The purpose of this study is to analyze the factors influencing liver function and to provide basic data of exercise prescription for prevention of fatty liver in 10 middle - aged men by comparing body composition of abnormal liver function indicator and normal. We measured fat mass and muscle mass in 5 normal and 5 abnormal patients according to liver function enzymes. The results of the analysis were as follows: First, the fat mass of the middle - aged men was significantly lower than that of the normal group in the right arm and left arm by 37.5% and 40.1%, respectively. The right leg was 9%, the left leg 9.59%, and the trunk 16.8% lower. Second, the analysis of muscle mass of middle - aged men showed that the right arm was 10.68% and the left arm was 16.98% lower than the abnormal group. Legs were significantly higher in the normal group than in the abnormal group (22.67% on the right side and 21.46% on the left side). The body was 7.32% higher than normal. Taken together, these results indicate that the liver function index normal group has higher muscle mass than the abnormal liver muscle group, especially the leg muscle mass. Therefore, the relationship between muscle mass and hepatic enzymes in middle - aged men is closely related, and muscle mass is thought to be very important. In order to improve and prevent liver function, enhancement of muscle mass in the body composition is considered to be very important factor. Therefore, it is considered that a program for improving muscle mass for liver function improvement and prevention should be developed.
자금정(紫金錠)이 간암세포주 HepG2의 세포고사 및 세포주기에 미치는 영향

Objectives : Jageum-Jung is used as an anti-cancer agent in oriental medicine, but the mechanism by which it induces cell death in cancer cells is still unclear. The purpose of this study was to investigate the effects of Jageum-Jung on apoptosis and cell cycle arrest in HepG2 hepatoma cells. Methods : Various cancer cell lines including HepG2, C6 glioma, SH-SY5Y, PANC-1, and MCF-7 cells, were used. Apoptosis was determined by DAPI nuclei staining and flow cytometry in HepG2 cells treated with various concentrations (from 25 to 200 ${\mu}g/ml$) of $H_2O$ extract of Jageum-Jung (JGJ) for 48 hrs. Expression of cell cycle arrest mediators including Rb, p53, p21, cyclin B1, cdk4, and cyclin E proteins were measured by Western blot analysis. To estimate intracellular hydrogen peroxide levels and intracellular nitric oxide levels, HepG2 cells were stained with DCFH-DA dye and DAF dye, subjected on flow cytometric analysis. Results : 1. Jageum-Jung decreased the viability of HepG2 cells in a dose-dependent manner. 2. Jageum-Jung induced the catalytic activation of caspase-3 in HepG2 cells. 3. Jageum-Jung increased the intracellular hydrogen peroxide and NO in HepG2 cells. 4. Jageum-Jung increased the expression of Rb, p53 and p21 in HepG2 cells. 5. Jageum-Jung induced the expression of cyclin B1, cdk4, and cyclin E in HepG2 cells. Conclusions : Taken together, we suggest that Jageum-Jung exhibits cytotoxic effects on HepG2 cells, causing apoptosis and cell cycle arrest. The results showed that Jageum-Jung may do so by regulating the expression of specific target molecules that promote efficient apoptotic cell death following $G_2$/M phase arrest in a dose-dependent manner.

Objectives : The purpose of this study is to investigate the effect of Ulmi Cortex(UC) on the cisplatin-induced cell death. Materials and Methods : I examined several kinds of cell populations such as $CD4^+$ T cells, $CD8^+$ T cells, macrophages and dendritic cells in spleen. Result : When cisplatin was injected to mice, UC recovered total number of cells in spleen and also the number of T cells, macrophages and dendritic cells. UC also effected the activation of $CD4^+$ and $CD8^+$ T cells such as $CD25^+$, $CD69^+$ cells. To further investigate the effect of UC on the cisplatin-induced cell death, I examined the death of splenocyte and total T cells. UC inhibited cisplatin-induced cell death. Conclusion : Taken together, my results suggest that UC may be a beneficial oriental medicine for side effects during anti-tumor therapy.

본 논문에서는 귀의 형태와 색을 통해서 질병을 자가진단 후 귀의 특정 부위를 자극하는 이침요법을 위한 시술 보조 시스템을 제안한다. 제안 시스템은 피시술자의 귀의 이미지 정보와 질병에 대한 정보를 처리하여 이침을 위한 혈자리를 귀 이미지에 표시해 주는 시스템을 구현하였다. 특히 귀를 인식하는 부분에 있어서, Haar-like feature와 Adaboost알고리즘을 사용하는 OpenCV내의 함수를 사용하였고 인식된 귀영역을 그리드 영역으로 나누고 질병에 대한 사전 정보에 따라서 그리드 영역내의 이침혈 자리 시스템을 표시하는 시스템으로 구성하였다.

Objectives : This study aimed to investigate the underlying protective mechanism of Rhizoma Arisaematis(RA) on 3-NP-induced Cytotoxicity in rat C6 glial cells. Methods : We investigated treatment ofC6 cells with 20mM 3-NP and pretreatment with RA to cause loss of cell viability. and morphological change. which was associated with elevation of ROS level. increase in Bax/Bcl2 ratio and HIF-a protein expression Results : RA inhibited 3-NP-induced cell death in C6 glial cells and inhibited the changes of the : MMPT (mitochondria membrane potential transition) and inhibited the decrease of mitochondria complex II activity and 3-NP-induced ROS generation in C6 cells. And RA decreased the activity of HIF-a and Bax. and increased the activity of $Bcl_2$ in C6 glial cells Conclusions : RA markedly protects C6 glial cells from 3-NP-induced oxidative injury.
LC-MS/MS를 이용한 인체 혈장에서 Ginsenoside Rb1의 분석법 검증

LC-MS/MS를 사용함으로써 인체 혈장에서 ginsenoside Rb1의 분석법을 개발하고 검증하였다. 유속 0.9 mL/min에 이동상 0.1% formic acid가 첨가된 water와 methanol을 사용하여 기울기 용리 조건으로 설정하였으며 사용한 분석 칼럼은 C 18(4.6 mm×150 mm, particle size 5 μm)을 사용하여 분리하였다. MRM(multiple reaction monitoring) 방법의 전기 분무 이온화 이온 분석기로 모니터링 하여 분석하였다. 인체 혈장 샘플은 액체-액체 추출방법에 의해 acetone과 water가 섞인 용액으로 추출하였다. 이 분석의 검량선 범위는 10~500 ng/mL이며 상관계수는 0.9995를 나타냈다. 일내, 일간의 정밀성 농도범위는 상관계수 5.8% 그리고 정확성은 96.0~104.6%로 나타났다. 이 LC-MS/MS를 이용한 인체 혈장의 ginsenoside Rb1의 연구가 약동학 연구에 적용할 수 있을 거라 생각한다. A new liquid chromatographic tandem mass spectrometric (LC-MS/MS) assay for the quantification of ginsenoside Rb1 in human plasma was developed and validated. The separation was performed on a Agilent C18 column (4.6 mm×150 mm, particle size 5 μm) with a gradient elution of 0.1% formic acid in water and 0.1% formic acid in methanol and a flow rate of 0.9 mL/min. The analyte was determined using electrospray positive ionization mass spectrometry in the multiple reaction monitoring (MRM) mode (m/z 1131.714→365.303). Human plasma samples were extracted with acetone : water (50:50) by the liquid-liquid extraction method. The method was linear over the dynamic range of 10~500 ng/mL with a correlation coefficient of r=0.9995. The intra-and inter-day precision over the concentration range of ginsenoside Rb1 was lower than 5.8% (correlation of variance, CV), and the accuracy was between 96.0~104.6%. This LC-MS/MS assay of ginsenoside Rb1 in human plasma is applicable for quantification in a pharmacokinetic study.
고창 복분자의 기능성원료 표준화를 위한 지표성분으로서 Ellagic Acid의 분석법 개발

고창 복분자의 개별인정형 건강기능식품 기능성 원료로 개발하기 위하여 지표성분 표준화를 위한 ellagic acid의 분석법 설정과 분석법에 대한 밸리데이션을 실시하였다. 1% formic acid가 첨가된 water와 acetonitrile을 이동상으로 하고 Symmetry®(C18, 4.6×250 ㎜, 5.0 ㎛) 칼럼을 사용하여 기울기용리(gradient elution) 방법으로 분석하였다. 본 연구에서는 분석법을 확립하고 분석법에 대하여 특이성, 직선성, 정확성과 정밀성 그리고 회수율에 대하여 확인하였다. Ellagic acid의 검량선은 R2=0.9996으로 좋은 선형성을 보였으며 LLOQ와 LOD는 각각 1 ㎍/mL와 0.3 ㎍/mL였다. 일내와 일간 분석에서 상대표준편차(RSD)는 각각 2.28%와 2.84% 미만으로 나왔다. 회수율 측정결과에서는 89.17~97.92%로 나왔고, RSD는 0.05~0.14%였다. 그러므로 HPLC를 이용한 ellagic acid의 분석법이 고창 복분자 추출물의 기능성원료 표준화를 위한 지표성분 분석을 위한 적합한 시험법임이 검증되었다. 본 시험법에 따라 분석한 고창 미숙과 복분자 추출물 내의 ellagic acid의 함량은 세 lot를 3번씩 분석하였을 때 약 1.92 ㎍/㎎(0.192%)이 나왔으며 RSD값은 2.36% 이하였다. 따라서 본 연구를 통하여 확립된 ellagic acid의 분석법이 고창 복분자의 개별인정형 건강기능식품 기능성 원료개발을 위한 기초자료로 활용될 것으로 본다. Method development and validation of ellagic acid for the standardization of Gochang Bokbunja as a functional ingredient and health food were accomplished. A Symmetry® (C18, 4.6×250 ㎜, 5.0 ㎛) column was used with a gradient elution system of 1% formic acid in water and acetonitrile. This method was validated according to specificity, linearity, accuracy, precision test, and recovery test. Specificity was confirmed with identical retention time, and calibration curves of ellagic acid showed good linear regression (R2>0.9996). Relative standard deviations (RSD) of data from the intra- and inter-day experiments were less than 2.28% and 2.84%, except in the low limit of quality control (LLOQ, 1 ㎍/mL) sample. The results of the recovery test were from 89.17% to 97.92% with RSD values from 0.05 to 0.14%. Therefore, we performed analysis of ellagic acid as a marker compound in Gochang Bokbunja extracts. The amount of ellagic acid in Gochang Bukbunja was about 1.918 ㎍/㎎ (0.192%) in the three times analysis, and RSD was less than 2.36% by the validated method. These results suggest that the developed HPLC method is simple, efficient, and could contribute to the quality control of Gochang Bokbunja extract as a functional ingredient.
