Phytoplankton community was studied in relation to a typhoon at Bok-gyo Bridge area in Juam Lake, Korea. In August 31, 2000, a typhoon (Prapiroon) was passed by Juam Lake with great power enough to destroy summer stratification of Juam Lake. Destratification resulted in temporal mixing of the whole water column and changed the physical and chemical properties of water bodies, and caused the changes of the biological properties. The transparency decreased from 195 cm before the typhoon to 84 cm after the typhoon with the resuspension of the bottom sediment. In the vertical distribution of the phytoplankton population, the maximum population was measured at depth of 2 m before the typhoon. However, immediately after the typhoon, the population distributed evenly throughout the entire water layers. The carbon biomass of the phytoplankton was also highest at the depth of 2 m before the typhoon, but immediately after the typhoon, it was uniformly distributed throughout the whole water layers. The vertical profiles of the concentrations of chlorophyll a, however, did not show a significant difference before and after the typhoon. The typhoon induced destratification and restratification altered the taxa of the phytoplankton. The major dominant phytoplankton taxa before the typhoon was diatoms including Aulacoseira granulata, but the green algae overwhelmed the diatoms in cell number and biomass after the typhoon. The object of this research is to study effect of the waste vegetable oil on mycelium and exopolysaccharide production from Pleurotus sajor-caju PSC 14. Among various culture temperatures, the mycelium and exopolysaccharide production were highest at 29°C. However, they decreased over and below 29°C. When waste vegetable oil 2.0g/L was used for 5 days, the maximum mycelium production was obtained. On the other hand, in the case of exopolysaccharide production, it was obtained at waste vegetable oil of 1.0g/L. Among various surfactants, Tween 80 was the best one for effect on the mycelium and exopolysaccharide production from submerged liquid fermentation of Pleurotus sajor-caju PSC 14. Fed batch fermentations were carried out in 20L airlift bioreactor containing 10L working volume. When Tween 80 was used, the maximum mycelium and exopolysaccharide concentration were respectively 5.62 g/L and 0.67g/L after 8 days of culture. The product yield from oil was 0.12g/g consumed carbon source. These results show that Tween 80 can be useful for the mycelium and exopolysaccharide production from submerged liquid fermentation of Pleurotus sajor-caju PSC 14 using waste vegetable oil.

This research investigated extraction and antioxidant activity of polysaccharides produced from liquid culture of Pleurotus sajor-caju PSC-14. When the ratio of water to sample was increased from 10:1 to 40:1 (mL/g), the polysaccharides extraction yields were increased from 47.2 to 100%. On the other hand, when the ratio increased from 50:1 to 60:1, the yields of polysaccharides extraction were decreased from 80.2% to 62.3%. Extraction yields of polysaccharides were increased with extraction temperature. Especially, when the extraction temperature was increased from 40 to 80 oC, extraction yields of polysaccharides were increased from 41.2 % to 82.6%. On the other hand, at the extraction temperature of 100 oC, there was a little decrease in polysaccharides yield. Maximum extraction yield of polysaccharides was obtained at the extraction temperature of 90 oC. To investigate effect of polysaccharides concentration on antioxidant activity, DPPH scavenging activities were measured. When the polysaccharides concentration was 15.0 mg/mL, the maximum DPPH radical scavenging activity was obtained, 69.25%.

The work on the eutropication of Youngsan river was conducted from Jul. 1997 to Jun. 1998. The value of water temperature in the study area varied from 4℃ to 32℃ with an annual mean of 17.5℃. DO varied from 4.0㎎/ℓ to 13.5㎎/ℓ with an annual mean 10.2 ㎎/ℓ. BOD varied from 0.8㎎/ℓ to 6.4㎎/ℓ with an annual mean 3.1㎎/ℓ. COD varied from 4.6㎎/ℓ to 9.0㎎/ℓ with an annual mean 6.38㎎/ℓ. Nitrate varied from 1.58㎎/ℓ to 6.77㎎/ℓ with an annual mean of 3.475㎎/ℓ. Total-nitrogen varied from 2.14㎎/ℓ to 8.38㎎/ℓ with an annual mean of 5.083㎎/ℓ. Total-phosphate varied from 0.035㎎/ℓ to 0.588㎎/ℓ with an annual mean of 0.128㎎/ℓ. Chl.a varied from 1.8㎍/ℓ to 75.0㎍/ℓ with an annual mean of 19.55㎍/ℓ. Studies on physico-chemical factors and phytoplankon were conducted in the Kwang yang Bay from September 1986 to June 1987; Results are as followes; water temperature was varied from 5.0℃ to 24.0℃,; dissolved oxygen (D.O) from 7.21㎎/ℓ, to 14.80㎎/ℓ with an annual average of 10.11㎎/ℓ; salinities was from 23.0‰ to 34.0‰ with an annual average of 29.5‰ ; pH range was from 7.4 to 8.0 with an annual average of 7.8; suspended solides (S.S) was from 9.00㎎/ℓ to 34.08㎎/ℓ with an annual average of 20.27㎎/ℓ; particulate organic matters (POM) was from 1.31㎎/ℓ to 24.71㎎/ℓ with an annual average of 5.77㎎/ℓ; transparency was the range of from 1.0 to 5.1m. ; ammonia was from 0.20㎍/ℓ to 4.50㎍/ℓ; nitrite was from 0.06㎍/ℓ to 9.35㎍/ℓ; phosphate was from 1.68㎍/ℓ to 137.20㎍/ℓ . Phytoplankton species were identified total 137 taxa composed of 4 phylum, 10 class, 24 order, 24 family, 11 var. species, the succession of dominant species was showed Chaetocerous curvisetes in November, Eucampia zoodiacus in Febulary, Stephanopyxis nipponica in April and Rhizossolnia stolterforthii in June. Phytoplankton standing crop was from 71 to 1,521×10 exp (3) cells/ℓ with an annual mean of 333×10 exp (3)cells/ℓ.
