        Brevibacterium ammoniagenes의 영양구성 變異株에 依한 5-IMP 生成

        Brevibacterium ammoniagenes ATCC 6872로부터 N.T.G. 처리하여 adenine-guanine 영양요구성 변이주, 즉 Brevibacterium ammoniagenes No. 9를 얻었으며, 이 균주는 그 生合成경로상의 효소, SAMP synthetase와 IMP dehydrogenase나 XMP aminase가 결여된 것으로 여겨진다. 本 實驗에서 이 菌株의 IMP 醱酵과정에서는 배양 4日경에 최대의 IMP를 生成하였고 그 때에 Hx는 減少하였으므로 이는 salvage合成에 의해 Hx의 IMP로 전환된 것으로 생각된다. 또한 배지中의 adenine과 guanine의 최적농도는 150 ㎎/㎖로서 adenine과 guanine의 培地中의 농도가 IMP醱酵에 크게 영향을 미쳤다. In attempts to obtain IMP Producting strains, Brevibacterium ammoniagenes ATCC 6872 was treated with N.T.G. Adenine-guanine requiring mutants were obtained from Brevibacterium ammoniagenes ATCC 6872, and then a strain of them was selected for production of IMP and named Brevibacterium ammoniagenes No. 9(ade, gu ). The production of IMP by Brevibacterium ammoniagenes No. 9 was about 3㎎/㎖ for 4 day of culture. The optimal concetration of adenine and guanine was 150㎎/㎖.

        표고버섯 배양액으로부터 단백다당류의 추출 및 정제 방법

        L. edodes의 배양액 및 균사체로부터 항암작용이 있는 단백 다당류의 추출 및 정제방법에 관한 실험을 실시하였다. 단백다당류 추출에서 비이온성 계면 활성제인 2% Triton X-100 용액으로 추출할 경우 상온에서 추출하여도 통상의 100℃ 열수 추출을 통해 얻을 수 있는 것보다 수율이 높았다. 그러나 계면활성제 용액에 4N NaCl을 첨가하여 추출하면 ethanol 침전과정에서 단백다당류의 침전물을 얻을 수 없었다. 열수 추출에 있어서 추출 온도는 100℃가 효과적이고 추출 시간은 4시간 정도면 충분하였다. 열수 추출 결과 ethanol 침전 및 동결건조를 통하여 얻어진 조단백 다당류의 정제 실험에서는 alumina를 충전제로 하는 gel filtration보다 sephadex에 의한 gel filtration 또는 DEAE-cellulose, DEAE-sephadex를 이용한 ion exchange chromatography가 보다 효과적이었다. The extraction and separation methods of protein-bound polysaccharides from the mycelium and culture broth of L. edodes were investigated. The use of 2% solution of surface active agent, Triton X-100 was effective for extraction of the protein-bound polysaccharide from the mycelium. The extraction of the protein-bound polysaccharides from mycelium with hot water was achived by 4 hours extraction at 100℃. For the separation and partial purification of the protein bound polysaccharides the column chromatography using DEAE-Cellulose, DEAE-Sephadex and Sephadex proved to be effective.

        일부 여대생의 체형 타입과 관련된 요인에 관한 연구

        This study was designed to examine body shape control and its related factors, including BMI(body mass index), desire for weight control, satisfaction with body shape, dissatisfaction with specific parts of body shape, starting period of concern about body shape, as well as experience, method, result, counsel, effort, expense and knowledge of body shape control. The survey was completed by 180 female university students in Gyeongsanbukdo through a self-administered questionnaire during September and October of 2006. The subjects were assigned to one of three groups based on self-estimated figure: 47.8% of the students estimated their figure as 'normal', while 38.3% of the students estimated themselves as 'thin' and 13.9% of the students as 'fat'. The results were as fellows. The mean age of the subjects was $19.45{\pm}1.67$ with a height and weight of $161.15{\pm}4.99cm$ and $52.63{\pm}10.78\;kg$, respectively. Their self-estimated figure was significantly related to BMI, desire for weight control, satisfaction, dissatisfaction with a specific part, and experience, counsel and effort in body shape control. This research suggests that students need to know how to properly control body shape in health and beauty.

        한국산 영지버섯에서 분리된 항응고성 다당

        식용버섯을 대상으로 항응고 활성을 검색한 결과 영지버섯 알칼리 추출물이 가장 높은 활성을 보였다. 영지버섯으로부터 추출한 조다당 획분인 GL-I은 1N NaOH로 8시간 추출물(GL-0)을 methanol 환류, 에탄올 침전을 거쳐 투석, 동결건조하여 조제하였다. GL-I 획분은 periodate 산화에 의해서는 활성이 크게 변하지만 pronase 소화시 활성의 변화가 거의 없는 것으로 보아 항응고 활성의 본체는 주로 당과 관계되는 것으로 추정된다. GL-I의 구성당은 glucose:galactose:xylose:mannose:arabinose가 19.3 : 3.0 : 2.3 : 1.3 : 1.0 : 0.3의 molar ratio로 구성되어 있다. GL-I 획분을 DEAE-Toyopearl 650C(GL-Ⅰa→Ⅰf)와 Sephadex G-100(GL-Ⅰc-Ⅰ→GL-Ⅰc-Ⅱ)을 이용하여 부분 정제하였다. The anticoagulant polysaccharide was screened from the ediable mushrooms. Among them, alkali extract of Ganoderma lucidum showed the highest activity in aPTT. A crude polysaccharide fraction (GL-I) was prepared from the lN NaOH solution extract of Ganoderma lucidum followed by methanol-reflux, precipitation with ethanol, dialysis and lyophilization. GL-I inhibited the intrinsic pathway in blood coagulation pathway and exhibited concentration dependent anticoagulation effects. The anticoagulant activity of GL-I was decreased greatly by periodate oxidation, but was not changed by pronase digestion. These suggest that carbohydrate moiety may be related to the anticoagulant activity. GL-I consisted of glucose, galactose, fucose, xylose, mannose, arabinose in a molar ratio of 19.3 : 3.0 : 2.3 : 1.3 : 1.0 : 0.3. GL-I was partially purified on the DEAE-Toyopearl 650℃(GL-Ia→GL-If) and on the Sephadex G-100(GL-Ic-I→GL-Ic-Ⅱ).

        버섯 천연조미료 제조에 관한 연구

        This study was conducted to provide basic data for the development of natural seasoning. The quality characteristics of water extracts from Pleurotus ostreatus and Lentinus edodes were determined. Also, savory taste, fragrance, color and overall preference was evaluated. The preference test on the seasonings prepared from Pleurotus ostreatus and Lentinus edodes with the addition of sea tangle was also performed. The crude protein content of dried, Lentinus edodes and Pleurotus ostreatus was 20.4% and 30.4% respectively. The total organic acid contents were higher in Pleurotus ostreatus than in Lentinus edodes extracts. The total free amino acid and total nucleotide contents were higher in Pleurotus ostreatus than in Lentinus edodes extracts. Three nucleotide, adenosine monophosphate(AMP), guanosine monophosphate (GMP), xanthine monophosphate(XMP) were found in mushroom seasonings while inosine monophosphate (IMP) was not detected. Pleurotus ostreatus extract showed higher score in savory taste and overall acceptability than Lentinus edodes extract, Pleurotus ostreatus seasoning added with 196 sea tangle and Lentinus edodes seasoning added with 1.5% sea tangle were rated as high in overall quality. In a sensory test of foods cooked with mushroom seasoning. It was shown that panels in their twenties well used to the taste of monosodium glutamate while the people in their thirties preferred the specific flavor of the mushroom seasoning.

        Brevibacterium sp.로부터 유도된 PFP 내성 변이주에 의한 L-Tyrosine 생성

        Brevibactericum속 균주로 부터 N.T.G. 처리하여 1차로 140개의 변이주를 선별하였다. 이들 균주로 부터 2차 확인후 50균주를 선택 생성량을 보았다. 그 중 가장 생성량이 많은 균주 APT-104(phe-) 균주를 다시 N.T.G. 처리하여 PFP 내성 균주 13균주를 분리하여 그 중 가장 생성량 높은 균주를 다시 N.T.G. 처리하여 PFP 농도가 높은 곳에서 내성이 강한 균주를 5균주 선택 하였다. 이상 선택한 균주의 L-tyrosine 생성량을 비교 검토한 결과 PFP-106 균주에서 가장 높은 생성을 보였다. (50mg/l) 따라서 이 균주의 배지조건을 검토하였다. 그 결과 탄소원에서의 영향은 Sucrose 나 glucose 첨가시 76mg/l, 50mg/l 로서 효과적이었으며 그 농도는 10% 농도가 가장 효과적 이었다. 질소원에서는 (NH_4)_2SO_4첨가시가 50mg/l 로 높았고 다른 질소원은 오히려 낮은 생성을 보였다. 그 농도는 3%가 적당하였다. 아미노산의 영향은 glutamic acid와 L-tryptophane 생성에서 각각 106mg/l, 108mg/l로서 가장 효과적이었다. 또한 L-phenylalanine 의 농도는 100mg/l 첨가시 L-tyrosine 생성량이 50mg/l로서 가장 효과적이었다. This study was attempted to investigate the production of L-tyrosine by Brevibacterium flavum ATCC 14067. To select the strain which produce more L-tyrosine, mutants were induced by N-methyl-N'-nitro-nitrosoguanidine(NTG) treatment and phenylalanine auxotrophic mutants were induced by NTG and penicillin treatments. PFP resistant mutant was isolated from a phenylalanine auxotroph by retreatment with NTG and screened for increase of L-tyrosine production. PFP-326 mutant resistant to PFP (100㎍/ml) was derived from phenylalanine auxotroph by mutagenesis with NTG and PFP-106 mutant resistant to PFP (120㎍/ml) was derived from PFP-326 by mutagenesis with NTG. The composition of media for L-tyrosine production in strain PFP-106 was studied. PFP-106 mutant strain produced 50mg/l of L-tyrosine while the the parent strain produced 0.56mg/l of L-tyrosine. The optimum composition of medium for L-tyrosine by strain PFP-106 was 10% sucrose as carbon source, 3% ammonium sulfate as nitrogen source. The optimum cultural condition for producing L-tyrosine by strain PFP-106 was L-phenylalanine at a concentration of 100㎍/mg.

      • Corynebacterium glutamicum의 Ethionine 耐性變異株에 依한 Methionine 生成에 關한 硏究

        This study was attempted to evaluate the formation of methionine by mutants of Corynebacterium glutamicum. STA 19 strain (thr-), threonine requiring mutant by NTG (N-methyl-N´-nitro-nitrosogua-nidine) treatment to Corynebacterium glutamicum was selected. With NTG retreatment to this strain, better production of methionine was found in ethionine resistent mutants (thr-, ethr) than wild strain and highest amount of methionine was produced in ER-108 (thr-, ethr) strain. The influence of the composition of media and several elements of the culture condition to the production of methionine were investigated. The results were as follows; 1. The Production of methionine by ER-108 strain was 800㎍/ml, and was increased to 950㎍/ml in case that cysteine was added. 2. The effect of vitamins addition on the formation of methionine revealed that sharp increase was existed only by addition of vitamin B12. In the cultivation with the addition of vitamin B12 20㎍/ml the formation of methionine was increased up to 1,500㎍/ml, 2 times as high as nonadded vitamin culture. 3. The optimal composition of media for producing methionine by ER-108 was glucose 10%, ammonium sulfate 2%, magnesium sulfate 0.1%, thereonine 400㎍/ml, cytosine 0.5mg/ml. 4. The optimal temperature, pH and time of culture for the production of methionine by ER-108 strain were 30°C, pH 7 and 72 hr, respectively.

        먹물버섯 에탄올추출물이 Benzo(a)pyrene 투여에 의한 마우스의 간 손상에 미치는 영향

        간 독성물질인 benzo(a)pyrene을 투여한 마우스에서 먹물버섯 에탄올 추출물의 간 손상 억제에 미치는 영향을 살펴본 결과, 마우스에 B(a)P 투여로 증가된 혈청 aminotransferase의 활성, 간 조직의 과산화지질 함량과 cytochrome P-450의 활성은 먹물버섯 에탄올 추출물 투여에 의해 유의적으로 감소하였다. 반면에 B(a)P의 투여에 의해 활성이 감소된 glutathions S-transferase와 glutathione 함량은 먹물버섯 에탄올 추출물 투여에 의해 다시 증가되었다. 또한 superoxide dismutase, catalase 그리고 glutathione peroxidase의 활성은 B(a)P의 투여시 그 활성이 각각 유의적으로 증가하였다가, 먹물버섯 에탄올 추출물 투여로 인하여 감소하는 경향을 나타내었다. 이상의 결과에서 먹물버섯 에탄올 추출물은 B(a)P 투여로 감소된 glutathione 함량과 glutathione S-transferase의 증가와 함께 B(a)P 투여로 증가된 지질과산화물 생성의 감소와 여러 항산화적 방어계의 작용을 가진 효소 활성에 영향을 주어 지질과산화 및 간 손상 억제 효과를 가지는 것으로 사료된다. This study was carried out to investigate the inhibiton effects of Coprinus comatus ethanol extract of edible mushroom on liver damage in benzo(a)pyrene (B(a)P)-treated mice. The activities of serum aminotransferase, cytochrome P-450 and hepatic content of lipid peroxide after B(a)P-treatment were increased than those of control, but those levels were significantly decreased by the treatment of Coprinus comatus ethanol extract. Whereas, the hepatic glutathione content and glutathione S-transferase activity were decreased by B(a)P-treatment than those of control, but those were increased by the treatment of Coprinus comatus ethanol extract. Also the activities of superoxide dismutase, catalase and glutathione peroxidase after B(a)P-treatment were markedly increased than those of control, but those levels were decreased by the treatment of Coprinus comatus ethanol extract. These results suggest that Coprinus comatus ethanol extract have a protective effect on liver damage by benzo(a)pyrene through the mechanisms of decreasing lipid peroxide and activities of free radical generating enzymes.
