        벼 종자 배아함몰도의 품종간 변이

        근래 쌀의 건강기능성에 대한 관심이 고조되면서 백미상태에서도 배아의 일부가 부착된 배아미의 이용을 위한 기술적 품종적 연구가 착수되고 있다. 본 연구는 다양한 종자형태를 보이는 49개의 품종에 대하여 현미 상태에서의 배아함몰도 및 종자형태와의 관련성에 대해 조사 분석한 것이다. 배아함몰도는 0.27(큰눈벼)~0.59(대립벼1) mm의 변이를 보였다. 49개의 품종 중 진부, 진보, 흑설, 오봉, 운광, 청남 등이 배아함몰도가 상대적으로 높은 품종이었다. 배아함몰도는 현미의 폭과는 정의 상관관계가 있었고 장폭비와는 부의 상관관계를 보여주었다. 배아함몰도가 높은 품종 육성을 위한 방향을 논의하였다. As the market demand on functionality rice has been increasing, embryo rice in which embryo residue remains even after milling has come to comsumers' attention because rice embryo contains several functionality components. Consequently, development of rice varieties for higher rate of embryo adhesion to grains after milling has become one of the breeding objectives for quality improvement. In this study, we observed embryo dent of 49 commercial varieties and analyzed the relationship between embryo dent and grain size and shape. Embryo dent of rice grains varied 0.27 (Keunnun)~0.59 (Daerip 1) mm. Varieties Jinbu, Jinbo, Heugseol, Obong, Unkwang, and Cheongnam showed relatively deeper embryo dent, suggesting that they will be applicable in breeding for embryo rice. Embryo dent was correlated positively with grain width (r=0.53 ** ) and grain size (r=0.34 * ), and negatively with grain width/length ratio (r= -0.38 ** ). Strategies for breeding embryo rice were discussed in relation to embryo dent, grain size and shape.

        Identification of a novel candidate gene for rolled leaf in rice

        A semi-narrow and adaxially rolled leaf mutant, rl15(t), was induced from Korean japonica rice cultivar Ilpum by chemical mutagenesis using ethyl methanesulfonate. We characterized the mutant and identified the novel gene causing the mutant phenotype. Cytological analysis of mutant leaves indicated that the adaxial leafrolling phenotype is due to the reduced size and number of bulliform cells in the mutant. Genetic analysis showed that the rolled leaf trait is controlled by a single recessive gene, designated rl15(t). Using an F2 mapping population generated from a cross between Milyang23 and the mutant, we mapped the candidate region to a 174 kb interval on the long arm of chromosome 1 near the centromeric region. Through whole genome sequencing in bulk and MutMap analysis, we identified the causal SNP within the candidate region. The results of RT-PCR analysis indicated that a splicing error occurred due to a base change from G to A at the beginning of the fifth intron of LOC_Os01g37837, which encodes a putative seryl-tRNA synthetase, resulting in the mutant phenotype. Further study of the rl15(t) gene will facilitate analysis of leaf architecture and morphogenesis in rice plants.

        해외소비자의 국내육성 벼 품종에 대한 선호도 및 식미관련 형질 분석

        This study aimed to assess the preferences of African, American, and Asian panels, in comparison to a Korean panel, for various Korean rice varieties, including three japonica, two indica, and one Tongil-type, all developed by the Rural Development Administration in Korea. Regarding rice appearance, most panelists, except for the Koreans, favored long and slender rice varieties like ‘Hanyeol’, ‘Hyangyeol’, and ‘Amissal’. In contrast, the Koreans preferred wider varieties like ‘Sindongjin’ and ‘Deuraehyang’. Notably, the overseas panelists consistently favored rice varieties with high appearance quality in sensory evaluations of cooked rice. Both overseas and Korean panelists strongly preferred the indica rice variety ‘Hanyeol’ in terms of appearance quality and sensory evaluations. However, Korean rice varieties like ‘Amissal’ and ‘Sindongjin’ scored high in appearance quality with overseas panelists but received low ratings in sensory aspects, including shape, stickiness, and taste, compared to those for ‘Hanyeol’. This suggests that considering sensory characteristics is important when exporting Korean rice varieties. Regarding taste characteristics preferred by the panelists derived from the correlation analysis between taste traits, Africans preferred rice with a smooth texture and no stickiness, especially favoring long-grain rice. Americans leaned towards rice with a slightly firm texture, some stickiness, and late aging characteristics. Asians preferred rice with a smooth texture, low stickiness, and long-grain varieties. In contrast, Koreans favored round rice with a glossy appearance, a slightly firm texture, late aging traits, and some stickiness. These research findings can serve as valuable data for the development of rice varieties for overseas markets and are expected to contribute to securing competitiveness in international markets.

        Identification of Heterosis QTLs for Yield and Yield-Related Traits in Indica-Japonica Recombinant Inbred Lines of Rice (Oryza sativa L.)

        Supplying sufficient rice to growing populations is a global challenge. Hybrid indica rice varieties exploiting heterosis have increased yields, but inter-subspecific crosses between indica and japonica varieties are hampered by sterility. Examination and genetic understanding of yield heterosis in indica/japonica crosses addressing yield barriers are basic requirements. In this study, QTLs for heterosis of yield traits were identified in indica-japonica recombinant inbred lines (RILs) using a total of 178 RILs originating from Dasanbyeo (indica) × TR22183 (japonica) (DT-RILs) and their backcrossed populations. Nine of sixty-six major quantitative trait loci (QTLs) identified in DT-RILs exhibited heterosis. Heterosis QTLs clustered with other traits on chromosomes 1, 4, and 8, and clusters were conserved between different RILs. The clusters contained several known yield enhancement genes/QTLs. Specific heterotic allele combinations contributed to four major heterosis QTLs, particularly for panicle and spikelet number traits. Heterosis for yield and yield-related traits was explained by the harmonized effects of overdominance, dominance, and epistatic interactions in inter-subspecific breeding populations.

      • ASE를 이용한 식품원료용 생약재의 잔류농약 분석방법 연구 및 모니터링

        생약재의 잔류농약을 분석하기위한 새로운 겋사방법의 개발과 업무의 효율화를 재고하기 위한 방안의 일환으로 가속용매추출장·치(Accelerated solvent extractor,ASE)를 이용한 분석방법 확링을 시도하였다. AS도의 운용조건은 acetone을 추출용매로 하여 1,000psi, 4et에서 운용하는 것이 최적의 조건이었고 이들 추출액의 정제는 prewashing 용매로 hexane을 Smf 사용하여 Florisil 카트리지를 활성화 한 후 10m4의 20% acetone 포화 hexane으로 용출하는 것이 정제효과면에서 가장 좋았다. 생약재에 있어서 대부분의 시료에서 70%이상의 회수율을 나타내었으며 최저 검출한계치는 0,O04~o.016r9/me로 매우 낮았다. 또한 위와 같은 분석방법을 이용하여 165종의 생약재에서 모니터링을 한 결과 7건에서 농약이 검출되었으나 그양은 0.118-0.045ppm으로 극미량 이었다. This study was carried out to improve the analytical method and monitoring of pesticides in natural medicines using Accelerated Solvent Extractor(ASE). Optimiui~ extractconotions for ASE were at 40'Cr 1000psi with acetone. Extracts were puffied through SPEflorisil cartridge with 10ee of 20% acetone saturated hexaRe. Recoveries from samples weregreater than 70% most of the natural medicines. The limit of detections(LODs) in all matrixwere 0.O04~o.016 #g/mf.The residual pesticides ·rrere detected in 7 samples of 165 natural medicines. However, thedetected pesticides were very small amount ranged from 0.045 to 0.118 #g/ma.
