        고온가열처리에 의한 바다방석고둥 ( Omphalius pfeifferi capenteri ) 의 식품성분 변화

        바다방석고둥을 원료로 한 신제품 개발에 관한 일련의 기초 연구로 저장성 부여를 위하여 시도하는 고온가열처리 조건 (F_0 5분, F_0 10분, F_0 15분 및 F_0 20분)에 따른 바다방석고둥의 성분변화에 대하여 살펴보았다. 바다방석고둥 통조림은 살균을 5분 이상 처리하는 경우 저장 중 팽창관이 발생하지 않았고, 또한 생균수가 검출되지 않아 저장성이 있었다. 고온가열처리 정도가 증가할수록 바다방석고둥의 일반성분 중 수분함량은 감소하였고, 조단백질 함량은 증가하였으며, 조지방 및 조회분 함량은 거의 변화가 없었다. 또한 바다방석 통조림은 고온가열처리 정도가 증가할수록 이취에 관여하는 휘발성염기질소 함량, pH와 색조에 관여하는 갈변도는 증가하였고, 영양에 관여하는 구성아미노산, 무기질과 맛에 관여하는 유리아미노산, TMAO, total creatinine 및 수율 둥은 감소하였다. 이들을 색조, 조직감 및 맛에 대하여 관능검사한 결과 자숙 처리한 시료에 비하여 모든 항목에서 F_0 10분까지는 차이가 없었으나, 그 이상 가열처리한 경우 품질이 저하하였다고 인지되었다. 이상의 결과로 미루어 보아 바다방석고둥을 고온가열처리하여 신제품을 개발하고자 하는 경우 F_0 값이 10분 이상이면 바람직하지 않은 것으로 판단되었다. The top shell, Omphalius pfeifferi capenteri meat vacuum-packed in can (diameter× height, 74.1 ㎜ × 50.7 ㎜) were heated at 115℃ up to F_0 values of 5 min, 10 min, 15 min and 20 min, and the changes in food components were studied. After 14 days storage at 37℃ and 55℃, no growth of microorganism and panelling were recognized from the canned meats which were sterlized at 115℃ with F_0 value of 5 min and over. In the case of proximate composition of the canned meats, the moisture content decreased with the increase of F_0 value, while crude protein increased. The increase of volatile basic nitrogen content, pH and degree of browning and the decrease of mineral, total amino acid, free amino add, trimethylamine oxide, total creatinine contents and yields were observed during thermal processing. In sensory evaluation on color, texture and taste in the canned meats, no significant difference was observed among a boiled sample and the canned meats heated at F_0 value of 10 min and below. But, in the canned meats heated at F_0 value of over 15 min, its sensory scores decreased with the increase of F_0 value. From these results, the reasonable F_0 value for preparation of the heat-treated top shell meats was in the range of 5∼10 min.

        새우 부산물을 이용한 기능성 소스의 제조

        The functional sauce from shrimp byproducts (heads, shells and tails) was prepared and examined for its characterization. The results of volatile basic nitrogen (VBN) suggested that shrimp byproducts were suitable materials for preparing functional sauce. The shrimp hydrolysate, which was incubated with Alcalase for 30 min, showed excellent yield and ACE inhibitory activity. The concentrated sauce from shrimp byproduct was high in crude protein, while low in VBN content and salinity when compared to commercial shrimp sauce. The total amino acid content (23,095.2 mg/100 mL) of concentrated sauce from shrimp byproduct was higher than that (4,582.5 mg/100 smL) of commercial shrimp sauce; also, the major amino acids were glutamic acid, aspartic acid, arginine and lysine. The free amino acid content and taste value of concentrated sauce from shrimp byproduct were 2,705.5 mg/100 mL and 81.0, respectively. The results on the taste value of concentrated sauce from shrimp byproducts suggested that the major taste active compounds among free amino acids were glutamic acid and aspartic acid. 새우 가공부산물을 효율적으로 이용할 목적으로 소스를 제조하였으며, angiotensin I converting enzyme(ACE) 저해능과 성분특성에 대하여 조사하였다. 새우 가공부산물로부터 제조한 소스의 선도는 휘발성염기질소의 함량으로부터 적합성이 인정되었다. Alcalase로 30분 반응시켜 제조한 새우 소스의 수율은 53.5%를 나타내었고, ACE 저해능은 70.1%를 나타내었다. 새우 가수분해물 농축 소스는 시판 액젓과 비교하여 단백질 함량이 18.7%가 높았고, 휘발성염기질소 함량 및 염도의 경우 각각 24.0 mg/100 mL 및 9.0%가 낮았다. 새우 가수분해물 농축 소스의 총 아미노산 함량은 23,095.2 mg/100 mL이었고 주요 아미노산으로는 glutamic acid, aspartic aicd, arginine 및 lysine 등이었다. 유리아미노산 함량은 새우 가수분해물 농축 소스(2,705.5 mg/100 mL)가 시판 액젓(1,407.7 mg/100 mL)에 비하여 높았으나, taste value는 새우 가수분해물 농축 소스(81.0)와 시판 액젓(81.7) 간에 차이가 없었다. Taste value의 결과로 보아 새우 가수분해물 농축 소스의 맛에 관여하는 주요 아미노산은 aspartic acid와 glutamic acid로 판단되었다. 이상의 결과로부터 새우 가수분해물 농축 소스는 고가의 새우 액젓의 대용품으로 이용 가능하리라 판단되었다.

        붕장어 부산물로 제조한 붕장어탕의 식품학적 특성

        붕장어 가공부산물인 머리와 frame를 효율적으로 이용하기 위한 일련의 연구로 붕장어 부산물을 이용한 붕장어탕의 개발을 시도하였고, 아울러 이의 식품성분 특성에 대하여도 살펴보았다. 살균조건 및 가온일수에 따른 붕장어탕의 저장성 부여를 위한 최적 살균조건은 F? value=8분으로 판단되었다. 붕장어탕의 일반성분은 수분의 경우 90.7%, 조단백질의 경우 4.8%, 조지방의 경우 2.6%, 조회분의 경우 1.5%를 나타내었다. 붕장어탕의 엑스분 질소 함량은 243.1 ㎎/100 g으로 시판 추어탕의 208.0 ㎎/100 g보다 높았다. 붕장어탕의 총 아미노산 함량은 4,310 ㎎/100 g이었고, 주요 구성아미노산은 glutamic acid(637.3 ㎎/100 g, 14.8%), glycine(409.1 ㎎/100 g, 9.5%) 및 alanine(404.4 ㎎/100 g, 9.3%)등이었다. 붕장어탕은 관능적인 비린내는 거의 감지되지 않았고, 맛은 우수하였으며, 항산화능이 인정되었으나 ACE 저해능은 크게 기대할 수 없었다. For the effective use of the conger eel by-products, such as head and frame, Tang, which is the Korean-type soup, from conger eel by-products (TCEB) was developed and its food component characterization was compared with that of commercial Chueotang, loach Tang. According to the results of viable cells and coliform group of TCEB heated at 115oC for various times, the reasonable F0 value was 8 min. The proximate composition of TCEB was 90.7% for the moisture, 4.8% for the protein, 2.6% for the lipid, and 1.5% for the ash. The extractive-nitrogen content of TCEB was 243.1 ㎎/100 g, which was higher than that of commercial Chueotang, 208.0 ㎎/100 g. The total amino acid content of TCEB was 4,310 ㎎/100 g and its major amino acids were glutamic acid (637.3 ㎎/100 g, 14.8%), glycine (409.1 ㎎/100 g, 9.5%) and alanine (404.4 ㎎/100 g, 9.3%). TCEB was not felt in the sensual fish odor and its sensual taste was good. The health functional properties for health of TCEB were 1.29 as a PF (protection factor) for antioxidative activity and 39.4% for angiotensin Ⅰ converting enzyme (ACE) inhibiting activity.

        This study was conducted to optimize the processing of high quality surimi gel from unmarketable cultured bastard halibut Paralichthys olivaceus. According to endogenous enzyme activity and processing optimization, high quality surimi gel from unmarketable cultured bastard halibut was prepared by mixing 3.0% (w/w) salt, 2.4% (w/w) starch, 5.0% (w/w) egg white and 4.8% (w/w) ice water in a Stephan mixer, set at 5˚C for 24 h, followed by boiling for 30 min, and finally cooling for 30 min. The strength of the surimi gel from unmarketable cultured bastard halibut prepared by the above processing method was 1,257 g×cm, which was 33% higher than that of a commercial surimi gel from Alaska pollock, grade SA.

        자숙 가다랑어뼈로부터 칼슘제의 제조 및 특성

        As a part of investigation for utilizing of canned tuna processing by-products as a food resource, we examined the processing conditions and characteristics (extraction methods and ashing condition) of a calcium powder from skipjack tuna bone. Among ashing, autoclaving, and shaking methods for extraction of calcium powder from skipjack tuna bone, ashing method was superior to other methods on the aspect of fish odor, white index, and calcium recovery of calcium powder. Based on the results of white index and soluble calcium ratio, the optimal ashing temperature and time for preparation of a calcium powder from skipjack tuna bone was at 900℃ for 15 min. Cohesive ratio of calcium powder by shaking at pH 7.0 was increased up to 16 hrs, but after that almost unchanged. Cohesive ratio of calcium powder by shaking for 24 hrs was increased at neutral and alkaline conditions (pH 6-8 and pH 9-11), but almost unchanged at acidic conditions (pH 2-5). For the effective utilization of the calcium powder from skipjack tuna bone, a suitable treatment is needed for improvement of calcium solubility at neutral condition.

        참치 자숙액 가수분해물을 이용한 건강 기능성 조미 소스의 제조

        참치가공 부산물의 효율적 이용을 위하여 건강 기능성이 고려된 참치 자숙액 조미 소스의 제조를 시도하였고, 그 특성에 대하여도 살펴보았다. 참치 조미 소스는 단백질이 11.8%로 시판 조미 소스의 약 2배에 해당하였고, pH, brix 및 염도는 각각 $577,348^{\circ}$ 및 11.9%이었다. 관능검사 결과, 참치 조미 소스는 시판 조미 소스에 비하여 맛, 냄새 및 색조에 있어 차이가 없었다. ACE 저해능은 참치 조미 소스가 시판 조미 소스(9.9 mg/mL)에 비하여 37% 정도 높았고, 항산화능은 20 mM ascorbic acid에 비하여는 약하였으나 인지는 되었다. 참치 조미 소스는 총 유리아미노산 함량이 1,905.2 mg/100 mL로 시판 조미 소스(712.7 mg/100 mL)에 비하여 약 2.7배 높았으며, 주요 유리아미노산은 taurine, glutamic acid, histidine 및 anserine이었다. 참치 조미 소스는 total taste value가 58.65로 시판 조미 소스의 34.30에 비하여 맛의 강도가 훨씬 강하리라 추정되었고, 맛에 크게 기여하는 주요 아미노산으로는 glutamic acid 및 histidine 등이었다. 총 아미노산 함량은 참치 조미 소스가 10,965 mg/100 mL로 시판 조미 소스의 4,818 mg/100 mL에 비하여 높았고, 참치 조미 소스의 주요 구성 아미노산으로는 glutamic acid, proline, histidine 및 glycine 등이었다. 참치 조미 소스의 섭취에 의한 무기질 강화 효과는 기대하기 어려우리라 판단되었다. The enzymatic hydrolysate of skipjack tuna cooking drip with good functionality was prepared by incubation with Alcalase for 30 min. For the preparation of functional seasoning sauce with enzymatic hydrolysate (SSE), the additives, such as concentrated enzymatic hydrolysate (100 mL), yeast extract powder (0.7 g), lactose (0.4 mL), liquid smoke (0.3 g) and sea tangle powder (1.4 g), were added to the enzymatic hydrolysate and boiled before filtration. The proximate composition of SSE was 11.8% for crude protein, 5.77 for pH and 11.9% for salinity. The SSE was higher in the crude protein, while lower in the salinity than commercial seasoning sauce. ACE inhibitory activity ($IC_{50}$) and antioxidative activity (PF) of SSE were 6.2 mg/mL and 1.14, respectively, which were superior to those (9.9 mg/mL in IC50 and 0.91 in PF) of commercial seasoning sauce. The free amino acid content (1,905.2 mg/100 mL) and taste value (58.65) of SSE were higher than in those (712.7 mg/100 mL and 34.30, respectively) of commercial sauce. Total amino acid content of SSE (10,965 mg/100 mL) was higher than that (4,818 mg/100 mL) of commercial sauce. The major amino acids of SSE were glutamic acid (12.2%), proline (11.0%), histidine (10.7%) and glycine (9.9%). The results suggested that SSE could be commercially sold.

        굴 스파게티 소스의 개발

        굴을 보다 효율적으로 이용하고자 신세대 기호에 맞는 굴 스파게티 소스의 제조를 시도하였다. 유기산, 색조, 점도 및 관능검사의 결과로 미루어 보아 스파게티 소스의 굴 최적 첨가량은 11%로 판단되었고, 저장성 부여를 위한 최적 F? value는 4분이라고 판단되었다. 최적조건에서 제조한 굴 스파게티 소스의 수분, 단백질, 지방 및 회분은 각각 71.2%, 2.8%, 6.9% 및 3.2%이었다. 굴 스파게티 소스의 관능검사결과 향과 조직감의 경우 시판 스파게티 소스보다 좋았으며, 색의 경우 차이가 없었다. 굴 스파게티 소스의 총 아미노산 함량은 2,532.2 ㎎/100 g이었고 주요 아미노산으로는 glutamic acid, aspartic acid 및 phenylalanine 등이었다. 굴 스파게티 소스는 칼슘/인의 비율이 1.88로 칼슘이 흡수되기 좋은 비율로 구성되었다. 유리아미노산 함량은 시판 스파게티소스(1,165.5 ㎎/100 g)가 굴 스파게티 소스(1,040.2 ㎎/100 g)에 비하여 높았고, taste value 또한 시판 스파게티 소스(189.35)가 굴 스파게티 소스(151.26)보다 높았다. Taste value의 결과로 미루어 보아 맛에 관여하는 주 아미노산은 aspartic acid와 glutamic acid로 판단되었다. The study was carried out to prepare spaghetti sauce with oyster (SSO) and the food components character-istics of the SSO were also compared to those of commercial spaghetti sauces (CSS). The optimal addition ratio of oyster for preparing SSO was 11% based on 100 g of SSO according to the results of organic acid content, Hunter color value, viscosity, and sensory evaluation. The reasonable F? value for the keeping storage of SSO was about 4 min. The proximate composition of SSO prepared under the optimal processing condition was 71.2% moisture, 2.8% protein, 6.9% crude lipid, and 3.2% crude ash. The results of sensory evaluation suggested that the quality of SSO was superior to that of CSS. However, there was no significant difference (p<0.05) in sensory evaluation on color between CSS and SSO. The total amino acid content (2,532.2 ㎎/100 g) of SSO was higher than that of CCS (2,305.7 ㎎/100 g). The contents of calcium and phosphorus of SSO were 25.7 ㎎/100 g and 48.7 ㎎/100 g, respectively. The calcium content/phosphorus content showed a suitable ratio for absorbing calcium. The total free amino content and the taste value were 1,040.2 mg/100 g and 151.26, respectively. The major taste?active amino acids were glutamic acid and aspartic acid.

        헬스케어용 가다랑어(Katsuwonus pelamis) 통조림의 제조 및 특성

        This study was conducted to prepare canned skipjack tuna Katsuwonus pelamis in the mixture of isotonic beverage and tomato paste (ST-S) for use as a health food. An analysis of the time-temperature profile and viable cells showed that a reasonable F0 value for ST-S preparation was 4 min. The proximate composition of ST-S was 76.8% moisture, 20.2% crude protein, 0.8% crude lipid, 1.7% ash and 0.5% carbohydrate. The calorie content of ST-S was 94.8 kcal, which is 47.4% lower than that of commercial canned skipjack tuna in oil (ST-O) and 2.2% lower than that of commercial chicken breast in water (CB-B). The total amino acid content of ST-S was 18.54 g/100 g, which is 31.4% lower than that of ST-O and 7.9% lower than that of CB-B. The major amino acids in ST-S were aspartic acid and glutamic acid. An enrichment effect due to such minerals as phosphorus, potassium and iron would be expected on consuming 100 g of ST-S. The major fatty acids in ST-S were 16:0 (27.4%), 18:1n-9 (14.3%) and 22:6n-3 (27.8%), which are different from those in ST-O and CB-B. The major free amino acids in ST-S were glutamic acid (8.1%), histidine (38.6%) and its related dipeptide, such as anserine (15.7%). In an evaluation of taste, flavor and color, ST-S was found to be superior to ST-O and CB-B.

        어류 알의 Protease Inhibitor 활성 분포

        To identify and examine the distribution of proteolytic inhibitory activity in crude extracts from fish eggs, and to determine the applicability of these protease inhibitors as anti-degradation agents in surimi-based products and fish meat, we compared the inhibitory activities of various extracts from fish eggs to those of commercial proteases, such as trypsin and papain. We used the optimal conditions for the screening of trypsin activity: 30 ug/uL of 0.1 % trypsin and 0.6 mM Na-benzoyl-L-arginine-p-nitroanilide (BAPNA) with a pH of 8.0 at 40℃ for 60 min. The activities of papain and four commercial proteases were investigated after mixing with 100 ug/uL enzymes and 0.3% casein with a pH of 8.0 at 40℃ for 60 min. We performed a screening assay to detect the inhibitory activity (%) of crude extracts from eight species of fish eggs against the target proteases trypsin and papain. The assay revealed a wide distribution of trypsin and papain inhibitors in fish eggs. The specific inhibitory activities (11.6-28.6 U/mg) of crude extracts from fish eggs against trypsin and BAPNA substrate were higher than that (0.64 U/mg) of egg whites, used as a commercial inhibitor. The inhibitory activities of crude extracts from fish eggs against trypsin, and of egg whites against casein substrate (1.94-4.51 U/mg), were higher than those of papain (0.24-1.57 U/mg) and commercial protease (0.04-0.32 U/mg). The extracts from fish eggs were rich in protease inhibitors that exhibited strong inhibitory activity against trypsin, a serine protease, and papain, a cysteine protease.

        생식용 굴(Crassostrea gigas) 작업장의 위생안전성에 대한 모니터링

        This study assessed the safety of raw oysters (Crassostrea gigas) for consumption during processing in a processing plant. Bacterial contamination (e.g., viable cell counts, coliform groups, and pathogenic bacteria) and chemical contamination (e.g., heavy metals and shellfish toxins) were measured on raw oysters, a processing equipment, employees, and work areas. No total mercury, lead, paralytic shellfish poison, diarrheic shellfish poison, or norovirus was detected in any post-harvested oyster samples. However, the cadmium level ranged from 0.1-0.2 mg/kg. The viable cell count and Escherichia coli and coliform group levels in post-harvested oysters ranged from 4.00-4.54 log CFU/g, ND-210 MPN/100 g, and 110-410 MPN/100 g, respectively. The viable contaminating cell counts on employees, equipment, and work areas were in the range of 0.90-3.46 log CFU/100 cm<sup>2</sup>. Airborne bacteria in the work areas ranged from 0.60 to 1.81 log CFU/plate/15 min. Thus, no significant health risks were detected in the processing plant.
