및 LDH양의 증가에 의하여 생쥐의 배양 대뇌신경세포에 독성을 나타냈다. 청심연자탕(淸心蓮子湯)은 hydrogen peroxide의 산화적 손상에 대한 신경독성에 대하여 Iipid peroxidation의 감소에 유의한 효과를 보였다. 청심연자탕(淸心蓮子湯)은 hydrogen peroxide의 산화적 손상에 의한 신경독성에 대하여 LDH양의 감소에 유의한 효과를 보였다. 이상의 결과로 보아 hydrogen peroxide는 생쥐에서 분리한 대뇌신경세포에 산화적 손상에 의한 신경독성을 나타냈으며 청심연자탕(淸心蓮子湯)이 hydrogen peroxide와 같은 산소자유기의 산화적 손상에 대한 방어에 효과적인 것으로 사료된다. 1. Purpose : The purpose of this study was to determine the effects of Chungsimyeunjatang on the cerebral neurons injured by hydrogen peroxide($H_2O_2$). 2. Methods : I observed cell viability in mouse cerebral neurons exposed to hydrogen peroxide by NR assay and MTT assay and determined lipid peroxidation and amounts of LDH release in mouse cerebral neurons exposed to hydrogen peroxide. After administration of Chungsimyeunjatang water extracts, I observed significant changes of cell viability, lipid peroxidation and amounts of LDH release in mouse cerebral neurons exposed to hydrogen peroxide. 3. Results : Hydrogen peroxide showed neurotoxicity. Cell viability in mouse cerebral neurons exposed to hydrogen peroxide decreased in NR assay and MTT assay. Lipid peroxidation and amounts of LDH release in mouse cerebral neurons exposed to hydrogen peroxide increased. Chungsimyeunjatang was very effective in blocking hydrogen peroxide-induced neurotoxicity.

        태음조위탕(太陰調胃湯)이 Glucose Oxidase에 의해 손상된 대뇌피질(大腦皮質) 신경세포(神經細胞)에 미치는 영향(影響)

        1. 연구목적 본 실험은 태음조위탕이 대뇌신경세포의 산화적 손상에 대한 효능을 밝히기 위한 것이다. 2. 연구방법 신생생쥐에서 순수 분리한 대뇌신경세포(大腦神經細胞)를 배양(培養)하여 glucose oxidase (GO)에 노출시킨 후 이의 독성효과(毒性效果)를 측정(測定)하였으며, 또한 GO에 의하여 유도(誘導)된 독성(毒性)에 대한 태음조위탕(太陰調胃湯)의 효과(效果)를 조사(調査)하였다. 3. 결과 및 결론 산소자유기(酸素自由基)인 GO는 MTTassay에 의한 세포생존율(細胞生存率)의 유의한 감소(減少)를 보였다. 산소자유기(酸素自由基)인 GO는 NR assay에 의한 세포생존율(細胞生存率)의 유의한 감소(減少)를 보였다. GO의 산화적(酸化的) 손상(損傷)에 의한 신경독성(神經毒性)에 대하여 태음조위탕(太陰調胃湯) 투여로 총단백질양의 유의한 증가(增加)를 보였다. GO의 산화적(酸化的) 손상(損傷)에 의한 신경독성(神經毒性)에 대하여 태음조위탕(太陰調胃湯) 투여로 lipid peroxidation의 유의한 감소(減少)를 보였다. 이상(以上)의 실험결과(實驗結果)는 태음조위탕(太陰調胃湯)이 대뇌신경세포(大腦神經細胞)의 산화적손상(酸化的損傷)에 대하여 유의성(有意性) 있는 방어적(防禦的) 작용(作用)을 나타낸 것으로, 이와 관련된 노화억제(老化抑制)나 뇌허혈(腦虛血), 다발성경화증(多發性硬化症), 치매 등 뇌기능손상(腦機能損傷)이나 저하(低下)로 유발되는 질환(疾患)에 대한 임상응용(臨床應用)에 대해서도 지속적(持續的)인 연구(硏究)가 필요(必要)하다고 사료(思料)된다. 1. Purpose : The purpose of this study was to determine the effects of Taeyeumjoweetang on the cerebral neurons injured by glucose oxidase(GO). 2. Methods : I observed cell viability in mouse cerebral neurons exposed to glucose oxidase by NR assay and MTT assay and determined lipid peroxidation in mouse cerebral neurons exposed to glucose oxidase. After administration of Taeyeumjoweetang water extracts, I observed significant changes of cell viability, lipid peroxidation in mouse cerebral neurons exposed to glucose oxidase. 3. Results : GO induced cell degeneration such as the decrease of cell viability was measured by MTT and NR assay in the cultured mouse cerebral cortical neurons. Taeyeumjoweetang was effective in the increase of total protein of neurons inhibited by GO. Taeyeumjoweetang was effective in the decrease of lipid peroxidation of neurons produced by GO.

        수종 한약재가 혈관에 미치는 영향

        Rhizoma Arisaematis, Lignum Akebiae, Rhizoma Zedoariae, Cortex Eucommiae, Folium Perillae, Radix Sophorae Subprostratae, Radixi, Radix Ledeboutriellae, Rhizoma Atractylodis, Herba Ephedrae, Radix Puerariae and Radi Aconitx Bupleuri have been used in Korea for many centuries as a treatment for various disease. The purpose of the present study is to determine the effect of several herbs on norepinephrine(NE) induced blood vessel contraction in rabbits and pigs. Rabbit(2 kg, male) were killed by $CO_2$ exposure and a segment (8-10mm) of each rabbit was cut into equal segments and mounted in a tissue bath. Contractile force was measured with force displacement transducers under 2-3 g loading tension. The dose of norepinephrine(NE) which evoked 50% of maximal response $(ED_{50})$ was obtained from cumulative dose response curves for NE $(10^{-6}{\sim}10^{-3}M)$. Contractions evoked by NE $(ED_{50})$ were inhibited significantly by Rhizoma Arisaematis, Lignum Akebiae, Rhizoma Zedoariae, Cortex Eucommiae, Folium Perillae, Radix Sophorae Subprostratae and Herba Ephedrae in abdominal aorta. Contractions evoked by NE $(ED_{50})$ were inhibited significantly be Lignum Akebiae, Rhizoma Zedoariae, Cortex Eucommiae, Herba Ephedrae, Radix Puerariae and Radix Bupleuri in femoral artery. Contractions evoked by NE $(ED_{50})$ were inhibited significantly by Radix Sophorae Subprostratae, Radix Aconiti and Herba Ephedrae in renal artery. These results indicate that each herb can relax NE induced contraction of rabbit and pig blood vessel selectively, and that this relaxation relates to Gui-Gyung(歸經).

      • 梔子大黃湯과 茵蔯梔子湯 Extract가 白鼠의 肝損傷에 미치는 影響

        An attempt was made to evaluate the effects of Chijadaewhangtang extract (CDE) and Inchinchijatang extract (ICE) on the recovery of hepatotoxicity induced acute treatment of carbon tetrachloride and results were as follows: 1. When rats of carbon tetrachloride-intoxication were treated with CDE and ICE, the serum aspartate aminotransferase activities released from liver were decreased. In ICE treated group, there were significant decrement after 12 hours and more fast return to normal values in comparison with CDE treated group. 2. In CDE and ICE-treated group, as compared with only carbon tetrachloride-treated group there were inhibition of serum alanine aminotransferase elevation and significant decrement was observed in ICE treated group after 48 hours and CDE treated group after 72 hours. 3. Serum alkaline phosphatase values were significantly diminished in CDE and ICE-treated group compared with only carbon tetrachloride intoxicated group. Meaningful diminution was observed in ICE administered group after 48 hours and CDE administered group after 96 hours. 4. Significant decrement of serum lactate dehydrogenase elevation after 96 hours was observed in CDE and ICE-treated group and more fast recover to normal values. 5. Administration of CDE and ICE shortened prothrombin time of plasma and significant recovery was showed in ICE treated group after 72 hours and CDE treated group after 96 hours. From these results, the recuperative effects of CDE and ICE on carbon tetrachloride-induced hepatotoxicity could be verified experimentally.

      • 淸肺散 煎湯液이 家兎의 腎臟機能에 미치는 影響

        To evalute effect of Chungpyesan(淸肺散) Water Extract on the renal function, blood pressure and heart rate, experiment performed in rabbits. The results obtained were as follows : 1. Intravenous administration of Chungpyesan Water extract at the doses of 0.75㎖/㎏ significantly increased renal plasma flow, glomerular filtration rate, urinary excretion of electrolytes and urinary excretion of free water clearance, in particular, urine volume. 2. Urine volume, glomerular filtration rate and urinary excretion of electrolytes were increased remarkably after intravenous administration of Chungpyesan Water extract at the doses of 1.5㎖/㎏. 3. Intravenous administration of Chungpyesan Water extract at the doses of 0.75㎖/㎏ significantly increased heart rate. 4. Intravenous administration of Chungpyesan Water extract at the doses of 1.5㎖/㎏ significantly decreased blood pressure, whereas heart rate remakably increased. These results suggest that Chungpyesan Water extract has a diuretic effects, which may be caused by urinary excretion of electrolytes, renal hemodynamics.

      • 滋賢丸 煎湯液이 無麻醉 家兎의 腎臟機能에 미치는 影響

        This study was carried out to evaluate the effects of Jasinhwan(滋腎丸) water extract on the renal function and arterial blood pressure. The results of this study were as follows : 1. Urine volume, renal plasma flow, and glomerular filtration rate were significant increased after intravenous administration of Jasinhwan water extract(0.2㎖/㎏). 2. Urine volume, renal plasma flow, glomerular filtration rate, and urinary excretion of electrolytes were significant increased after intravenous administration of Jasinhwan water extract(0.4㎖/㎏). 3. Heart rate was significant decreased after intravenous administration of Jasinhwan water extract(0.4㎖/㎏). These results suggest that the increased of ruine volume after intravenous administration of Jasinhwan water extract was related with the changed of renal hemodynamics and the increased of urinary excretion of electrolytes.
