        Effects of MTNR1B variants on fasting glucose levels in a Korean population

        Fasting glucose level is the most basic and widely used indicator of diabetes. Several genome wide association studies have reported that the gene encoding melatonin receptor 1B (MTNR1B) exerts a major effect on serum fasting glucose levels. We tested for the association between single nucleotide polymorphisms (SNPs) in the MTNR1B gene and fasting glucose levels in a Korean population consisting of 8,229 subjects taken from two community-based cohorts. The mean age of the subjects in the study population was 51.9 years. For this study, we selected 363 SNPs located in the MTNR1B gene,which is located on chromosome 11. Multivariate linear regression models were used to test for genotypic effects on fasting glucose levels while adjusting for age and sex under an additive model. The MTNR1B SNP most highly associated with fasting glucose levels was rs10830962 (p=1.95×10-5), followed by rs3847554 (p=3.16×10-4). Replication of these initial findings is important to better understand the correlation between MTNR1B variations and their effects, especially in Asian populations.

        Human LZIP induces monocyte CC chemokine receptor 2expression leading to enhancement of monocytechemoattractant protein 1/CCL2-induced cell migration

        Chemokines and chemokine receptors play a role in inflamation. Human LZIP is an uncharacterized tran-scription factor and is known to participate in leuko-tactin (Lkn)-1/CCL15-induced cell migration. We in-vestigated the role of human LZIP in expression of C chemokine receptors (CCRs) and its involvement in monocyte migration. RNase protection analysis showed that LZIP increased mRNA expression of CCR2 and CCR1 in THP-1 cells. Surface expressions of both CCR2 and CCR1 were also increased by LZIP. oretic mobility shift assay showed that LZIP binds to the C/EBP element in the CCR2 promoter. LZIP also enhanced the chemotactic activities of monocyte chemoatractant protein-1/CCL2 and Lkn-1. These results suggest that LZIP regulates expression of chemokine receptors that are involved in monocyte migration.

      • Proteomic analysis of differential protein expression in neuropathic pain and electroacupuncture treatment models

        Acupuncture has long been used for pain relief. Although recent studies have shown that acupuncture can reduce neuropathic pain, the mechanism of this effect is not clear and little information is available regarding proteins that are involved in the development of neuropathic pain and the effects of acupuncture. We have developed an animal model for neuropathic pain using young adult male Sprague-Dawley rats. The model was confirmed by behavioral tests. Electroacupuncture (EA) treatment was applied to Zusanli (ST36) of neuropathic pain model to examine the analgesic effect of EA. The protein expression profile of the hypothalamus in both neuropathic pain and EA treatment models was analyzed using two-dimensional electrophoresis-based proteomics. We detected thirty-six proteins that were differentially expressed in the neuropathic pain model compared with normal rats and that restored to normal expression levels after EA treatment. Twenty-one of these proteins were identified in the MSFiT database and are involved in a number of biological processes, including inflammation, enzyme metabolism and signal transduction. Potential applications of our results include the identification and characterization of signaling pathways involved in EA treatment and further exploration of the role of selected identified proteins in the animal model.

        대한임상검사과학회지 50년사: 명칭과 KCI 등재에 대하여

        The Korean Society for Clinical Laboratory Science publishes the Korean Journal of Clinical Laboratory Science (KJCLS) as the official journal for the Korean Association of Medical Technologists. This year, 2017, marks the 50th anniversary of KJCLS. The original name was "the Korean Journal of Medical Technologists" (Volume 1, 1967~Volume 26, 1994), which was replaced with the "Korean Journal of the Clinical Laboratory Science" (Volume 27, 1995~Volume 35, 2003) and "the Korean Journal of Clinical Laboratory Science" (Volume 26, 2004~Present). The purpose of this study was to provide a framework for further development of KJCLS. In 1998, the National Research Foundation of Korea began the journal assessment service. The major domestic databases are KCI, KOFST, and KAMJE; the major international databases are SCI Index; SCI Core, SCI-Extended), SCOPUS, MEDLINE, and PUBMED. KJCLS has been registered with KCI of the National Research Foundation of Korea since 2014. More than 1,300 articles have been published, which all have made tremendous contributions to the field. To index KJCLS in the journal databases, we measured the value to understand the vocational level and social position. KJCLS is faced with a great challenge; immense effort will be needed to accomplish the original goal of KJCLS to become an internationally recognized journal. Ultimately, the goal of the Korean Society for Clinical Laboratory Science and the Korean Association of Medical Technologists should be to register KJCLS in databases such as KCI and SCOPUS.

        Triglyceride (TG) down-regulates expression of MCP-1 and CCR2 in PMA-derived THP-1 macrophages

        Triglycerides (TGs) are implicated in the development of atherosclerosis. A key contributing factor for atherosclerosis is the migration of macrophages to atherosclerotic lesions. MCP-1 is a major chemoattractant for macrophages to atherosclerotic lesions. We examined the expression profile of MCP-1 and CCR2 in THP-1macrophages in response to TG treatment by RT-PCR analysis. Chemical inhibitors were used to identify cell signaling pathway(s) involved in regulation of MCP-1 and CCR2 expression. We found that treatment of THP-1macrophages with TG down-regulated MCP-1 expression in a time and dose-dependent manner. PMA treatment alone did not affect MCP-1 expression. Using chemical inhibitors of cell signaling pathways, we found that the NFjB inhibitor inhibited TG-induced down-regulation of MCP-1. CCR2 expression decreased after TG treatment in THP-1 macrophages and the PKC inhibitor alleviated TGinduced down-regulation of CCR2. Our results provide further insights into the role of TG on macrophages during atherosclerosis.

        A Correlation Analysis of Serum Creatinine Based eGFR and Serum Cystatin C Based eGFR with Thyroid Dysfunction Patients

        Serum creatinine-based eGFR and serum cystatin C-based eGFR are the most popular methods for measuring renal function. Thyroid hormone is known to affect serum creatinine-based eGFR and serum cystatin C-based eGFR; however, the clinical significance of thyroid dysfunctional patients of renal function evaluation has not been fully elucidated to date. This study examined the effect of thyroid hormone on serum creatinine-based eGFR and serum cystatin C-based eGFR. Moreover, we also evaluated the correlation analysis between serum creatinine-based eGFR and serum cystatin C-based eGFR in patients with thyroid dysfunction. A total of 442 patients with hypothyroidism and 284 patients with hyperthyroidism were investigated. A correlation analysis between thyroid hormone and serum creatinine- (and cystatin C-) based eGFR was performed. A correlation analysis between thyroid hormone and serum cystatin-C based eGFR indicated that serum cystatin-C based eGFR is more of an independent biomarker than serum creatinine-based eGFR in thyroid dysfunction patients. Therefore, serum cystatin C-based eGFR more accurately reflects renal function than serum creatinine-based eGFR in thyroid dysfunction patients.

        Decision Tree를 이용한 효과적인 유방암 진단

        최근 의료분야에서는 대규모의 데이터를 빠르게 검색 및 추출이 가능하게 의사결정트리 기법에 대한 연구들이 진행되고 있다. 현재 CART, C4.5, CHAID 등 여러 기법이 개발되었는데, 이러한 클레시파이 기법들은 몇몇 의사결정 나무 알고리즘이 이진분리로 분류를 하는데, 나머지 데이터의 결과가 손실될 우려가 있다. 그중 C4.5는 엔트로피의 측정값에 높고 낮음으로 트리 모양을 구성해 가는 방식이고, CART 알고리즘은 엔트로피 매트릭스를 사용하여 범주형 자료나 연속형 자료에 적용할수가 있다. 이에 본 논문에서는 클래시파이 기법 중 C4.5와 CART를 유방암 환자 데이터에 대해 적용하여 실험하여, 그 결과 분석을 통한 성능 평가를 수행하였다. 실험에서는 교차검증을 통해 그 결과에 대한 정확성을 측정하였다. Recently, decision tree techniques have been studied in terms of quick searching and extracting of massive data in medical fields. Although many different techniques have been developed such as CART, C4.5 and CHAID which are belong to a pie in Clermont decision tree classification algorithm, those methods can jeopardize remained data by the binary method during procedures. In brief, C4.5 method composes a decision tree by entropy levels. In contrast, CART method does by entropy matrix in categorical or continuous data. Therefore, we compared C4.5 and CART methods which were belong to a same pie using breast cancer data to evaluate their performance respectively. To convince data accuracy, we performed cross-validation of results in this paper.

        보건의료직종별 SBT 국가시험체제에 대한 타당성 연구

        Purpose: The purposes of this research were to study the validity of the National Examination based on smart based test(SBT) by 7 types of healthcare occupation. Methods: We conducted a questionnaires survey among 1,219 people including 210 professors for all 7 types of healthcare occupation and 573 clinical specialists ; 436 students from random sampling participated and the reliability of the questionnaire was tested using Cronbach’s alpha. Results: Professors from 7 types of healthcare occupation were investigated for their opinions on the introduction of the SBT system ; 65.9% of the professors answered positively on the introduction of the SBT National Examination System, while 40.1% of the clinical experts answered likewise. In particular, professors and the clinical experts recognized altogether that the multimedia questionnaire, being effective for the evaluation of clinical and practical competence of the subjects, enabling the evaluation of various areas and strengthening the school education in clinical practices, the 42.0% of the students from 7 departments answered that the future direction of the national test is in the SBT test system with significant difference (p=.000) found in students majoring in order of Emergency Medical Services, Radiology and Physical therapy, compared to students from other majors, showing the positive response to the SBT system. Conclusion: The results of this study comprehensively evaluated the positive recognition of development of multimedia items and SBT system in national examination of 7 types of healthcare occupation in professors, field specialists and students.

        Development of TaqMan Quantitative PCR Assays for Duplex Detection of Dirofilaria immitis COI and Dog GAPDH from Infected Dog Blood

        Dirofilaria immitis (D. immitis) is a filarial nematode that causes cardiopulmonary dirofilariasis in dogs. In the late stages of infection, infected dogs show one or more symptoms and advanced heart disorder with perivascular inflammation. To detect D. immitis specifically and efficiently in the early stages of infection, a duplex TaqMan qPCR assay was developed based on previous studies using primers and probes specialized to detect D. immitis cytochrome c oxidase subunit I (COI ) and dog glyceraldehyde-3-phosphate dehydrogenase (GAPDH). As positive controls, plasmid DNAs were constructed from D. immitis COI or dog GAPDH and a TA-cloning vector. Simplex and duplex TaqMan qPCR assays were performed using the specific primers, probes, and genomic or plasmid DNA. The duplex reaction developed could detect D. immitis COI and dog GAPDH in the same sample simultaneously after optimization of the primer concentrations. The limit of detection was 25 copies for the simplex and duplex assays, and both showed good linearity, high sensitivity, and excellent PCR efficiency. The duplex assays for pathogen detection reduce the costs, labor, and time compared to simplex reactions. Therefore, the duplex TaqMan qPCR assay developed herein will allow efficient D. immitis detection and quantification from a large number of samples simultaneously.
