We already reported that genetically engineered resveratrol-enriched rice (RR) showed to down-regulate skin melanogenesis. To be developed to increase the bioactivity of RR using calli from plants, RR was adopted for mass production using plant tissue culture technologies. In addition, high-pressure homogenization (HPH) was used to increase the biocompatibility and penetration of the calli from RR into the skin. We aimed to develop anti-melanogenic agents incorporating calli of RR (cRR) and nanoparticles by high-pressure homogenization, examining the synergistic effects on the inhibition of UVB-induced hyperpigmentation. Depigmentation was observed following topical application of micro-cRR, nano-calli of normal rice (cNR), and nano-cRR to ultraviolet B (UVB)-stimulated hyperpigmented guinea pig dorsal skin. Colorimetric analysis, tyrosinase immunostaining, and Fontana-Masson staining for UVB-promoted melanin were performed. Nano-cRR inhibited changes in the melanin color index caused by UVBpromoted hyperpigmentation, and demonstrated stronger anti-melanogenic potential than micro-cRR. In epidermal skin, nanocRR repressed UVB-promoted melanin granules, thereby suppressing hyperpigmentation. The UVB-enhanced, highly expressed tyrosinase in the basal layer of the epidermis was inhibited by nano-cRR more prominently than by micro-cRR and nano-cNR. The anti-melanogenic potency of nano-cRR also depended on pH and particle size. Nano-cRR shows promising potential to regulate skin pigmentation following UVB exposure.

This paper describes the feasibility of improving the braking performance of sports utility vehicle (SUV) by controlling rear wheels with electrorheological (ER) valve pressure modulator. Braking force distribution generated by controlling the slip of rear wheels makes suitable braking force according to the variation of the vehicle load. As a first step, principal design parameters of the ER valve and hydraulic booster are appropriately determined by considering the braking pressure variation during the braking system operation and then an electrically controllable pressure modulator is constructed. The governing equation for ER valve pressure modulator is derived and the system modeling for a rear wheel slip and yaw rate control is composed. Computer simulation shows that the proposed baking system can eliminate the braking distance increase in a laden condition and the steering stability during cornering is improved.
Modulation of Melanin Synthesis by Amaranthus spp. L Seed Extract in Melan-a Cells

Anti-melanogenic effects of amaranth (AT), one of the key source of squalene, were investigated in melanocytes. Amaranth seed powder was extracted with water and melan-a cells were treated with various concentrations of AT. By using HPLC, content of myo-inositol, one of potential active components, was measured in the crude extract of AT.AT reduced the melanin content in melan-a melanocytes and down-regulated melanogenic enzyme activity such as tyrosinase, TRP-1 and TRP-2. By regulating melanogenic enzyme activity, AT may be a potential natural source for whitening agent. Myo-inositol was detected in AT by HPLC and may be one of the active compounds from AT involved in the regulation of anti-melanogenesis. In this study, we demonstrated that AT has anti-melanogenesis properties. This new function of amaranth may be useful in the development of new skin-whitening products and its value as food.

Irritable bowel syndrome (IBS) is a functional gastrointestinal disease with complex pathophysiology involving the brain-gut axis. To assess the effects of Wasabia koreana (WK) on IBS, we employed a mouse model of colonic zymosan injection presenting with diarrhea-predominant IBS-like symptoms. Oral WK administration significantly diminished stool score, suppressed colon length and weight change, and minimized body weight loss without affecting food intake. In WK-treated mice, the submucosal thickening and epithelial lining of the colon were inhibited and were similar to those of naïve mice. Infiltration of mast cells into the colon and serum tumor necrosis factor-α levels were markedly suppressed. These effects were comparable to those of sulfasalazine, an anti-inflammatory drug. Furthermore, the number of visceral pain-related behaviors was significantly decreased, and locomotion activities measured in the elevated plus maze and open field tests were significantly increased by WK in a dose-dependent manner compared with amitriptyline, an antidepressant. These changes were accompanied by reduced FosB2 expression in the brain. Taken together, these data suggest that WK may have potential as a medicinal food for IBS by acting on inflammatory diarrhea and neural activity.

This study was designed to evaluate the pharmacological effects of Vaccinium bracteatum Thunb. methanol extract (VBME) on microglial activation and to identify the underlying mechanisms of action of these effects. The anti-inflammatory properties of VBME were studied using lipopolysaccharide (LPS)-stimulated BV-2 microglial cells. We measured the production of nitric oxide (NO), inducible NO synthase (iNOS), cyclooxygenase (COX)-2, prostaglandin E2 (PGE2), tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and interleukin-6 (IL-6) as inflammatory parameters. We also examined the effect of VBME on intracellular reactive oxygen species (ROS) production and the activity of nuclear factor-kappa B p65 (NF-κB p65). VBME significantly inhibited LPS-induced production of NO and PGE2 and LPS-mediated upregulation of iNOS and COX-2 expression in a dosedependent manner; importantly, VBME was not cytotoxic. VBME also significantly reduced the generation of the pro-inflammatory cytokines TNF-α, IL-1β, and IL-6. In addition, VBME significantly dampened intracellular ROS production and suppressed NF-κB p65 translocation by blocking IκB-α phosphorylation and degradation in LPS-stimulated BV2 cells. Our findings indicate that VBME inhibits the production of inflammatory mediators in BV-2 microglial cells by suppressing NF-κB signaling. Thus, VBME may be useful in the treatment of neurodegenerative diseases due to its ability to inhibit inflammatory mediator production in activated BV-2 microglial cells.

Background: Hericium erinaceus is considered a functional food and potential medicinal source. The present study was conducted to examine the potential antioxidant and anti-inflammatory activities of carried out with water and ethanol extracts of Hericium erinaceus grown on germinated green rice (HEGR-W and HEGR-E, respectively) and the water and ethanol extracts of germinated green rice (GR-W and GR-E, respectively) as potential medicinal resources or antioxidant and anti-inflammatory agents. Methods and Results: The total phenolic and flavonoid contents, DPPH, and ABTS activity, reducing power, DNA protective activity, cell viability, and NO production were investigated. The total phenolic and flavonoid contents were highest in HEGR-E ($66.53{\pm}2.40 mg{\cdot}GAE/100g$ and $82.12{\pm}7.10mg{\cdot}CE/100g$ respectively). HEGR-E exhibited high DPPH ($44.70{\pm}1.28%$) and, ABTS ($44.70{\pm}1.28%$) activity and reducing power (0.219). HEGR and GR extracts showed protective activity against DNA damage. The cytotoxicity of HEGR and GR in RAW264.7 cells and LPS-induced RAW264.7 cells was low. HEGR-E and GR-W exhibited anti-inflammatory effects through a 28% inhibition of NO production in LPS-induced RAW264.7 cells. Conclusions: These results suggested that the extracts of Hericium erinaceus grown on germinated green rice could be a potential medicinal material with natural antioxidant and NO inhibitory properties.
