      • 스마트 건설안전 기술의 현장 적용 활성화를 위한 제도 개선 연구

        최근 건설업의 높은 재해율을 저감하기 위한 노력의 일환으로 스마트 안전관리 기술의 현장 도입이 시도되고 있으며, 이와 관련한 제도적인 개선이 일부 이뤄지고 있다. 본 연구에서는 이러한 개선된 제도의 효용성 파악 및 스마트 안전관리 기술의 현장 적용 확산을 위한 추가 개선점을 도출하기 위해 수행되었다. 건설업에 종사하는 참여자를 대상으로 설문조사를 수행하였고, 개선된 제도의 인지 및 건설현장에서 스마트 안전 기술을 확산하기 위해 필요한 항목을 조사하였다. 조사 결과 스마트 건설안전의 현장 적용에 대한 높은 관심을 확인할 수 있었고, 안전관리비의 및 현장 안전 서류업무 중복 문제 등을 제기하였다. 본 연구를 통해 도출된 의견은 향후 스마트 건설안전 기술의 도입·확산을 위한 제도적 개선의 기초자료로 활용될 것이다. Recently, as part of an effort to reduce the high accident rate in the construction industry, the smart construction safety technology is being introduced, and some institutional improvements are being made simultaneously. This study has been carried out to identify the effectiveness of such an improved institution and to draw additional opinions for improvement to spread the field application of smart safety technology. A survey was conducted for participants in the construction industry, and awareness of the improved institution and additional requirements for spreading smart safety technology in the construction site were investigated. As a result of the investigation, high interest in the field application of smart construction safety technology has been confirmed, and some issues such as safety management costs and duplication of on-site safety paperwork have been raised. The opinions derived through this study will be used as baseline data for future institutional improvements for the introduction and diffusion of smart construction safety technology in the construction site.

      • 10주간 산림 운동이 여성 노인의 체력 요인과 혈중 지질 성분 변화에 미치는 영향

        Purpose: The purpose of this study was to investigate the effects of 10 weeks of exercise in the forest health fitness factors and blood lipids profiles in elderly women. Method: Twenty-four elderly individuals aged 65 years or older, residing in C city, were randomly selected and divided into two groups: a control group (CO, n=11) and a forest exercise group (EX, n=13). The exercise group did in forest exercise three times a week for a duration of 10 weeks, with an exercise intensity aimed at burning 200-250 calories. Measurements of body composition, physical fitness tests, and blood lipid profile were taken at the beginning of the study and after 10 weeks. Results: As a result, the EX group showed a significant decrease in fat mass compared to the CO group after 10 weeks (p<.05). In addition, in the EX group, after 10 weeks, physical fitness factors, left hand grip strength (p<.05), Chair sit to stand (p<.000), and 2min step test (p<.05), Timed up & go (p<.000), WC8 (p<.000) showed a statistical difference compared to CO group. Blood lipid profile of LDL-C were decreased in the EX group compared to the CO group after 10 weeks (p<.05). Conclusion: Consequently, the results of this study Regular foreste exercise was found to be effective in reducing body fat, improving blood lipids, and improving basic physical strength in elderly women. Interdisciplinary Research Implications: In the future, it is judged that studies analyzing various physiological variables by analyzing and comparing various groups such as male elderly and elderly with diseases are necessary.

        IPA 기법을 활용한 공군사관생도의 교양체육 인식연구

        The purpose of this study was to find improvements in physical education and provide basic data for efficient education at the Republic of Korea Air Force Academy. Research methods for this study were as follows. The study surveyed 210 cadets and the importance and satisfaction of physical education recognized by cadets were analyzed using difference verification and IPA matrix analysis. As a result of the study, the first grade of the factors that cadets recognize as important in physical education was the "professor"s communication ability," which was also the third grade in satisfaction. On the contrary, the first grade of the factors that cadets recognize as satisfaction in physical education is the "professor"s sincerity," which was also the third grade in important. In addition, the factors that showed higher satisfaction than importance in difference verification in importance and satisfaction of physical education were "professor"s ability," "royalty" and "stress relief" while the factors that showed higher importance than satisfaction were "sports facilities". The IPA matrix analysis result as follow, quadrant I was mostly "professor"s ability", quadrant II were "exchange promotion", "requirements to facilities", and factors shown in quadrant III were not current considerations. In conclusion, this study showed that the school education system has been doing well.

        Escherichia coli O157:H7의 단크론성 항체 생산과 효소면역분석법의 개발

        본 연구에서는 최근 식품이나 농축산물 등에서 빈번하게 검출되고 있는 장관출혈성 대장균인 E. coli O157:H7에 대한 단크론성 항체를 생산하고, 신속ㆍ정확하게 검색할 수 있는 간접효소면역분석법을 개발한 후 시료적용가능성을 확인하였다. 먼저 LPS, HKEC 및 FKEC를 E. coli O157:H7의 면역원으로 준비하여 mouse 면역에 사용하였고, 세포융합과 cloning을 실시하여 단클론성 항체를 생산하였으며, 생산된 항체 중 높은 역가를 나타낸 HKEC 4G8-5 항체로 ID-ELISA법을 개발하였다. 개발된 IDELISA법의 최저검출한계는 10? cell/㎖이었고, S. aureus와 Sal. Typhimurium에 대해서 약간의 반응성을 나타내었지만 E. coli O157:H7에 대해 보다 강한 반응성을 나타내었기 때문에 E. coli O157:H7에 매우 특이적인 분석법인 것으로 확인되었다. 개발된 ID-ELISA법을 이용하여 돼지고기, 소고기, 닭고기 및 새싹채소에 대한 검출률 향상을 위해 각 시료를 증균한 결과, 3종의 육류와 새싹채소는 각각 증균 6시간과 2시간부터 검출이 가능한 것으로 나타나 기존의 증균법보다 신속하게 시료 속 E. coli O157:H7을 분석할 수 있을 것으로 판단된다. 따라서 본 연구에서 개발된 IDELISA법을 적용하여 농축산물 중에 존재하는 E. coli O157:H7을 분석한다면 보다 신속하고 간편하게 분석할 수 있을 것으로 판단된다. Escherichia coli O157:H7 causes hemolytic uremic syndrome and hemorrhagic colitis in humans. The objectives of this study were to produce monoclonal antibody(MAb) against E. coli O157:H7 and to develop an enzyme linked immunosorbent assay(ELISA) for the rapid detection of E. coli O157:H7 in agri-stockbreeding. The characterization of MAb produced from hybridoma cell (HKEC 4G8-5) was validated by ELISA and Western blot. The produced MAb was specific to E. coli O157:H7 and showed weak cross-reaction to Staphylococcus aureus. The detection limit of ELISA based on 4G8-5 MAb was 10? cell/㎖. Although the ELISA could not detect E. coli O157:H7 in the meat and sprout samples inoculated with 1×10¹ cell/10 g without enrichment, the same samples after enrichment for 6 hr were confirmed to be positive by ELISA. These results indicated that the ELISA combined with short enrichment (6 hr) is useful tool for rapid screening of E. coli O157:H7 in various samples.

        육류 및 새싹채소에 오염된 Escherichia coli O157:H7의 신속검출을 위한 면역크로마토그래피법의 개발

        E. coli O157:H7은 요독증후군, 출혈성 대장염 및 설사 등을 유발하는 원인균으로, 전세계적으로 이들의 오염에 따른 식중독 발생이 증가하고 있어, 본 연구에서는 E. coli O157:H7을 신속ㆍ정확하게 분석할 수 있는 면역크로마토그래피법을 개발하였고, 보다 신속한 검출을 위하여 육류와 새싹시료를 대상으로 최소 증균시간을 확인하였다. 먼저, 면역크로마토그래피법의 개발을 위해 colloidal gold와 EC MAb를 합성하여 EC MAb-gold conjugate를 제작한 다음, 접합여부를 확인한 결과, test line과 control line에 각각 처리되는 EC MAb와 anti-mouse IgG에 특이적으로 반응하였다. 제작된 EC MAb-gold conjugate를 이용하여 개발된 면역크로마토그래피법의 검출한계는 1×10<sup>5</sup> CFU/mL 수준까지 검출이 가능한 것으로 확인되었고, 다른 대장균속 및 주요 식중독균과의 교차반응성은 나타나지 않았다. 개발된 면역크로마토그래피법을 이용하여 임의로 E. coli O157:H7을 오염시킨 돼지고기, 소고기, 닭고기 및 새싹채소를 증균하는 동안 분석한 결과, 돼지고기와 닭고기는 6시간, 소고기는 10시간, 특히 새싹채소는 2시간 후부터 1×10 CFU/100 μL 수준까지 검출이 가능한 것으로 나타났다. 따라서 본 연구에서 개발된 면역크로마토그래피법을 이용하여 육류와 채소 등에 오염된 E. coli O157:H7을 최대 10시간 이내로 신속히 분석할 수 있을 것으로 판단된다. Escherichia coli O157:H7 has been considered as a significant food-borne pathogen since its role in causing hemorrhagic colitis and hemolytic uremic syndrome in humans was recognized. In this study, we developed an immunochromatography (ICG) assay for the detection of E. coli O157:H7. E. coli O157:H7 monoclonal antibody (EC MAb) and colloidal gold were conjugated and its specificity was determined by the ICG treated with EC MAb and anti-mouse IgG at test and control lines, respectively. The detection limit of the ICG was 1×10<sup>5</sup> CFU/mL, and no cross-reactivity was observed to other E. coli strains and major food-borne pathogens. To determine the minimum enrichment time for the ICG, meats and sprouts were inoculated with 1×10 CFU/100 μL of E. coli O157:H7. After enrichment time of 10 and 2 h for meats and sprouts, respectively, up to 1×10 CFU/100 μL of E. coli O157:H7 could be detected by ICG.

        농산물 생산 환경에서 분리된 Staphylococcus aureus의 항생제 내성 및 독소 유전자 확인

        Objectives: This study was undertaken to analyze Staphylococcus aureus from cultivation environments for agricultural products and to confirm antibiotic resistance and enterotoxin genes for the isolated S. aureus. Methods: A total of 648 samples were collected from apple, peach, ginseng and balloon flower farms. S. aureus was isolated from soil, agricultural water, personal hygiene elements (hands, gloves and clothes) and work utensils (boxes). Results: S. aureus was detected in a total of 25 samples and 72 strains were isolated. The resistance rate of the isolated S. aureus strains was confirmed at 33.3%, with 24 resistant strains among the total of 72. Fourteen different patterns types were found, and three pattern types (NV, OX, VA) were confirmed most frequently. As result of the detection of enterotoxin gene type, four gene types (sea: 1, sed: 4, seg: all isolated S. aureus, sei: all isolated S. aureus) were analyzed among a total of nine types. Conclusions: This study demonstrates that personal hygiene techniques should be properly managed, such as washing and sterilization before or after work, because agricultural contamination by S. aureus frequently developed through improper management.
