Current status of the novel food ingredient safety evaluation system

Increasing demand for new foods, technological development, and vegan market growth have led to an increase in new food ingredients, so the need for safety assessment of these ingredients is important. Representative safety assessment systems are the Generally Recognized as Safe (GRAS) notification of the Food and Drug Administration in the USA and the novel food system of the European Food Safety Authority in the European Union. GRAS is a notification system for information on food ingredients, food additives and functional foods under the responsibility of the applicant, while the novel food system assesses the safety of food ingredients excluding food additives. In Korea, a safety evaluation system is established for temporary food ingredients, which includes food ingredients without a domestic intake history. However, safety assessment systems for novel foods from other countries and food ingredients produced by the application of new technology need to be improved.
Food-derived coagulase-negative Staphylococcus as starter cultures for fermented foods

Food safety is of significant concern to consumersand is a major issue for the food industry. As such,the industry is aware of the importance of safety assessmentsof starters used in the production of fermented foods. Coagulase-negative staphylococci (CNS) are the predominantbacteria found in fermented foods worldwide. Becauseof this, food-derived CNS are used as starters for cheeseand meat fermentation, and have been investigated for useas starters in soybean fermentation. Although food-derivedCNS are generally considered non-pathogenic, their safetyremains uncertain following the isolation of CNS from skininfections in humans and animals, and because they belongto the same genus as the highly pathogenic species Staphylococcusaureus. This review explores what is knownabout the safety of food-derived CNS, focusing on antibioticresistance, enterotoxin genes, and biogenic amineproduction, to aid in the selection of starter candidates.

Enterococcus faecium strain DMEA02 was isolated from meju, a traditional Korean fermented soybeans. The genome of strain DMEA02 is 2,742,551-bp long with a G + C content of 38.29% and it consists of a single circular 2,573,925-bp chromosome and a single circular 168,626-bp plasmid. No genes associated with resistance to eight antibiotics (chloramphenicol, clindamycin, erythromycin, gentamicin, kanamycin, streptomycin, tetracycline, and vancomycin), biofilm formation, or hemolysis were identified from the chromosome and plasmid of strain DMEA02. The obtained genetic information supports the safety of strain DMEA02 as a potential fermentation starter culture candidate for fermented soybean food productions.
Complete genome sequence of Lactiplantibacillus plantarum KM2 from low-temperature aging beef

We previously isolated Lactiplantibacillus plantarum strain KM2 from low temperature aging beef. Its complete genome sequence was determined in the current study to understand KM2’s genetic background. The genome contains a single circular 3,233,034 bp chromosome and five plasmids. The genome G + C content is 44.24%. Significantly, strain KM2 contains a plantaricin A gene cluster, an antifungal determinant, and exopolysaccharide biosynthetic genes. The complete genome sequence of strain KM2 will contribute to understanding the genetic background of antimicrobial activities.

크론병은 다양한 기관을 침범하는 장외 증상을 유발할 수 있으며 크론병의 폐 침범은 매우 드물다고 알려져 있다. 본 증례는 복강 내 농양 및 장피누공을 동반한 크론병으로 azathioprine 및 corticosteroid를 투약하던 환자에서 Enterococcus faecium 및 Mycobacterium tuberculosis의 동시 감염으로 인한 복강 내 농양으로 항결핵제제와 항생제로 치료하던 중 발생한 폐결핵에 동반한 크론병의 폐 침범 사례로 국내에 보고된 바가 없다. 본 증례의 28세 남자환자는 10년전 크론병을 진단받고 같은 해 충수 및 소장 절제술을 시행하고 장피누공을 동반한 소장 및 대장 크론병(Montreal classification of Crohn’s disease A2L3B3, Crohn’s disease activity index 154)으로 melsalazine, prednisolone, azathioprine을 복용하던 분으로 복부에 만져지는 종괴로 본원에 내원하였다. 내원 당시 시행한 복부 전산화 단층촬영 및 경피적 농양 배액술의 결핵균 핵산증폭검사상 복강 내 결핵성 농양을 진단 받은 후 prednisolone, azathioprine은 중단하고 항결핵제제와 항생제 투약을 시작하였다. 결핵 약제 시작한지 한달 후 장피 누공의 치료를 위해 infliximab을 투약하였으며 당시 흉부 엑스선 검사는 정상 소견을 보였다. 항결핵제제 및 infliximab을 투여하던 중 3개월 후 환자는 2주동안 지속되는 기침, 가래로 내원하였고 흉부 엑스선 검사 및 흉부 전산화 단층촬영상폐렴 및 농양 소견을 보였다. 경기관지폐생검 및 기관지폐포세척검사 상 조직 괴사와 비건락성 육아종을 동반한 활동성 염증 소견 및 결핵균 핵산증폭검사(TB-PCR)상 양성 소견을 보였으나 객담 도말 및 배양검사에서 항산균은 검출되지 않았다. 기존의 결핵 치료에 악영향을 미칠 가능성이 있고 추적 복부 전산화 단층촬영상 장피 누공에 특이 변화를 보이지 않아 infliximab을 중단하고 크론병의 폐 침범으로 생각되어 기존의 항결핵제제를 유지한 상태에서 고용량corticosteroid 치료를 시작하였다. 기존 항결핵제제와 고용량 corticosteroid 투약한지 약 1개월 후 시행한 흉부 엑스선 검사 및 흉부 전산화 단층촬영상 폐 병변은 호전되었다. 이에 저자들은 크론병으로 치료 중인 환자에서 폐결핵을 동반한 크론병의 폐 침범 1예를 경험하였기에 이를 보고하는 바이다.
Complete genome sequence of Staphylococcus xylosus strain DMSX03 from fermented soybean, meju

Staphylococcus xylosus strain DMSX03 was isolated from fermented soybean, meju, and its complete genome sequence was determined. It contains a single circular 2,825,246-bp chromosome and a 48,816-bp plasmid. The G + C content of the genome is 32.76%. Strain DMSX03, having protease and lipase activities, encodes 44 protease and 8 lipase genes. Thus, the genetic background corroborates the enzymatic activities reported for strain DMSX03, making it a potential starter candidate.

메주로부터 분리한 Enterococcus faecalis DM01 균주는 8종의 항생제(ampicillin, chloramphenicol, ciprofloxacin, erythromycin, gentamicin, penicillin G, tetracycline, vancomycin) 저항성, 바이오필름 및 용혈현상을 나타내지 않았다. GC 함량 37.68%, 2,785,968 bp 크기의 단일 chromosome을 보유하고 있는 DM01 균주의 유전체 정보는 콩발효식품 제조용 종균 적용에 요구되는 항생물질 저항성, 바이오필름 및 용혈현상 관련의 안전성을 뒷받침하는 것으로 확인되었다. Enterococcus faecalis strain DM01 was isolated from meju, a traditional Korean fermented soybean product. The strain did not exhibit resistance to eight antibiotics (ampicillin, chloramphenicol, ciprofloxacin, erythromycin, gentamicin, penicillin G, tetracycline, and vancomycin), biofilm formation, and hemolytic activity. The genome of strain DM01 consists a single circular 2,785,968-bp chromosome with a G + C content of 37.68%. The complete genomic sequence of strain DM01 provides genetic information to support the absence of antibiotic resistance, biofilm formation, and hemolytic activity.
