This study aimed to investigate the in vivo relevance of P-glycoprotein (P-gp) in the pharmacokinetics and adverse effect of phenformin. To investigate the involvement of P-gp in the transport of phenformin, a bi-directional transport of phenformin was carried out in LLC-PK1 cells overexpressing P-gp, LLC-PK1-Pgp. Basal to apical transport of phenformin was 3.9-fold greater than apical to basal transport and became saturated with increasing phenformin concentration (2-75 mM) in LLC-PK1-Pgp, suggesting the involvement of P-gp in phenformin transport. Intrinsic clearance mediated by P-gp was 1.9 mL/min while passive diffusion clearance was 0.31 mL/min. Thus, P-gp contributed more to phenformin transport than passive diffusion. To investigate the contribution of P-gp on the pharmacokinetics and adverse effect of phenformin, the effects of verapamil, a P-gp inhibitor, on the pharmacokinetics of phenformin were also examined in rats. The plasma concentrations of phenformin were increased following oral administration of phenformin and intravenous verapamil infusion compared with those administerd phenformin alone. Pharmacokinetic parameters such as Cmax and AUC of phenformin increased and CL/F and Vss/F decreased as a consequence of verapamil treatment. These results suggested that P-gp blockade by verapamil may decrease the phenformin disposition and increase plasma phenformin concentrations. P-gp inhibition by verapamil treatment also increased plasma lactate concentration, which is a crucial adverse event of phenformin. In conclusion, P-gp may play an important role in phenformin transport process and, therefore, contribute to the modulation of pharmacokinetics of phenformin and onset of plasma lactate level.
우편수송DSS를 위한 수송 MODULE 구축에 관한 연구

본 연구는 우편물의 효율적인 수송을 위해 복합연계수송 개념을 도입한 Multi-commodity, Multi-modal Network Model과 이를 토대로 정식화하고 이를 적용 한 LHGO programming Model를 제안하고자 한다. 구축된 LINGO Model은 응용 Application내에 장착되어지는 Module로 구성가능하며 이를 GIS Tool인 GEOmania를 이용한 우편물 수송 DSS을 위한 Application 개발에 활용하고자 한다.

The concentrative nucleoside transporter CNT1 and equilibrated nucleoside transporter ENT1 mediate the cellular uptake of naturally occurring pyrimidine and purine nucleosides and many structurally diverse anticancer and antiviral nucleoside analogs, thereby regulating drug responses or toxicity at the target site. The objectives of this study were to analyze interindividual variations in the cellular accumulation of gemcitabine and to examine the correlation between the uptake of gemcitabine and expression levels of CNT1 and ENT1 transporters. Gemcitabine was a substrate for both CNT1 and ENT1 with higher affinity to CNT1 than to ENT1. The difference in gemcitabine uptake was 4.8-fold in peripheral blood mononuclear cells (PBMCs) from 10 subjects. Among these, the CNT1- and ENT1-mediated uptake of gemcitabine was 14.3- and 16.5-folds, respectively. CNT1-mediated gemcitabine uptake showed a higher correlation with the CNT1 expression level than did ENT1-mediated uptake with ENT1 expression level. In conclusion, CNT1 seemed to be a major contributing factor to gemcitabine uptake in PBMCs and showed 14.3-fold inter-individual variations. However, ENT1-mediated uptake of gemcitabine might compensate for the total uptake of gemcitabine; therefore,the variation in the apparent accumulation of gemcitabine was smaller than that of the individual transporters.
Platinum Transporters and Drug Resistance

Cisplatin, a platinum coordinated complex, is a widely used antineoplastic agent for the treatment of metastatic tumors of the testis, metastatic ovarian tumors, lung cancer, advanced bladder cancer and many other solid tumors. The cytotoxic action of the drug is often thought to be associated with its ability to bind DNA to form cisplatin-DNA adducts. The development of resistance to cisplatin during treatment is common and constitutes a major obstacle to the cure of sensitive tumors. Although to understand the clinically relevant mechanisms of resistance, many studies have been aimed at clarifying the biochemical/molecular alterations of cisplatin-resistance cells, these studies did not conclusively identify the basis of cellular resistance to cisplatin. In this review, cisplatin resistance was discussed in terms of the relevant transporters, such as copper transporters (CTRs), organic cation transporters (OCTs) and multi-drug resistance related transporters (MDRs). These transporters seem to be contributed to cisplatin resistance through the reduction of drug accumulation in the cell. Better understanding the mechanism of cisplatin resistance associated with transporters will provide the useful informations for overcoming the cisplatin resistance.
Multistage opto-neuromorphic process system based on commercial optoelectric devices

This paper proposes a structure for opto-neuromorphic processes by using multistage connections of phototransmitters. The basic unit consists of a light-emitting diode and a photoresistor for a non-inverting photo signal processor. By constructing a cascade structure of these basic units, multistage photo transmission of input light signal is examined in pulse signals with various periods and duty cycles as the input to the first optoneuromorphic stage. This approach verifies that optical signals can be transmitted through our proposed structure and that by controlling the amount of the transmitted light between the basic units, the gain between stages can be controlled. It is demonstrated that this structure can be used for opto-neuromorphic processes by transmitting light signals as if transmitting signals between neurons with different weights and activation patterns. The proposed structure can be modified with an inverter circuit to build a photo-inverter and by using a combination of photo-transmitter and photo-inverters, it can serve as the basic structure to build more complicated opto-neuromorphic process systems.

This paper examines Im Sun-deuk’s two short stories, “Iryoil” (Sunday, 1937) and “Nazuke oya” (A Godmother, 1942). Im, an active participant in Korea’s socialist movement in the 1930s, turned to literature as her career at a time when Japanese colonial authorities had tightened their control over Korea to further the Japanese empire’s colonial expansion into China. In the process, the Japanese implemented an assimilation policy to mobilize the Korean people into the Japanese war effort. By employing the concept of confession in “Iryoil” and “Nazuke oya,” Im projects an autobiographical self onto her protagonists, both of whom are modern women striving to preserve their identities as socially conscious intellectuals committed to nationalism, socialism, and feminism. By positioning her protagonists as confessors to other characters in the texts, Im’s short stories portray intellectuals in colonial Korea as in conflict or harmony with her protagonists. Im’s protagonists embody female subjects who find a new kind of womanhood under Japanese colonial rule. However, in “Nazuke oya,” Im’s protagonist displays a more flexible and open attitude toward Koreans with attitudes and beliefs different from her own. In this sense, the act of confession as presented in Im’s texts is an attempt to build a bond and sense of community among the Korean people from a woman’s perspective, and at a time when Korean identity risked becoming obsolete.
