        Reliability and validity of rasterstereography measurement for spinal alignment in healthy subjects

        Objective: The Back Mapper is one type of Rasterstereography and it can be used in the clinic without radiation exposure. The purpose of our study was to prove the reliability and validity of the Back Mapper and to compare it with the Spinal Mouse, whichis an assessment tool for spinal curvatures using a wheeled mouse, and the Cobb angle by X-ray. Design: Cross-sectional study. Methods: Twenty healthy adults participated in the test to investigate for the inter-rater reliability, intra-rater reliability, and concurrentvalidity. The tests were performed with assessment devices for scoliosis such as the Back Mapper, Spinal Mouse andCobb’s angle. Data was analyzed by an intraclass correlation coefficient (ICC) value and a standard error of measurement for reliabilityand correlation analysis for validity. Results: Intra-rater reliability of the Back Mapper was good (Cronbach’s α=0.821-0.984, ICC=0.696-0.969) except for assessingthe lordotic angle. Inter-rater reliability was good (Cronbach’s α=0.870-0.958, ICC=0.770-0.919) in assessment for trunk imbalance,rotation of scapulae, thoracic angle, lumbar angle, and kyphotic angle. The kyphotic angle in the Spinal Mouse had a significantcorrelation icompared with the Back Mapper (r=0.510, p<0.05), and the Cobb’s angle from an X-ray had a significant correlationwith trunk inclination (r=0.532, p<0.05). Conclusions: These findings provide good intra-reliability of the Back Mapper in healthy subjects, but the Back Mapper requiresmore experienced practice to have good inter-reliability. Also, the variables of the Back Mapper does not seem as appropriate comparedwith the Cobb angle by X-ray

        백선피(白鮮皮) 추출물이 DNCB로 유발된 NC/Nga 생쥐의 아토피 피부염에 미치는 영향

        Objectives Dictamni Radicis Cortex extracts (DRC) has been known to suppress allergic reaction, however the cellular target of DRC and its mode of action remain unclear. The purpose of this study is to investigate the effects of Dictamni Radicis Cortex extracts on DNCB induced atopic dermatitis-like skin lesions of NC/Nga mouse. Methods This study was designed to investigate the effects of DRC extract in the DNP-IgE-induced activation of MC/9 murine mast cell lines in vitro and in the DNCB-induced activation of NC/Nga mouse in vivo. For this investigation, We examined IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$ and GM-CSF mRNA expression by Real-time PCR, IL-13, MIP-$1{\alpha}$ production by ELISA analysis and manifestations of NFAT1, NFAT2, AP-1 and NF-${\kappa}B$ p65 transcription factors by western blotting in vitro. Then, we examined WBC, eosinophil and neutrophil in NC/Nga mouse, IL-5, IL-13 in serum, IFN-${\gamma}$, IL-4 in the spleenocyte culture supernatant, the absolute cell numbers of $CD4^+$, $CD8^+$, $^+Gr-1^+CD11b$, $B220^+CD23^+$ in the ALN, PBMCs and dorsal skin, IL-5, IL-13 in the dorsal skin by Real-time PCR and the distribution of mast cells by H&E and toluidine blue. Results In vitro the mRNA expression of IL-4, IL-5, IL-6, IL-13, TNF-${\alpha}$, GM-CSF and IL-13, MIP-$1{\alpha}$ production by ELISA analysis were completely abolished by DRC and the western blot analysis decreased the expression of mast cell-specific transcription factors including NFAT-1, NF-${\kappa}B$ p65. In vivo DRC oral adminstration also decreased the counts of WBC, eosinophils and inflammatory cytokines such as IL-13 and IgE in the serum. DRC oral adminstration elevated IL-4 level in the spleenocyte culture supernatant. DRC oral adminstration decreased total ALN cells, total skin cells, cell numbers of $CD4^+$, $B220^+CD23^+$ in the ALN, $^+Gr-1^+CD11b$ in the PBMCs and $CD4^+$, $CD8^+$ in the dorsal skin. The mRNA expression of IL-5, IL-13, thickness of epidermis, inflammation immune cells and mast cells were abolished by DRC in the dorsal skin. Conclusions Histological examination showed that infiltration levels of immune cells in the skin of AD-induced NC/Nga mouse were much improved by DRC oral adminstration. These results, therefore, suggest that DRC can regulate molecular mediators and immune cells that are functionally associated with atopic dermatitis induced in NC/Nga mouse, and may play an important role in recovering AD symptoms.

        Roles of GASP-1 and GDF-11 in Dental and Craniofacial Development

        Purpose: Growth and differentiation factor (GDF)-11 is a transforming growth factor-β familymember that plays important regulatory roles in development of multiple tissues whichinclude axial skeletal patterning, palatal closure, and tooth formation. Proteins that have beenidentified as GDF-11 inhibitors include GDF-associated serum protein (GASP)-1 and GASP-2. Recently, we found that mice genetically engineered to lack both Gasp1 and Gdf11 have anincreased frequency of cleft palate. The goal of this study was to investigate the roles of GDF-11 and its inhibitors, GASP-1 and GASP-2, during dental and craniofacial development andgrowth. Methods: Mouse genetic studies were used in this study. Homozygous knockout mice for Gasp1(Gasp1-/-) and Gasp2 (Gasp2-/-) were viable and fertile, but Gdf11 homozygous knockout (Gdf11-/-) micedied within 24 hours after birth. The effect of either Gasp1 or Gasp2 deletion in Gdf11-/- miceduring embryogenesis was evaluated in Gasp1-/- ;Gdf11-/- and Gasp2-/- ;Gdf11-/- mouse embryosat 18.5 days post-coitum (E18.5). For the analysis of adult tissues, we used Gasp1-/- ;Gdf11+/-and Gasp2-/- ;Gdf11+/- mice to evaluate the potential haploinsufficiency of Gdf11 in Gasp1-/- andGasp2-/- mice. Results: Although Gasp2 expression decreased after E10.5, Gasp1 expression was readilydetected in various ectodermal tissues at E17.5, including hair follicles, epithelium in nasalcavity, retina, and developing tooth buds. Interestingly, Gasp1-/- ;Gdf11-/- mice had abnormalformation of lower incisors: tooth buds for lower incisors were under-developed or missing. Although Gdf11+/- mice were viable and had mild transformations of the axial skeleton, nospecific defects in the craniofacial development have been observed in Gdf11+/- mice. However,loss of Gasp1 in Gdf11+/- mice occasionally resulted in small and abnormally shaped auricles. Conclusions: These findings suggest that both GASP-1 and GDF-11 play important roles indental and craniofacial development both during embryogenesis and in adult tissues.

        Objective : This study was designed to analyze the clinical studies on Tic disorder in traditional Korean medicine and traditional Chinese medicine. Methods : To figure out the Tic disorder, Korean medical studies from Oriental Medicine Advanced Searching Integrated System (OASIS) and Chinese Medical Journals from Wangfang data and China National Knowledge Infrastructure (CNKI) which published in 2008 to 2012 were analyzed. Results & Conclusions : 1. DSM-IV(7 studies), CCMD(3 studies) were frequently used in the diagnosis of tic disorder. 2. The herbs used for the treatment of tic disorder are Uncariae ramulus et Uncus(釣鉤藤), Paeoniae Radix Alba(白芍藥), Poria(白茯?), Buthus martensi Karsch(全蝎) etc. 3. Acupuncture points frequently used were Paek'oe(GV20), Pungji(GB20), Naegwan(PC6), Sasinchong(EX-HN1), T'aech'ung(LR3), Joksamni(ST36), Hapgok(LI4) and so on. 4. The cause of Tic disorder is connected with liver, heart, spleen, kidney, gall bladder among the internal organs and Fire(火), Wind(風), Dampness(濕) of external causes.

        가감양격산(加減凉膈散)이 RBL-2H3 비만세포와 OVA/alum에 감작된 생쥐에 미치는 항알레르기 효과

        Objectives: Gagamyanggyeoksan (G-YGS) has been used to suppress allergic reaction, however, the cellular target of G-YGS and its mode of action remain unclear. The present study was designed to investigate the effect of extracted G-YGS on the PMA and lonomycin (PI)-induced activation of RBL-2H3. Methods: For this investigation, We examined IL-4, IL-13 mRNA expression by Real-Time PCR, IL-4, IL-13 production by ELISA analysis and manifestations of GATA-1, GATA-2, NF-AT1, NF-AT2, AP-1 and NF-${\kappa}B$ p65 transcription factors by western blotting, OVA-specific IgE, IL-4, IL-13 by mouse be sensitive to OVA. Results: Here we showed that treatment of RBL-2H3 mast cells with G-YGS, suppressed PI-induced production of Th2 cytokines including IL-4 and IL-13 in a dose dependent manner. The mRNA expression of IL-4 were completely abolished by G-YGS at the concentration of $100{\mu}g/ml$. Data from a stable cell lines consistently expressing IL-4. And the mRNA expression of IL-13 were abolished by G-YGS at the $200{\mu}g/ml$. But there is no difference between the $50{\mu}g/ml$, the $100{\mu}g/ml$ and the comparison. Results from the western blot analysis of transcription factors involving IL-4 and IL-13 expression indicated that it prominently decreased the expression of mast cell specific transcricption factors including GATA-1, GATA-2, NF-AT2, c-Jun, NF-${\kappa}B$ p65 but not c-Fos. And G-YGS suppressed IgE, IL-4, IL-13 in mouse be sensitive to OVA. Conclusions We suggested the anti-allergic activities of G-YGS might be mediated by down-regulation of Th2 cytokines such as IL-4 and IL-13 through the regulation of transcription factors as GATA-1, GATA-2, NF-AT2, c-Jun, NF-${\kappa}B$ p65.

        Development of Educational Content for Nurses to Prevent Safety Accidents of Hospitalized Children: Delphi Approach

        Purpose: The purpose of this study was to develop content for safety education to prevent hospital safety accidents among hospitalized children and to investigate the status of safety education performed by nurses. Methods: First, the Delphi method was used, with 18 experts, to develop educational contents for preventing safety accidents. Second, an exploratory survey was performed of the actual status of safety education for preventing safety accidents among hospitalized children using a questionnaire developed based on the Delphi method. The participants of this study were 159 nurses with at least 6 months of work experience. Results: The educational content developed through the Delphi method for preventing safety accidents among hospitalized children contained seven domains (falls, injury, electric shocks/burns, suffocation/aspiration, poisoning/abuse, kidnapping, medical devices) with 44 topics. The item mean of nurses'perceptions of the importance of child safety education was4.18, and the actual performance score was 3.72, which was a statistically significant difference (t=11.58, p<.001). Conclusion: These seven comprehensive domains of accident prevention education for hospitalized children are expected to be useful for interventions to support the safety of hospitalized children.
