        We investigated the effect of safflower-seed on healing of the open tibial fracture in rats. A total eighty male rats (13 weelts,450-500 g) were divided into two groups and immediately afterfracturing the tibia, normal or 10% safflower-seed diet were giver arbitrarity. This treatment wasgiven for 8 weeks. Five er six rats were killed from each group at t, 3. 5. 7 days and 2, 4, 6, 8 weeksafter fracture. Fracture healing was evaluated by eramining the degree of wound healingmacroscopically, radiographf, bone histomorphonletry and histologfcal examination. After 1, 3, 5, 7days. the wound healiug was accelerated in safflower- seed diet group. Radiography does not reveal thedifference in fracture healing between two groups. After 2 weeks, safflower- seed had aB significant,stiinutatory effect on exterBal callus formation (p <0.05). But after 4, 6,8 weeks, no difference was otserved between normal and safflower-seed diet group in callus size. After 2,4,6 and 8 weeks, the hist☞logical study also revealed that the area of cartilage over the fracture site with bone on each side wasdecreased by endochondral ossification in safflower-seed diet group faster than in normal diet group. loconclusion, safflower- see4 stimulate fracture healing during the initial healing phase by enhancing thecallus formation.

        Quinones (menadione, benzoquinone, 및 2,3-dimethoxy-1,4-naphthoquinone)의 혈소판 세포독성

        Our previous studies demonstrated that quinone (menadione) is cytotoxic to rat platelets. In an attempt to assess the relative contributions of redox cycling and/or arylation in quinone-induced cytotoxicity, we have studied three quinones with different mechanisms: 2, 3-dimethoxy-1, 4-naphthoquinone (DMNQ; pure redox cycler), menadione (both redox cycler and arylator), and 1, 4-benzoquinone (pure arylator). The order of redox cycling capacity in platelet rich plasma (PRP) isolated from rats was menadione>DMNQ>1, 4-benzoquonone, which was consistent with the previous studies using isolated hepatocytes. 1, 4-Benzoquinone was more toxic to rat platelets than menadione, while DMNQ did not cause cell death at all. Lactate dehydrogenase inhibition studies revealed that 1, 4-benzoquinone inhibited significantly in a time-dependent manner, while menadione and DMNQ did not at all. These results suggested that arylation by quinone compounds might play a critical role in quinone-induced cytotoxicity in rat platelets.

        Menadione에 의한 흰쥐 혈소판 세포독성에서 nitric oxide의 역할

        Nitric oxide, a physiological transmitter, is reported to mediate cellular injury in various tissues. Its reactivity to free radical is believed to be one of the reasons for its involvement in cytotoxicity. Menadione, a representative quinone, is cytotoxic to several cell systems including isolated hepatocyte, endothelial cell and red blood cells. Its toxic mechanism is related to oxidative stress, mediated by toxic free radicals. Our previous studies demonstrated that menadione induced cell lysis and increase of oxygen consumption in platelets. It has been reported that platelets have nitric oxide producing enzyme, nitric oxide synthase. Thus, we have investigated to manifest the role of nitric oxide.in menadione-induced cytotoxicity in rat platelets. Menadione induced cytotoxicity in platelets was unaffected by $N^G$-nitro-arginine methyl ester (L-NAME), selective and competitive inhibitor of nitric oxide synthase. We also invesitgated the role of extracellular nitric oxide in menadione-induced cytotoxicity of platelets by addition with sodium nitroprusside (SNP). SNP did not affect platelet cytotoxicity by menadione. These results suggested that nitric oxide which was generated endogeneously or exogeneously might have a negligible role in menadione-induced cytotoxicity in rat platelets.

      • 천연물의 항염증작용 검색에 관한 연구

        본 연구에서는 여러가지 in vivo 동물모델을 이용한 항염증제의 효력 검색법을 확립하여 항암증제의 효력시험 수탁고시에 따른 시험법을 확립하고, 비교적 부작용이 적은 천연물을 대상으로 항염증작용 검색 및 ㅡㄱ 기전을 연구하고자 하였다. 대표적인 급성 염증 모델인 rat에서의 carrageenan 및 PAF 유도 족척부종법, 염증반응 1기의 대표적인 현상인 백혈구 유주도를 측정하는 모질인 meuse에서의 zymosan운토 백혈구 유주 억제도 측정법, 염증만응 3기의 섬유화 현상을 측정하는 모델인 rat에서의 cotton pellet유도 육아종 형성 측정법, 만성 염즈으이 대표적 모델인 ratd에서의 adjubant 유도 관절염법 등 6종으 in vivo호력 시험법을 표준화하고 당귀와 원방풍 물 , 추출액, 은행엽메탕올 추출액, 죽염 수영액의 항염증작용을 검색하엿다. 당귀 물 추출액(1g/kg), 은행엽 메탄올 추출액 (1g/kg),죽염(1.5g/kg)은 carrageenan 및 PAF 유도 부종을 유의하게 억제하였고, 이중 은해엽 메틴올 추출액은 모세혈관 투과도를, 죽염은 육아종 형성을 유의하게 억제하였다. 검색한 천연물 중 zyinosan 유도 백혈굴 육주 및 adjuva띠:유도 관절염에 대해 항염증 작용을 갖는 물질은 얼었다. Carrageenan 유도 족척부종에 대한 항염증 작용 중은행13탄을 추출액과 죽염은 초기 및 후기 부종 전시간대(0.511r~6hr)에 걸쳐 항투종 작용이 있었으나 당귀 물 추출액은 carrageenan 유』A 부종 쵸기(1시간 이내)와 PAF 유도 부종에 대해 항루종 작용이 있어 carrageenan유도 분종의 초기 단계는 매개물질로 PAF가 관련한다는 점을 고려한다면 당귀의 항염증작용은 염증 매개 물질인 PAf의 작용을 억제하여 나'타날 가능성이 있을 것으로 사료된다. 이상의 실험을 통하여 염증반응 각 단계에 대한 항염증작용 겋색법을 화립하여 항염증제의 효력시험 수탁 고시에 따른 시혐법을 표준화하였고, 비교적 부작용이 적은 수종 천연물을 대상으로 항영킁제로의 이용 가능성롸 그 기전을 연구하였다. The antiinflammatory effects of fngeff-ca gigat fedeDoHriefBa sefefoifer, eiHfgo fifaD3 and Bamboo salt (Jukyom ) were investigated by in ri:same administration of methanolic extract of Ginkgo folium also inhibited the vascular permeability in - mice. The aqueous solution of Bamboo salt inhibited the formation of granuloma in SD rat at a oral dose -of Igfg. ABgelica gigantis rad;ix seems to have antiinflammatory effect by inhibition of PAF.

        천연물의 항염증작용에 관한 연구

        These studies were conducted to investigate the antiinflammatory effects and the mechanism of action of natural products. We used the methods of carrageenan induced foot edema , PAF (platelet activating factor) induced foot edema , inhibition test of vascular permeability, inhibition test of white blood cell migration, formation of granuloma and adjuvant induced arthritis to examine the antiinflammatory effects of Angelica gigas, Ledebouriella seseloides, Ginkgo biloba and Bamboo salt (Jukyom). The oral administration of the water extract of Angelica gigantis radix, the methanolic extract of Ginkgo folium and the aqueous solution of Bamboo salt showed antiinflammatory effect on carrageenan and PAF induced foot edema in SD rat at a dose of 1 g/kg. The same administration of methanolic extract of Ginkgo folium also inhibited the vascular permeability in mice. The aqueous solution of Bamboo salt inhibited the formation of granuloma in SD rats at a oral dose of 1 g/kg. Angelica gigantis radix seems to have antiinflammatory effect by inhibition of PAF.
