Quantitative Analysis of Single Cell Adhesion and Migration Using μ-PIV and TIRF

In order to quantify the single cell adhesion and migration, we propose a novel method using total internal reflection florescence (TIRF) microscopy and μ-PIV. A living cell has focal contacts on the substrate so that cell adhesion depends upon the formation of adhesive contacts between the cell and the substrate. Determination of the cell-substrate contact area (or the number of contact points) is necessary in order to understand how biomaterial properties influence cell adhesion. By using TIRF microscopy, we can obtain the image of near-focal contact (sub-micrometer scale). From the obtained images, it is determined whether the cell adheres properly. The cell movement which is caused by changing the hydrodynamic condition is also observed. The moving focal contacts are measured quantitatively with μ-PIV. This combination of TIRFM and μ-PIV proves to be very useful for analyzing cell viability in a new environment.

기존 전력망이 스마트그리드로 진화하면서 발전, 송·변전, 배전분야의 각 기기간 정보를 교류하기 위하여 기존에 운영중인 시스템은 서로 다른 통신 프로토콜을 변환하기 위해 게이트웨이가 필요하다. 게이트웨이는 기밀성, 무결성, 가용성을 향상시키기 위한 보안기술이 요구된다. 특히 전력계통분야에서 많이 사용하고 있는 DNP 3.0 프로토콜과 IEC 61850 프로토콜은 보안성 적용시 구조적 차이가 있다. 본 논문에서는 국제표준 에 따른 프로토콜 매핑과 보안을 적용하여 개발한 게이트웨이를 소개한다.

We carried out a time-series analysis of images which were obtained while a living cell was forming its focal contact at the bottom of the gap between the cell and the substrate. In order to visualize the contact area selectively, we adopted total-internal-reflection-fluorescence (TIRF) method, which can illuminate specimen volume within only several hundred nano-meters above the substrate. From the fluorescent intensity of the TIRF image, we could calculate the distance of the cell surface from the substrate. As a result, we visualized the change of cell-contact type from the close-contact into focal-contact with information of its vertical displacement, and presented the three-dimensional evolution process of the cell-surface-profile near the contact area during this metamorphosis.
기존 운동기기 부착용 운동중량/횟수 자동측정기기의 성능평가

본 연구는 기존 운동기기에 부착 가능한 새로운 운동중량, 횟수 자동측정기기의 개발을 위하여 센서모듈과 운동정보획득 알고리즘을 이용하여 운동의 양을 측정할 수 있는 시스템의 운동 계측성능 평가에 관한 것이다. 개발된 시스템은 기존 사용되고 있는 운동기기에 손쉽게 장착할 수 있도록 설계하였으며 측정된 데이터는 스마트폰에 블루투스로 전달 가능하도록 하였다. 실험결과 운동습관이 다른 3명의 사용자로부터 무게 및 횟수를 작은 오차로 검출할 수 있었다. 본 시스템은 사용자의 운동을 측정하여 운동데이터 활용이 가능한 각종 기기에 전달함으로써 개인 맞춤형 헬스케어 시스템 및 재활치료 시스템에도 적용이 가능하다는 장점을 가지고 있다. In this study, a performance evaluation was carried out to develop a new add-on device with an algorithm for automatically measuring the exercise weight and repetitions when using an existing fitness machine. The developed device was designed to be simply attached to an exercise machine to measure the exercise weight and repetitions. The measured data are transferred to a smartphone using Bluetooth. As result, it is possible to detect the weights and repetitions from three users with different exercise habits with little error. The system can measure the user"s motion and transfer the data to various devices capable of utilizing the workout data, giving it the advantage of being applicable to personalized health care and rehabilitation therapy.

Bacterial chemotaxis may have a significant impact on the study of structure and function of bacteria. Quantification of chemotactic motion is necessary to identify chemoeffectors and to determine the bacterial transport parameters used in prediction models of chemotaxis. Although many studies have accumulated the knowledge about chemotaxis for many years, the motion of a single bacterium has not been studied yet. In this study we propose a device microfabricated by soft lithography and consisting of microfluidic channels, which can generate a gradient of chemoeffectors in the main channel so that a single bacterium is injected into this channel by hydrodynamic focusing. This microfluidic assay offers superior performance to measure a single cell or a group of cells and to quantify their motion, and it also builds a delicate gradient of chemoeffectors to raise accuracy of measuring the bacterial motion.

전반사형광현미경법과 타원체맞춤 방법을 이용한 고체벽면 근처에서의 3 차원 박테리아 경로 추적

The motion of flagellated bacteria close to surfaces is relevant to understanding the early stage of Biofilm formation. When Bacterium cells are close to a surface, the cells often swim parallel to the surface in a circle for some time. The simplest and most effective way of understanding this motion is the individual cell tracking. In the present study, TIRFM is used to generate the evanescent wave near the surface so that it emits a green light due to eGFP gene transfection and illuminates a small specimen volume of about 100-㎚ depth. Also, using the PTV method, we can obtain the 3D motion data of bacteria near the surface. However, most of current 3D tracking methods, which are designed for spherical particles, is not optimized to track a bacteriumlike cell which is of a prolate ellipsoid. In this study, using the PTV method and proposing the ellipsoidal fitting to model the shape of bacteria realistically, we obtain the more accurate data of bacteria motion near the wall.
스마트그리드를 위한 IEC61850과 DNP3를 적용 보안게이트웨이 개발

현재 변전소(SUBSTATION)에서는 전력설비들간 상호운용성을 위하여 IEC 61850 표준이 적용된 기기들이 설치 및 운영되고 있으며, 전력 설비를 감시하고 제어하는 SCADA(Supervisory Control and Data Acquisition)는 DNP3(Distributed Network Protocol)을 적용되고 있다. 본 논문에서는 DNP3와 IEC 61850를 변환을 위하여 게이트웨이를 통해서 상호 정보를 전송하고 있는데, SCADA와 게이트웨이 DNP3 통신 구간, 게이트웨이와 IEC 61850의 IED(Intelligent Electric Device) 간 통신 구간에서 적용이 가능하도록 구현한 보안기능을 소개하고자 한다.
