Most of the discussion on the final stress avoidance phenomenon in recent metrical stress theory and the Optimality-Theoretic(OT) approach has focused on the prosodic status of a single, word-final syllable. If we extend this to the final two syllables of a word, however, we discover never-seen-before patterns of word-final stress avoidance, which include an asymmetry in typology between iambic and trochaic stress systems. Within the framework of the OT approach, this study explores the relationship between the stress patterns of the penultimate and ultimate syllables and NONFINALITY constraints in terms of final stress avoidance phenomenon. In particular, I note that there is no consistency in the way NONFINALITY(FT) constraint deals with final stress avoidance since it encompasses two unrelated stress phenomena, such as antepenultimacy and Foot Extrametricality. I argue that the double functions of the NONFINALITY(FT) constraint be taken over partly by a more specific NONFINALITY(σweak-in-foot) constraint and partly by a new alignment constraint, ALIGN-R(RIGHTMOSTNONHD(WD), PRWD), respectively.       • Aptamer-Directed Covalent Conjugation of DNA to an Intact Protein

        A specific DNA conjugated to an active protein is not only used to identify, manipulate and functionalize the protein but also, in itself, a programmable linker that can connect the protein to other molecules. However, maintaining a protein activity after the conjugation remains as a critical issue. In this work, we propose the use of guanine analogue for the aptamer-directed covalent conjugation of DNA to an unmodified protein. The guanine analogue, formed by nitrosation of guanine, is able to mediate activation-free crosslinking with nearby proteins. We invented a synthetic way to prepare this reactive base-containing aptamer and successfully confirmed that the newly synthesized aptamer forms a covalent linkage to the aimed location of protein with exceptionally high specificity. As a model, we demonstrate that a heparin-binding domain of thrombin is site-specifically conjugated with the selectively bound aptamer wherein the reactive base is included in a sequence-specific manner.

      • 사람의 간암 세포주에서 p53 유전자의 기능과 p21^(CIP1) 유전자 발현의 상관관계에 관한 연구

        Expression of p21^(CIP1) gene has been known to be regulated by p53. We have examined whether basal expression of p21^(CIP1) is directly correlated to the status of p53 gene expression in seven human hepatoma cell lines. Six hepatoma cell lines except Hep3B expressed significant amount of p53 and Rb transcripts. However, only three of the six cell lines(HepG2, SK-HEP-1, HT-17) showed detectable basal expression of p21^(CIP1) transcripts, whereas three other Hub cell lines(Huhl, 4 and 7) originated from Japan did not. Neither p21^(CIP1) nor p53 transcripts were observed in the Hep3B cell line. There, was a considerable correlation between the status of functional p53 and p21^(CIP1) gene expression in these hepatoma cells, since loss of p21^(CIP1) gene expression was observed in the Huh7 cells which have mutated p53 and in the Huhl and Huh4 cells which have HBV containing HBX gene. Treatment of HepG2 hepatoma cells with 25 nM testosterone resulted in higher mitogenic activities by the ³H-thymidine incorporation assay. However, expression of p21^(CIP1) gene did not seem to be involved in mitogenic enhancement by testosterone in these cells.

        트레드밀 훈련과 Lee Silverman Voice Treatment(LSVT)-BIG 프로그램이 파킨슨병 환자의 균형 및 보행능력에 미치는 효과 : 단일사례연구

        This study aimed to investigate the effect of treadmill training and Lee Silverman Voice Treatment(LSVT)-BIG program on the balance and gait functions of a patient with Parkinson's disease. A participant was a person who suffered from idiopathic Parkinson's disease. We used a single-subject experimental research design with reversal phases (A-B-A). Experimental session was eight sessions in three phases (a total of 24 sessions): (1) baseline phase (A), (2) intervention phase (B), and (3) withdrawal phase (A'). During all phases, a participant underwent daily routine rehabilitation therapy, including mat exercise, flexibility training, and therapist-guided functional activity. Furthermore, treadmill training and LSVT-BIG was additionally performed for 70 minutes, once per day in the intervention phase, which consisted of warm-up (5 minutes), LSVT-BIG (30 minutes), treadmill training (30 minutes), and cool-down (5 minutes). During the intervention phase, the TUG and 10MWT scores improved by 3.75 seconds (36.89 seconds to 33.14 seconds) and 2.28 seconds (26.07 seconds to 23.79 seconds) in average, respectively. And gains were maintained after the intervention. Furthermore, the BBS and DGI scores improved by 17 points (4 points to 21 points) and 11 points (6 points to 17 points) in average, respectively. These findings suggest that treadmill training and LSVT-BIG may be helpful to enhance balance and walking functions in a person with Parkinson’s disease. Further studies need to support these results.

      • Role of Prostaglandin E in Modulating Immune Response in Lymph Nodes Draining the Uterus during the Implantation Period in Rats

        본 교실의 연구결과에 의하면 토끼와 흰쥐에서 수정란 착상시기에 peripheral lymphocyte와 thymocyte의 활성도가 저하될 뿐만 아니라 착상기간중 생성되는 prostaglandin E(PGE)의 생리적인 농도로도 peripheral lymphocyte와 thymocyte의 활성도가 억제되었다. 그러므로 본 연구에서는 흰쥐의 착상시기에 전신적인 면역기능 뿐만 아니라 국소적으로 DLN lymphocyte의 활성도가 억제되는지를 관찰하고 PGE가 어떠한 기전으로 모체의 면역기능을 억제하는가를 관찰하여 다음과 같은 결과를 얻었다. 1. 흰쥐의 착상시기에 DLN lymphocyte의 활성도가 임신하기 않은 흰쥐의 DLN lymphocyte에 비하여 통계적으로 유의하지는 않으나 저하되었으며 이러한 저하현상은 100% 흰쥐에서 관찰되었다. 2. 착상시기의 DLN lymphocyte에 prostaglandin 합성억제제인 indomethacin(ID)를 처리하면 DLN lymphocyte의 활성도가 통계적으로 유의하게 증가하였다. 그러나 임신하지 않은 흰쥐의 DLN lymphocyte의 활성도는 증가되어 있으나 $PGE_2$를 전처리하면 DLN lymphocyte의 활성도가 유의하게 억제되며 $PGE_2$를 전처치한 후 ID를 처리하면 DLN lymphocyte의 활성도가 $PGE_2$로 전처치하지 않고 ID를 처리한 경우에 비하여 유의하게 증가하였다. 그러나 $PGE_2$ 대신estradiol, progesterone 및 hCG를 전처치하였을 경우에는 ID 처리로 DLN lymphocyte의 활성도가 증가하지 않았다. 3. 임신하지 많은 흰쥐의 DLN lymphocyte에 $PGE_2$를 전처리하면 PGE-producing cell이 유도되어 PGE 생성이 증가하는지를 확인하기 위하여 $PGE_2$를 전처리하고 Con A를 처리한 후 배양액의 PGE를 정량한 결과 PGE를 전처리하지 않은 DLN lymphocyte에 비하여 유의하게 PGE 생성이 증가하였다. 이상의 결과로 보아 흰쥐의 착상시기에는 모체의 DLN lymphocyte의 활성도가 저하되며, 특히 PCE는 PGE-producing cell을 유도함으로써 착상시기의 모체의 면역기능에 영향을 주는 것으로 생각된다. In our previous studies, it was found that activities of maternal peripheral lymphocytes and thymocytes were depressed during the implantation period in rats and rabbits. This study was therefore attempted to clarify further this immunosuppression locally by determining lymphocyte response in lymph nodes draining the uterus (DLN) and to elucidate the mechanism by which prostaglandin E (PGE) modulates immune response during the implantation process in rats. As compared with non-pregnant rats, the response of DLN lymphocytes to concanavalin A (Con A) was depressed during the implantation period in 100% of rats studied. The activity of DLN lymphocytes depressed on day 8 of pregnancy was, however, restored partially by the treatment of indomethacin (ID), indicating that prostaglandin (PG) might be one of factors responsible for immunomodulation during the process of implantation. DLN lymphocyte activity in non-pregnant rats was suppressed if PGE was pre-treated prior to Con A and this suppression was partially restored by the treatment of ID. Furthermore, DLN lymphocytes pre-treated with PGE produced PGE in vitro and this PGE production was blocked by the treatment of ID, suggesting that PGE induced PGE-producing cells. However, the pretreatment of estradiol, progesterone, and hCG at doses enough to suppress lymphocyte activity was ineffective in inducing PGE-producing cells. From these results, it is suggested that PGE induces PGE-producing suppressor cells, thereby increasing PGE concentration and PGE in turn depresses maternal local immune response as well as systemic immune response during the implantation period in rats.

        운율음운론에 암묵적으로 드러난 강세의 독립성(taxonomy)을 재검토하기 위해 리듬기반 강세와 어휘강세를 비교한다. 강세의 독립성은 패러다임 기반의 강세 경우 심각한 도전을 받는데 스페인어의 동사체계가 그렇다. 본 논문에서는 스페인어의 동사강세가 순수하게 어휘 엑쎈트 체계로 분석될 수 있으며 따라서 이는 강세의 독립성을 위협하는 경우가 아니라고 결론지으며 이를 스페인어 비동사의 리듬 강세와 대조시킨다.
