        인터루킨 - 2 정량을 위한 효소면역 측정법

        To establish an enzyme-linked immunosorbent assay technique to quantitate interleukin-2 (IL-2), five different monoclonal antibodies against human recombinant IL-2 were prepared. A pair of monoclonal antibodies recognizing different epitopes on the IL-2 molecule were selected for sandwich ELISA. The procedure involved utilization of one specific antibody immobilized on a microtiter plate and the other labelled with alkaline phosphatase. A linear relationship was achieved, using concentrations of IL-2 up to 100 ng/ml, when the absorbance of the reaction mixture was measured on a spectrophotometer. The whole assay procedure can be completed within five h. This assay technique for IL-2 would provide a simple and rapid alternative to other assays such as bioassay using IL-2 dependent CTLL cells.

        과학 전시물의 조도 적정성 평가 지표에 관한 연구- 국립중앙과학관 자연사관 전시물을 중심으로 -

        The lighting of the exhibition hall in a museum or science museum plays an auxiliary role in showing the purpose and characteristics of the exhibition and is a factor that strengthens the concentration and emotion of the exhibits. In order to evaluate the appropriateness of lighting and illumination for exhibits, 77 factors were selected and surveyed by 67 employees of the National Science Museums and 3 exhibition companies. 19 factors were selected by statistical analysis. Selected factors of average scores were more than 8, and the p-value of those were 0.05 or less compared to 40% of total factors. 15 science museum exhibits were evaluated by 19 factors and statistically analyzed. 10 factors were significantly different between more than every 3 exhibits in both the upper(average scores : more than 7) and lower(average scores : lower than 5) evaluation results. The 10 selected factors were investigated as efficient and essential factors for the evaluation of the lighting of the exhibits and the illumination of the exhibits. The 10 factors are 1) the distance between the lighting fixtures and the exhibits, 2) the installation location of the lighting fixtures, 3) the illuminance of each exhibit, 4) the uniformity of illuminance, 5) differential illuminance (internal illuminance of the exhibit > illuminance on the panel > external illuminance of the exhibit), 6) light focus on the exhibits, 7) lighting display effect, 8) readability, 9) visibility, and 10) ease of repair. As the results of evaluating 15 individual exhibits with 10 and 19 factors, the excellent exhibits for illuminance appropriateness were 1) endemic plants of Korean Peninsula, 2) grypania, the first eukaryote, 3) bacteria kingdom, etc., 4) mesozoic fish fossils of Korean Peninsula, 5) leading fossils, 6) Ediacara fossil fauna, and 7) moon rock, rock from the moon(only in 10 factors evaluation) and were significantly different from 1) defensive features of herbivorous dinosaurs, 2) weapons of carnivorous, and 3) Cenozoic mammals in Korean Peninsula within 95% confidence. The evaluation results of these three exhibits were significantly lower in more than 10 factors of 19 factors and 9 factors of 10 selected factors. Evaluation factors of illumination can be used for the design and production stage, but they did not give a specific answer on the appropriate illuminance for the exhibition in different conditions. Therefore, in the next study, we plan to prepare and present a standard for an appropriate illuminance for the surrounding lighting environment.

        과학 영상 콘텐츠 평가 지표에 관한 연구 - 유튜브 채널에 공개된 과학 영상물을 중심으로 -

        Currently, a large number of science video contents are being produced and provided. However, it is difficult to judge successful video contents in the planning, shooting, and editing stages because there are no evaluation method. The purpose of this study is to develop evaluation factors that can be used by anyone and can be used in all stages of planning, shooting, and editing. The appropriate factors were selected through the following process. First, appropriate evaluation factors were found for the development of evaluation factors targeting science videos. Second, evaluation factors were verified through the evaluation of scientific video. Third, appropriate evaluation factors were determined after comparing and analyzing the evaluation results and the number of video views. After a survey, 51 excellent factors were selected with a mean value of 7.5 or higher and a significant difference from the bottom 25% of 196 factors with a 95% confidence level. 20 science videos were evaluated with 51 excellent factors, and each factor was statistically analyzed. 16 selected evaluation factors that could statistically distinguish between good and poor with 95% confidence level were selected. The evaluation results of 16 factors and the number of video views were compared and analyzed, but some science videos are a large difference in the rankings of the two results. Therefore, these videos showing different results were analyzed, and additional factors were discovered by identifying the characteristics of the video. 20 science videos were evaluated again with 20 renewed factors, and each factor was analyzed. 7 evaluation factors that could statistically distinguish between good and poor with 95% confidence level were selected. The final selected factors were: (1) arousing and resolving curiosity and wonder, (2) voice actors and voice delivery, (3) fun and interest, (4) interesting title, (5) empathy, communication and understanding, (6) immersion and concentration, and (7) attention. The selected 7 evaluation factors can be used as a predictive index for excellent quality and high number view’s videos, and the developed evaluation index is necessary to reduce time, manpower, and cost for video production. However, there is a disadvantage that under exceptional conditions, the number of views may be higher or lower than expected. For example, videos that are highly relevant, such as timely topics, issues, unknowns, challenges, and space, will be higher views, and have relatively lower for the fun and interest factor will be lower views.

        hCG 단일클론 항체의 생산 및 특성 연구

        Pure subunits of human chorionic gonadotropin (hCG) were obtained by chromatographic separation using DEAE-Sephadex A-25 and Sephadex G-75 for the immunization. The spleen cells obtained after immunization of either w-hCG or β-subunits were fused with myeloma cells and twelve hybridoma cell lines were established. The results showed that the fusion efficiency and antibody producibility varied depending on the myeloma cell lines. The cell line P3×63-Ag8.653 was found to be a better fusion partner than SP2/0 for the production of antibodies from hybridoma cells. For the formation of ascitic fluid in mice, Incomplete Freund`s Adjuvant(IFA) was a better priming agent than pristane. The yield of specific monoclonal antibodies by the immuno-affinity purification method was 1.6-3.4 ㎎/㎖ ascitic fluid. Seven clones were studied for the specificity to w-hCG and its subunits using ^(125)I-labeled tracer and the clones were classified into 3 groups. The equilibrium affinity constants of the seven antibodies ranged from 5×10^7 M^(-1) to 8×10^8 M^(-1).

      • Production and Characterization of Monoclonal Antibodies Specific to human Chorionic Gonadotropin and its Subunits

        hCG subunit를 순수 분리정제하여 w-hCG와 ${\beta}$-subunit를 각각 Balb/c 생쥐에 면역 주사하여 얻은 비장세포를 myeloma 세포와 융합하여 12개의 하이브리도마 세포주를 얻었다. Myeloma 세포주에 따른 세포융합의 효율과 hCG 항체 생성율을 조사했을 때 $P3{\times}63$-Ag 8.653이 SP2/0 보다 우수했으며, priming agent에 따른 복수생성율을 조사했을 때 IFA가 pristane 보다 우수함을 알았다. 하이브라도마 클론에서 유발된 복수액을 immuno-affinity 정제했을 때 특이 항체의 양은 클론에 따라 복수 ml 당 1.6-3.4 mg을 얻을 수 있었으며 모두 $IgG_1$으로 나타났다. 이 중 7개의 단일클론 항체클론에 대해 w-hCG 및 ${\alpha}$와 ${\beta}$-subunit에 대한 친화항수를 조사했을 때 $5{\times}10^7/M$에서 $8{\times}10^8/M$로 나타났으며 이 특이 친화성에 의해 항체를 3개의 그룹으로 분류할 수 있었다.         사지단지증아의 1례

        저자들은 사지단지증의 1예를 경험하였기에 문헌고찰과 함께 보고하는 바이다. A case of achondroplasia which is a rare congenital anomaly was presented. Four fifths of achondroplastic patients occur as a fresh mutation, older fathers having an increased risk of having children with this condition. Once originated, the deformity can be transmitted to half the affected person`s offspring6.15. The literatures on the achondroplasia are briefly reviewed.
