        As copyright infringement is exploded and related industrial damage become huge by illegal file sharing developed by internet file sharing technology like P2P, three-strikes out rule for extermination illegal file sharing has been introduced in copyright law by individual country. Proposed or introduced three-strikes out law is different in each country by enforcement agent, target, and intensity, and it can be categorized in three types of France, US and Korea. Our three-strikes out law distinguished by shut down a website and enforcement by administrative order without judical decision caused great concern and criticism. While other nations have been through hard times in legalization internet three-strikes out rule facing heavy opposition of citizen's and court, three-strikes out law become more sophisticated. Illegal file sharing can be solved fundamentally by reasonable reward system of market. Korea which was enforced three-strikes out law already need to improve three-strikes out law to minimize problems and side effects and fulfill expected effectiveness of law by various stakeholders' continuous attention and discussion. For this, Strengthen independency and autonomy of Korea copyright commission, minimization of ISP's burden, Provision of balanced-opinion to internet user are needed.

        Gastroprotective Activities of Sennoside A and Sennoside B via the Up-Regulation of Prostaglandin E2 and the Inhibition of H+/K+-ATPase

        Sennoside A (erythro) and sennoside B (threo) are dianthrone glycosides and diastereomers. We investigated their abilities to prevent the gastric lesions associated with diseases, such as, gastritis and gastric ulcer. To elucidate their gastroprotective effects, the inhibitions of HCl•EtOH-induced gastritis and indomethacin-induced gastric ulcers were assessed in rats. It was observed that both sennoside A and sennoside B increased prostaglandin E2 (PGE2) levels and inhibited H+/K+-ATPase (proton pump). In a rat model, both compounds reduced gastric juice, total acidity and increased pH, indicating that proton pump inhibition reduces gastric acid secretion. Furthermore, sennoside A and B increased PGE2 in a concentration-dependent manner. In a gastric emptying and intestinal transporting rate experiment, both sennoside A and sennoside B accelerated motility. Our results thus suggest that sennoside A and sennoside B possess significant gastroprotective activities and they might be useful for the treatment of gastric disease.

      • 휘발성 유기용매의 In vitro 대사속도 측정 장치의 개발

        Species, doses and routes extrapolation can be sucessfully carried out by using a physiologically-based pharmacokinetic (PBPK) approach. And PBPK approach to assess risk of hazardous chemicals is reasonable whatever the exposure scenarios are happened. Both partitioning coefficients of chemical between tissue and blood and enzymatic metabolic rate constants are key parameters to build up the PBPK model. In this study, we tried to estimate in vitro metabolic rate constants using a special apparatus instead to measure the in vivo constants which are used to PBPK simulation since the in vitro tests are less expensive and more convenient than in vivo tests. For the purpose, we designed and tested the new system to measure continuously the headspace concentration of VOC. The newly designed system is composed with a diffusion chamber which generates gaseous substrate, a reaction vessel with a recirculating pump to establish a closed system, an autbmatic sampler from a gas phase, a gas chromatography to analyze the headspace. In addition, a cold water condenser is attached between the reaction vessel and pump to reduce the content of gaseous moisture which interferes with chemical analysis. To validate the newly developed methodology, in vitro metabolic rate constants of trichloroethylene (TCE) as a prototype VOC were estimated by simulating observed results with an ACSL program. The simulated results are consistent to those estimated by the other research groups. This finding suggests that our newly designed closed system may be a useful apparatus to estimate in vitro metabolic rate constants for VOC.

        pVHL-Mediated Transcriptional Repression of c-Myc by Recruitment of Histone Deacetylases

        The biological functions of Myc are to regulate cell growth, apoptosis, cell differentiation and stem-cell self-renewal. Abnormal accumulation of c-Myc is able to induce excessive proliferation of normal cells. von Hippel-Lindau protein (pVHL) is a key regulator of hypoxia-inducible factor1 (HIF1), thus accumulation and hyperactivation of HIF1 is the most prominent feature of VHL-mutated renal cell carcinoma. Interestingly, the Myc pathway is reported to be activated in renal cell carcinoma even though the precise molecular mechanism still remains to be established. Here, we demonstrated that pVHL locates at the c-Myc promoter region through physical interaction with Myc. Furthermore, pVHL reinforces HDAC1/2 recruitment to the Myc promoter, which leads to the auto-suppression of Myc. Therefore, one possible mechanism of Myc auto-suppression by pVHL entails removing histone acetylation. Our study identifies a novel mechanism for pVHL-mediated negative regulation of c-Myc transcription.

        The use of subordination in Korean EFL student writing

        Learning context has been recognized as a significant factor for learning a language in the field of second language acquisition and ‘study-abroad’ context is a new environment from ESL or EFL settings. Even though many Korean EFL college students have study-abroad experiences, research into improvement in study-abroad students’ competence, especially writing competence, is rare. This study examined argumentative writings of Korean EFL college students who had studied abroad for two semesters as visiting or exchange student and compared them with those of students who stayed in Korea. The written texts of the students were analyzed, focusing on the use of subordinate clauses since subordination indicates syntactic maturity and complexity of a text. The results of the analysis of subordination demonstrated that the two groups showed no statistically significant differences, except for the reduced adverb clauses. Since the study-abroad group did not show a better command of using subordinate clauses in their academic writings, it can be assumed that the experience abroad did not necessarily benefit students in their grammatical competence in writing. This result implies that for a better syntactic complexity in Korean EFL college students’ writings, more explicit instruction on subordination is needed, regardless of study-abroad experience.

        살균제 Tricyclazole에 대한 안전성 평가 (I)

        Tricyclazole을 수확전 $90{\sim}40$일에 $2{\sim}3$회 처리한 작물과 토양중에서의 수확시 잔류되는 tricyclazole의 양은 각각 현미가 $0.37{\sim}0.46ppm$, 백미가 $0.29{\sim}0.39ppm$, 볏짚이 $0.14{\sim}0.30ppm$ 및 토양이 $0.15{\sim}0.19ppm$이었다. 담수조건하에서의 tricyclazole의 분해 양상은 처리후 $2{\sim}3$주까지 비교적 빠른 속도로 분해가 진행되다가 그 이후로는 안전한 속도로 분해되었으며 그 때의 반감기는 $140{\sim}180$일이었다. 또한 토양 미생물의 생균수에 미치는 영향은 거의 없었으며 Salmonella, Saccharomyces 등을 이용한 돌연변이성 유무 실험에서는 대조구와 별차이가 없었다. Fate of tricyclazole in rice paddy system was studied. The effect on soil microorganism as well as the mutagenicity of the compound was also examined. The residues of tricyclazole in crops and soil with two times application before harvest were 0.37 in unpolished rice, 0.29 in polished rice, 0.14 in rice straw, and 0.15 ppm in paddy soil. With three times of application the residues were increased to 0.46, 0.39, and 0.19 ppm, respectively. Until $2{\sim}3$ weeks after treatment of pesticide the degradation of tricyclazole was progressed comparatively but very slowly afterward and the half life of that was about $140{\sim}180$ days. There was no effect for viable count of soil microorganisms and for mutagenic test by Salmonella and Saccharomyces systems.

        Glutamate에 의한 세포내 칼슘농도변화와 세포독성과의 관계

        Pathophysiological elevation of intracellular calcium concentration ([Ca^(2+)]_i) in the neuron has been considered as an important responsible factor in the neuronal cell damages. However, the mechanism of increase of [Ca^(2+)]_i and the relationship between [Ca^(2+)]_i level and cytotocixity have not been fully demonstrated. In the present study, real-time alteration of [Ca^(2+)]_i and cellular response (cell damages) in the pheochromocytoma cells (PC12) stimulated by glutamate were investigated. Glutamate dose dependently decreased cell viability determined propidium iodide fluorescence method and morphology change. Choversely related with cell damages, glutamate dose dependently increased the level of [Ca^(2+)]_i. To investigate the mechanism of glutamate-induced increase of [Ca^(2+)]_i, [Ca^(2+)]_i was first measured in the cells cultured in calcium free media and in the presence of dantrolene, an inhibitor of calcium release from ryanodine receptor located in endoplasmic reticulum (ER). Similar to the increase [Ca^(2+)]_i in the calcium-containing media, glutamate does dependently increased [Ca^(2+)]_i in the cells cultured in free calcium media. However, pretreatment (2hr) with 20~50μM dantrolene substantial lowered glutamate-induced increase of [Ca^(2+)]_i, suggesting that release of calcium from ER may be major sourse of increase of [Ca^(2+)]_i in PC12 cells. Dantrolene-induced inhibition of [Ca^(2+)]_i resulted in recovery of cytotoxicity by glutamate. Relevance of N-methyl-D-aspartate (NMDA) receptor, a type of glutamate receptor on glutamate-induced increase of [Ca^(2+)]_i, [Ca^(2+)]_i was also determined in the cells pretreated (2hr) with NMDA receptor antagonist MK-801. Glutamate-induced increase of [Ca^(2+)]_i was reduced by MK-801 dose dependently. Furthermore, glutamate-induced cytotoxicity was also provented by MK-801. These results demonstrate that glutamate increase [Ca^(2+)]_i dose dependently and thereby cause cytotoxicity. The increase of [Ca^(2+)]_i may release from ER, especially through ryanodine receptor and/or through NMDA receptor. Alteration of calcium homeostasis through disturbance of ER system and/or calaium influx through NMDA receptor could contribute glutamate-induced cell damages.

        해상 유류 유출 사고 후, 유류 해양 오염에 따른 주요 해산 어ㆍ패류내 PAHs 추적

        Marine organisms such as fish (Sebastes schlegeli) and mussels were cultured in sea water tanks placed at Dukpo area which was contaminated by the two oil spill accidents. Results showed that PAHs concentrations in fish tissue were higher than in fish liver. This was explained by the cytochrome P450 IA induction in fish liver after PAHs exposure. Other studies showed that higher PAHs levels were detected in mussels cultures in oil contaminated area than in control site. From these results, we concluded that Dukpo area is still polluted by oil including PAHs and it takes a long time to recover of oil contamination after the oil spill accidents.
