        생쥐 췌조직내 Protein Methylase에 대한 자율신경계약물의 영향

        It was reported that protein carboxymethylation is involved in amylase secretion of parotid gland by isoproterenol. It was also suggested that a small part of the total cellular protein carboxymethylation is directly involved in pancreatic enzyme secretion. On the contrary, other authors reported that there is no relationship between protein carboxymethylation and secretion in pancreas and parotid gland. In recent study, it was proposed that a methyl acceptor protein plays a limited modulatory role in the coupling of cytosolic Ca++ accumulate on and exocytosis. In this study, the effects of cholinergic and adrenergic agents on the activities of protein methylase II in pancreatic tissues were examined to test the relationship between protein methylation and pancreatic secretion. The results are as follows. The activity of amylase was slightly increased at the concentration of 10-5M of isoproterenol and norepinephrine. The activities of protein methylase I and II were decreased by isoproterenol and norepinephrine, but the activities of protein methylase III were hardly changed. The cholinergic stimulants acetylcholine and carbachol at a concentration of 10-5M increased the activities of protein methylase I and decreased the activities of protein methylase III compared with control.

        닭 췌장 Protein Methylase II의 분리정제 및 성질

        Protein methylase II (S-adenosyl-L-methionine:protein carboxyl-0-methyltransferase; EC 2.1.1.24., PM II) was purified from chicken pancreas by subcellular fractionation, DEAE-cellulose chromatography, QAE-Sephadex A-50 chromatography, Sephadex G-75 chromatography, and Sephadex G-75 rechromatography. The purified PM II gave a single band upon polyarcrylamide gel electrophoresis both in the presence of SDS and in Tris glycine buffer without SDS. The pI value of purified PM II was identified as 5.7 on isoelectric focusing gel. Properties and activities of PM II were studied and the following results were obtained. 1) PM II from chicken pancreas was purified approximately 221-fold with a yield of 1.3%. 2) The purified PM II appear constituted of a single polypeptide chain of a molecular weight 46,800 daltons. 3) Hemoglobin exhibited the highest of methyl-accepting activity among the substrates tested. 4) The purified PM II has a Km of 4.67 X 10-6M and a Vmax of 37.5 pmoles of methyl-14C/min/mg enzyme for SAM-14CH3 as methyl donor in the presence of histone type II-As. 5) It is found that S-adenosyl-L-homocysteine is a competitive inhibitor for PM II with KI value of 3.23 X 10-5M.

      • 천연물의 항염증작용 검색에 관한 연구

        본 연구에서는 여러가지 in vivo 동물모델을 이용한 항염증제의 효력 검색법을 확립하여 항암증제의 효력시험 수탁고시에 따른 시험법을 확립하고, 비교적 부작용이 적은 천연물을 대상으로 항염증작용 검색 및 ㅡㄱ 기전을 연구하고자 하였다. 대표적인 급성 염증 모델인 rat에서의 carrageenan 및 PAF 유도 족척부종법, 염증반응 1기의 대표적인 현상인 백혈구 유주도를 측정하는 모질인 meuse에서의 zymosan운토 백혈구 유주 억제도 측정법, 염증만응 3기의 섬유화 현상을 측정하는 모델인 rat에서의 cotton pellet유도 육아종 형성 측정법, 만성 염즈으이 대표적 모델인 ratd에서의 adjubant 유도 관절염법 등 6종으 in vivo호력 시험법을 표준화하고 당귀와 원방풍 물 , 추출액, 은행엽메탕올 추출액, 죽염 수영액의 항염증작용을 검색하엿다. 당귀 물 추출액(1g/kg), 은행엽 메탄올 추출액 (1g/kg),죽염(1.5g/kg)은 carrageenan 및 PAF 유도 부종을 유의하게 억제하였고, 이중 은해엽 메틴올 추출액은 모세혈관 투과도를, 죽염은 육아종 형성을 유의하게 억제하였다. 검색한 천연물 중 zyinosan 유도 백혈굴 육주 및 adjuva띠:유도 관절염에 대해 항염증 작용을 갖는 물질은 얼었다. Carrageenan 유도 족척부종에 대한 항염증 작용 중은행13탄을 추출액과 죽염은 초기 및 후기 부종 전시간대(0.511r~6hr)에 걸쳐 항투종 작용이 있었으나 당귀 물 추출액은 carrageenan 유』A 부종 쵸기(1시간 이내)와 PAF 유도 부종에 대해 항루종 작용이 있어 carrageenan유도 분종의 초기 단계는 매개물질로 PAF가 관련한다는 점을 고려한다면 당귀의 항염증작용은 염증 매개 물질인 PAf의 작용을 억제하여 나'타날 가능성이 있을 것으로 사료된다. 이상의 실험을 통하여 염증반응 각 단계에 대한 항염증작용 겋색법을 화립하여 항염증제의 효력시험 수탁 고시에 따른 시혐법을 표준화하였고, 비교적 부작용이 적은 수종 천연물을 대상으로 항영킁제로의 이용 가능성롸 그 기전을 연구하였다. Thmethods of "carrageenan induced foot edema", "PAF (platelet activating (actor) induced foot edema","inhibition test of vascular perra☞ability", "inhibition test of white blood cell migration", "formation egranuloraa" and "adjuvant indLiced arthritis" were estabilished. The antiinflammatory effects of fngeff-ca gigat fedeDoHriefBa sefefoifer, eiHfgo fifaD3 and Bamboo salt (Jukyom ) were investigated by in ri:same administration of methanolic extract of Ginkgo folium also inhibited the vascular permeability in - mice. The aqueous solution of Bamboo salt inhibited the formation of granuloma in SD rat at a oral dose -of Igfg. ABgelica gigantis rad;ix seems to have antiinflammatory effect by inhibition of PAF.

        췌조직과 성장 발육에 따른 흰쥐 조직내 S-Adenosylmethionine Synthetase 활성도 및 S-Adenosyl-L-methionine의 분포

        S-Adenosyl-L-methionine synthetase (ATP: methionine S-Adenosyltransferase, EC 2.5.1.6; AdoMet synthetase) catalyzes the biosynthesis of S-Adenosyl-L-methionine(AdoMet) from methionine in the presence of ATP. To elucidate the role of transmethylation reaction in the pancreatic tissues, we examined AdoMet synthetase and isozyme activities, and AdoMet contents in the various tissues. The activities of AdoMet synthetase marked the highest in the kidney, and the lowest in the testis among the various tissues of rat. Considerable amounts of AdoMet synthetase activities were detected in the pancreatic tissues of various animals except for those of frog. The level of ${\alpha}$ and ${\gamma}$ isozyme activities were present in the pancreatic tissues of various animals, while ${\beta}$ isozyme activities were detected as trace. AdoMet synthetase activities of rat brain, liver, testis were decreased with growth. In the rat pancreatic tissues, AdoMet synthetase activities were increased during 16 days after birth and then decreased between 16 and 47 days of age. Levels of AdoMet contents of rat brain and testis were decreased with growth. However, AdoMet contents of rat pancreas were decreased until 26 days of age, and then increased thereafter. AdoMet synthetase isozyme patterns did not vary with growth in the pancreas and testis. But, in the liver, ${\beta}$ form is strikingly increased with growth.

      • Porcine Liver Protein Methylase Ⅱ의 分離 精製 및 性質

        Abstract-Protein methylase II(S-adenosyl-L-methionine: protein carboxyl-O-methyltransferase; 2.1.1.24) was purified from porcine liver by subcellular fractionation, ammonium sulfate precipitation, gel filtration, and ion exchange chromatography, and the properties of the enzyme were studied. The molecular weight of the enzyme was estrimated to be 21,000 daltons determined by Sephadex G-75 chromatography. The K_m value of the enzyme showed 2.4×10^-6 M and V_max 13 pmole/min/mg for S-adenosyl-L-methionine. K_i of S-adenosyl-L-homocysteine for the enzyme was measured as 5.5×10^-7 M. PM II activities for histone type II-A were dependent on the amount of the purified enzyme over the range of 8-40㎍ protein.

        Abstract-The effects of cholinergic and adrenergic agents on the activities of SAM-synthetase [ATP : L-methionine S-adenosyltransferase EC 2.5.1.6] in pancreatic tissues were examined to test the relationship between SAM-synthetase activity and pancreatic secretion. Furthermore, S-adenosyl-methionine concentrations were measured by HPLC in pancreatic tissues to test the relationship between SAM-synthetase and protein methylation. The results are as follows 1) Cholinergic agents (acetylcholine and carbachol) at concentration of 10^-5 M increased amylase secretion and SAM-synthetase activity compared with those of unstimulated control. These results were blocked by 10^-5 M of atropine. 2) Adrenergic agents(isoproterenol, epinephrine and norepinephrine) hardly changed amylase secretion and SAM-synthetase activities. 3) The SAM contents were similar to those of control in case of acetylcholine treated group, but slightly increased in isoproterenol treated group.
