        해양생물 크릴(Euphausia superba)이 흰쥐의 혈청 유리지방산 및 전해질 농도에 미치는 영향

        본 연구는 식이조성에 동결 건조한 krill (Euphausia superba) meal을 첨가 하였을 때 Sprague Dawley계 수컷 흰쥐의 유리지방산(non-esterified fatty acid, NEFA), blood urea nitrogen (BUN), creatinine, uric acid 및 전해질에 미치는 영향에 대하여 조사를 하였다. 이조성은 기본식이를 급여한 대조군인 GC군(krill meal 미함유)을 비롯하여 기본식이에 10.0%, 20.0%, 30.0%의 krill meal 급여군을 각각 G10군, G20군, G30군으로 구분하였다. 5주간 실험 사육한 결과, 흰쥐의 NEFA와 BUN 농도는 krill meal 급여군(G10군, G20군, G30군)에서 대조군(GC 군)보다 유의적으로 감소된 결과를 보였고, creatinine과 uric acid 농도는 G10군에서는 GC군과 유의적 차이가 없었으나, G20군과 G30군에서 유의적으로 감소하였다(p<0.05). 전해질 농도에서 total calcium (T-Ca)은 G20군과 G30군에서 유의 적으로 증가하였으며, 인(Pi) 농도는 각 군간에 유의적 차이가 없는 것으로 관찰되었다(p<0.05). The purpose of this study was to investigate the change of hematology and serum chemistry values on Sprague-Dawley rats, while used krill (Euphausia superba) meal diet for 5 weeks. Seven-week-old male rats were divided into four groups (n=6) and fed experimental diets containing three different krill meal contents and control group; 10.0% krill meal (G10), 20.0% krill meal (G20), 30.0% krill meal (G30), and control group (GC). Concentrations of non-esterified fatty acid (NEFA), blood urea nitrogen (BUN) in serum were significantly lower in the G10, G20, G30 than GC group (p<0.05). Statistically significant differences, at the confidence level of 95%, for the creatinine, uric acid, electrolyte (T-Ca) parameters in the sera were observed in G20 group, G30 group. The concentration of electrolyte (Pi) in serum was no significant difference among the groups (p<0.05). The results indicate that a krill meal diet was effectively reduce the NEFA.

        와송(Orostachys japonicus A. Berger) 추출물의 총 페놀, 플라보노이드 함량 및 항산화 활성 비교

        The purpose of this study was to measure the bioactivity and antioxidant activity of Orostachys japonicus A. Berger. Orostachys japonicus A. Berger have been known to contain functional materials such as kaempferol, hydroquinone, methyl gallate, quercetin, gallic acid etc. To identify the main functional materials of Orostachys japonicus A. Berger, the contents of flavonoid and phenol were measured. We extracted Orostachys japonicus A. Berger powder from four solvents such as chloroform:methanol (CM, 2:1, v/v), distilled water (DW), 70% methanol, 70% ethanol. After that, this study determined tannin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power and reducing power of Orostachys japonicus A. Berger extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of Orostachys japonicus A. Berger extracts was higher in the order of 70% ethanol, 70% methanol, DW and CM (p<0.05). The results showed that antioxidant activity of Orostachys japonicus A. Berger extracts supposed to affect by the total phenol and flavonoid contents.

        치자(Gardenia jasminoides Ellis) 과피의 용매별 추출물의 Flavonoid 함량 및 항산화 활성 비교

        The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides fructus Ellis (GJE) in Namhae, Korea, following some established methods. CM (Chloroform:Methanol, 2:1, v/v), 70% ethanol, and n-butanol extracts were collected. Flavonoid content and value as a functional food ingredient of GJE peel was investigated through assessing antioxidant [DPPH (1,1`-diphenyl-2-picrylhydrazyl), ABTS (2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid)], and hydroxyl radical scavenging activities; superoxide dismutase like ability; ferrous ion-chelating capacity; and tannin content by solvent extraction. Solvent extract antioxidant activities significantly increased (p<0.05) at increasing concentrations (0.2, 0.4, 0.6 mg/mL). GJE peel extracts were less active than the positive control [ascorbic acid, BHA (butylated hydroxyanisole), and EDTA (ethylenediaminetetraacetic acid disodium salt dihydrate)]. Based on the results of this study, GJE peel could be used as a natural antioxidant source due to its high antioxidant activity and bioactive compound content.

        블랙 초크베리 추출물의 항산화 활성 및 아질산염 소거 활성

        The purpose of this study was to measure the antioxidant activity and bioactivity of black chokeberry (Aronia melanocarpa). The black chokeberry was known to contain many physiologically active substance, such as ascorbic acid, anthocyanin, quercetin. We extracted black chokeberry powder from four solvents such as chloroform:methanol(CM, 2:1, v/v), distilled water (DW), 70% ethanol, 70% methanol. After that, we determined anthocyanin, total phenol, flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power and Nitrite scavenging activity of black chokeberry extracts and as results of comparing each extract. respectively. From the above results shows that antioxidant activity and bioactivity of black chokeberry extracts was the highest in 70% methanol (p<0.05). The results suggest that black chokeberry can be used as nutraceutical foods and natural antioxidant.

        치자(Gardenia jasminoides Ellis fructus) 껍질 추출물의 항산화 활성

        치자 껍질의 항산화 활성을 측정하기 위하여 70% methanol, distilled water (DW) 및 ethylacetate (EA)의 3가지 용매를 사용한 추출물의 용매 별 flavonoid 함량 및 항산화 활성, 금속 chelating 능력 측정을 통하여 치자 껍질의 기능성 식품 재료로서의 가치를 검토한 결과, 치자 껍질의 proanthocyanidin 함량은 48.165±0.811 mg CE/g DW로 나타났으며, 추출 수율은 70% methanol (36.26%), DW (39.87%), EA (2.88%) 로 나타났다. 추출 용매 별 항산화 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 증가할수록 유의적으로 증가하였으며, control로 사용된 ascorbic acid, BHA, EDTA 보다는 낮은 활성이 관찰되었다. Flavonoid 함량(mg QE/g)은 70% methanol (0.416), DW (0.225), EA (0.212) 순으로 확인되었으며 다른 항산화 분석 및 금속 chelating 능력 실험에서도 이와 유사하게 관찰되어 모든 분석에서 70% methanol 추출물이 가장 활성이 강한 것으로 나타났다. 이상의 결과를 미루어 볼 때 치자 껍질의 용매 별 flavonoid 함량에 따라 항산화능, 금속 chelating 능력이 증가하는 것으로 사료되며, EA 추출물을 제외한 다른 추출물에서의 수율과 항산화 활성은 높은 수준으로 관찰되었다. 따라서 치자의 껍질은 proanthocyanidin과 flavonoid 화합물을 다량 함유하고 있으며 그로 인한 높은 항산화 활성과 생리활성을 가지고 있어 기능성 식품 및 천연항산화제로서의 가치가 매우 기대된다. The purpose of this study was to measure the bioactivity and antioxidant activity of peel from Gardenia jasminoides Ellis fructus (GJE). GJE have been known to contain functional materials such as crocin, crocetin, geniposide, gardenosid, geniposidic acid, iridoid glycosides etc. We were separated into GJE peel. After that, we determined proanthocyanidin. GJE peel were extracted by 70% methanol, ethyl acetate (EA) and distilled water (DW) of three solvents. To investigate by the solvent extract of flavonoid content and value as a functional food ingredient of GJE peel through antioxidant activities (DPPH radical scavenging activity, ABTS radical scavenging activity, superoxide dismutase like ability, hydroxyl radical scavenging activity, ferrous ion-chelating capacity) were performed. Solvent extract antioxidant activity of increasing concentrations (0.2, 0.4, 0.6 mg/mL) were significantly increased (p<0.05). GJE peel extracts showed lower activity than positive control (ascorbic acid, BHA, EDTA). These results, by a solvent of peel were found that the relationship with the increase of flavonoid content increased physiological activity. The antioxidant activity of the extract from the other except for the EA extract on peel was observed at a high level. The results suggest that GJE peel can be used as nutraceutical foods and natural antioxidant.

        치자(Gardenia jasminoides Ellis) 씨 추출물의 질소산화물 소거능 및 지질과산화 저해능에 관한 연구

        치자 씨 추출물이 항산화력 및 지질과산화 저해능에 미치는 영향을 알아 보고 치자의 기능성식품 소재로서의 가치를 검토하기 위하여 실험을 수행한 결과, 치자 씨의 anthocyanin 함량을 측정한 결과 2.201±0.516 mg/100 g DW로 나타났으며, 치자 씨의 용매 별 추출 수율은 chloroform:methanol (CM, 2:1, v/v) 36.39%, 70% ethanol (27.32%), n-butanol (26.23%) 로 확인되었다. 추출 용매 별 항산화 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 증가할수록 유의적으로 증가하였으며 positive control로 사용된 ascorbic acid, BHA, trolox 보다는 낮은 활성이 관찰되었다. 치자 씨의 total phenol 함량(mg CAE/g)은 CM (32.50), 70% ethanol (30.09), n-butanol (11.07) 추출물 순으로 n-butanol 추출물에서 가장 적은 함량을 보였으며, Nitric oxide (NO) radical 소거능에서는 CM (76.97~84.24%), 70% ethanol (74.10~79.99%), n-butanol (30.66~37.15%) 추출물 순으로 관찰되었다. Nitrite (NO2) 소거능은 CM (33.53~43.23%), 70% ethanol (32.40~35.98%), n-butanol (24.72~28.14%) 순으로 관찰되었다. β-carotene 탈색 저해능은 CM (23.73~44.70%), 70% ethanol (22.03~41.32%), n-butanol(16.00~27.87%) 순으로 확인되었다. Reducing power (optical density)는 70% ethanol (0.073~0.182), CM (0.057~0.154), n-butanol (0.028~0.079) 순으로 관찰되었다. 지질과산화 저해능은 씨 추출물 중 CM (53.26~76.56%), 70% ethanol (52.97~76.56%), n-butanol (38.54~53.33%) 순으로 나타났다. 이에, 치자씨 추출물은 천연 항산화제로서 기능성 식품의 가치가 높을 것으로 판단된다. The object of this study was to measure the bioactivity and antioxidant activity of seed from Gardenia jasminoides Ellis fructus (GJE). GJE seeds were performed the extraction of them by chloroform:methanol (CM, 2:1, v/v), 70% ethanol and n-butanol. Sequentially, total phenol content, nitrogen oxide scavenging activity, antioxidant activity and lipid peroxidation inhibition activity of the extracts were investigated. Solvent extract bioactivity of increasing concentrations (0.2, 0.4, 0.6 mg/mL) were significantly increased (p<0.05). GJE seed extracts showed lower activity than positive control (ascorbic acid, BHA, trolox). The highest concentration of CM extracts was obtained in the same manner as the results of analysis of the total phenol contents of the GJE seed, and 70% ethanol extract showed the highest activity of reducing power. The water soluble carotenoids crocin and flavonoid were effective. As a result of this experiment. the seeds of GJE showed excellent antioxidant, and lipid peroxidation inhibitory properties.

        남해산 치자(Gardenia jasminoides Ellis fructus) 껍질의 생리활성 및 지질과산화 저해 활성에 미치는 영향

        치자 껍질의 phytic acid 함량과 CM (chloroform:methanol, 2:1, v/v), n-부탄올 및 70% 에탄올을 사용한 추출물의 용매(3가지) 별 총 페놀 함량 및 항산화 활성 능력 등을 통하여 치자 껍질의 효용 가치를 검토한 결과, phytic acid 함량은 4.966±0.996 mg PAE/g DW (dry weight)로 나타났으 며, 용매 별 생리 활성은 농도(0.2, 0.4, 0.6 mg/mL)가 높아질수록 유의적으로 증가하였으며(p<0.05), 양성대조군으로 사용된 ascorbic acid, BHA, trolox 보다는 각 농도에서 낮은 활성이 관찰되었다. 치자 껍질의 총 페놀 함량은 CM, n-부탄올, 70% 에탄올 추출물 순으로 CM 추출물에서 39.74±0.15 mg CAE/g으로 가장 높았으며, nitric oxide (NO) 라디칼 소거능과 지질과산화 저해능은 용매 별로 CM > 70% 에탄올 > n-부탄올, nitrite (NO 2 ) 소거능은 n-부탄올 > CM > 70% 에탄올, β-carotene 탈색 저해능과 환원력은 CM > n-부탄올 > 70% 에탄올 순의 활성이 나타났다. 이상의 결과, 치자 껍질의 용매 별 총 페놀 함량 순과 항산화력, 환원력은 일치 하였으며, 질소산화물 소거능과 지질과산화 저해능 분석에서는 일치하지 않은 것으로 확인되었다. 이는 질소산화물 소거능과 지질과산화 저해능은 페놀 성분 보다 다른 생리활성물질이 더 큰 영향을 주는 것으로 추정된다. 이에, 치자 껍질은 질소산화물 소거능, 항산화능 및 지질과산화 저해능 등이 우수한 것으로 나타나 기능성 식품 소재로서의 가치가 있는 것으로 판단된다. The object of this study was to measure the bioactivity and lipid peroxidation inhibition activity of peel from Gardenia jasminoides Ellis fructus (GJE) in Namhae Korea. The amount of phytic acid was also determined. Extraction was performed using three solvents, CM (choloform:methanol, 2:1, v/v), n-butanol and 70% ethanol. To investigate by the solvent extract of total phenol content and value as a functional food ingredient of GJE peel through nitrogen oxide scavenging activity, antioxidant activity, reducing power and lipid peroxidation inhibition were performed. The bioactivities of the extract solvents increased significantly with increasing concentrations (0.2, 0.4, 0.6 mg/mL, p<0.05). The total phenol contents of GJE peel extracts were highest in CM (39.74±0.15 mg CAE/g) extract. The order of total phenol contents, antioxidant activity and reducing power of the solvents in the GJE peel were the same, in the analysis of nitrogen oxides scavenging activity and lipid peroxidation inhibition, it was confirmed the results were inconsistent. As a result, the GJE peel showed excellent bioactivities. Considering the extraction yield and various physiological activities, it is considered that efficiency is better when extracted from CM and 70% ethanol extracts.

        추출 용매에 따른 아사이 베리(Euterpe oleracea Mart.)의 생리활성 및 항산화 활성 비교

        The acai berry (Euterpe oleracea Mart.) contains vitamin B complex, vitamin C, anthocyanin and so on. Especially acai berry was seen as nutritionally comparable to blueberry and related berries. The acai berry has significant aging-reducing properties. Compounds have been found to have anti-aging and antioxidant components. Acai berry was extracted with 70% methanol, 70% ethanol and CM (chloroform:methanol=2:1, v/v). After sample and reagents of each experiment was reacted, DPPH radical scavenging activity and ABTS radical scavenging activity, ferric reducing antioxidant power, reducing power were measured to determine the antioxidant capacity, and as results of comparing each extract. Ethanol (70%) extraction was measured highest. Anthocyanin, total phenol, flavonoid also appeared similar to the results. In addition, the antioxidant activities of the extraction solvents were increased significantly with increasing concentrations, but showed lower antioxidant activity than the positive control (ascorbic acid). As a result, antioxidant activities of sample supposed to affect by the anthocyanin, phenol and flavonoid contents.
