        In order to use as a starter after drying, optimum growth conditions of Str. thermophilus and L. bulgaricus were investigated. The results were as follows. The growth of Str. thermophilus was reached at stationary phase after 12 hours incubation and L. bulgaricus was after 6 hours. Str. thermophilus was grown well, in the medium containing 10% of skim milk, 2%5 of sucrose, 0.1% of KCl and 0.015%(v./v.) of soy-sauce. In the case of L. bulgaricus, 10% of skim milk, 2% of glucose, 0.5% of bacto-peptone, 0.1% of K₂HPO₄ and 0.02% of pyridoxine was suitable for the maximum growth.

      • 배위체 조건 변화에 따른 성숙 및 미성숙 백서 심장 Na^+-K^+ ATPase의 Digitoxin 억제도 변화에 대한 생화학적 연구

        The inhibition rates of digitoxin for Na^+-K_+ ATPases from adult and infant rat heart tissues were determined for a particular set of binding ligands. When ligand condition was changed, specific activities of Na^+-K^+ ATPases of whole homogenates from adult rat heart tissues were following. For 15 mM ATP 0.25 ml, 50 f mM MgCl2 0.25 ml, 1.0 M NaCl 0.25 ml, 0.1 M KCI 0.25 ml, that is [Mg/ATP/Na/K], specific activity (micromol/ Pi/mg protein/hr) was 0.741 : [Mg/ATP/K], NaCl was excluded, 0.631 : [Mg/ATP/Na], KCI was excluded, 1.126 : [Mg/ATP/Na/1M KCI], 0.1 M KCI was replaced by 1.0 M KCI, 0.148. The inhibition rates of digitoxin for Na^++K_+ ATPases from adult rat heart were less than that of infant rat. When ligand conditions were [Mg/ATP/Na], [Mg/ATP/K], the rates of inhibition were remarkably changed. Our following experimental data was showed that the specific activity of Na ATPase was inhibited the more rate in the infant rat heart tissues than adult by digitoxin. When the concentration of the Na^+ in the adult rat heart tissues was increased, binding rate of digitoxin with ATPase was decreased. When K_ in the adult rat heart tissues was increase, binding rate of digitoxin with ATPase was increase. In the views of the subunit of the Na^++K_+ ATPase in the rat heart tissues, our results suggest that the activity of ATPase and inhibition pattern by digitoxin in the infant rat was showed the same pattern of the other mammals, but that in the adult rat was showed the opposite pattern.

        Monascus sp. J101을 이용한 적색색소의 Extractive Fermentation

        Monascus sp.의 변이주인 색소생성능이 높은 Monascus sp. J101을 이용한 적색색소 생산의 발효특성을 분석하고 extractive fermentation을 시도하여 색소의 생산성을 높이고자 하였으며 Amberlite XAD-7 resin을 첨가한 경우에 대한 동력학적 분석을 수행하였다. Monascus sp. J101이 생산하는 적색색소는 균체의 색소의 생산량이 총 색소생산량의 약 17%에 해당하였으며, 색소의 생산은 product에 의해 regulation되는 것을 확인하였다. 한편, 균체의 생육은 48시간부터 정지기에 도달하였으나 색소의 생산은 그 이후에도 계속되어 mixed growth-associated의 양상을 보였다. 균체의 배양중에 permeabilizing agents를 첨가하여 extractive fermentation을 수행한 결과, 배양 48시간에 Triton X-100 0.05%를 첨가했을 때 약 18%의 색소생산의 증가를 보였으며, ethyl acetate를 0.25% 첨가한 경우는 약 12%, deoxycholate 0.05%인 경우는 약 15% 증가하였다. 균체의 배양중에 nonionic adsorbent인 Amberlite XAD-7을 첨가하고 extractive fermentation을 행한 결과, 배양 48시간에 Amberlite XAD-7을 12.0% 첨가하였을 때 첨가하지 않은 control에 비해 색소의 생산이 약 48.9%가 증가하였다. The characteristics of monascus fermentation using a hyperpigment-producing mutant, Monascus sp. J101, were analyzed, and the extractive fermentations employing permeabilizing agents and resin were carried out to increase the productivity of red pigment. And the kinetic analysis was also carried out in case of the monascus fermentation using Amberlite XAD-7. The extracellular content of the red pigment produced by Monascus sp. J101 was about 17% of the total, and the production of pigment was regulted by its own product. The cell growth reached a stationary phase at 48 hours after inoculation, whereas the pigment production continued up to 110 hours, which showed the pattern of a mixed growth-associated type. During the fermentation, various permeabilizing agents were added to the culture medium and their effects on pigment production were examined. By adding 0.05% Triton X-100 at 48 hours of cultivation, about an 18% increase in pigment production was accomplished as compared to the control, 12% for 0.25% ethyl acetate and 15% for 0.05% deoxycholate, respectively. When a nonionic adsorbent, Amberlite XAD-7 was added to theh culture medium at a concentration of 12.0% at 48 hours of cultivation, the pigment production was enhanced by about 48.9% as compared to the control.

      • 기존 교육과정에서의 성과바탕 의학교육 이론 적용

        Objectives: This paper aims to describe an outcome-based curriculum development process at a medical school that has difficulty in advancement from the higher stage outcomes to the individual lesson outcomes, and to propose a way to implement it practically. Methods: We reviewed the objectives, strategies and previous products of the school’s taskforce activities and suggested the principle of bidirectional approaches of outcome based curriculum development. Results: The developing strategy identified such as firstly, the evaluation of present curriculum and then, the review of the outcomes developed previously with considering the nation-wide environmental change in medical education. Then, we selected one example course which was focused the resources of the school to, and finally the product of the example course was propagated to the other courses with central monitoring. Conclusion: Bidirectional model of ‘Top-down’ plus ‘Bottom-up’ approaches could be an efficient way to develop the outcome-based curriculum in a medical school, which has difficulties to advance the developing process due to various reasons including limited resources.

        한국 각막이식의 실태

        Purpose: To report an epidemiologic study of keratoplasty in which we analyze basic data of corneal transplants and corneal donations in Korea. Methods: From June 2004 through October 2004, questionnaires were distributed to 25 hospitals. The questionnaires were about the characteristics of patients registered in eye banks for keratoplasty from May 2001 to April 2003, and about the results of keratoplasties performed from May 2002 to April 2003. Results: The leading corneal diseases in patients registered for keratoplasty were infectious keratitis (22.0%), trauma (21.0%), and pseudophakic and aphakic bullous keratopathy (13.8%), in that order. The leading indications in corneal recipients were: trauma (15.5%), pseudophakic and aphakic bullous keratopathy (13.7%), and infectious keratitis (10.7%), in that order. In 233 cases of keratoplasties, penetrating keratoplasty was performed in 229 cases (98.3%) and lamellar keratoplasty was performed in 4 cases (1.7%). The most common combined surgery was cataract extraction. Common causes of graft failure were rejection of donor cornea and infection. Imported donor corneas were used in sixty-one cases (26.2%) among the 233 total keratoplasties. Conclusions: We constructed an epidemiologic data base of patients registered for keratoplasty, corneal donation, and the results of keratoplasties throughout Korea.
