Expression of CD30 in Testis and Epididymis of Adult Mice

CD30 is a member of tumor necrosis factor receptor (TNFR) superfamily and has pleiotropic functions including cell activation, proliferation, differentiation, and death, depending on cell types and stage of differentiation. Although CD30 expression has been described mainly in hematopoietic tissues, several types of nonhematopoietic tumors including embryonic carcinoma and germ-cell tumors express CD30. We examined CD30 distribution in the testis and epididymis from wild type and CD30-deficient mice. In the testis, spermatogonia, spermatocytes and Sertoli cells expressed CD30, but not in spermatids. Spermatogonia and spermatocytes near the basement membrane strongly reacted to anti-CD30. In the epididymis, CD30 expression was exclusively observed in luminal epithelia and some interstitial cells. Taken together, these results show a spatio-temporal regulation of CD30 expression in mouse testis and epididymis and suggest a possible role of CD30 in spermatogonia and spermatocytes.
생쥐난자의 체외수정과 배발달에서 다양한 정자농도의 효과에 관한 연구

체외수정율에 영향을 미치는 생쥐난자의 체외수정시 체외수정용 배양액소적의 양과 정자농도가 체외수정율 및 배발달율에 미치는 영향을 조사하였으며, 그 결과는 아래와 같았다. 1. 25,100 및 400㎕의 체외수정용 배양액소적에서의 체외수정율은 각각 44.0,83,3 및 50,3%였다. 2. 1×10^4,1×10^6 및 1×10^8의 정자농도를 이용하여 체외수정을 유도한 결과 체외수정율은 각각 34.1,77.1 및 50.6%였다. 3. 1×10^4,1×10^6 및 1×10^8에서 체외수정된 수정란의 정상적인 배반포기까지의 배발달율은 각각 0,40.0 및 30.0%였다. This study was carried out investigate whether the insemination drop volume and sperm concentration has been influenced upon the rates of in vitro fertilization, and to elevate utilizating efficiency and improve the fertilization efficiency of mouse oocytes in vitro. The results obtained in these experiments were summarized as follows: 1. The percentage of in vitro fertilization on differented insemination drop volume for 25, 100, 400㎕ was 44.0,83.3,50.3%respectively. 2. In order to determine more accurately at what sperm concentration during in vitro fertilization of oocytes can be fertilization by spermatozoa the oocytes were collected at 1416 hr times after hCG injection and inseminated for various sperm concentration. 3. The percentage of oocytes developed to 2cell stage following sperm concentration for 1×10^4,1×10^6 833, 960, 80%, repectively.

Production of therapeutic monoclonal antibodies (mAbs) using a plant platform has been considered an alternative to the mammalian cell-based production system. A plant-derived mAb CO17-1AK (mAbP COK) can specifically bind to various types of cancer cell lines. The target protein of mAbP COK is the epithelial cell adhesion molecule (EpCAM) highly expressed in human epithelial cancer cells, including breast and colorectal cancer cells. It has been hypothesized that its overexpression supports tumor growth and metastasis. A ganglioside is extended well beyond the surfaces of the various cell membranes and has roles in cell growth, inflammation, differentiation, and carcinogenesis. However, the regulation of EpCAM gene expression in breast cancers and the role of gangliosides in oncogenesis are unclear. Here, the purpose of this study was to determine the effects of mAbP COK on human breast cancer cell proliferation, apoptosis, and ganglioside expression patterns. Our results show that treatment with mAbP COK suppressed the growth of breast cancer cells and induced apoptotic cell death. It also upregulated the expression of metastasis-related gangliosides in breast cancer cells. Thus, treatment with mAbP COK may have chemo-preventive therapeutic effects against human breast cancer.

The human colorectal carcinoma-associated GA733antigen epithelial cell adhesion molecule (EpCAM) was initially described as a cell surface protein selectively expressed in some myeloid cancers. Gangliosides are sialic acid-containing glycosphingolipids involved in inflammation and oncogenesis. We have demonstrated that treatment with anti-EpCAM mAb and RAW264.7 cells significant inhibited the cell growth in SW620 cancer cells, but neither anti-EpCAM mAb nor RAW264.7 cells alone induced cytotoxicity. The relationship between ganglioside expression and the anti-cancer effects of anti-EpCAM mAb and RAW264.7was investigated by high-performance thin-layer chromatography. The results demonstrated that expression of GM1 and GD1a significantly increased in the ability of anti-EpCAM to inhibit cell growth in SW620cells. Anti-EpCAM mAb treatment increased the expression of anti-apoptotic proteins such as Bcl-2, but the expression of pro-apoptotic proteins Bax, TNF-α,caspase-3, cleaved caspase-3, and cleaved caspase-8were unaltered. We observed that anti-EpCAM mAb significantly inhibited the growth of colon tumors, as determined by a decrease in tumor volume and weight. The expression of anti-apoptotic protein was inhibited by treatment with anti-EpCAM mAb, whereas the expression of pro-apoptotic proteins was increased. These results suggest that GD1a and GM1 were closely related to anticancer effects of anti-EpCAM mAb. In light of these results, further clinical investigation should be conducted on anti-EpCAM mAb to determine its possible chemopreventive and/or therapeutic efficacy against human colon cancer.
Ganglioside GT1b Mediates Neuronal Differentiation of Mouse Embryonic Stem Cells

It has been reported that ganglioside GT1b is expressed during neuronal cell differentiation from undifferentiated mouse embryonic stem cells (mESCs), which suggests that ganglioside GT1b has a direct effect on neuronal cell differentiation. Therefore, this study was conducted to evaluate the effect of exogenous addition of ganglioside GT1b to an in vitro model of neuronal cell differentiation from undifferentiated mESCs. The results revealed that a significant increase in the expression of ganglioside GT1b occurred during neuronal differentiation of undifferentiated mESCs. Next, we evaluated the effect of retinoic acid (RA) on GT1b-treated undifferentiated mESCs, which was found to lead to increased neuronal differentiation. Taken together, the results of this study suggest that ganglioside GT1b plays a crucial role in neuronal differentiation of mESCs.

Cancer, which is expressed in various forms, is one of the leading causes of human death. Soamsan (SAS) is composed of ten medicinal herb, the prescription was made according to the principles of Oriental traditional medicine based on the concept of synergic effects and interaction of among the components. SAS has been used for the cancer therapy, but the mechanism of it's effect is not well known. In the present study, the cytotoxic effect of the SAS water extract on cancer cell lines was investigated by the method of MTT in A549 cell lines and the anti-angiogenic effect was shown in the assay of chorioallantoic membrane (CAM) and in the cornea of rat administerd orally with SAS water extraction. The viability of A549 cell lines was not affected by the whole extract of SAS but the n-Hexan fraction of SAS water extract showed strong cytotoxicity which was not seemed to be done by the apoptotic mechanism. SAS water extract showed inhibition effects of angiogenesis induced in the cornea of rat and CAM assay. As the above results, it is suggested that SAS can be a candidate for new prescription for cancer therapy.
